Description
本発明は、徐放性タンパク質治療剤の製造、組成物、および使用に関する。具体的には、本発明は、水性環境または生理学的環境中でタンパク質を経時的に長期にわたってかつ均一に放出するための複数のポリマーコーティングタンパク質マイクロスフェアの製造、組成物、および使用に関する。
癌、心血管疾患、血管状態、整形外科障害、歯科障害、創傷、自己免疫疾患、胃腸障害、および眼疾患を含む多くの異なる状態を処置するためには、生物学的標的、例えば、網膜もしくは腫瘍に投与された、または非経口投与された治療タンパク質が徐放されることが望ましい。制御されかつ長期にわたる薬物の送達のための生体適合性かつ生分解性のポリマーが何十年もわたって用いられてきた。ポリマーが経時的に分解するにつれて、治療薬物がゆっくりと放出される。

様々なポリマーを使用した。または、様々なポリマーは微粒子のポリマー表層の製造において使用されることが意図される。これらのポリマーには、特に、エチルセルロース(「EC」)、ポリオルトエステル(「POE」)、ポリ-D,L-ラクチド-コ-グリコリド(「PLGA」)、およびポリ-ε-カプロラクトン(「PCL」)が含まれる。
微粒子化VEGF Trap粒子を、酢酸エチルに溶解した50mg/mLエチルセルロースの溶液中に約50mg/mL VGTの濃度で懸濁した。本明細書では「VGT-50-EC懸濁液」と呼ぶ。

微粒子化VEGF Trap粒子を、酢酸エチルに溶解した50mg/mL PCLの溶液中に約50mg/mL VGTの濃度で懸濁する。本明細書では「VGT-50-PCL懸濁液」と呼ぶ。
実施例2(前記)に従って作製した、それぞれのVGTポリマー懸濁液を、BUCHI Mini Spray Dryer B-290(Buchi Labortechnik AG, Flawil, CH)を用いて噴霧乾燥に供した。簡単に述べると、それぞれの懸濁液を微粒化して微小滴を形成した。その後に、微小滴を加熱乾燥して溶媒を除去し、ポリマーコーティングされたタンパク質微粒子を形成した。懸濁液を12.5mL/minで噴霧乾燥機に注入した。アスピレーター空気およびスプレーガスの流速はそれぞれ約530L/hおよび35m3/hであった。入口温度を90°に設定し、出口温度を約54℃に設定した。
例示されたプロセスに従って製造された、噴霧乾燥され、ポリマーコーティングされたタンパク質粒子は、約2.5μm~約65μmの範囲の円相当径を有する複数の微粒子を生じる(図2)。サイズのばらつきは主にポリマー表層の厚みを反映しているが、タンパク質コアの直径は、ある程度、サイズのばらつきの一因となり得る。
VEGF-Trapタンパク質の安定性を、定量サイズ排除クロマトグラフィー(SE-UPLC)を用いて評価した。定量サイズ排除クロマトグラフィー(SE-UPLC)を用いると、無傷の単量体に対する小さな分解産物および大きな凝集産物の定量が可能になる。結果を表3に記載した。本質的に、噴霧乾燥およびスプレーコーティングプロセスの全体を通してタンパク質は安定なままであった。
1コーティングされていないVEGF Trapを除去するために1時間、再構成した後に、抽出されたVEGF Trapに基づく。
2SE-UPLCによるパーセントネイティブの平均(n=4)。
3RP-HPLCによるポリマーに対するVGT装填材料の重量:重量パーセントの平均(n=4)。
様々な微粒子バッチを緩衝液(10mMリン酸、0.03%ポリソルベート20、pH7.0)中に懸濁し、37℃でインキュベートしながら、経時的に溶液に放出されたタンパク質の量および質を測定することによって微粒子からのタンパク質の放出を確かめた。1~2週間の間隔をおいて微粒子を穏やかな遠心分離によってペレット化し、放出されたタンパク質を含有する上清の80%を後の分析のために収集した。等量の新鮮な緩衝液を交換し、穏やかにボルテックスすることによって微粒子を再懸濁し、37℃のインキュベーションチャンバーに戻した。上清中のタンパク質の量および質をサイズ排除クロマトグラフィーによって評価した。
粒径分布をポリマー濃度および微粒化スプレーガス流によって制御した。ポリマー濃度を高めると大きな粒子に分布がシフトした(200mg/mL PLGA、45mmスプレーガス流対100mg/mL PLGA、45mmスプレーガス流;表5を参照されたい)。同様に、微粒化スプレーガス流を弱めると液滴が大きくなり、従って、粒子が大きくなった(100mg/mL PLGA、25mmスプレーガス流対100mg/mL PLGA、45mmスプレーガス流;表5を参照されたい)。
VEGF TrapまたはIgGを、低分子量(202S)ポリ(乳酸)(PLA-LMW)、高分子量(203S)ポリ(乳酸)(PLA-HMW)、ポリ酸無水物であるポリ[1,6-ビス(p-カルボキシフェノキシ)ヘキサン](pCPH)、ポリ(ヒドロキシ酪酸-コヒドロキシ吉草酸)(PHB-PVA)、PEG-ポリ(乳酸)ブロック共重合体(PEG-PLA)、およびポリ-D,L-ラクチド-コ-グリコリド(PLGA)でスプレーコーティングした。25mg/mLの噴霧乾燥済タンパク質を50~100mg/mLポリマーと組み合わせた。インビトロ放出アッセイを10mMリン酸緩衝液、pH7.2中、37℃で行った。結果を表6に示した。
VEGF TrapおよびIgGをそれぞれのポリマーコートから抽出し、SE-UPLCによって純度について測定した。結果を表7にまとめた。一般的に、タンパク質は、試験されたポリマーのスプレーコーティングプロセスと適合した。タンパク質を放出し続けたポリマーについて、タンパク質は少なくとも14日間、安定なままであった。

