トロンボポイエチン組成物
発明の背景
造血は、骨髄中の多能性の幹細胞から、血球が発生し、分化する過程である。
この過程は標的細胞上の膜結合受容体を経て作用するポリペプチド成長因子(サ
イトカイン)の複雑な相互作用を伴う。サイトカイン作用は、しばしば系統特異
的及び/または段階特異的である特定のサイトカイン対する反応を伴う、細胞増
殖及び分化を生じる。幹細胞からの単細胞型、たとえば、血小板の発生は、適当
な連鎖中で作用する多数のサイトカインの調和して機能する作用を必要とするか
もしれない。
種々のサイトカインが治療剤として開発された。たとえば、赤血球の発生を刺
激するエリトロポリエチンは腎不全から生じる貧血の治療に用いられる。患者の
免疫システムの回復の速度を速めるためにがんの化学療法とともにいくつかのコ
ロニー刺激因子が、用いられてきた。インターロイキン−2、α−インターフェ
ロン及びγ−インターフェロンが特定のがんの治療に用いられる。
巨核球新生及び血小板新生を刺激する活性が血小板減少症の動物の体液中に同

ヒトトロンボポイエチン(TPO)は332アミノ酸残基の70kDの糖タンパク質である
。それは約152残基のアミノ末端成長因子ドメインと炭水化物に富むカルボキシ
末端ドメインを含有する。アミノ末端ドメインを含む TPOの端を切り取った形は
、試験管内及び生体内において生物活性を示す。非−ヒト TPOは科学文献中にも
記載されている(たとえば、Lok他の同書、Bartley他の同書及びShimada他の「B
lood」84(10補遺1):326a,1994年)。
トロンボポイエチンは、タンパク質分解を受けやすいようであり、異種の形ま
たは、分解した形で単離された(Bartley他の同書、de Sauvage他の同書)。し
たがって、科学文献中に報告されたトロンボポイエチンの標品は、組成及び種々
の分子種の相関的な活性がよく特徴づけられていない。けれども、少なくともい
くつかのタンパク質分解生成物が生物的に活性である。しかしながら、現在まで
トロンボポイエチンの大規模生産に関してほとんど研究がなされなかった。そし
て、医薬用途に適切な TPOの組成物についての、当分野においてニーズがある。
このような製剤は貯蔵について安定で用い易くあるべきである。
発明の概要
本発明の目的は、貯蔵において安定な、水性組成物を含む、TPOの医薬組成物
を供給することである。
本発明の更なる目的は、TPOの医薬組成物の調製及び包装で用いられる、貯蔵
びん及びフィルターの表面を含む表面への TPOの吸着を減少させる方法をもたら
すことである。
本発明の追加の目的は水溶液及び凍結乾燥粉末を含む、TPOの医
薬組成物の安定化方法をもたらすことである。
一面では本発明は、TPO、生理的に許容し得る緩衝液、非イオン界面活性剤及
びポリオールからなる群から選択される表面吸着抑制剤並びに等張量の生理的に
許容し得る塩を含む組成物をもたらす。
TPOはヒト TPOまたは非−ヒト(たとえば、ラット、マウス、犬または非−ヒト
霊長目の動物)TPOであってもよい。本発明の特定の態様の範囲内で、組成物は
