Description
1.1 配列番号10、配列番号11、配列番号12及び配列番号13のトランスフェクションの後のNRARPの遺伝子発現レベル
以下の配列、すなわち、配列番号10(SYL136001)、配列番号11(SYL136005)、配列番号12(SYL136003)及び配列番号13(SYL136004)のうちの1つからなるセンス鎖と、併せて、相補的なアンチセンス鎖とからなる19bpの平滑末端dsRNAのうちの1つの100nMを、トランスフェクト剤としてLipofectamine 2000を用いて、ヒトHeLa細胞にトランスフェクトした。各配列番号の後のSYL参照番号は、参照するdsRNA化合物を指す。これらの実施例全体を通して(そうでないことが文脈から明らかでない限り)、特定の配列番号の投与又はトランスフェクションに言及する場合、このことは、図2及び図3に示すその配列番号からなるセンス鎖と相補的なアンチセンス鎖とからなる19bpのdsRNAを投与又はトランスフェクトしたことを示すことに留意されたい。トランスフェクションは全て、製造元の標準的な使用説明に従って実施した。同じ実験において、siRNAのスクランブル配列を、干渉の特異性の対照として使用した。細胞ペレットを収集し、処理して、siRNAの作用機構の結果としてのmRNAレベルの変動の可能性を評価した。RNAのレベルを、リアルタイムPCRにより、相対定量法、すなわち、比較の閾値を2-ΔΔCTとする方法を使用して定量化した。{Livak及びSchmittgen、2001年}。リアルタイム定量的PCRの実験は全て、三つ組で実施し、3回の独立した実験において繰り返した。平均及びSEMを計算し、図に示した。図4に示すように、NRARPのmRNAレベルが、ヒトHeLa細胞中で、研究した3つの時点において、配列番号10、配列番号11、配列番号12及び配列番号13のトランスフェクションに応答して顕著に(50~60%)減少した。予想通り、siRNAのスクランブル配列は、研究した時点のいずれにおいても、NRARPの発現レベルを調節しなかった。
本発明のsiRNAのトランスフェクションの後の細胞生存率を分析するために、ヒトHeLa細胞中で、トランスフェクト剤としてLipofectamine 2000を用いて、以下の配列、すなわち、配列番号10、配列番号11、配列番号12及び配列番号13のうちの1つの100nMのトランスフェクションを行った後に、in vitroでの毒性を研究した。トランスフェクションは全て、製造元の標準的な使用説明に従って実施した。同じ実験において、siRNAのスクランブル配列を、干渉の特異性の対照として使用した。細胞ペレットを、トランスフェクションの24、48及び72時間後に収集し、処理して、細胞生存率の変動の可能性を評価した。Promega社製のCellTiter 96(登録商標)Aqueous Non-Radioactive Cell Proliferationアッセイを使用して、細胞生存率を測定した。この方法は、生きている細胞の、MTSテトラゾリウムをホルマザンに還元する能力に基づく。ホルマザンの量を、490nmにおける光吸収を測定することによって定量化する。平均及びSEMを計算し、図5にプロットした。図5は、以下の配列、すなわち、配列番号10、配列番号11、配列番号12及び配列番号13のうちのいずれについても、そのトランスフェクションに応答する細胞生存率の変化はなかったことを示している。要約すると、本発明のsiRNAは、無毒性であることが見出され、十分に忍容されている。
本発明のsiRNAの安定性を改善するため、及び免疫原性の活性化を起こさないことを確保するために、種々のsiRNA最適化化学的改変を、規準配列である配列番号10(SYL136001)に導入し、したがって、以下の化学的に改変された新しい実体、すなわち、配列番号19、配列番号23、配列番号25、配列番号27、配列番号29、配列番号31、配列番号35、配列番号37及び配列番号39を得た。新しい化学的改変配列を、ヒト、マウス及びラットの細胞中に個々にトランスフェクトして、NRARPのmRNAレベルを低下させるそれらの能力を分析した。化学的改変を、図3に詳述する。ヒトHeLa細胞、ヒトBEAS-2B細胞及びマウスC2C12細胞を選択した。その理由は、これらは、NRARPを顕著なレベルで発現する細胞であり、siRNAの、NRARPの発現に対する作用を試験するための良好なモデルとなると考えられるからである。