- 【特許請求の範囲】 1.(a)以下のリン脂質を含有するリン脂質混合物:(i)約20から約95 モルパーセントのホスファチジルコリン;(ii)約2.5から約50モルパー セントのホスファチジルエタノールアミン;(iii)約2.5から約50モル パーセントのホスファチジルセリン;および(iv)約0から40モルパーセン トのホスファチジルグリセロールおよび(b)リン脂質混合物1mgにつき約0 .1μgから約3μgの組織因子を含有するリポソ−ム組成物を含有する組織因 子に基づくプロトロンビン時間試薬。 2.約0.5パーセントから約1.5パーセントのグリシンをさらに含有する第 1項記載のプロトロンビン時間試薬。 3.トレハロース、マルトース、ラクトース、グルコースおよびマニトールから なる群から選択される炭水化物低温保存剤をさらに含有する第1項記載のプロト ロンビン時間試薬。 4.炭水化物低温保存剤が約50mMから250mMのトレハロースである第3 項記載のプロトロンビン時間試薬。 5.リン脂質混合物が、約5から約15モルパーセントのホスファチジルエタノ ールアミン;約5から約20モルパーセントのホスファチジルセリン、約10か ら25モルパーセントのホスファチジルグリセロールおよびその残りのホスファ チジルコリンを含有する第1項記載のプロトロンビン時間試薬。 6.リン脂質混合物が、約8から約12モルパーセントのホスファチジルエタノ ールアミン、約3から約10モルパーセントのホスファチジルセリン、約14か ら約20モルパーセントのホスフアチジルグリセロールおよび約58から約75 モルパーセントのホスファチジルコリンを含有する第1項記載のプロトロンビン 時間試薬。 7.リン脂質混合物が、約12モルパーセントのホスファチジルエタノールアミ ン、約3から約10モルパーセントのホスファチジルセリン、約14から約20 モルパーセントのホスファチジルグリセロールおよび約58から約75モルパー セントのホスファチジルコリンを含有する第1項記載のプロトロンビン時間試薬 。 8.組織因子がリコンビナント組織因子である第7項記載のプロトロンビン時間 試薬。 9.約0.6から約1.2パーセント(w/v)のグリシンを含有する第4項記 載のプロトロンビン時間試薬。 10.以下のステップ:(a)リン脂質混合物、組織因子およびキャリアータン パクを洗剤で共溶解する; (b)洗剤を除く;そして (c)カルシウム塩を添加する を有するリン脂質小胞の脂質二重層と会合した活性組織因子を含有するプロトロ ンビン時間試薬の調製方法。 11.リン脂質混合物が約20から約95モルパーセントのホスファチジルコリ ン、約2.5から約50モルパーセントのホスファチジルエタノールアミン、約 2.5から約50モルパーセントのホスファチジルセリンおよび約0から約40 モルパーセントのホスファチジルグリセロールを含有する第10項記載の方法。 12.ステップ(a)において約0.5から約1.5モルパーセントのグリシン をさらに共溶解性とする、第10項記載の方法。 13.組織因子がリコンビナント組織因子である第12項記載の方法。 14.洗剤がチャプス(CHAPS)、オクチルベーターD−グルコピラノシド およびオクチルベーターD−チオグルコピラノシドからなる群から選択される第 11項記載の方法。 15.洗剤が透析、タンジェンシャル・フロー・ディアフィルトレーション(t angential flow diafiltration)およびクロマト グラフ法からなる群から選択される手法により除かれる第11項記載の方法。 16.(a)以下のリン脂質を含有するリン脂質混合物;(i)約20から約9 5モルパーセントのホスファチジルコリン;(ii)約2.5から約50モルパ ーセントのホスファチジルエタノールアミン;(iii)約2.5から約50モ ルパーセントのホスファチジルセリン:および(iv)約0から40モルパーセ ントのホスファチジルグリセロール;(b)リン脂質混合物1mgにつき約0. 1μgから約3μgの組織因子;および(c)洗剤 を含有するリン脂質ミセルを含有する組織因子に基づくプロトロンビン時間試薬 。 17.約0.5から1.5パーセントのグリシンをさらに含有する第16項記載 のプロトロンビン時間試薬。 18.トレハロース、マルトース、ラクトース、グルコースおよびマニトールか らなる群から選択される炭水化物低温保存剤をさらに含有する第16項または1 7項記載のプロトロンビン時間試薬。 19.炭水化物低温保存剤が約50mMから250mMのトレハロースである第 18項記載のプロトロンビン時間試薬。 20.リン脂質混合物が、約5から約15モルパーセントのホスファチジルエタ ノールアミン;約5から約20モルパーセントのホスファチジルセリン、約10 から25モルパーセントのホスファチジルグリセロールおよびその残りのホスフ ァチジルコリンを含有する第19項記載のプロトロンビン時間試薬。 21.リン脂質混合物が、約8から約12モルパーセントのホスファチジルエタ ノールアミン、約3から約10モルパーセントのホスファチジルセリン、約14 から約20モルパーセントのホスファチジルグリセロールおよび約58から約7 5モルパーセントのホスファチジルコリンを含有する第1項記載のプロトロンビ ン時間試薬。 22.リン脂質混合物が、約12モルパーセントのホスファチジルエタノールア ミン、約3から約10モルパーセントのホスファチジルセリン、約14から約2 0モルパーセントのホスファチジルグリセロールおよび約58から約75モルパ ーセントのホスファチジルコリンを含有する第16項または17項記載のプロト ロンビン時間試薬。 23.組織因子がリコンビナントの組織因子である第22項記載のプロトロンビ ン時間試薬。 24.約0.6から約1.2パーセント(w/v)のグリシンを含有する第19 項記載のプロトロンビン時間試薬。 25.洗剤がアルキルグルコピラノシドである第16項または17項記載のプロ トロンビン時間試薬。 26.以下のステップ:(a)リン脂質混合物、組織因子およびキャリアータン パクを洗剤で共溶解する; (b)洗剤を除く;そして (c)カルシウム塩を添加する を有するリン脂質ミセルと会合した活性組織因子を含有するプロトロンビン時間 試薬の調製方法。 27.洗剤が、オクチルベーターD−グルコピラノシドおよびオクチルベーター D−チオグルコピラノシドからなる群から選択される第26項記載の方法。 28.リン脂質混合物が約20から約95モルパーセントのホスファチジルコリ ン、約2.5から約50モルパーセントのホスファチジルエタノールアミン、約 2.5から約50モルパーセントのホスファチジルセリンおよび約0から約40 モルパーセントのホスファチジルグリセロールを含有する第27項記載の方法。 29.ステップ(a)において約0.5から約1.5モルパーセントのグリシン をさらに共溶解性とする、第28項記載の方法。 30.洗剤がアルキルグルコピラノシドを含有する第27項または第29項記載 の方法。 31.洗剤がオクチルベーターD−グルコピラノシドおよびオクチルベーターD −チオグルコピラノシドからなる群から選択される第30項記載の方法。