pH5.0〜7.0、好ましくは 5.5〜6.5 の水溶液である。代りの態様では組成物は凍
結乾燥された粉末である。さらなる態様では、組成物はトロンボポイエチンの凝
集を減少させるのに十分な量のヒスチジンを含む。他の態様では、組成物は、ト

発明の詳細な説明
用語「等張」は、この明細書ではその慣用の意味(すなわち、血液の張度に等
しい張度であって、NaClの 0.9%溶液に等しい)について用いられる。「等張量
」の塩は、等張溶液を作成または凍結乾燥された製剤の再形成に関する等張溶液
を生成するのに必要な量である。
濃度は本明細書ではモル単位または液体組成物のw/v%で特定化されている
。組成物が凍結乾燥された粉末の形である時、各々の成分の濃度は、粉末の再形
成について特定化された濃度をもたらすようなものとなるだろう。
本発明の組成物は表面吸着、凝集、他の物理的もしくは化学的分解またはこれ
らの過程の組合せによる TPO活性の喪失を減少させるように処方される。本発明
者により、非イオン界面活性剤及びポリオールが表面、たとえばフィルターまた
はガラスびんへの TPOの吸着を減少させることができることを見い出した。
本発明によって、 TPOを緩衝液、非イオン界面活性剤及びポリオ
ールからなる群から選択される表面吸着抑制剤並びに等張量の生理的に許容し得
る塩と組合せる。組成物は水溶液または凍結乾燥された粉末として処方し得る。
後者は使用の前に医薬として許容し得る希釈剤、たとえば注射用の滅菌水で再形
成される。
本発明で用いるための緩衝液は、pH範囲 5.0〜7.0 の範囲で有効な、低イオン
強度の生理的に許容し得る緩衝液を含む。このような緩衝液は、リン酸塩、酢酸
塩、クエン酸塩、コハク酸塩及びヒスチジン緩衝液を含む。「低イオン強度」は
5〜500mM、好ましくは10〜100mM、最も好ましくは約20mMを意味する。リン酸ナ
トリウム及びカリウム緩衝液並びにヒスチジン緩衝液が好ましい。組成物のpHは
、5.5〜6.5 が好ましく、約 6.0が最も好ましい。特に好ましい緩衝液は20mMの
リン酸ナトリウム緩衝液、pH6.0(16mMのリン酸モノ塩基性ナトリウム+4 mMのリ
ン酸ジ塩基性酸ナトリウム)である。
本発明で有用な表面吸着抑制剤は非イオン界面活性剤及びポリオールを含む。
非イオン界面活性剤は、ポリオキシエチレンソルビタン脂肪酸エステル、たとえ
ばポリソルベート20(ポリオキシエチレンソルビタン モノラウレート)、ポリ
ソルベート80(ポリオキシエチレンソルビタン モノオレエート)等を含む。こ
れに関して有用な他の非イオン界面活性剤はポリエチレンオキシド、ソルビタン
エステル、ポリオキシエチレンアルキルエーテル、並びにグリセリルモノオレエ
ート及びグリセリルモノステアレートを含む脂肪酸のグリセリドを含む。本発明
で用いるポリオールは、ポリエチレングリコール(たとえばPEG3350)、マンニト
ール、キシリトール、ソルビトール、イノシトール及びグリセロールを含む。一
般に、表面吸着抑制剤は濃度 0.001%〜5%、好ましくは0.01%〜 1.0%、より
好ましくは約0.05%で含まれるだろう。特に好ましい表面吸着抑制剤は濃度約0.
