Claims (1)
- 【特許請求の範囲】 1.試料中の被分析核酸を定量する方法であって、 −各々が被分析核酸から区別し得るが被分析核酸と同時に増幅され得る種々の 核酸構築物をそれぞれ異なる量で試料に添加するステップ; −被分析核酸及び核酸構築物の両方と反応し得る増幅試薬を使用し、試料に核 酸増幅処理を実施するステップ; −被分析核酸及び各核酸構築物から誘導された増幅物の相対量を検出するステ ップ;及び −前記相対量から被分析核酸の量を計算するステップを含む方法。 2.前記核酸構築物が、各核酸構築物に固有の区別可能な配列を除き、前記被分 析核酸と同じ配列を有することを特徴とする請求項1に記載の方法。 3.前記固有の区別可能な配列が、約20ヌクレオチドを含むランダムな配列で あることを特徴とする請求項2に記載の方法。 4.前記核酸構築物を、相互に一定係数ずつ異なる一連の量で添加することを特 徴とする請求項1から3のいずれか一項に記載の方法。 5.前記核酸構築物を、相互に係数10ずつ異なる一連の量で添加することを特 徴とする請求項1から4のいずれか一項に記載の方法。 6.核酸単離処理を実施する前に前記核酸構築物を添加した既知量の試験液に核 酸単離処理を実施することにより試料を調製することを特徴とする請求項1に記 載の方法。 7.前記試験液に核酸単離処理を実施することにより調製した試料に既知量の単 離対照配列を添加することを特徴とする請求項6に記載の方法。 8.試料を1つ以上の反応アリコートに分割し、それぞれに前記核酸構築物を異 なる量範囲で添加することを特徴とする請求項1から7のいずれか一項に記載の 方法。 9.前記各反応アリコートに、同じ核酸構築物を添加することを特徴とする請求 項8に記載の方法。 10.前記量範囲が重複しないことを特徴とする請求項8または9に記載の方法 。 11.1つ以上の試料において被分析核酸を定量する方法であって、各試料に請 求項1に記載の方法を実施するが、ここで全ての試料に同じ核酸構築物を使用し 、それぞれの特定の核酸構築物の量を変える方法。 12.各試料において1種の核酸構築物を量ゼロで使用することを特徴とする請 求項11に記載の方法。 13.一群の試験液において被分析核酸を定量する方法であって、 −ある量の前記試験液を既知の係数ずつ希釈し、所定範囲内にあると推定され る量の被分析核酸を含む前記希釈試験液から試料を得るステップ;及び −前記試料に、試料に添加する核酸構築物の量が前記範囲内である請求項1に 記載の方法を実施するステップを含む方法。 14.更に −前記量範囲より少ない量の被分析核酸を含む試料を同定するステップ; −前記試料を調製した試験液に、試験液がより低い係数で希釈される請求項1 3に記載の方法を実施するステップを含む請求項13に記載の方法。 15.更に、 −前記量範囲より多い量の被分析核酸を含む試料を同定するステップ; −前記試料を調製した試験液に、試験液がより高い係数 で希釈される請求項13に記載の方法を実施するステップを含む請求項13に記 載の方法。 16.試料中の被分析核酸を定量する方法であって、 −前記試料中に存在する被分析核酸の推定量と同じ範囲内にあると推定される 量の核酸構築物を試料に添加するステップ; −前記被分析核酸及び核酸構築物の両方と反応し得る増幅試薬を使用し、前記 試料に核酸増幅処理を実施するステップ; −被分析核酸及び核酸構築物から誘導された増幅物の相対量を検出するステッ プ; −前記相対量から前記試料中に存在する被分析核酸の量を推算するステップ; 及び −前記試験液から得た第2試料に、添加する核酸構築物の量が第1試料中に存 在した被分析核酸の前記推算量と同じ範囲内にある請求項1に記載の方法を実施 するステップを含む方法。 17.実施例1に記載の方法に従って試料中の核酸を定量するためのキットであ って、種々の核酸構築物、前記核酸構築物及び被分析核酸と反応し得る増幅試薬 、増幅された 被分析核酸用の検出プローブ、及び増幅された種々の核酸構築物にそれぞれ特異 的な異なる検出プローブを含むキット。
