本発明は、単離ポリヌクレオチドおよびそのポリヌクレオチドによりコードさ
れたタンパク質、およびコガネムシ科(Scarabaeidae)の制御におけるその使用
に関する。加えて、本発明は、これらのタンパク質の製造法に関する。
コガネムシ科、例えばコフキコガネ(Melolontha melolontha)およびウッド
・チェイファー(Melolontha hippocastani)、特にその幼虫(地虫)は、農業や
林業の収穫高に深刻な被害をもたらし得る。化学的殺虫剤による制御は困難で、
環境的に有害であるので、生物学的な手段を用いてそれらの昆虫の繁殖や蔓延を
制御しようとする試みが増えている。例えば、EP633 936A1には、Bacillus t
huringiensisの特定株の栄養細胞、胞子またはタンパク質結晶を使用して、コガ
ネムシ科を制御する方法が記載されている。しかし、この方法の効果は満足のい
くものではない。
WO87 05928に、コガネムシの幼虫の生物学的制御に、Bacillus popilliae(
B.popilliae)胞子の使用が提案されている。
B.popilliaeは、コフキコガネや他のコガネムシ科の幼虫に、いわゆる乳化病
を引き起こす生物体である。Bacillusが寄生した幼虫は、血リンパ中に高濃度の
B.popilliaeの栄養細胞および胞子嚢を有し、その結果、幼虫は乳白色へと変色
する。
B.popilliaeは、最初、米国においてマメコガネ(Popillia japonica)の乳化
病の原因としてDutkyにより記載され(Journal of Agricultural Research 61(19
40)p.57-68、「マメコガネの乳化病の原因となる、2つの新しい胞子形成細菌」
)、後に、コフキコガネ(Melolontha melolontha)の地虫において、Hurpinおよ
びVagoおよびWilleにより同定された(B.HurpinおよびC.Vago:Entomophaga 3(1
958)p.285-330、「Les maladies du hanneton commun(Melolontha melolontha L
.(Col.、Scarabaeidae)」;H.Wille:Mitteilungen.Schweizerishe Entomolog i
sche Gesellshaft 29(1956)p.271-282:「Bacillus fribourgensis n.sp.,Erre
ger einer“milky disease”im Engerling von Melolontha melolontha L.」)。
B.popilliae細菌の特徴は、とりわけ、カタラーゼを形成しないこと、ほとんど
の単離株が抗生物質バンコマイシンに耐性であり、胞子形成の間、実際の胞子に
隣接した紡錘形の胞子嚢内に配置され、特徴的なタンパク質結晶を形成すること
である。コガネムシ科の病原体としての効力については、B.popilliaeは高度の
特異性をもつ。異種のコガネムシから単離されたB.popilliae亜種は、成長の特
徴、タンパク質結晶の組成およびそのプラスミドについてかなり差がある場合が
ある。
B.popilliaeによる甲虫幼虫の寄生は、胞子嚢の経口摂取によりなされる。胞
子は幼虫の消化管で発芽し、栄養細菌細胞が消化管上皮および基底膜を通って血
リンパに浸透し、そこで3〜4週間増殖する。次いで、B.popilliae細胞は胞子
を形成し、最終的に甲虫幼虫は死滅する。
しかしながら、他のBacillus種と比較して、インビトロの条件下で、B.popill
iaeは優先的に栄養細胞を形成し、例外的にしか胞子を形成しない。WO 87 05
928では、インビトロで胞子を得る方法(これは、B.popilliaeの栄養細胞を特定
の培地で培養し、最後に特定の補助剤を加えて刺激し胞子を形成させる)につい
て記載しているが、この方法では胞子形成率はわずか約80%である。従って、
生物学的制御に十分な量の感染性胞子物質を調達するには、装置、人員および費
用の点でかなりの量を投資する必要があり、それ故、この方法は経済的に無理で
