限がある。
体としては例えば大腸菌に12株が公知であり、この際
外膜中での欠失のためにペリプラズム蛋白質もしくはべ
りプラズム蓄積蛋白質が培地中に達する。これは非特異

白質の遺伝情報と結合すると、熱シヨツク応答及びフィ
ラメント状成長?介してペリプラズム蛋白質の培地への
分泌が生じる。しかしながら、このシステムは熱シヨツ
ク応答により付加的にATP−依存グロチアーゼLaが
生産される限りでは問題である。こうして所望の蛋白質
の蛋白質分解が生じる。従って、La−欠失株(Ion

このような菌株は従来製造されていない。

いうように実施することもできる。この種の突然変異体
は指示蛋白質により確かめられるので、ここで取り扱か
う本発明方法の変法によりプラスミド不含にされ、これ
によりこのプラスミド不含突然変異的を工業的生産が宅
1れる蛋白質の構造遺伝子を包含する他のハイブリッド
プラスミドで形質転換する。指示反応としての多糖加水
分解(デンプン加水分解)に関するその能力が容易なス
クリーニング全可能とするので、指示蛋白質の例はCG
Tである。CGTの使用においてエキソ蛋白質全コード
するDNAフラグメントヲ独自の、搬出に必須のシグナ
ル配列と共に、簡単にかつ目的にあわせてその活性を制
御することのできる強力なプロモーターの制御下に、例

W7261の組換えプラスミドでの転換の後、化学的突
然変異誘発の受容を行ない、かつ引き続@膜及び細胞壁
活性物質を作用させる。D−サイクロセリンに対する耐
性は本発明により改良された排泄のための使用可能な指
示である。細胞壁活性物質での処理は有利に複数回繰り
返す。潜在性分泌突然変異体は多数の得られた突然変異
体から簡単なスクリーニングシステムで選択される。突
然変異体の定量評価は酵素テスト又はイムノブロッティ
ング法(Immunoblotting )により行な
われる。
、細胞壁活性物質での処理、スクリーニング、プラスミ
ド除去、細菌の培養、並びに蛋白質獲得に関しては導入
部に記載した文献のような関連技術文献に記載されてい
る。
模でエキソ蛋白質を生産する。
ソ蛋白質は安価で、かつ短時間で精製されるか、もしく
は更に使用される。
”封入体”に凝集して細胞内に貯蔵するのに対し、本発
明による突然変異体は過剰に生産したエキソ蛋白質を定
量的に培地中に排泄する。こうして、細胞破壊及び”封
入体″の変性といった時間がかかり、かつ1部複雑な工
程がな(なる。”封入体”はしばしば搬出すべき蛋白質
のプレ型からなり、こうしてなおシグナル配列全有し、
こうして完全な触媒機能を示すことはない。僅かな場合
にのみ変性が満足のいく程度に達せられるので、本発明
による突然変異体を用いるエキソ蛋白質生産は著しく方
法全容易にした。エキソ蛋白質は培地からのみ(場合に
より濾過により)精製すればよいか、又はエキソ酵素の
場合には高い収率のために直接所望の酵素反応に使用さ
れる。