トランスフェクト剤として、Lipofectamine(HeLa細胞)、Mirus Transit-X2(BEAS-2B)、Dharmarfect 3(C2C12)及びsiPORT NeoFX(ARL6)を用いて、以下の配列、すなわち、配列番号19、配列番号23、配列番号25、配列番号27、配列番号29、配列番号31、配列番号35、配列番号37及び配列番号39のうちの1つの100nMを個々に、細胞にトランスフェクトした。トランスフェクション後の3つの時点(24、48及び72時間)において、遺伝子の発現を分析した。トランスフェクションは全て、製造元の標準的な使用説明に従って実施した。同じ実験において、siRNAのスクランブル配列を、干渉の特異性の対照として使用した。RNAのレベルを、リアルタイムPCRにより、相対定量法、すなわち、比較の閾値を2-ΔΔCTとする方法を使用して定量化した{Livak及びSchmittgen、2001年}。リアルタイム定量的PCRの実験は全て、三つ組で実施し、3回の独立した実験において繰り返した。平均及びSEMを計算し、プロットし、図6~図9に示すグラフを得た。図6に、HeLa細胞において得られた結果を示す。この細胞系では、配列番号10(SYL136001)は、NRARPのmRNAレベルを、トランスフェクションの24時間後に60%、かつトランスフェクションの48及び72時間後に50%低下させた。化学的に改変させた配列番号19は、NRARPのmRNAレベルを、トランスフェクションの24時間後に60%、かつトランスフェクションの48時間後に40%低下させ;トランスフェクションの72時間後には、基礎レベルが完全に回復した。配列番号23は、NRARPのmRNAレベルを、トランスフェクションの24~48時間後に40%、かつトランスフェクションの72時間後に10%低下させた。配列番号25は、NRARPのmRNAレベルを、トランスフェクションの24~48時間後に50~60%、かつトランスフェクションの72時間後に20%低下させた。配列番号27は、NRARPのmRNAレベルを、トランスフェクションの24~48時間後に30%低下させ、トランスフェクションの72時間後には、基礎レベルが回復した。配列番号29は、NRARPのmRNAレベルを非常に有効に低下させた。およそ80%の低下が、この配列に応答して、トランスフェクションの24~48時間後に観察され、その後、mRNAレベルは増加したが、依然として、トランスフェクションの72時間後でも基礎レベルを40%下回った。配列番号31は、NRARPレベルを有効に低下させ、研究した3つの時点で、基礎レベルと比して60%の低下が観察された。配列番号35は、NRARPのmRNAレベルを、トランスフェクションの24~48時間後に60%低下させ、トランスフェクションの72時間後に、mRNAレベルの急激な回復が見出され、したがって、NRARPのmRNAレベルは、基礎レベルを10%下回った。配列番号37は、NRARPのmRNAレベルの最も大きな低下を引き起こす化合物であった。この化合物のトランスフェクションの24、48及び72時間後に、90%、80%及び70%の低下が観察された。配列番号39は、NRARPのmRNAレベルを、トランスフェクションの24時間後に70%、かつトランスフェクションの48時間後に50%低下させ、その後、mRNAレベルは急激に増加したが、トランスフェクションの72時間後でも、基礎レベルは完全には回復しなかった。図7に、ヒトBEAS-2B細胞において得られた結果を示す。この細胞系では、配列番号10(SYL136001)及び配列番号19は、NRARPレベルを、トランスフェクションの24時間後に70%低下させ、その後、mRNAレベルは緩慢に増加したが、mRNAレベルは、トランスフェクションの48時間後に基礎レベルを50%下回り、トランスフェクションの72時間後に基礎レベルを40%~50%下回ったものの、依然として、基礎レベルが、NRARPのmRNAレベルに関する研究の時間枠内で完全に回復することはなかった。配列番号23及び配列番号27は、ヒトBEAS-2B細胞において、NRARPのmRNAレベルを若干低下させ、研究した3つの時点で、低下はおよそ20~30%であった。トランスフェクションの72時間後でも、基礎レベルは完全には回復しなかった。配列番号25及び配列番号39は、NRARPのmRNAレベルを徐々かつ時間依存性に低下させ、トランスフェクションの72時間後に、50%~60%の最大の低下に達した。配列番号31及び配列番号35は、NRARPのmRNAレベルを有効に低下させ、mRNAレベルは、研究した3つの時点で、いずれの化合物にも応答して基礎レベルをおよそ60~70%下回った。