Description

【発明の詳細な説明】 〔産慎上の利用分野〕 本発明は活性型ビタミンＤ，類の安定性が著しく優れた
活性型ビタミンＤ，類製剤用組成物に関する。
〔従来の技術及び発明が解決しようとする謀題〕
１α−ヒドロキシビタミンＤ，、１α，２５ージヒドロ
キシビタミン０３等の活性型ビタミン０，類は、くる病
、骨軟化症等の骨疾患に優れた治療薬として知られてい
る。しかし、活性型ビタミンＤ，類は、非常に不安定な
物質で室温においても分解し、製剤化しても長期保存は
不可能であった。
従来、これを安定化する方法としては、活性型ビタミ．
ンＤ，類をトリグリセライドに溶解し組成物とする方法
（特開昭５２−１３０９０５号）アミノ酸類を安定化剤
として添加する方法（特開昭６２−１７号）、プロピレ
ングリコール脂肪酸エステル類に溶解し組成物とする方
法《特開昭６３−５４３２３号）等が知られている。
しかし、これらの方法も必ずしも満足し得るものでなく
、更に優れた活性型ビタミンＤａ類の安定化法の開発が
望まれていた。
〔課題を解決するための手段〕
斯かる実状において、本発明者は鋭意研究を行った結果
、ポリビニルアセタールジェチルアミノアセテート及び
ヒドロキシプ口ピルセルロースが活性型ビタミンＤ，類
に対し優れた安定化効果を有することを見出し、本発明
を完成した。
すなわち、本発明は、活性型ビタミンロ。類とポリビニ
ルアセタールジエチルアミノアセテート又は／及びヒド
ロキシブ口ビルセルロースを含有することを特徴とする
活性型ビタミンＤ，類製剤用組成物を提供するものであ
る。
本発明に用いられる活性型ビタミン０３類としては、例
えば１α−ヒドロキシビタミンＤ３（１α−０１１−Ｄ
ａ）　、１α，２４−ジヒドロキシビタミンＤ，（１α
．　　２　４　−　（ＯＨ）２　−Ｄａ）１α，２５−
ジヒドロキシビタミンＤ，（１α．２　５　ｙ　（０｝
１）２　−ｏ＠）　、１α．２４．２５−トリヒドロキ
シビタミンＤ，（１α，２４．２５＜ＤＨ）ｓ　　Ｏｓ
）などの１α位に水酸基を有するもの、また２４−ヒド
ロキシビタミン［］ｓ（２　４　　０｝１−［１３＞　
　　２　５−ヒドロキシビタミンＤａ（２５　　０Ｈ一
口，）などの１α位に水酸基を有しない活性型ビタミン
Ｄ，などがあげられる。
本発明において、ポリビニルアセタールジエチルアミノ
アセテーｌ・及びヒドロキシブ口ビルセルロースはそれ
ぞれ単独でも、また両者を混合して配合してもよい。こ
れらの配合量は、活性型ビタミンＤ，類の１　−　１０
０．　０００重量倍、特に１０〜１０００重量倍が好ま
しい。
本発明の活性型ビタミンＤ，類製剤用組成物は、活性型
ビタミンＤ，類とポリビニルアセクールジエチルアミノ
アセテート又は／及びヒドロキシプ口ビルセルロースを
均一に混合することにより製造されるが、好ましい製造
法としては次の方法があげられる。すなわら、活性型ビ
タミンＤ，を、これを溶解し得る溶媒、例えばエタノー
ル、プロバノール、イソブロパノール等に溶解し、これ
にポリビニルアセタールジエチルアミノアセテート又は
／及びヒドロキシプロビルセルロースを添加し、よく攪
拌して溶解させた後、溶媒を笛去すれば本発明の活性型
ビタミン０，類製剤用組成物が得られる。
このようにして得られる組成物中の活性型ビタミン０３
類は安定性が著しく改善されているので、通常の製剤化
工程に付して長期間の保存においても安定な活性型ビタ
ミンＩ］ｓ類の製剤を得ることができる。尚この製剤化
にあたっては、一般に使用される製剤化の補助剤を適宜
使用できると共に、他の有効成分を配合することもでき
る。
〔実施例〕
次に本発明を実施例により更に詳細に説明する。
実施例１ １α一ＯＨ　　Ｄａ　　２１１ｇをエタノール５０ｍｍ
’に溶解し溶液とし、これにポリビニルアセタールジエ
チルアミノアセテート２ｇを加え溶解させた後、エタノ
ールを減圧下に留去し、乾燥して組成物１　９　０　０
　ｍｇを得た。この組成物中の１α−０１ｔ−　０３の
含量は、０．１重量％であった。
実施例２ 実施例１と同様にして、１α−１］＋１−Ｌ　　２ｒａ
ｇとヒドロキシプ口ビルセルロース２ｇの混合物からな
る組成物を製造した。
実施例３ 実施例１と同様にして、１α−旧＋−Ｌ　　２ｍｇとポ
リビニルアセタールジエチルアミノアセテート１ｇ及び
ヒドロキシブ口ビルセルロース１ｇの混合物から成る組
成物を製造した。
試験例１ 実施例１〜３で得た組成物を５０℃で保存し、２週間及
び４週間後の１α一旧１−Ｄ，の残存率を調べた。対照
としては、１α一旧１−０コｔ　＋ｓｇをエタノールに
溶解ｊ５た溶液に乳糖１ｇを加えエタノールを減圧下に
留去し乾煙したものを用いた。
本発明の組成物と対照品中の１α一旧１−Ｄ３残存率の
経時変化を第１表に示した。第１表より明らかなように
、本発明の組成物中の１α一旧１−口，は対照品中のと
比較して、極めて安定性に優れているのが分る。
第１表 乳糖　　　　　　　　　　７８．　０５ｇ結晶セルロー
ス　　　　　４．　２５ｇステアリン酸　　　　　　１
．　７０ｇこの錠剤は、１錠中に１α一ＯＨ−Ｄａを約
１μｇ含有するものである。
〔発明の効果〕
本発明によれば、活性型ビタミンＤ，類の安定性が著し
く改善された組成物が得られ、これは製剤用として極め
て有用である。

  1. 【特許請求の範囲】 １　活性型ビタミンＤ＿３類とポリビニルアセタールジ
    エチルアミノアセテート又は／及びヒドロキシプロピル
    セルロースを含有することを特徴とする活性型ビタミン
    Ｄ＿３類製剤用組成物。 ２　ポリビニルアセタールジエチルアミノアセテート又
    は／及びヒドロキシプロピルセルロースの量が、活性型
    ビタミンＤ＿３類の１〜１００，０００重量倍である請
    求項１記載の活性型ビタミンＤ＿３類製剤用組成物。
