Claims (1)
- 【特許請求の範囲】 1.治療目的の産物を発現できる一次あるいは二次細胞であって、 a)治療目的の産物をコードする、あるいはそれ自体が治療目的の産物である外 因性DNA、およびb)トランスフェクト一次あるいは二次細胞において、該外 因性DNAを発現するのに充分な、非レトロウイルス由来のDNA配列が そのゲノムへ安定して組み込まれた、脊椎動物(例えば哺乳類)由来のトランス フェクトされた一次あるいは二次細胞。 2.その中に組み込まれた治療目的の産物をコードする外因性DNA、該外因性 DNAはトランスフェクト一次あるいは二次細胞において、その外因性DNAを 発現するのに充分なDNA配列に効果的に結合しているもの、を安定して発現す ることができるトランスフェクトされた脊椎動物(例えば哺乳類)由来の一次あ るいは二次細胞。 3. a) 1)取得時の一次あるいは二次細胞において、生産あるいは含有されていない治 療目的の産物をコードする外因性DNA、 2)取得時の一次あるいは二次細胞において、異常に少量あるいは不完全な形で 生産あるいは含有される治療目的の産物をコードする外因性DNA、および3) 取得時の一次あるいは二次細胞において、生理学的に通常の量を生産あるいは含 有する治療目的の産物をコードする外因性DNA、 からなる群より選択された治療目的の産物をコードする外因性DNA、および、 b)(a)の外因性DNAを発現するのに充分な非レトロウイルス由来のDNA 配列、ここで(a)の該外因性DNAは(b)のDNA配列に効果的に結合して いる、がそのゲノムへ安定して組み込まれた、脊椎動物(例えば哺乳類)由来の 一次あるいは二次細胞。 4.細胞内のエピソーム中に存在する(a)および(b)の配列 a)治療目的の産物をコードする、若しくはそれ自体が治療目的の産物である外 因性DNA、及びb)一次あるいは二次細胞において該外因性DNAが発現する のに充分な非レトロウイルス由来のDNA配列でトランスフェクトされた脊椎動 物(例えば哺乳類)由来の一次あるいは二次細胞。 5.(a)線維芽細胞、角化細胞、上皮細胞、内皮細胞、ダリア細胞、神経細胞 、血液細胞成分、筋細胞、肝細胞およびそれらの前駆体からなる群、あるいは( b)ヒト一次細胞、ヒト二次細胞、マウス一次細胞、マウス二次細胞、ウサギ一 次細胞、及びウサギ二次細胞からなる群、から選択された前記の請求項のうちい ずれかの一次あるいは二次細胞。 6.(a)該外因性DNAが酵素、サイトカイン、ホルモン、抗原、抗体、凝固 因子、調節タンパク質、リボザイム、転写タンパク質、レセプター、アンチセン ス核酸配列およびタンパク質からなる群から選択された治療目的の産物をコード する又は(b)該外因性DNAがトランスフェクト一次若しくは二次細胞中のタ ンパク質若しくは核酸のキレート化合物形成に充分なDNA配列、細胞性調節タ ンパク質と結合するDNA配列、染色体の二次若しくは三次構造を変化させるD NA配列、および転写を調節する成分であるDNA配列からなる群から選択され た、それ自体が治療目的の産物である前記の請求項のうちいずれかの一次あるい は二次細胞。 7.選択しうるマーカーをコードするDNAを付加的に含む前記の請求項のうち いずれかの一次あるいは二次細胞。 8.その中に組み込まれた治療目的の産物をコードする外因性DNAを発現しう る脊椎動物(例えば哺乳類)由来のトランスフェクト二次細胞のクローン細胞株 。 9.外因性DNAがトランスフェクト二次細胞のゲノムDNAに安定して組み込 まれている請求項8のクローン細胞株。 10.(a)該外因性DNAが、酵素、サイトカイン、ホルモン、抗原、抗体、 凝固因子、調節タンパク質、リボザイム、転写タンパク質、レセプター、アンチ センス核酸配列およびタンパク質からなる群から選択された治療目的の産物をコ ードする又は(b)該外因性DNAが、トランスフェクト一次若しくは二次細胞 中のタンパク質若しくは核酸のキレート化合物形成に充分なDNA配列、細胞性 調節タンパク質と結合するDNA配列、染色体の二次若しくは三次構造を変化さ せるDNA配列、および転写を調節する成分であるDNA配列からなる群から選 択された、それ自体が治療目的の産物である、請求項8若しくは請求項9のクロ ーン細胞株。 