05%のポリソルベート80である。表面吸着抑制剤の組
合せ(たとえばポリソルベート80+PEG3350)も用いることができる。
等張量の生理的に許容し得る塩は、非イオン界面活性剤を含有する水溶液中に
ある場合、TPOの表面吸着を更に減少させることが見い出された。これに関して
好ましい塩は塩化物、たとえば、NaCl,KCl,CaCl2及びMgCl2を含む。NaClは特
に好ましい。当業者に明らかなように、等張性を達成するのに必要な塩の実際の
量は選択された特定の塩及び組成物の他の成分のような因子に依存するだろう。
組成物中に上記で開示された特定の成分の組合せを含ませることは好都合であ
る。たとえば、非−ヒスチジン緩衝化組成物にヒスチジンを添加すると組成物中
の TPOをさらに安定化することができる。ヒスチジンが TPOの凝集を減少させ、
pH>5.5 での非−タンパク質分解性分解(たとえば、脱アミド化または酸化)も
減少させ得ることを実験的な証拠が示している。したがって、ヒスチジンを、た
とえばリン酸塩緩衝化 TPO組成物中に、濃度5〜500mM、好ましくは10〜100mM、
より好ましくは約20mMで含むことができる。ポリエチレングリコールまたは他の
ポリオールは非イオン界面活性剤を含有する組成物に、さらに表面吸着を減少さ
せるために加えることができる。ヒスチジン及びポリオールは凍結乾燥された組
成物に凝集を減少させるために有利に含ませることができる。
アルブミンも本発明の組成物に包含させることができる。非−ヒト血清アルブ
ミンが試薬級組成物または獣医用を目的とするもの(動物実験用を含めて)で用
いることができるが、ヒト血清アルブミンがヒト用を目的とする医薬組成物中の
含有物のために好ましい。アルブミンは凍結乾燥組成物中の賦形剤として有用で
あり、濃度0.01〜 1.0%、好ましくは約0.25%で含まれる時、安定剤として作用
する。
1または複数の保存剤も、本発明の組成物、特に多用途のために包装された組
成物中に含ませることができる。本発明の組成物に用いることができる保存剤は
医薬製剤中に普通に用いられるもの、たとえば、メチルパラベン、プロピルパラ
ベン、ベンジルアルコール、m−クレゾール、チオサリチル酸エチル水銀、フェ
ノール、チメロサール等を含む。
糖を本発明の組成物中で、増量剤、安定剤、または等張性調整剤として含んで
よい。糖は凍結乾燥組成物の増量剤及び安定剤として特に重要である。適当な糖
は、デキストロース、ラクトース、マルトース、スクロース、トレハロース等を
含む。
TPOの医薬組成物は、慣用の方法により、非経口、特に静脈内または皮下デリ
バリーのために処方される。製剤化方法は当業界で周知であって、たとえば「レ
ミントンの医薬の科学(Remington's Pharmaceutical Sciences)」(Gennaro編、
Mack Publishing Co.,Easton PA,1990年。これは参照により本明細書に組入れ
る)に開示されている。医薬組成物では、もっと高濃度の溶液が供給され、使用
の前に希釈することができるが、一般的には TPOの濃度は 0.1〜10mg/mlの範囲
内である。この好ましい範囲外の TPO濃度の組成物も、たとえば研究試薬として
用いるのに製造することができる。上記開示されたものに加えて、当業界で公知
目的、たとえば凍結乾燥の前に加える増量剤、のための成分を含むことができる
。これらの組成物はさらに他のサイトカイン、特に初期に作用するサイトカイン
、たとえば幹細胞因子、IL−3,IL−6,IL−11またはGM−CSF を含有すること
ができる。
医薬用途を目的とす るTPO組成物は無菌で、発熱因子がないであろうし、許容
された医薬的方法にしたがって、製造及び包装される
だろう。組成物は単位用量または多数回用量で包装することができる。組成物は