あり、従って実用的な目的には不適当である。
従って、本発明の目的は、これらの害虫を満足に制御でき、大量生産が技術的
に簡単で経済的であるコガネムシの生物学的制御方法を提供することである。
この目的は、Bacillus popilliaeに特徴的な結晶タンパク質と同一または少な
くともそれに関連したタンパク質をコードし、配列表の配列番号1に示されたヌ
クレオチド配列全部または一部、またはそれに関連した、置換、欠失、挿入およ
び/または逆位によりそれから誘導されたヌクレオチド配列、または完全にまた
は部分的にそれとハイブリダイズするヌクレオチド配列を有するポリヌクレオチ
ドを供給することにより達成される。
今回驚くべきことに、B.popilliae由来のタンパク質結晶は、コガネムシ幼虫
の摂食阻害に決定的な役割を果し、このタンパク質結晶を経口投与後に成虫は死
滅することが確認された。本発明に記載のポリヌクレオチドの供給により、初め
て、実質的に無制限な、すなわち特にバイオテクノロジー的な大量規模で、これ
らのB.popilliaeタンパク質結晶を得るまたは製造することが可能となる。
本発明に記載のポリヌクレオチド自体は、天然源から得られるか、または合成
的または半合成的に製造することができる。
それらは、特に、Bacillus popilliaeに特徴的な結晶タンパク質、またはBaci
llus popilliaeに特徴的なタンパク質結晶、またはそれに関連したタンパク質を
原核細胞または真核細胞で発現することのできる組換えDNAベクター分子の成
分として存在する。このベクター分子は、とりわけ、どのような細胞にでも、例
えば、市販の平易に培養できる大腸菌またはBacillus thuringiensisの細菌培養
物に導入でき、すでにその細胞中に存在しているか共に挿入したプロモーターで
制御し、複製し発現させることができる。適当なベクター分子は、特に、グラム
陽性細菌を起源とするプラスミドである。
バイオテクノロジー的に、本発明に記載のポリヌクレオチドによりコードされ
た結晶タンパク質を得るために、宿主細胞に天然に存在するか、または組換えの
結果存在するプロモーターに連結させた本発明に記載のポリヌクレオチドを含む
形質転換宿主細胞を使用することが提案されている。例えば形質転換、形質導入
または接合により、本発明に記載のポリヌクレオチドを宿主生物体(すなわち、
微生物、ウイルス、原生動物、植物細胞またはその他)に導入し、これらの細胞
またはウイルスの遺伝物質に組み込み、発現させる。
特に適していることが判明した宿主細胞はBacillus thuringiensis、例えばBa
cillus thuringiensis亜種kurstakiの栄養細胞である。Bacillus thuringiensis
は非常に詳細に研究されており、このシステムでは大量のB.popilliaeの結晶タ
ンパク質を製造することが比較的容易である。
本発明に記載のタンパク質は、単独で、または、少なくとも1つの他の物質と
併用してコガネムシを制御するための、すなわち摂食活動阻害および/または成
虫および/または幼虫コガネムシ、特にMelolontha種およびそれに関連した種を
死滅させるための生物学的殺虫剤として使用することができる。本明細書の「物
質」は、化学的および生物学的物質を含み、微生物を含む。他の病原体、例えば
とりわけウイルス、リケッチア、細菌、真菌および微胞子虫と組み合わせること
により、結晶毒素の作用を有利に増大させることが可能である。
本発明の好ましい態様において、本発明に記載のタンパク質はBacillus popil
liaeおよび/またはBacillus thuringiensisの胞子と共に使用する。Bacillus s
phaericusの胞子もまた非常に適している。
別の同じように大変有利な方法では、本発明に記載のタンパク質を、コガネム
シの消化管上皮に、細胞融解タンパク質および/またはレセプタータンパク質と
組み合わせて、好ましくは融合タンパク質の形で使用する。
さらに別の方法では、本発明に記載のタンパク質は真菌の胞子と組み合わせて
使用する。
植物に有害な土壌伝播性の生物体および/または真菌および/または伝染病の
