Description

【発明の詳細な説明】 （産業上の利用分野） 本発明はニトログリセリンを有効成分として含有するス
プレー剤に関する。
（従来の技術） ニトログリセリン噴霧製剤としては例えば、ニトログリ
セリン、プロパンジオール、無水エタノール及びジシク
ロロジフロロメタン（以下フロンガスという）などの噴
霧剤からなる組成物をガス圧式噴霧容器に封入したもの
（特公昭４１−１０７１８）またニトログリセリンの溶
媒としてカプリツクカプロイツクグリセリン脂肪酸エス
テルを用いて、フロンガス等の噴霧剤とともにガス圧式
噴霧容器に封入したものなどが知られている。
（発明が解決しようとする問題点） 従来はニトログリセリン噴霧剤は、ニトログリセリンの
吸収性が悪い、口内粘膜に対する刺激性が強い、噴霧回
数が重なって、容器中に噴出剤の減量に伴いニトログリ
セリンの噴出量が低下するなどの欠点がある。これらの
欠点は医薬品製剤としては極めて好ましくはないが今だ
改良されていない。
（問題点を解決する為の手段） 本発明者らは上記欠点の改良されたニトログリセリンの
スプレー剤について種々検討の結果、ニトログリセリン
を水とエタノールの混液に溶解することによって、ニト
ログリセリンの口腔内粘膜吸収は良好であり、また、水
の存在により刺激性は低下し、更にpHを２．４〜６．７
より好ましくは３．０〜６．５、更に好ましくは３．５
〜６．０に調整することによって保存安定性が高まり、
なおかつ液圧式定量噴霧容器に薬液を封入する事によっ
て常時一定した噴霧量を与えるスプレー剤が得られる事
を見出し、本発明を完成した。
即ち、本発明はニトログリセリンを含み、pHが約２．４
〜約６．７であるエタノール水溶液を液圧式噴出容器に
封入したスプレー剤に関するものである。
本発明のエタノール水溶液におけるニトログリセリンを
エタノール水溶液全量に対して約０．０５〜約２w／v
％、好ましくは約０．３〜１．２w／v％、より好ましく
は約０．５〜約w／v％である。
エタノール含量は、エタノール水溶液全体に対して約１
０〜約８５v／v％、好ましくは約３０〜約７５v／v％、
より好ましくは約５０〜７０v／v％である。残部は通常
水であり、本発明のスプレー剤はフロンガス等の噴霧剤
を実質的に含有しない。
エタノール水溶液中でのニトログリセリンの安定性はpH
への依存性が高いので、適当な酸もしくはアルカリでpH
の調整を行えばよいが、通常、塩酸もしくは水酸化ナト
リウム等で行うのが好ましい。
液圧式スプレー容器に封入されるニトログリセリンを含
むエタノール水溶液は、場合により味覚などの改良のた
め、糖類、アミノ酸類及び香料などを含んでもよいが、
実質的にニトログリセリン、エタノール及び残部の水か
らなり、他の成分を含まない組成物の安定性は良いので
通常は他の成分を含む必要はない。
噴霧器による１回の噴霧量はエタノール水溶液中のニト
ログリセリン濃度により変るので一概には云えないが、
ニトログリセリン量が０．３〜０．４mg程度になるよう
にするのが好ましい。通常は、約２０mg〜７００mg、好
ましくは約３０mg〜約４００mg程度である。
次に液圧噴霧容器の材質はプラスチック、ガラス及びア
ルミニウムのいずれかでも良い。例えばプラスチックで
は、高密度ポリエチレン、ポリエチレンテレフタレー
ト、ポリ塩化ビニール及びポリプロピレン等が挙げられ
る。
（本発明の効果） 本発明のスプレー剤は、以下の実験例で示す様に、ニト
ログリセリンの保存安定性が良く、口内粘膜に対する刺
激性が少なく、吸収性が良い製剤である。
（実験例） 1) 実験試料 表１に示す処方の内容液（実施例１記載の方法もしくは
それに準じて調整した。）を容量１０ml、１回のスプレ
ー液量約４０mgのポリエチレン製液圧式定量噴霧容器に
封入した。但しNo.11は市販品（カプリツクカプロイツ
クグリセリン脂肪酸エステルを溶媒とし、フロンガスを
噴霧剤とし、ガス圧式噴霧器に封入したもの）。
2) 評価方法 実験試料を５０℃の恒温室に保存し、ニトログリセリン
の含有率の変化をしらべた。また、各試料を１０名のパ
ネラーが口腔内に１回ずつスプレーし、刺激性を知覚的
に試験した。更にビーグル犬の舌下に各試料を１回ずつ
スプレーし、ニトログリセリンの吸収性を血中濃度の測
定によりしらべた。
3) 実験結果 表１に結果を示す。本発明のスプレー剤は口腔内噴霧時
の刺激性が特に強くなく、悪味も感じることなく、ま
た、保存時のニトログリセリンの安定性が良く、舌下粘
膜からの吸収性が高く満足すべき条件を備えている。
また、本発明用品No.４と比較例No.１１（ジクロロジフ
ロロメタン封入、ガス圧式）を各々２００回連続噴出さ
せたところ、前者はコンスタントに約４０mg近辺の噴出
量であったが、後者はガス圧の低下に従って若干量（５
０mg近辺→４５mg近辺）の噴出量の低下が認められた。
次に、本発明を実施例によって説明するが、これら実施
例により本発明は制約されない。
実施例１ ６０v／v％エタノール水にニトログリセリンを０．７５
w／v％の濃度に溶解し、０．０１Ｎ塩酸を用いてpHを
５．５に調整する。この溶液１０mlを１回スプレー液量
４３mgの液圧式スプレーノズルを備えたポリエチレン瓶
に充てんし密封する。以上により、１回スプレー当り、
ニトログリセリンを約０．３mg口腔内に投与されるスプ
レー剤を得る。
実施例２ ５v／v％エタノール水にニトログリセリンを０．０４６
w／v％の濃度に溶解し、０．０１Ｎ塩酸でpHを５．０に
調整する。この溶液２０mlを、１回スプレー量６５０mg
の液圧式スプレーノズルを備えたポリエチレン瓶に充て
んし、密封する。以上により、１回スプレー当り、ニト
ログリセリンを約０．３mg口腔内に投与されるスプレー
剤を得る。
実施例３ 実施例１と同様の製法でエタノールの濃度が８０v／v％
pHが５．７のスプレー剤を得る。

    に対して0.3 〜1.2w/v％含み、pHが3.0 〜6.5 であり、
    エタノール濃度が30〜85v/v ％であるエタノール水溶液
    を液圧式噴霧容器に封入したニトログリセリンスプレー
    剤
