- 【特許請求の範囲】 1.(a)腫瘍関連抗原(TAA)または感染性生物に関連する抗原と結合す る抗体成分を含み、該抗体成分が可溶の免疫原性担体タンパク質と複合している 第一のワクチンを哺乳動物に投与するステップと、 (b)該TAAまたは該感染性生物に関連する抗原のエピトープによく似る抗 イディオタイプ抗体成分を含み、該抗イディオタイプ抗体成分が可溶の免疫原性 担体タンパク質と複合している第二のワクチンを該哺乳動物に投与するステップ と を含む腫瘍関連抗原(TAA)を発現する腫瘍または感染性生物に起因する疾患 に対する哺乳動物の体液性免疫応答および細胞性免疫応答を誘導する方法。 2.上記ステップ(a)の抗体成分が、 (a)マウス・モノクローナル抗体と、 (b)マウス・モノクローナル抗体に由来するヒト化抗体と、 (c)ヒト・モノクローナル抗体と、 (d)(a)、(b)または(c)に由来する抗体フラグメントと からなる群から選ばれる請求項1に記載の方法。 3.該抗体フラグメントがF(ab’)2、F(ab)2、Fab’、Fab、 Fv、sFv、および最少認識単位からなる群から選ばれる請求項2に記載の方 法。 4.該抗イディオタイプ抗体成分が、 (a)ポリクローナル抗体と、 (b)マウス・モノクローナル抗体と、 (c)(b)に由来するヒト化抗体と、 (d)ヒト・モノクローナル抗体と、 (e)類人霊長類抗体と、 (f)(a)、(b)、(c)、(d)または(e)に由来する抗体フラグメ ントと からなる群から選ばれる請求項1に記載の方法。 5.該抗体フラグメントがF(ab’)2、F(ab)2、Fab’、Fab、 Fv、sFv、および最少認識単位からなる群から選ばれる請求項4に記載の方 法。 6.上記方法が、 (c)該第二のワクチンの投与前および投与中にインターフェロンγを投与す るステップ をさらに含む請求項1に記載の方法。 7.上記方法が、 (c)該第二のワクチンの投与前および投与中にインターロイキン2を投与す るステップ をさらに含む請求項1に記載の方法。 8.上記方法が、 (c)該第二のワクチンの投与前および投与中にインターロイキン2およびイ ンターフェロンγを同時投与するステップ をさらに含む請求項1に記載の方法。 9.(a)TAAまたは感染性生物に関連する抗原と結合する抗体成分を含み 、該抗体成分が可溶の免疫原性担体タンパク質と複合しているワクチンを哺乳動 物に投与するステップと、 (b)可溶の免疫原性担体タンパク質とは複合していないが、TAAまたは感 染性生物に関連する抗原と結合する抗体またはその抗原結合性フラグメントを投 与するステップと を含む、TAAを発現する腫瘍または感染性生物に起因する疾患に対する哺乳動 物の体液性免疫応答および細胞性免疫応答を誘導する方法。 10.上記ステップ(b)の抗体または抗体フラグメントがビオチンと複合し 、上記方法が、 (c)アビジンを投与して該ビオチン化抗体または該ビオチン化抗体フラグメ ントの循環濃度を低下させるステップ をさらに含む請求項9に記載の方法。 11.上記方法が、 (d)上記ステップ(a)のワクチンの第二回投与を行うステップ をさらに含む請求項10に記載の方法。 12.上記方法が、 (e)該第二回のワクチン投与前および投与中にインターフェロンγを投与す るステップ をさらに含む請求項11に記載の方法。 13.上記方法が、 (e)該第二回のワクチン投与前および投与中にインターロイキン2を投与す るステップ をさらに含む請求項11に記載の方法。 14.上記方法が、 (e)該第二回のワクチン投与前および投与中にインターロイキン2およびイ ンターフェロンγを同時投与するステップ をさらに含む請求項11に記載の方法。 15.(a)患者からT細胞を得るステップと、 (b)キメラ免疫グロブリン/T細胞レセプターまたはキメラ免疫グロブリン /CD3タンパク質のいずれかをコードするDNA分子を含み、該DNA分子の 該免疫グロブリンをコードしている部分がTAAまたは感染性生物に関連する抗 原と結合する抗体の可変部をコードしている発現ベクターをT細胞に導入し、ト ランスフェクトしたT細胞を得るステップと、 (c)該トランスフェクトしたT細胞の増殖を刺激して、トランスフェクトし たT細胞数を増加させるステップと、 (d)数の増加した該トランスフェクトしたT細胞集団を患者に戻すステップ と を含む、TAAを発現する腫瘍または感染性生物に起因する疾患に対する患者の 体液性免疫応答および細胞性免疫応答を誘導する方法。 16.上記方法が、 (e)該トランスフェクトしたT細胞を該患者に戻した後、インターフェロン γおよびインターロイキン2からなる群から選ばれるサイトカインのうち少なく とも一種を投与するステップと、 (f)該キメラ免疫グロブリン/T細胞レセプターまたは該キメラ免疫グロブ リン/CD3タンパク質の免疫グロブリン部分と結合し、可溶の免疫原性担体タ ンパク質と複合している抗イディオタイプ抗体成分を含むワクチンを該患者に投 与するステップと をさらに含む請求項15に記載の方法。 