Claims (1)
- 【特許請求の範囲】 1.(a)少なくとも1種のカチオン性脂質を抗原決定基をコードするポリヌ クレオチドと混合してカチオン性脂質−ポリヌクレオチド複合体を形成させ、つ いで (b)該脂質−ポリヌクレオチド複合体を動物に投与する ことを特徴とする、動物において免疫応答を引き起こさせる方法。 2.該カチオン性脂質がリポフェクタミンTMである請求項1に記載の方法。 3.該カチオン性脂質がリポフェクテースTMである請求項1に記載の方法。 4.該カチオン性脂質が1−プロパンアンモニウム,N−[2(2−ブロモ) エチル]−N,N−ジメチル−2,3−ビス(9−オクタデセニルオキシ)ブロマ イドである請求項1に記載の方法。 5.該カチオン性脂質がDORI−エーテルである請求項1に記載の方法。 6.該カチオン性脂質がDORI−エーテルリゾ脂質である請求項1に記載の 方法。 7.該ポリヌクレオチドがRNA分子であり、該RNA分子が免疫原をコード する請求項1に記載の方法。 8.該ポリヌクレオチドが、 (a)プロモーター、および (b)免疫原をコードするDNA配列、その際、該DNA配列の転写は該プロモ ーターの制御下にある、 を含む組換えDNA分子である請求項1に記載の方法。 9.該ポリヌクレオチドが、 (a)プロモーター、および (b)免疫原をコードするRNA配列、その際、該RNA配列の発現は該プロモ ーターの制御下にある、 を含む組換えRNA分子である請求項1に記載の方法。 10.該組換えDNA分子が発現ベクターである請求項8に記載の方法。 11.該プロモーターがSV40プロモーターである請求項10に記載の方法 。 12.該プロモーターがRSVプロモーターである請求項10に記載の方法。 13.該プロモーターがCMVプロモーターである請求項10に記載の方法。 14.(a)少なくとも1種のカチオン性脂質を免疫原をコードするポリヌク レオチドと混合してカチオン性脂質−ポリヌクレオチド複合体を形成させ、 (b)該脂質−ポリヌクレオチド複合体を動物に投与し、ついで (c)該動物からポリクローナル抗体を単離する ことを特徴とする、動物において免疫原に対するポリクローナル抗体を製造する 方法。 15.(a)少なくとも1種のカチオン性脂質を、免疫原をコードするDNA 配列を含むポリヌクレオチドと混合して脂質−ポリヌクレオチド複合体を形成さ せ、 (b)該脂質−ポリヌクレオチド複合体を少なくとも1のマウスに投与し、 (c)該免疫したマウスからBリンパ球を取り出し、 (d)該免疫したマウスからのBリンパ球をミエローマ細胞と融合させてハイブ リドーマを生成させ、 (e)該ハイブリドーマをクローニングし、 (f)抗免疫原抗体を産生する陽性クローンを選択し、 (g)該抗免疫原抗体を産生するクローンを培養し、ついで (h)抗免疫原抗体を培養液から単離する ことを特徴とする、モノクローナル抗体の製造方法。 16.タンパク質分子のエピトープをマッピングする方法であって、 (a)該タンパク質分子をコードするDNA分子をランダムに断片化し、 (b)これらDNA断片を発現ベクター中にサブクローニングし、 (c)少なくとも一つのカチオン性脂質を各発現ベクターサブクローンと混合し て各発現ベクターサブクローンとのカチオン性脂質−発現ベクター複合体を生成 させ、 (d)該カチオン性脂質−発現ベクター複合体をマウスに投与し、ついで (e)どのDNA断片がマウスにおいて抗体を産生させることができるかを決定 する ことを特徴とする方法。 17.(a)少なくとも1種のカチオン性脂質を、感染性疾患の病原体である 微生物の抗原決定基をコードするポリヌクレオチドと混合してカチオン性脂質− ポリヌクレオチド複合体を形成させ、ついで (b)該脂質−ポリヌクレオチド複合体を動物に投与して該感染性疾患に対する 能動免疫を生じさせる ことを特徴とする、動物における感染性疾患に対する能動免疫を生じさせる方法 。
