- 【特許請求の範囲】 (1)配列表中SEQ ID NO.1〜56及び137に規定された核酸配列を含むかまたは配 列表中SEQ ID NO.57〜136に規定された核酸配列を含む、但しおのおのの核酸はD NA-またはRNA-型構造を有する、トランスフォーミング成長因子−β(TGF-β) をコードする遺伝子領域とハイブリダイズするアンチセンス−オリゴヌクレオチ ド類またはそれの有効な誘導体。 (2)前記核酸がトランスフォーミング成長因子−β1、−β2及び/または−β3 をコードする遺伝子領域とハイブリダイズする請求項1記載のアンチセンス−オ リゴヌクレオチド類。 (3)前記核酸がTGF-βをコードする遺伝子部分の複数の領域及び/またはTGF- βをコードするおよびコードしない遺伝子部分の複数の領域とハイブリダイズす る請求項1及び/または2記載のアンチセンス−オリゴヌクレオチド。 (4)アンチセンス−オリゴヌクレオチドがフォスフォロチオエート オリゴデ オキシヌクレオチドである請求項1〜3のいずれか1項に記載のアンチセンス− オリゴヌクレオチド。 (5)ヌクレオチド鎖を3'-5'方向に成長させることによる亜リン酸トリエステル 化学を用いる固相合成法、但し、前記合成法中、各ヌクレオチドが固相に共有結 合している第1のヌクレオチドと結合し、前記合成法は −原料ヌクレオチドの5'DMT保護基を切断し、 −鎖増殖用原料ヌクレオチドを添加し、 −亜リン酸基を修飾し、次いで身反応5'−水酸基を封鎖し、かつ −固相支持体から切断し、 −次いで合成生成物を得る の工程を含む、により得られる請求項1〜4のいずれか1項に記載のアンチセン ス−オリゴヌクレオチド。 (6)SEQ ID NO.1〜56及び137のオリゴヌクレオチド類がTGF-β1のアンチセン ス−オリゴデオキシヌクレオチド類であり、SEQ ID NO.57〜136のオリゴヌクレ オチドがTGF-β2フォスフォロチオエート−アンチセンス−オリゴデオキシヌク レオチドである請求項1〜5のいずれか1項に記載のアンチセ ンス−オリゴヌクレオチド。 (7)請求項1〜6のいずれか1項に記載のアンチセンス−オリゴヌクレオチド とGCの量が同一であるナンセンス−コントロール−ヌクレオチドであるオリゴ ヌクレオチド。 (8)請求項1〜6のいずれか1項に記載のアンチセンス−核酸を含有する医薬 組成物。 (9)TGF-βの発現が病原性及び/または病気に起因するアンギオゲネシス(ang iogenesis)の抑制に関連する腫瘍の治療用の請求項8に記載の医薬組成物の製 造のための、請求項1〜6のいずれか1項に記載のアンチセンス−オリゴヌクレ オチド類の使用。 (10)TGF-βの免疫抑制効果の治療用、細胞障害性のリンパ球の増殖の増大、TG F-βの内因性発現過多の治療用、胸部腫の治療用、神経線維腫の治療用、膠芽腫 を含む悪性神経膠腫の治療用、及び、食道癌及び胃癌の治療と同様に皮膚癌の治 療及び予防用の請求項8に記載の医薬組成物の製造のための請求項9記載の使用 。 (11)請求項1〜6のいずれか1項に記載のアンチセンス−TGF-β−オリゴヌク レオチド類の有効投与量の投与により、TGF-βを抑制し、かつ、それによる免疫 抑制および/または病気に起因するアンギオゲネスの抑制の減少により、TGF-β の発現が病原性に関係する腫瘍の治療方法。 (12)免疫抑制の減少が、アンチセンス−TGF-β−オリゴヌクレオチド類の投与 前の状態に比較して細胞障害性のリンパ球の増殖の増大を伴い、その結果、始ま った細胞障害性の活動が腫瘍細胞の数を減らす請求項11に記載の治療方法。
