Abstract

NEW MATERIAL:A 5-fluorouracil nucleoside of formula I (R1 is H or OH; R2 is OH; either of R3 and R4 is OH, and the other is H; either of R5 and R6 is H, methyl or hydroxymethyl group, and the other is H).
EXAMPLE: 1-(β-D-Ribopyranosyl)-5-fluorouracil.
USE: A medicine having an antitumor activity with low toxicity.
PROCESS: A solubilized 5-fluorouracil, e.g. 2,4-bis-0-trimethylsilyl-5-fluorouracil, is reacted with a reactive derivative wherein the OH group of sugar expressed by formula II (R is acyloxy group; Ra 1 is H or OH; Ra 2 is OH; either of Ra 3 and Ra 4 is H, and the other is methyl group, etc.), is protected, e.g. polyacyl sugar, in the presence of a Lewis acid catalyst, e.g. stannic chloride, in an inert solvent. The reaction product is then hydrolyzed with an alkali to give the compound of formula I free of the protecting group.