制御において、本発明に記載のタンパク質を単独で、または少なくとも1つの他
の物質を組み合わせて使用することが可能である。制御は、不働化、特に摂食活
動阻害、および/または問題としている土壌伝播性生物体の死滅を含む。
本発明に記載のタンパク質を獲得または製造するために、1つまたはそれ以上
の本発明に記載のポリヌクレオチドを微生物(例えば、細菌、ウイルス、真菌ま
たは原生動物)、または動物細胞または植物細胞培養物に導入(形質転換)し、
プロモーター、好ましくは調節可能なプロモーター(これは、該微生物または細
胞に天然に存在するか、または組換えの結果存在する)で調節および制御し、発
現させる。特に好ましい別法は、ポリヌクレオチドをBacillus thuringiensis種
の細菌に導入する。
本発明はまた、コガネムシにより食べられるのを防ぐために、本発明に記載の
ポリヌクレオチドを植物または植物の一部に接種することを含む。別の言葉で言
えば:本発明はまた、全部またはいくつかの植物組織において、Bacillus popil
liaeに特徴的な結晶タンパク質と同一またはそれに類似し、摂食活動阻害および
/または幼虫および/または成虫コガネムシ、特に、Melolontha種およびそれに
緊密に関連した種の死滅に適した結晶タンパク質を合成することのできるトラン
スジェニック植物の調製における、および/または植物に有害な土壌伝播性生物
体および/または真菌の不働化および/または死滅および/または伝染病におけ
る、本発明に記載のポリヌクレオチドの使用を含む。
終わりに、本発明に記載の異種ポリヌクレオチドを含み、それを発現して、Ba
cillus popilliaeに天然に存在する特徴的な結晶タンパク質と同一または類似の
タンパク質を生じる組換え遺伝物質をもつ、植物体または植物組織または植物生
殖物質の製造法を提案する。本発明に記載の方法では、植物細胞または植物組織
を、本発明に記載のポリヌクレオチドと更に植物細胞においてそのポリヌクレオ
チドの安定な組込みと発現を引き起こすことのできる調節ヌクレオチド配列を含
む組換えDNAを用いて形質転換し;植物体またはその生殖物質またはその両方
を、異種DNAで形質転換した植物細胞から、または対応する組織から再生し;
望ましい場合には、再生した植物体またはその生殖物質またはその両方を生物学
的に再生産する。
本発明はまた、該方法または別の方法を用いて製造した遺伝子操作した形質転
換細胞に関するものであり、そのゲノムにはBacillus popilliaeに天然に存在す
る特徴的な結晶タンパク質と同一または類似のタンパク質をコードする本発明に
記載のポリヌクレオチドを含む(組換え)異種DNAが安定に組込まれており、
細胞のポリメラーゼにより認識され細胞に天然に存在するか、または組換えの結
果存在するプロモーターの制御下でそのタンパク質は発現される。
本発明は、態様に関して、下記でより詳細に記載する。実施例1:
本発明に記載のポリヌクレオチドの調製
Bacillus popilliae亜種melolonthae株をMelolontha melolonthaの幼虫から単
離する。この株の細菌細胞から、初めに全DNA物質を現行の方法を用いて調製
し、次に、結晶タンパク質を単離精製し、そのアミノ酸配列を決定する(市販の
タンパク質シークエンサーを用いる)。このタンパク質の2つの末端領域の部分
配列に対応するオリゴヌクレオチドを、結晶タンパク質遺伝子のDNAプローブ
の合成用の複製連鎖反応(PCR)におけるプライマーとして用いる目的で合成
する。PCRをBacillus popilliae亜種melolonthae由来の全DNAを用いて行
う。あるいは、初めに、単離された結晶タンパク質を短いフラグメントに切断し
、そこで、これらのいくつかのフラグメントのアミノ酸配列が決定でき、当業者
に既知のBacillus thuringiensis由来のCryIIAタンパク質のアミノ酸配列と比
較することにより、そのタンパク質の2つの末端領域に由来すると考えられる2
つのフラグメントを選択できる。次いで、これらのフラグメントに対応するDN
A配列をオリゴヌクレオチドとして調製し、PCR合成のプライマーとして使用