11.トランスフェクト二次細胞が、(a)トランスフェクト二次線維芽細胞、 トランスフェクト二次角化細胞、トランスフェクト上皮細胞、トランスフェクト 内皮細胞、トランスフェクトグリア細胞、トランスフェクト神経細胞、トランス フェクト血液細胞成分、トランスフェクト筋細胞、トランスフェクト肝細胞、お よびそれらのトランスフェクト前駆体からなる群、あるいは(b)トランスフェ クトヒト二次細胞、トランスフェクトマウス二次細胞、およびトランスフェクト ウサギ二次細胞からなる群から選択されたものである、請求項8若しくは請求項 9のクローン細胞株。 12.外因性DNAが、トランスフェクト二次細胞中のエピソームに存在する請 求項8、10若しくは11いずれかのクローン細胞株。 13.トランスフェクト二次細胞が選択しうるマーカーをコードするDNAを付 加的に含む請求項8ないし12いずれかのクローン細胞株。 14.治療目的の産物を発現できる不均一細胞株であって、a)治療目的の産物 をコードする外因性DNA及びb)トランスフェクト一次あるいは二次細胞にお いて該外因性DNAが発現するのに充分な非レトロウイルス由来のDNA配列 がそれらのゲノムへ安定して組み込まれた、脊椎動物(例えば哺乳類)由来のト ランスフェクト一次あるいは二次細胞の不均一細胞株。 15.(a)トランスフェクト線維芽細胞、トランスフェクト角化細胞、トラン スフェクト上皮細胞、トランスフェクト内皮細胞、トランスフェクトグリア細胞 、トランスフェクト神経細胞、トランスフェクト血液細胞成分、トランスフェク ト筋細胞、トランスフェクト肝細胞およびそれらのトランスフェクト前駆体から なる群、あるいは(b)トランスフェクトヒト一次細胞、トランスフェクトヒト 二次細胞、トランスフェクトマウス一次細胞、トランスフェクトマウス二次細胞 、トランスフェクトウサギ一次細胞、およびトランスフェクトウサギ二次細胞か らなる群、より選択されたトランスフェクト一次又は二次細胞である請求項14 の不均一細胞株。 16.(a)DNAが酵素、サイトカイン、ホルモン、抗原、抗体、凝固因子、 調節タンパク質、リボザイム、転写タンパク質、レセプター、アンチセンス核酸 配列および新規タンパク質からなる群から選択された治療目的の産物をコードす る又は(b)外因性DNAがトランスフェクトされた一次若しくは二次細胞中の タンパク質若しくは核酸のキレート化合物形成に充分なDNA配列、細胞性調節 タンパク質と結合するDNA配列、染色体の二次若しくは三次構造を変化させる DNA配列、並びに転写を調節する成分であるDNA配列からなる群から選択さ れた、それ自体が治療目的の産物である、請求項14又は請求項15の不均一細 胞株。 17.選択しうるマーカーをコードするDNAを付加的に含む請求項14、15 並びに16のうちいずれかの不均一細胞株。 18.治療目的の産物をコードする外因性DNA、該外因性DNAは細胞中のエ ピソームに存在するもの、を発現することのできるトランスフェクトされた一次 若しくは二次細胞の不均一細胞株。 19.ヒト成長ホルモン、エリスロポエチン、又はグルカゴン様ペプチド1類縁 ペプチドをコードする外因性DNAである請求項1若しくは請求項4の一次又は 二次細胞。 20.治療目的の産物がエリスロポエチン、又はグルカゴン様ペプチド1類縁ペ プチドである請求項8ないし18のうちいずれかの細胞株。 21.治療目的の産物がヒト成長ホルモンである請求項8ないし18のうちいず れかの細胞株。 22.グルカゴン様ペプチド1類縁ペプチドが、GLP−1(7−37)、GL P−1(7−36)、GLP−1(7−35)、GLP−1(7−34)、及び 切断(truncate)されたカルボキシル末端がアミド化されたGLP−1 の誘導体、及びアミノ酸置換、欠失、付加若しくは他の改変(例えば、非アミノ 酸成分の付加)を有し、切断(truncate)されたGLP−1の誘導体と 実質的に同等又は高い生物学的な活性若しくは血中安定性をもたらすGLP−1 の誘導体からなる群より選択されたグルカゴン様ペプチド1誘導体である、請求 項19若しくは請求項20による細胞又は細胞株。 