一般に、ポリテトラフルオロエチレンで裏打ちした栓及び適当なラベルを有する
密封したガラスびん中に包装されるだろう。凍結乾燥された組成物は、適当な量
の適切な希釈剤、たとえば注射のための水(WFI)または5%デキストロース WFI
溶液を含むキットとして包装することができる。
本発明のトロンボポイエチン組成物は、骨髄系統の細胞の増殖が望ましいとこ
ろではどこでも、たとえば、形成不全の貧血、骨髄形成異常症候群、化学療法ま
たは先天性の血球減少症により誘発される血球減少症の治療において、治療上で
用いることができる。TPOは特に、血小板生産、たとえば血小板減少症の治療に
有用である。血小板減少症は、その症状を単独または協力して作り出す、多様な
群の病気及び臨床状態に伴う。減少した血小板数は、たとえば、血小板生産にお
ける欠陥、異常な血小板分布、大量の輸血による希釈的な減量または異常な血小
板破壊によることがある。たとえばがん治療に用いられる化学療法薬は骨髄中の
血小板始原細胞の発生を抑制し、そして、生じた血小板減少症は化学療法を制限
し、輸血を必要とするかもしれない。さらに、特定の悪性疾患は血小板生産及び
血小板分配を損うことがある。悪性細胞を殺すのに用いられる放射線療法も血小
板始原細胞を殺す。血小板減少症は薬、新生児の同種免疫または血小板輸血同種
免疫により誘発される種々の血小板自己免疫疾患からも生ずるかもしれない。TP
Oは輸血の必要性を減少または取り除き、それにより、血小板同種免疫の発生を
減少させることができる。血小板の異常破壊は、(1)血管の移植片または傷つ
けられた組織における増加した血小板の消費または(2)たとえば薬物誘発血小
板減少症、特発性血小板減少性紫斑病(ITP)、自己免疫病、血液学病、たとえば
、白血病及びリンパ腫もしくは骨髄を含
む転移がんを伴う免疫メカニズムに起因することがある。他の TPOの兆候は形成
不全の貧血及びたとえば化学療法または AZTを用いる HIV感染の治療に起因する
、薬物誘発骨髄抑制を含む。
血小板減少症は、増加した出血、たとえば鼻−口付近、胃腸管からの粘膜の出
血、並びに傷、潰瘍または注射部位からの分泌として証明される。
治療用量は一般に1日当り患者の重量1kg当り 0.1〜 100μgの範囲で、好ま
しくは1日当り 0.5〜50μg/kg、より好ましくは1〜25μg/kg/日で、正確
な用量は、治療されるべき症状の性質及び程度、患者の特性等を考慮に入れて、
許容できる基準にしたがって臨床医が決定する。用量の決定は当業者のレベル内
である。TPOは一般に、化学療法もしくは骨髄移植後に28日までの期間または血
小板数が>20,000/mm3、好ましくは>50,000/mm3が達成されるまで投与される
だろう。より一般的には、TPOは1週間またはそれよりも少なく、しばしば1〜
3日の期間にわたって投与されるだろう。一般に、TPOの治療的に有効量は、リ
ンパ球または骨髄始原細胞の増殖及び/または分化の臨床的に有意な増加を生成
するのに十分な量であって、それは、成熟細胞(たとえば、血小板、赤血球また
は好中球)の循環レベルにおける増加として示されるだろう。血小板疾患の治療
は、したがって、血小板数が少なくとも20,000/mm3、好ましくは50,000/mm3に
到達するまで継続されるだろう。TPOは、他のサイトカイン、たとえばIL−3,I
L−6及びIL−11、幹細胞因子、エリトロポイエチン(EPO)、G−CSF 並びにGM−
CSF といっしょに投与することもできる。組合せ療法の養生法では、他のサイト
カインの毎日の用量は一般に、EPOでは< 150U/kg、GM−CSF では5〜15μg
/kg、IL−3では1〜5μg/kg及びG−CSFでは1〜25μg/kgであろう。EPO
との組合せ療法は、たとえば低
EPOレベルの貧血の患者で必要である。
TPOは体外、たとえば自己由来の骨髄培養でも用いることができる。簡単に言