この細胞系では、配列番号29及び配列番号37が、NRARPのmRNAレベルを低下させるのに最も有効な生成物であり、それぞれが、研究した3つの時点において、およそ80%及び90%の急激なかつ持続性の低下を引き起こした。予想通り、siRNAのスクランブル配列は、研究した時点のいずれにおいても、NRARPの発現レベルを低下させなかった。図8に、マウスC2C12細胞において得られた結果を示す。配列番号10(SYL136001)は、NRARPレベルを、トランスフェクションの24時間後に60%低下させ、その後、mRNAレベルは緩慢に増加した。しかし、トランスフェクションの72時間後に、NRARPのmRNAレベルは基礎レベルを40%下回ったが、依然として、この時点でも基礎レベルは完全には回復しなかった。配列番号19は、NRARPのmRNAレベルを、トランスフェクションの24時間後におよそ30%低下させ、トランスフェクションの48時間後に、80%の劇的な低下が見出され、その後、レベルは増加したが、依然として、トランスフェクションの72時間後でも、NRARPのmRNAレベルは基礎レベルを40%も下回った。配列番号23、配列番号25及び配列番号27は、NRARPのmRNAレベルを若干低下させ、これらの配列のそれぞれのトランスフェクションに応答して観察された低下は、研究した3つの時点でおよそ20~40%であった。配列番号29及び配列番号39は、NRARPのmRNAレベルを、トランスフェクションの24及び48時間後に有効に低下させ、それぞれのこれらの時点におけるレベルは、基礎レベルを50%及び60%下回った。トランスフェクションの72時間後の両方の配列に対する応答は、先行する時点に比して増加したが、この時点では依然として、基礎レベルは回復しなかった。配列番号31は、NRARPのmRNAレベルを、トランスフェクションの24時間後におよそ40%低下させ、トランスフェクションの48時間後に70%の大きな低下が見出され、その後、レベルは増加したが、依然として、トランスフェクションの72時間後でも、NRARPのmRNAレベルは基礎レベルを40%も下回った。この細胞系では、配列番号35及び配列番号37が、NRARPのmRNAレベルの最も大きな低下を引き起こす化合物であり、配列番号35及び配列番号37は、NRARPのmRNAレベルを、トランスフェクションの24時間後に60%、かつトランスフェクションの48時間後におよそ80%低下させた。トランスフェクションの72時間後に、基礎レベルの部分的な回復が生じた。C2C12細胞において、ここでも予想通り、siRNAのスクランブル配列は、NRARPのmRNAレベルを、研究した時点のいずれにおいても低下させなかった。図9に、ARL6ラット細胞において得られた結果を示す。配列番号10(SYL136001)は、NRARPのmRNAレベルを、トランスフェクションの24~48時間後に50%、かつトランスフェクションの72時間後に60%低下させた。配列番号37は、NRARPレベルを、トランスフェクションの24時間後に50%、トランスフェクションの48時間後に60%、かつトランスフェクションの72時間後に70%低下させた。
本発明のsiRNAのトランスフェクションの後の細胞生存率を分析するために、ヒトHeLa細胞及びヒトBEAS-2B細胞並びにマウスC2C12細胞中でそれぞれ、トランスフェクト剤として、Lipofectamine 2000、Mirus Transit-X2、Dharmafect 3を用いて、以下の配列、すなわち、配列番号10(SYL13600)、配列番号19、配列番号23、配列番号25、配列番号27、配列番号29、配列番号31、配列番号35、配列番号37及び配列番号39のうちの1つの100nMのトランスフェクションを行った後に、in vitroでの毒性研究を実施した。トランスフェクションは全て、製造元の標準的な使用説明に従って実施した。同じ実験において、siRNAのスクランブル配列を、干渉の特異性の対照として使用した。細胞ペレットを、トランスフェクションの24、48及び72時間後に収集し、処理して、siRNAのトランスフェクションの結果としての細胞生存率の変動の可能性を評価した。Promega社製のCellTiter 96(登録商標)Aqueous Non-Radioactive Cell Proliferationアッセイを使用して、細胞生存率を測定した。この方法は、生きている細胞の、MTSテトラゾリウム化合物をホルマザンに還元する能力に基づき、ホルマザンを、490nmにおける光吸収により測定する。