17.上記方法が (e)該キメラ免疫グロブリン/T細胞レセプターまたは該キメラ免疫グロブ リン/CD3タンパク質の免疫グロブリン部分と結合し、可溶の免疫原性担体タ ンパク質と複合している抗イディオタイプ抗体成分を含むワクチンを該患者に投 与するステップ をさらに含む請求項15に記載の方法。 18.(a)患者からT細胞を得るステップと、 (b)キメラ免疫グロブリン/T細胞レセプターまたはキメラ免疫グロブリン /CD3タンパク質のいずれかをコードするDNA分子を含み、該DNA分子の 該免疫グロブリンをコードしている部分がTAAのエピープまたは感染性生物に 関連する抗原のエピトープによく似る抗体の可変部をコードしている発現ベクタ ーをT細胞に導入してトランスフェクトしたT細胞を得るステップと、 (c)該トランスフェクトしたT細胞の増殖を刺激して、トランスフェクトし たT細胞数を増加させるステップと、 (d)数の増加した該トランスフェクトしたT細胞集団を患者に戻すステップ と を含む、TAAを発現する腫瘍または感染性生物に起因する疾患に対する患者の 体液性免疫応答および細胞性免疫応答を誘導する方法。 19.上記方法が、 (e)該トランスフェクトしたT細胞を該患者に戻した後、インターフェロン γおよびインターロイキン2からなる群から選ばれるサイトカインのうち少なく とも一種を投与するステップと、 (f)該キメラ免疫グロブリン/T細胞レセプターまたは該キメラ免疫グロブ リン/CD3タンパク質の免疫グロブリン部分と結合し、可溶の免疫原性担体タ ンパク質と複合している抗体成分を含むワクチンを該患者に投与するステップと をさらに含む請求項18に記載の方法。 20.上記方法が、 (e)該キメラ免疫グロブリン/T細胞レセプターまたは該キメラ免疫グロブ リン/CD3タンパク質の免疫グロブリン部分と結合し、可溶の免疫原性担体タ ンパク質と複合している抗体成分を含むワクチンを該患者に投与するステップ をさらに含む請求項18に記載の方法。 21.製薬的に許容できる担体、および可溶の免疫原性担体タンパク質と複合 している治療有効量の抗癌胎児性抗原(CEA)抗体成分を含む、癌胎児性抗原 (CEA)を発現する腫瘍を持つ患者の治療用ワクチン。 22.該抗CEA抗体成分が、 (a)マウス・モノクローナル・クラスIII抗CEA抗体と、 (b)マウス・モノクローナル・クラスIII抗CEA抗体に由来するヒト化抗 体と、 (c)ヒト・モノクローナル抗CEA抗体と、 (d)(a)、(b)または(c)に由来する抗体フラグメントと からなる群から選ばれる請求項21に記載のワクチン。 23.該抗体フラグメントがF(ab’)2、F(ab)2、Fab’、Fab 、Fv、sFv、および最少認識単位からなる群から選ばれる請求項22に記載 のワクチン。 24.製薬的に許容できる担体、および可溶の免疫原性担体タンパク質と複合 していて、CEAのエピトープとよく似ている、治療有効量の抗イディオタイプ 抗体成分を含む、CEAを発現する腫瘍を持つ患者の治療用ワクチン。 25.該抗イディオタイプ抗体成分が、 (a)クラスIII抗CEA抗体の可変部と結合するポリクローナル抗体と、 (b)クラスIII抗CEA抗体の可変部と結合するモノクローナル抗体と、 (c)(b)に由来するヒト化抗体と、 (d)クラスIII抗CEA抗体の可変部と結合する類人霊長類抗体と、 (e)クラスIII抗CEA抗体の可変部と結合するヒト・モノクローナル抗C EA抗体と、 (f)(a)、(b)、(c)、(d)または(e)に由来する抗体フラグメ ントと からなる群から選ばれる請求項24に記載のワクチン。 26.該抗体フラグメントがF(ab’)2、F(ab)2、Fab’、Fab 、Fv、sFv、および最少認識単位からなる群から選ばれる請求項25に記載 のワクチン。 27.(a)TAAまたは感染性生物に関連する抗原と結合する抗体成分を含 み、該抗体成分が可溶の免疫原性担体タンパク質と複合している第一のワクチン を哺乳動物に投与するステップと、 (b)可溶の免疫原性担体タンパク質と複合していないが、TAAまたは感染 性生物に関連する抗原と結合する抗体またはその抗原結合性フラグメントを投与 するステップと、 (c)該TAAまたは該感染性生物抗原のエピトープによく似ている、可溶の 免疫原性担体タンパク質と複合している抗イディオタイプ抗体成分を含む第二の ワクチンを該哺乳動物に投与するステップと を含む、TAAを発現する腫瘍または感染性生物に起因する疾患に対する哺乳動 物の体液性免疫応答および細胞性免疫応答を誘導する方法。 28.該第一のワクチンがクラスIII抗CEA抗体を含み、ステップ(b)の 該抗体がクラスIII抗CEA抗体であり、該第二のワクチンがクラスIII抗CEA 抗体の可変部と結合する抗体を含む請求項27に記載の方法。 29.結合する部分を有する二重特異性抗体を患者に投与するステップを含む 、CEAを発現する腫瘍を持つ患者の治療方法。
