Claims (1)
- 【特許請求の範囲】 1.精製された哺乳類トロンボポエチンであって、 a)変性条件下でSDS−ポリアクリルアミドゲル電気泳動により決定される場 合、Mr=70,000±10,000ドルトン;及び b)SDS−ポリアクリルアミドゲル電気泳動及び銀染色法により決定される場 合、汚染性タンパク質に対して少なくとも90%純粋であることによって特徴づけ られる精製されたトロンボポエチン。 2.Mr<55KDのTPO種を実質的に有さない請求の範囲第1項記載の精製された トロンボポエチン。 3.マウストロンボポエチンである請求の範囲第1項記載の精製されたトロン ボポエチン。 4.前記トロンボポエチンが残基45(Ser)から残基379(Thr)の、配列番号2に 示されるアミノ酸残基の配列を含んで成る請求の範囲第3項記載の精製されたト ロンボポエチン。 5.霊長類トロンボポエチンである請求の範囲第1項記載の精製されたトロン ボポエチン。 6.ヒトトロンボポエチンである請求の範囲第5項記載の精製されたトロンボ ポエチン。 7.前記トロンボポエチンが残基22(Ser)から残基353(Gly)の、配列番号4に 示されるアミノ酸残基の配列を含んで成る請求の範囲第6項記載の精製されたト ロンボポエチン。 8.生物学的流体からトロンボポエチンを精製するための方法であって、 MPLレセプターのリガンド−結合ドメインを含んで成る、固体支持体に結合さ れるポリペプチドに、トロンボポエチンを含む生物学的流体を適用し、それによ り前記トロンボポエチンが前記ポリペプ チドに吸着され; 吸着されていない物質を溶離するために前記ポリペプチドを洗浄し; 前記ポリペプチドから吸着されたトロンボポエチンを溶離し; 前記溶離されたトロンボポエチンをアニオン交換クロマトグラフィーにより分 別し;そして 前記分別されたトロンボポエチンを集めることを含んで成る方法。 9.前記生物学的流体がならし細胞培養培地又は乳汁である請求の範囲第8項 記載の方法。 10.前記生物学的流体が濃縮されたならし細胞培養培地である請求の範囲第8 項記載の方法。 11.生物学的流体をレセプターポリペプチドに適用する前、その生物学的流体 を濃縮する段階をさらに含んで成る請求の範囲第8項記載の方法。 12.前記固体支持体が架橋されたアガロースビーズである請求の範囲第8項記 載の方法。 13.前記集められたトロンボポエチンが、 a)変性条件下でSDS−ポリアクリルアミドゲル電気泳動により決定される場 合、Mr=70,000±10,000ドルトン;及び b)SDS−ポリアクリルアミドゲル電気泳動及び銀染色法により決定される場 合、汚染性タンパク質に対して少なくとも90%純粋であることによって特徴づけ られる請求の範囲第8項記載の方法。 14.前記集められたトロンボポエチンがMr<55KDのTPO種を実質的に有さない 請求の範囲第13項記載の方法。 15.前記トロンボポエチンがマウストロンボポエチンである請求の範囲第8項 記載の方法。 16.前記トロンボポエチンが霊長類トロンボポエチンである請求の範囲第8項 記載の方法。 17.前記トロンボポエチンがヒトトロンボポエチンである請求の範囲第8項記 載の方法。 18.前記ポリペプチドがマウス又はヒトMPLレセプターのリガンド−結合ドメ インを含んで成る請求の範囲第8項記載の方法。 19.前記ポリペプチドが配列番号7の残基27〜480から実質的に成る請求の範 囲第18項記載の方法。 20.医薬的に許容できるビークルと共にトロンボポエチンを含んで成る医薬組 成物であって、ここで前記トロンボポエチンが、変性条件下でSDS−ポリアクリ ルアミドゲル電気泳動により決定される場合、Mr=70,000±10,000ドルトンによ り特徴づけられ、そして前記組成物がMr<55KDのトロンボポエチン種を実質的に 有さないことを特徴とする医薬組成物。
