- 【特許請求の範囲】 1.異種タンパク質産物の発現のために有用な宿主細胞であって、親細胞に比 較して、有意に低下したレベルのメタロプロテアーゼを発現するために遺伝子的 に修飾されているものである、前記細胞。 2.酵母細胞である、請求項1に記載の宿主細胞。 3.サッカロミセス属の株、特にサッカロミセス・セレビシエである、請求項 2に記載の宿主細胞。 4.糸状菌である、請求項1に記載の宿主細胞。 5.請求項4に記載の宿主細胞であって、アクレモニウム属、アスペルギルス 属、カンジダ属、コクリオボルス属、エンドチア属、フザリウム属、フミコーラ 属、ニューロスポラ属、リゾムコル属、リゾプス属、サーモミセス属、トリコデ ルマ属、ポドスポラ属、ピリクラリア属及びペニシリウム属からなる群から選択 した株である、前記宿主細胞。 6.請求項5に記載の宿主細胞であって、アスペルギルス・オリザエ、アスペ ルギルス・ニガー、アスペルギルス・ニドラン、アスペルギルス・アワモリ、ア スペルギルス・フォエニシス、アスペルギルス・ジャポニクス、アスペルギルス ・フォエツス、フザリウム・グラミネアルム、フザリウム・オキシスポルム、フ ザリウム・ソラニ、フミコーラ・グリセア、ニューロスポラ・クラッサ、ペニシ リウム・クリソゲヌム、リゾムコル・メイヘイ、トリコデルマ・リーセイ及びト リコデルマ・ビリデからなる群から選択した株である、前記宿主細胞。 7.請求項1〜6のいずれか1項に記載の宿主細胞であって、その中の前記メ タロプロテアーゼがフザリウム属のメタロプロテアー ゼである、前記宿主細胞。 8.請求項7に記載の宿主細胞であって、その中の前記メタロプロテアーゼが 、フザリウム・オキシスポルムのメタロプロテアーゼである、前記宿主細胞。 9.請求項8に記載の宿主細胞であって、その中の前記メタロプロテアーゼが 、配列番号2として提示されたアミノ酸配列またはこれと相同の配列を有するフ ザリウム・オキシスポルムp45メタロプロテアーゼである、前記宿主細胞。 10.請求項1〜6のいずれか1項に記載の宿主細胞であって、その中の前記メ タロプロテアーゼが約pH6〜8の範囲の最適タンパク質分解活性を有する中性メ タロプロテアーゼである、前記宿主細胞。 11.請求項10に記載の宿主細胞であって、その中の前記メタロプロテアーゼが NpIまたはNpIIの群の中性のアスペルギルス属のメタロプロテアーゼである、前 記宿主細胞。 12.請求項11に記載の宿主細胞であって、その中の前記メタロプロテアーゼが 、配列番号4として提示された部分ヌクレオチド配列またはこれと相同の配列を 含むcDNA配列によりコードされる、アスペルギルス・オリザエ中性メタロプロテ アーゼI(NpI)である、前記宿主細胞。 13.請求項1〜12のいずれか1項に記載の宿主細胞であって、前記メタロプロ テアーゼをコードする構造領域または調節領域で遺伝子的に修飾されたものであ る、前記宿主細胞。 14.請求項13に記載の宿主細胞であって、特異的もしくはランダム変異誘発、 PCR生成変異誘発、部位特異的 DNA欠失、挿入及び/もしくは置換、遺伝子破壊 もしくは遺伝子置換技術、アンチセンス技術またはそれらの組合せにより遺伝子 的に修飾されたものである 、前記宿主。 15.請求項1〜14のいずれか1項に記載の宿主細胞であって、その細胞中で発 現されたメタロプロテアーゼのレベルが約50%よりも多く、好ましくは約85%よ りも多く、より好ましくは約90%よりも多く、最も好ましくは約95%よりも多く 低下しているものである、前記宿主細胞。 16.請求項1〜14のいずれか1項に記載の宿主細胞であって、本質的にいかな るメタロプロテアーゼ活性をも欠いている、前記細胞。 17.請求項1の宿主細胞中で異種タンパク質産物を生産する方法であって、そ の方法は、 (a)前記宿主細胞に前記タンパク質産物をコードしている核酸配列を導入し、 (b)適当な増殖培地中で、(a)段階の宿主細胞を培養し、そして (c)前記異種タンパク質産物を単離することを含むものである、前記方法。 18.前記宿主細胞が酵母細胞である、請求項17に記載の方法。 19.前記宿主細胞がサッカロミセス属、好ましくはサッカロミセス・セレビシ エである、請求項18に記載の方法。 20.前記宿主細胞が糸状菌である、請求項17に記載の方法。 21.請求項20に記載の方法であって、前記宿主細胞が、アクレモニウム属、ア スペルギルス属、カンジダ属、コクリオボルス属、エンドチア属、フザリウム属 、フミコーラ属、ニューロスポラ属、リゾムコル属、リゾプス属、サーモミセス 属、トリコデルマ属、ポドスポラ属、ピリクラリア属及びペニシリウム属からな る群から選択した株である、前記方法。 