変異体の製造のためにCGTに関する遺伝情報金含有す
るグラスミドpCM 7 r33及び大腸菌DS410
’を選択する。改良された酵素分泌を有する突然変異体
に関する指示体としてC()T’i選択した。これに応
じて、大腸菌DS410 (1)CM 703 ) ’
!i−完全培地の液体培地中で67°Cで培養した。光
学密度1が達せられた後、この培地懸濁液にN−メチル
−N′−二トロN−ニトロングアニジン(MNNG )
2 [1μI/ ml f添加し、その後更に30分
間37°Cで恒温保持した(約90係殺菌)。細胞を燐
酸塩緩衝/fi、(pH7,0)で洗浄することにより
、突然変異誘発物質の作用を中断した。細胞懸濁液の部
分i’kLB−プレート(デンプン2チ、ラクトース0
.5チ及びD−サイクロセリンの上昇濃度)にひろげる
。選択のために使用した抗生物質濃度はD−サイクロセ
リン200〜1000μI/ ml f有し、この際こ
の濃度は各突然変異誘発工程の後2倍になった。37°
Cで48時間培養した後、分泌されたCGTの活性に起
因するデンプン加水分解量の大きさにより突然変異体の
選択全行なった。グラスミド金コードする酵素の改良さ
れた潜在性の分泌を、指示プレート(アミロペクチンア
ズール0.5%)上に塗ることにより試験し、酵素テス
トにおいて確かめる。多数の突然変異体から最高細胞外
酵素活性を示す大腸菌株を選択した(大腸菌D8410
に5とする)。抗生物質不含培地中で多数回培養するこ
とにより、突然変異体をプラスミド不含にすることがで
きた。
0に5にプラスミドpCM 703(cGT’iコード
する)で再形質転換した。この組換え菌株をLB−液体
培地(ラクトース0.5係及びグルコース0.2%)中
で60℃で培養した。24時間、48時間もしくは72
時間恒温保持した後、そのつと細胞懸濁液の部分量を取
ジ出しf′c、oその後、細胞を遠心分離し、10チデ
ンプン浴液同量で希釈した培地上澄全40°Cで恒温保
持した。5分間の恒温保持の後、メタノール1.5容t
’を添加した。このことは未反応デンプンの沈殿に導ひ
いた。沈殿したデンプンを遠心分離し、上澄中のシクロ
デキストリン含量をHPLC装置で調べた。次の結果金
得た。
45〜50 180〜20072時間
55〜60 22[]〜240比較例 例2を繰り返したが、大腸菌HB101中にプラスミド
I)CM 703 k形質転換した。培養及び細胞外酵
素活性の測定を同様にして行なった。
0.52 例ろ(β−ラクタマーゼの分泌) 本発明による突然変異体である大腸菌D841 [1に
5全β−ラクタマーゼに関する遺伝情報′f!:有すル
フラスミドpAP 5で形質転換した。この組換プラス
ミド中ではβ−ラクタマーゼ遺伝子は例えばラクトース
での誘発によりβ−ラクタマゼの過剰表現に作用するt
ac−プロモーターの制御下にある。大腸菌D3410
K5(pAP 5 )の培養及び細胞外β−ラクタ
マーゼー活性の測定のための48時間恒温保持後の試料
採取は例2により行なった。細胞外β−ラクタマーゼ活
性は色原体基質、すなわちニトロセフイン金用いて、4
80nmにおける分光光度測定法により測定した; 3
iochemistry 、第26巻(1984年)、
第1282〜1287頁。
1(pAP 5 )全使用した。次の結果が得られた: 恒温保持時間 上澄中の酵素活性(相対数値) 48時間 0.1 例4 例1及び2による方法金繰り返したが、大腸菌DS41
0のかわりに大腸菌株BW7261(ton A 22
、ompF627)eプラスミドル0M 703と共に
使用した。これから得られた突然変異体全プラスミド不
含とし、プラスミドpCM 303で形質転換した。こ
の菌株をラクトース0.5%及びグルコース0.2チを
含有する液状完全培地(LB)中で30’Cで培讐した
。

Claims (1)
- 【特許請求の範囲】 1、(a)表現すべきエキソ蛋白質の構造遺伝子を、表
現のために好適なプラスミド中に自体公知法で統合して
ハイブリッドプラスミドとし、(b)この形成されたハ
イブリッドプラスミドで大腸菌を自体公知法で形質転換
し、 (c)この形質転換大腸菌株に自体公知法で突然変異誘
発を行ない、かつ (d)場合により、その後細胞壁活性物質にさらし、 (e)工程(c)及び場合により工程(d)を行なつた
細胞材料を、自体公知法で、使用した大腸菌株に対して
高められた蛋白質分泌を有する大腸菌突然変異体に関す
るスクリーニングにかけ、かつ (f)場合により、工程(e)により得られた高められ
た蛋白質分泌を有する大腸菌突然変異体を自体公知法で
、プラスミド不含にする、ことにより製造可能な、培地
中に多量の蛋白質を分泌するための大腸菌株。 2、(a)表現すべきエキソ蛋白質の構造遺伝子を、表
現のために好適なプラスミド中に自体公知法で統合して
ハイブリッドプラスミドとし、(b)この形成されたハ
イブリッドプラスミドで大腸菌を自体公知法で形質転換
し、 (c)この形質転換大腸菌株に自体公知法で突然変異誘
発を行ない、かつ (d)場合により、その後細胞壁活性物質にさらし、 (e)工程(c)及び場合により工程(d)を行なつた
細胞材料を、自体公知法で、使用した大腸菌株に対して
高められた蛋白質分泌を有する大腸菌突然変異体に関す
るスクリーニングにかけ、かつ (f)場合により、工程(e)により得られた高められ
た蛋白質分泌を有する大腸菌突然変異体を自体公知法で
、プラスミド不含にする、ことを特徴とする、請求項1
記載の大腸菌株の製法。 3、請求項1記載の大腸菌株を使用し、培地中に多量の
分泌を行なうことにより所望の蛋白質を製造するための
方法。