Claims (14)
- 生分解性ポリマーでコーティングされた治療タンパク質粒子を含む徐放性医薬組成物を製造する方法であって、
(a)水性治療タンパク質溶液を噴霧乾燥して治療タンパク質微粒子を形成する工程であって、噴霧乾燥機の入口温度が水の沸点より高い温度に設定され、および該噴霧乾燥機の出口温度が54~55℃で、該治療タンパク質微粒子の集団を含む粉末が形成され、該水性治療タンパク質溶液が2mL/min~15mL/minの速度で噴霧乾燥機に注入される、工程;
(b)該治療タンパク質微粒子の集団を含む粉末を、生分解性ポリマーおよび有機溶媒とを含む溶液中に懸濁し、懸濁液を形成する工程;および
(c)該懸濁液を噴霧乾燥し、生分解性ポリマーでコーティングされた治療タンパク質微粒子の集団を含む徐放性医薬組成物を形成する工程であって、該噴霧乾燥する工程が、該懸濁液を微粒化する工程、および次いで、該微粒化した懸濁液に、該有機溶媒の引火点より高い温度で熱を加え、有機溶媒を蒸発させ生分解性ポリマーでコーティングされた治療タンパク質微粒子の集団を形成する工程を含み、および該微粒子が、3重量%未満の水を含む、前記方法、
を含み、
該徐放性医薬組成物が、癌、心血管疾患、血管障害、整形外科障害、歯科障害、創傷、自己免疫疾患、胃腸障害および眼疾患からなる群から選択される症状を処置することができる、
方法。 - 該生分解性ポリマーが、ポリ乳酸(PLA)、ポリグリコール酸(PGA)、ポリ乳酸-ポリグリコール酸コポリマー(PLGA)、ポリ-D,L-ラクチド-コ-グリコリド(PLGA)、PLGA-エチレンオキシドフマレート、ポリエチレングリコール1000とエステル化したPLGA-α-トコフェリルスクシネート(PLGA-TGPS)、ポリ酸無水物であるポリ[1,6-ビス(p-カルボキシフェノキシ)ヘキサン](pCPH)、ポリ(ヒドロキシ酪酸-コヒドロキシ吉草酸)(PHB-PVA)、ポリエチレングリコール-ポリ(乳酸)コポリマー(PEG-PLA)、ポリ-ε-カプロラクトン(PCL)、ポリ-アルキル-シアノ-アクリレート(PAC)、ポリ(エチル)シアノアクリレート(PEC)、ポリイソブチルシアノアクリレート、ポリ-N-(2-ヒドロキシプロピル)メタクリルアミド(ポリ(HPMA))、ポリ-β-R-ヒドロキシブチレート(PHB)、ポリ-β-R-ヒドロキシアルカノエート(PHA)、ポリ-β-R-リンゴ酸、リン脂質-コレステロールポリマー、2-ジオレオイル-sn-グリセロ-3-ホスファチジルコリン/ポリエチレングリコール-ジステアロイルホスファチジルエタノールアミン(DOPC/PEG-DSPE)/コレステロール、ポリサッカライド、セルロース、エチルセルロース、メチルセルロース、アルジネート、デキストランおよびデキストランヒドロゲルポリマー、アミロース、イヌリン、ペクチンおよびガーゴム、キトサン、キチン、ヘパリン、ヒアルロン酸、シクロデキストリン(CD)ベースのポリロタキサンおよびポリ擬ロタキサン、ポリアスパラギン酸、ポリグルタミン酸、ポリロイシン、ロイシン-グルタミン酸コポリマー、ポリブチレンスクシネート(PBS)、ゼラチン、コラーゲン、フィブリン、フィブロン、ポリオルトエステル、ポリオルトエステル-ポリアミジンコポリマー、ポリオルトエステル-ジアミンコポリマー、潜在性酸(latent acid)を組み込んだポリオルトエステル、ポリ(エチレングリコール)/ポリ(ブチレンテレフタレート)コポリマー、ならびにその組み合わせおよびコポリマーからなる群より選択される、請求項1に記載の方法。