23.請求項1ないし請求項7若しくは請求項19若しくは請求項22の、トラ ンスフェクトされた一次若しくは二次細胞並ひに、トランスフェクトされていな い一次若しくは二次細胞から本質的に構成される細胞混合物。 24. a)一次細胞を含む脊椎動物(例えば哺乳類)由来の細胞混合物を生産し、 b)治療目的の産物並びに一次細胞において外因性DNAが発現するのに充分な 付加的DNA配列をコードする外因性DNAを含むDNA構築物で、(a)にお いて生産される一次細胞をトランスフェクトし、それによって治療目的の産物を コードする外因性DNAの発現が可能なトランスフェクトされた一次細胞を生産 80.相同組換えに適した条件下で、調節領域を含むDNA構築物を細胞に導入 することにより、その調節領域が挿入されるかまたは発現させようとする遺伝子 の調節領域の全てまたは一部と置換して、発現させようとする遺伝子と機能的に 結合され、これにより遺伝子の発現が可能な相同的に組換えられた細胞が生産さ れる、細胞(例えば、哺乳類由来)に存在するが、採取したままの細胞内では発 現されないか、採取したままの細胞内では有意な量は発現されない、発現させよ うとする遺伝子の発現を開始する方法。 81.該遺伝子が酵素、サイトカイン、ホルモン、抗原、抗体、凝固因子、調節 タンパク質、レセプターおよびタンパク質からなる群から選択されたものである 請求項80の方法。 82.該遺伝子がヒトのエリスロポエチン、成長ホルモン及びインスリン遺伝子 からなる群から選択されたものである請求項81の方法。 83. a) 1)脊椎動物由来の一次若しくは二次細胞で発現される産物をコードする外因性 DNA、 2)脊椎動物由来の一次若しくは二次細胞におけるゲノムDNA配列と相同した DNA配列、及び3)少なくともひとつの選択しうるマーカーをコードするDN A配列、 からなるDNA構築物を供給し、 b)(a)において供給されたDNA構築物を用いて一次若しくは二次細胞をト ランスフェクトし、これによって(a)において供給されたDNA構築物を含む 一次若しくは二次細胞を生産し、並びに c)ゲノムDNA配列と相同したDNA配列とゲノムDNA配列間で起こる相同 組換えに適した条件下で、(b)において生産された一次若しくは二次細胞を維 持し、これによって一次若しくは二次細胞のゲノムDNAへ組み込まれた(a) のDNA構築物を有する、脊椎動物由来の相同組換えされた一次若しくは二次細 胞を生産し、 d)(c)において生産された相同組換えされた一次若しくは二次細胞を、ター ゲティングDNA構築物中に存在する選択しうるマーカーを選択する選択薬剤に さらし、それによって、選択しうるマーカーが安定して組み込まれていない細胞 は殺され、選択しうるマーカーが安定して組み込まれている細胞は生存可能でコ ロニーを形成でき、並びに e)(d)で生産されたコロニーをスクリーニングして、相同組換えされた一次 若しくは二次細胞株を同定する工程からなる、脊椎動物(例えば哺乳類)由来の 一次若しくは二次細胞のゲノムDNAへDNA配列をターゲティングする方法。 84.ポジティブ選択マーカーがneoであって、選択薬剤がG418である請 求項83の方法。 85. a) 1)脊椎動物由来の一次若しくは二次細胞で発現される産物をコードする外因性 DNA、 2)脊椎動物由来の一次若しくは二次細胞におけるゲノムDNA配列と相同した DNA配列、及び3)宿主細胞ゲノムにおいてターゲティング配列と相同配列の 間の相同組換えによりポジティブ選択マーカーのターゲットされた組み込みが起 こるが、ネガティブ選択マーカーは組み込まれないようなコンフィグレーション で、少なくともポジティブおよびネガティブの選択マーカをそれぞれひとつコー ドするDNA配列からなるDNA構築物 を供給し、 b)(a)において供給されたDNA構築物を用いて一次若しくは二次細胞をト ランスフェクトし、これによって(a)において供給されたDNA構築物を含む 一次若しくは二次細胞を生産し、並びに