22.請求項21に記載の方法であって、アスペルギルス・オリザエ、アスペルギ ルス・ニガー、アスペルギルス・ニドラン、アスペルギルス・アワモリ、アスペ ルギルス・フォエニシス、アスペルギルス・ジャポニクス、アスペルギルス・フ ォエツス、フザリウム・グラミネアルム、フザリウム・オキシスポルム、フザリ ウム・ソラニ、フミコーラ・グリセア、ニューロスポラ・クラッサ、ペニシリウ ム・クリソゲヌム、リゾムコル・メイヘイ、トリコデルマ・リーセイ及びトリコ デルマ・ビリデからなる群から選択した株である、前記方法。 23.請求項17〜22のいずれか1項に記載の方法において、前記メタロプロテア ーゼがフザリウム属のメタロプロテアーゼである、前記方法。 24.請求項23に記載の方法であって、前記メタロプロテアーゼが、フザリウム ・オキシスポルムのメタロプロテアーゼである、前記方法。 25.請求項24に記載の方法であって、前記メタロプロテアーゼが、配列番号2 として提示されたアミノ酸配列、またはそれに相同の配列を有している、フザリ ウム・オキシスポルムp45メタロプロテアーゼである、前記方法。 26.請求項17〜22のいずれか1項に記載の方法であって、前記メタロプロテア ーゼが約pH6〜8の範囲の最適タンパク質分解活性を有する中性メタロプロテア ーゼである、前記方法。 27.請求項26に記載の方法であって、前記メタロプロテアーゼがNpIまたはNp IIの群の中性のアスペルギルス属のメタロプロテアーゼである、前記方法。 28.請求項27に記載の方法であって、前記メタロプロテアーゼが、配列番号4 として提示された部分ヌクレオチド配列またはそれと 相同の配列を含むcDNA配列によりコードされる、アスペルギルス・オリザエの中 性メタロプロテアーゼI(NpI)である、前記方法。 29.請求項17〜28のいずれか1項に記載の方法であって、前記宿主細胞が前記 メタロプロテアーゼをコードする構造領域または調節領域で遺伝子的に修飾され たものである、前記方法。 30.請求項29に記載の方法であって、前記宿主細胞は、特異的もしくはランダ ム変異誘発、PCR生成変異誘発、部位特異的 DNA欠失、挿入及び/もしくは置換 、遺伝子破壊もしくは遺伝子置換技術、アンチセンス技術またはそれらの組合せ により遺伝子的に修飾されたものである、前記宿主。 31.請求項17〜30のいずれか1項に記載の方法であって、宿主細胞により発現 されたメタロプロテアーゼのレベルが、50%よりも多く、好ましくは85%よりも 多く、より好ましくは90%よりも多く、最も好ましくは95%よりも多く低下して いる、前記方法。 32.請求項17〜30のいずれか1項に記載の方法であって、前記宿主細胞により 発現された産物が、本質的にいかなるメタロプロテアーゼ活性をも欠いている、 前記方法。 33.請求項17〜32のいずれか1項に記載の方法であって、前記タンパク質産物 が、真核の酵素、たとえばインスリン、成長ホルモン、グルカゴン、ソマトスタ チン、インターフェロン、PDGF、VII因子、VIII因子、ウロキナーゼ、EPO、キモ シン、組織プラスミノーゲン活性化剤または血清アルブミンである、前記方法。 34.請求項17〜32のいずれか1項に記載の方法であって、前記タンパク質産物 が真菌起源のタンパク質である、前記方法。 35.請求項34に記載の方法であって、前記タンパク質産物が、真菌の酵素、特 にアミロース分解酵素、たとえば、α−アミラーゼ、β−アミラーゼ、グルコア ミラーゼ、β−ガラクトシダーゼ、セル ロース分解酵素、脂質分解酵素、キシラン分解酵素、タンパク質分解酵素、酸化 還元酵素、たとえば、ペルオキシダーゼもしくはラッカーゼ、ペクチナーゼ、ま たはクチナーゼである、前記方法。 36.請求項17〜32のいずれか1項に記載の方法であって、前記タンパク質産物 が細菌のタンパク質である、前記方法。 37.請求項36に記載の方法であって、前記タンパク質産物が、細菌の酵素、特 にアミロース分解酵素、たとえば、α−アミラーゼ、β−アミラーゼ、グルコア ミラーゼ、β−ガラクトシダーゼ、セルロース分解酵素、脂質分解酵素、キシラ ン分解酵素、タンパク質分解酵素、酸化還元酵素、たとえば、ペルオキシダーゼ もしくはラッカーゼ、ペクチナーゼまたはクチナーゼである、前記方法。 38.請求項17〜37のいずれか1項に記載の方法であって、前記タンパク質産物 が、前駆体タンパク質、すなわち、チモーゲン、ハイブリッドタンパク質、プロ 配列もしくはプレ−プロ配列として得られた、または未成熟の形のタンパク質で ある、前記方法。