する。PCR産物をサザンブロット・ハイブリダイゼーションのプローブとして
使用する。Bacillus popilliaeゲノムの5.3kB EcoRIフラグメントをこの
方法で同定する。5.3kB EcoRIフラグメントを、例えば、プラスミドpB
CSK+に挿入し、大腸菌株XL1Blue MRFでクローン化する。市販のシー
クエナーゼ・テストシステム、例えばT7シークエンス・キットを用いてサンガ
ー鎖反応ターミネーション法により5.3kB EcoRIフラグメントをシークエ
ンスすると、配列表の配列番号1に示される結果が得られる。実施例2:
本発明に記載の遺伝子操作で調製した結晶タンパク質
実施例1で得られたBacillus popilliaeの5.3kB EcoRIフラグメントを
グラム陽性細菌のプラスミドに挿入し、Bacillus thuringiensis亜種kurstakiの
結晶非含有株の細菌細胞に導入する。プラスミドとして特に適当なものはBacill
us thuringiensisのクローニングベクターとして知られるプラスミド、例えばp
HT304、pHT315またはpHT370であり、その調製はO.Arantesお
よびD.Lerecius,Gene,148(1991),p.115-119に記載されている。この点で、こ
の文献を参照して、その内容を本明細書に取り込む。
組換え細菌を複製またはクローン化し、必要であれば、結晶タンパク質の合成
を刺激する。これは胞子形成を誘導することにより、好ましくは単に選択的な方
法で培養条件を修飾することによりなされる。次いで、タンパク質の遊離を、例
えば、培養条件をさらに修飾して、胞子発芽または胞子嚢の自己消化を起こすこ
とにより誘導する。遊離タンパク質を培養培地から分離し、精製し、必要であれ
ば、使用するまで、冷たい、乾燥した、暗い条件下で貯蔵する。実施例3:
Melolontha melolontha種の成虫コガネムシの制御における、本発明に記載の結
晶タンパク質の使用
成虫コフキコガネ(Melolontha melolontha)を自然に近い条件下で10日間
ケージにおく。2日後、本発明に記載のタンパク質の水性懸濁液を20匹の動物
に投与する。直ちに食物摂取が阻害される。わずか4日後に、10匹の動物(=
60%)が死亡する。実践的には、調製物を甲虫の食餌植物に噴霧する。実施例4:
Melolontha melolontha種のコガネムシ幼虫(地虫)の制御における、本発明に
記載の結晶タンパク質の使用
コフキコガネ(Melolontha melolontha)の地虫を開放地から掘り出し、実験
室の飼育室に個別に入れ、ニンジン片を食餌として与える。3週間後、水性懸濁
液中の本発明に記載のタンパク質を胞子と共に、または胞子無しで、10匹の動
物に食餌として与える。早くも1日後、食物の摂取が大いに減退阻害される。実
践的には、調製物を、可能であれば他の病原体と組み合わせて、好ましくは餌と
共に、土壌に混ぜる。

Claims (1)
- 【特許請求の範囲】 1.Bacillus popilliaeに天然に存在する結晶タンパク質と同一または関連し たタンパク質をコードし、および、配列表の配列番号1に示されるヌクレオチド 配列またはその部分配列を有するか、または、置換、欠失、挿入および/または 逆位によりそれから誘導されたヌクレオチド配列を有するか、または完全または 部分的にそれとハイブリダイズするヌクレオチド配列を有する、単離ポリヌクレ オチド。 2.ポリヌクレオチドを天然源から得るか、または合成的または半合成的に製 造する、請求項1に記載の単離ポリヌクレオチド。 3.請求項1または2に記載のポリヌクレオチドを含み、原核細胞または真核 細胞中で、Bacillus popilliaeに特徴的な結晶タンパク質、またはBacillus pop illiaeに特徴的なタンパク質結晶またはそれに関連したタンパク質またはタンパ ク質結晶を発現することのできる、組換えDNAベクター分子。 4.ベクター分子はグラムー陽性細菌にあるプラスミドである、請求項3に記 載の組換えDNAベクター分子。 5.