c)ゲノムDNA配列と相同したDNA配列とゲノムDNA配列間で起こる相同 組換えに適した条件下で、(b)において生産された一次若しくは二次細胞を維 持し、これによって一次若しくは二次細胞のゲノムDNAへ組み込まれた(a) のDNA構築物を有する、脊椎動物由来の相同組換えされた一次若しくは二次細 胞を生産し、 d)(c)において生産された一次若しくは二次細胞を、ターゲティングDNA 構築物中に存在するポジティブ選択マーカーを選択する選択薬剤にさらし、それ によって、ポジティブ選択マーカーが安定して組み込まれていない細胞は殺され 、ポジティブ選択マーカーが安定して組み込まれている細胞は生存可能でコロニ ーを形成でき、並びに e)さらに、(c)および(d)において生産された細胞を、ターゲティングD NA構築物中に存在するネガティブ選択マーカーに対抗して選択する選択薬剤に さらし、それによってネガティブ選択マーカーが安定して組み込まれている細胞 は殺され、(d)と(e)により相同的組換え細胞が選択される結果となるよう な工程からなる、脊椎動物(例えば哺乳類)由来の一次若しくは二次細胞のゲノ ムDNAへDNA配列をターゲティングする方法。 86.細胞が(a)線維芽細胞、角化細胞、上皮細胞、内皮細胞、グリア細胞、 神経細胞、血液細胞成分、筋細胞、肝細胞およびそれらの前駆体からなる群、あ るいは(b)ヒト一次細胞、ヒト二次細胞、マウス一次細胞、マウス二次細胞、 ウサギ一次細胞、及びウサギ二次細胞からなる群、から選択された脊椎動物(例 えば哺乳類)由来の一次若しくは二次細胞である、請求項78〜85いずれかの 方法。 87.発現させようとする遺伝子または外因性DNAが、(a)酵素、サイトカ イン、ホルモン、抗原、抗体、凝固因子、調節タンパク質、リボザイム、転写タ ンパク、受容体、アンチセンス核酸配列および新規タンパク質からなる群から選 ばれた治療目的の産物をコードし、あるいは(b)それ自体が、細胞においてタ ンパク質若しくは核酸のキレート形成(sequestration)に十分な DNA配列、細胞性調節タンパク質と結合するDNA配列、染色体の二次若しく は三次構造を変化させるDNA配列、および転写を調節する成分であるDNA配 列からなる群から選択された治療目的の産物である、請求項78〜86いずれか の方法。 88.請求項78〜82、85、86並びに87いずれかの方法によって生産さ れた、相同組換えされた一次若しくは二次細胞。 89.請求項80、81、82、85、86並びに87いずれかの方法によって 生産された、相同組換えされた二次細胞のクローン細胞株。 90.取得時の該細胞が不死化細胞である請求項80の方法91.二つのネガテ ィブ選択マーカーが用いられ、一つのネガティブ選択マーカーがgptであり、 一つのネガティブ選択マーカーがHSV−TK遺伝子である請求項85、86並 びに87の方法。 92.DNA構築物が少なくとも一つのポジティブ選択マーカー及び少なくとも 一つのネガティブ選択マーカーを付加的に含む請求項78〜82いずれかの方法 。 93.ポジティブ選択マーカーがneoであって、ポジティブ選択薬剤がG41 8である請求項78〜82、85、86並びに87いずれかの方法。 94.(a)ネガティブ選択マーカーがgptであり、ネガティブ選択薬剤が6 −チオキサンチン(6−thioxanthine)である、若しくは(b)ネ ガティブ選択マーカーがHSV−TK遺伝子であり、ネガティブ選択薬剤がガン シクロピル(gancyclovir)である、請求項78〜82、85、86 、89並びに93いずれかの方法。 95.請求項68、若しくは請求項72〜77、若しくは請求項88及び89い ずれか記載の細胞又は細胞株が培養され、充分な数の該トランスフェクトされた 細胞が供給される、有効量の治療目的の産物を哺乳類へ送達することのできるト ランスフェクトされた細胞の供給方法。 96.遺伝子治療に用いる治療目的の産物の製造のための、請求項68、若しく は請求項72〜77、若しくは請求項88及び89いずれか記載の細胞又は細胞 株の使用。 97.治療、例えば遺伝子治療、に用いるための、請求項68、若しくは請求項