うと、化学療法の前に患者から骨髄を除去し、任意に1または複数の他のサイト
カインと組合せた TPOで処理する。次いで、骨髄の回復の速度を早めるために化
学療法後に、処理された骨髄を患者に戻す。さらに、TPOは骨髄または末梢血液
始原(PBPC)細胞の体外増殖のために用いることができる。化学療法に先立って
、末梢循環中に早期に始原細胞を遊離させるために、骨髄を幹細胞因子(SCF)ま
たはG−CSFで刺激することができる。これらの始原体を末梢血液から捕集し、
濃縮し、次いで、任意に SCF,G−CSF,IL−3,GM−CSF,IL−6またはIL−11
を含むが、それらに限定されない1または複数の他のサイトカインと組合せた T
POで培養を処理して高密度の巨核球培養に分化及び増殖させることができ、次に
それを高用量化学療法後に患者に戻すことができる。
TPOは造血過程の研究における実験試薬としても有用である。本発明の組成物
及び方法は上記用途のためのトロンボポイエチンの貯蔵安定性製剤をもたらす。
本発明をさらに次の非限定的な例により説明する。
例
組換えヒト及びマウス TPOを赤ちゃんハムスターのトランスフェクトされた腎
臓細胞(BHK570細胞、ATCC CRL 10314)を用いて調製した。TPOを固定化された
MPL受容体上のアフィニティークロマトグラフによるか、または染料−リガンド
アフィニティークロマトグラフ、イオン交換クロマトグラフ及びヒドロキシアパ
タイトでのタンパク質汚染物の吸着の組合せを用いてクロマトグラフ的に細胞順
化培地から精製した。TPOの生物活性を、標的細胞として、ヒト M
PL受容体をコードする発現ベクターをトランスフェクトしたBaF3細胞(Vigon他「
Proc.Natl.Acad.Sci.USA」89.p.5640〜5644、1992年)を用いる有糸分裂誘
発アッセイで検定した。BaF3は、インターロイキン−3に依存する、マウス骨髄
から由来したプレ−リンパ球系統である(Palacios及びSteinmetz「Cell」41.p
.727〜734、1985年、Mathey-Prevot 他「Mol.Cell.Biol.」6.p.4133〜4135
、1986年)。細胞を3H−チミジンの存在する試験サンプルに暴露した。細胞 DN
Aに取り込まれた3H−チミジンの量をヒト TPOの標準曲線との比較により定量し
た。有糸分裂誘発アッセイで最大の半分の刺激を与える量は10U/mlと定義され
た。
ポリソルベート80(トゥイーン80)、NF(National Formuraly)級、をSpectrum
Chemical Mfg.Corp.(ニュージャージー州、ニューブルンスヴィク)から得
た。塩化ナトリウム(AR、米国薬局方)、リン酸ナトリウム、2塩基性、7水化
物(米国薬局方、TAC)、リン酸ナトリウム、1塩基性、1水和物(米国薬局方)
はMallinkrodt Chemical Corp.(ミズーリー州、チェスタフィールド)から得
た。
例1
精製されたトロンボポイエチンの表面吸着をフィルター回収率及びガラスビー
スとインキュベートした溶液からの回収率により評価した。TPOのサンプルを試
験のための適当な緩衝液(表参照)で希釈した。
ろ過吸着を注射器に取り付けられた 0.2μmのポリフッ化ビニリデン膜フィル
ター{Durapore(商標)フィルター、Millipore、マサチュセッツ州、ベッドフ
ォード}またはAcrodisc(商標)膜(Gelman Sciences、ミシガン州、アン ア
ーボア)を用いて測定した。 TPO溶液を手で押してフィルターを通した。サン
プルは、注射器に溶液を加える前及びフィルター通過後に採取した。組換えマウ
スTPOの回収率は、0.25%のヒト血清アルブミンを含有する製剤の方がアルブミ
ンを含まないものより高いことが見い出された(図1)。同様な結果が組換えヒ