宿主細胞に天然に存在するか、または組換えの結果存在するプロモーター に連結させた請求項1または2に記載のポリヌクレオチドを含み、そのポリヌク レオチドを発現して、Bacillus popilliaeに特徴的に存在する結晶タンパク質と 同一または類似の結晶タンパク質を合成できる、形質転換宿主細胞。 6.宿主細胞はBacillus thuringiensis、特にBacillus thuringiensis亜種ku rstakiである、請求項5に記載の形質転換宿主細胞。 7.請求項1に記載のヌクレオチド配列によりコードされた単離タンパク質お よび/または該タンパク質の部分。 8.配列表の配列番号2に示されるアミノ酸配列、または、それに関連し、置 換、欠失、挿入および/または逆位によりそれから誘導可能なアミノ酸配列の全 部または一部を含む、単離タンパク質。 9.摂食活動阻害および/または成虫および/または幼虫のコガネムシ、特に Melolontha種およびそれに緊密に関連した種を死滅させるために、単独でまたは 微生物を含む他の物質の少なくとも1つと併用した、請求項7または8に記載の タンパク質の使用。 10.不働化、特に摂食活動阻害、および/または植物に有害な土壌伝播性生 物体および/または真菌の死滅および/または伝染病のために、単独でまたは微 生物を含む他の物質の少なくとも1つと併用した、請求項7または8に記載のタ ンパク質の使用。 11.細菌胞子、特にBacillus popilliaeおよび/またはBacillus thuringie nsisの胞子を他の物質(の1つ)として使用する、請求項9または10に記載の 使用。 12.真菌胞子を他の物質(の1つ)として使用する、請求項9または10に 記載の使用。 13.細胞融解タンパク質および/またはコガネムシの消化管上皮のレセプタ ータンパク質を他の物質(の1つ)として、特に請求項7または8に記載のタン パク質との融合タンパク質の形態で使用する請求項9または10に記載の使用。 14.請求項1に記載の1つ以上のポリヌクレオチドを、微生物(例えば、細 菌、ウイルス、真菌または原生動物)に、または動物細胞または植物細胞培養物 に導入(形質転換)し、微生物または細胞に天然に存在するか、または組換えの 結果存在するプロモーター、好ましくは調節可能なプロモーターで調節および制 御し、発現させる、請求項7または8に記載のタンパク質の製造法。 15.ポリヌクレオチドをBacillus thuringiensis種の細菌に導入する、請求 項14に記載の方法。 16.全部またはいくつかの植物組織において、Bacillus popilliaeに特徴的 な結晶タンパク質と同一または類似であり、摂食活動阻害および/または成虫お よび/または幼虫コガネムシ、特にMelolontha種およびそれに関連した種の死滅 に適当である結晶タンパク質を合成することができる、トランスジェニック植物 の製造における、請求項1に記載のポリヌクレオチドの使用。 17.Bacillus popilliaeに天然に存在する特徴的な結晶タンパク質と同一ま たは類似の結晶タンパク質をコードする請求項1に記載のポリヌクレオチドを含 む(組換え)異種DNAを、ゲノムに安定に組込んで遺伝子操作した形質転換細 胞であって、その細胞のポリメラーゼにより認識される、細胞に天然に存在する か、組換えの結果存在するプロモーターの制御下でそのタンパク質が発現され形 質転換細胞。 18.その発現によりBacillus popilliaeに天然に存在する特徴的な結晶タン パク質と同一または類似のタンパク質を生成する請求項1に記載の異種ポリヌク レオチドを含む組換え遺伝物質をもつ植物体または植物組織または植物生殖物質 を製造する方法であって、それら植物体の細胞または組織を、請求項1に記載の ポリヌクレオチドおよび植物細胞においてポリヌクレオチドの安定な組込みと発 現をもたらし得る調節ヌクレオチド配列を含む組換えDNAで形質転換し、次い で、植物体またはその生殖物質またはその両方を、異種DNAで形質転換した植 物細胞から、または対応する組織から再生し、望ましい場合には、再生した植物 またはその生殖物質、またはその両方を生物学的に再生産する方法。