ト TPOについても得られた。試験した範囲(pH4〜8)にわたってpHの関数とし
て大きなフィルター回収の差はなかったが、回収は pH6.0で最大であった。組換
えヒト TPOの表面吸着も、製剤中に0.01%のポリソルベート80が存在する時に減
少した(図2)。等張性塩化ナトリウムはポリソルベート80の存在でさらに吸着
を減少させることが見い出された(図3)。
ガラスビーズを用いて、誇張した表面:面積比の下でさらに表面
ンプルを管ロッカー上で5℃で 1.0gのガラスビーズ(425〜 600μm)に暴露し
た。分裂促進作用を 1.5,5及び24時間後に測定して時間の関数として吸着を評
価した。図4に示すように、吸着は24時間の試験期間にわたって時間と共に増加
した。吸着はポリソルベート20よりもポリソルベート80によって大きい量で減少
した。回収率はポリソルベート80の濃度が 0.001%〜0.05%に増加するにつれて
増加した。ポリソルベート80と等張性塩化ナトリウムの組合せは、ポリソルベー
ト80単独よりも表面吸着を減少させるのに効果がある。
例2
溶液中の TPOの安定性に関するpHの影響をリン酸塩及びヒスチジ
ン緩衝液中で試験した。アフィニティ精製及びクロマトグラフ的に精製された組
換えマウス及びヒト TPOのサンプルを適当な緩衝液で希釈するか、またはNaClも
しくは HClを用いて所望のpHまで滴定した(表参照)。次いでサンプルを種々の
温度で貯蔵し、上記のように有糸分裂誘発アッセイ及び SDSポリアクリルアミド
ゲル電気泳動(SDS PAGE)、続いてウェスタンブロットにより断続的に検定した
。凝次1次分解速度定数(Kobs)を、適当な時に残存する分裂促進活性%対時
間の対数直線状適合度のスロープから評価した。
SDS PAGE/ウェスタンブロットは、アフィニティ精製した組換えヒト TPOはpH
及び時間に依存する風に、低分子量変異体へタンパク質分解を受ける。pH範囲6
〜8では、タンパク質分解は2つの低い分子量の変異体(ゲル電気泳動により35
及び18kD)に分裂を生じ、タンパク質分解は温度の関数としてpHの増加につれ増
加した。pH≦
よりも、わずかに低い分解生成物がはっきり見え、温度の関数として、pHの減少
につれて増加した。
クロマトグラフ的に精製した組換えヒト TPOは異なる分解プロフィルを示した
。SDS PAGE/ウェスタンブロット分析は、pH<5で2つの低分子量種(ゲル電気
泳動で30及び20kD)へと温度依存性の開裂を示し、タンパク質分解はpHの減少と
共に増加した。pH>6では、有意なタンパク質分解は観察されなかったが、pH及
び温度の上昇で高分子量凝集物が生成した。すべての分解生成物は親タンパク質
よりも低い生物活性を有していた。偽似一次分解速度(Kobs)を示すpH−速度
プロフィル(Kobs対pHで)は、温度の関数として、
種々の一連の酵素触媒反応(pathway)の相対的分解速度の差を示唆
を示した。生物活性の急速な喪失はpH>6で起こるが、肉眼で見え
るほどの変化はSDS PAGE及びウェスタンブロットによって明らかでなかった。こ
れらの結果は、pH>6では分解は主として脱アミド化、酸化または電気泳動によ
って検出されない、いくつかの他のメカニズムを介して起こる。5℃での分解速
度はpH<では相対的に一定で、このpH範囲における一連の触媒反応は大きな活性
化エネルギー(急勾酸の温度依存)を有し、この温度での TPOの総合損失に有意
に寄与しない。
リン酸ナトリウム及びカリウム緩衝液を用いた結果を比較した時、同等の分解
速度がpHの関数として見い出された。異なった濃度のリン酸カリウム緩衝液を用
いた時(20mM〜100mM,pH6.0)、有意な差は認められなかった。しかしながら、
pH範囲5〜6の緩衝液としてヒスチジンを用いた時に差が認められた。pH5.0で
の分解速度は、37℃でリン酸塩緩衝液よりもヒスチジン緩衝液で大きかったが、
pH5.5または 6.0では、分解速度は等しいか、ヒスチジン溶液において低い(図
5)。差異は重大ではないが、同様な結果が30℃で見られた。5℃では分解速度
は、pH5〜6でヒスチジン及びリン酸塩緩衝液で同等であった。これらの結果は
ヒスチジンの存在での低pH分解触媒反応を経る増加した分解と一致しているが、
ヒスチジンは高pH分解触媒反応を経る分解を減少させる。低pH分解触媒反応は温
度が減少すると阻止されるから、ヒスチジンの存在下の促進された分解は5℃で
は明らかでない。
説明の目的で本発明の特定の態様を明細書に記載したけれども、前記のものか
ら、本発明の精神及び範囲を離れることなしに種々の変更をなし得る。したがっ
て、本発明は添付された請求の範囲によって以外には限定されない。

- 【特許請求の範囲】 1.トロンボポイエチン、 生理的に許容し得る緩衝液、 非イオン界面活性剤及びポリオールからなる群から選択される表面吸着抑制剤 、並びに 等張量の生理的に許容し得る塩を含む組成物。 2.pH5.0〜7.0 の水溶液である請求項1に記載の組成物。 3.pHが 5.5〜6.5 である請求項2に記載の組成物。 4.凍結乾燥された粉末である請求項1に記載の組成物。 5.前記トロンボポイエチンがヒトトロンボポイエチンである記載の組成物。 6.前記緩衝液がリン酸塩、酢酸塩、クエン酸塩及びヒスチジン緩衝液から選 択されるものである請求項1に記載の組成物。 7.前記緩衝液がリン酸塩緩衝液であって、前記組成物がさらにトロンボポイ エチンの凝集を減少させるのに十分な量のヒスチジンを含む請求項6に記載の組 成物。 8.前記表面吸着抑制剤が非イオン界面活性剤である請求項1に記載の組成物 。 9.前記表面吸着抑制剤がポリオキシエチレンソルビタン脂肪酸エステルであ る請求項8に記載の組成物。 10.前記ポリオキシエチレンソルビタン脂肪酸エステルが、ポリソルベート20 またはポリソルベート80である請求項9に記載の組成物。 11.前記ポリソルベート20またはポリソルベート80が0.01%〜1%(w/v) の濃度で存在する請求項10に記載の組成物。 12.前記塩がNaCl,KCl,CaCl2またはMgCl2である請求項1に 記載の組成物。 13.さらにアルブミンを含む請求項1に記載の組成物。 14.さらに保存剤を含む請求項1に記載の組成物。 15.非経口用途に適切な請求項1に記載の組成物。 16.表面へのトロンボポイエチンの吸着を減少させる方法であって、前記表面 にトロンボポイエチンの水溶液を接触させる前に、前記溶液に、非イオン界面活 性剤及びポリオールからなる群から選択された有効量の表面吸着抑制剤を加える ことを含む前記方法。 17.前記表面吸着抑制剤が非イオン界面活性剤である請求項16に記載の方法。 18.前記表面吸着抑制剤がポリオキシエチレンソルビタン脂肪酸エステルであ る請求項17に記載の方法。 19.前記ポリオキシエチレンソルビタン脂肪酸エステルがポリソルベート20ま たはポリソルベート80である請求項18に記載の方法。 20.前記ポリソルベート20またはポリソルベート80を0.01%〜1%(w/v) の濃度まで加える請求項19に記載の方法。 21.トロンボポイエチンの組成物を安定化する方法であって、前記組成物に有 効量のヒスチジンを加えることを含む方法。 22.前記有効量のヒスチジンが10〜100mM である請求項21に記載の方法。 23.前記組成物が凍結乾燥された粉末の形にある請求項21に記載の方法。 24.水性医薬組成物であって、 トロンボポイエチン 10〜100mM のリン酸塩緩衝液 0.01〜 1.0%のポリソルベート20またはポリソルベート80、及び等張量の塩化 ナトリウムを含み、pHがほぼ 6.0である前記組成物。 25.さらに10〜100mM のヒスチジンを含む請求項24に記載の組成物。