それぞれの実験について、平均及びSEMを計算し、図10~図12にプロットした。図10、図11及び図12は、使用した細胞系全てにおいて、以下の配列、すなわち、配列番号10(SYL13600)、配列番号19、配列番号23、配列番号25、配列番号27、配列番号29、配列番号31、配列番号35、配列番号37及び配列番号39のうちのいずれについても、そのトランスフェクションに応答する細胞生存率の変化はなかったことを示している。更に、ARL6ラット細胞においてもまた、以下の配列、すなわち、配列番号10(SYL136001)及び配列番号37のうちの1つに対して、トランスフェクト剤としてsiPORT NeoFXを使用して、トランスフェクションに応答する細胞生存率のレベルを評価した。図13は、ARL6細胞において、試験した配列にいずれについても、それに応答する細胞生存率の顕著な変化はなかったことを示している。したがって、本発明のsiRNAは全て、無毒性であり、十分に忍容されていると結論付けることができる。
血管新生に対する本発明のsiRNAの作用を研究するために、HUVEC細胞を用いて実験し、これらの細胞中のNRARPの発現を分析した。
血管新生は、既存の静脈からの新しい静脈の形成である。このプロセスは、発生の過程及びいくつかその他の生理学的な過程において不可欠であり、眼において、このプロセスに調節不全が生じると、種々の種類の網膜疾患、例として、加齢黄斑変性(AMD)及び糖尿病性網膜症(DR)に至る恐れがある。ヒト臍帯静脈内皮細胞(HUVEC)は、最初の供給物質から1~3回継代した後に保存されている初代細胞である。これらの細胞は、血管新生性の物質、例として、VEGFに応答して、誘発されて、管状構造を形成することができる。

i)細胞
HUVEC細胞(参照番号:CC-2519/バッチ番号:000191772)を、Lonza社から得た。全ての実験を、継代6~9回時の、75~85%の細胞密度で培養した同じバッチの細胞を用いて実施した。トランスフェクションを、siPORT NeoFX(商標)を使用して、標準的な手順に従って実施した。電気穿孔については、「Hernandez JLら、2004年、Angiogenesis.7:235~241頁」に公開されている手順に従った。手短に述べると、cell manipulator(登録商標)600(BTX社)を使用して、1200μFで20ミリ秒のパルスを適用することによって、1×106個の細胞に電気穿孔を行った。電気穿孔はそれぞれ、2μgの遺伝子材料を使用して実施した。
(1)RNAの単離及び逆転写
全RNAを、細胞培養物から、RNeasy RNA抽出キット(Invitrogen社、CA、米国)を使用して単離した。4μgの全RNAを、High-Capacity cDNA Archiveキット(Applied Biosystems、Inc.社、Foster City、CA、米国)を使用して、製造元の使用説明に従って逆転写した。
qPCRを、Stepone plus検出システム(Applied Biosystems社)を使用して実施した。TaqMan 2× Universal Master Mix中で、50ナノグラムの各試料を、以下、すなわち、95℃、10分間;続いて、95℃、15秒間の40サイクル;及び60℃、1分間の条件下で増幅した。
電気穿孔を行って、siRNAをHUVEC細胞内に導入して、首尾よい成果を得たことから、この方法を使用して、その後の研究を行った。この場合、マトリゲル上での毛細血管構造の形成、遊走、創傷治癒及び増殖についてのアッセイにおいて、NRARPの改変siRNA、すなわち、配列番号37の役割を評価した。
(1)アッセイの設定
4時間の期間にわたり欠乏状態となした細胞を、マトリゲルで被覆した96ウエルプレート上に15,000個の細胞/ウエルで蒔いた。補充物質を有しない内皮細胞用基本培地(EBM)を、負の(構造を形成しない)対照として使用し、一方、補充物質(hEGF、ヒドロコルチゾン、ウシ脳抽出物、ゲンタマイシン、ヘパリン)及び10%のFCSを有する完全培地を、最多数の管状構造を誘発する陽性の対照として使用した。アッセイは、三つ組で、3~6回の独立した実験において実施した。24時間平板培養してから、培養物を分析した。
細胞を、電気穿孔によりトランスフェクトし、完全培地中で24時間の期間にわたり回復させ、更に2時間にわたり血清が欠乏する状態となし、その後、細胞を、マトリゲルで被覆した96ウエルプレート上に、30,000個の細胞/ウエルの細胞密度で蒔いて、構造を形成させた。播種してから6時間及び24時間後に、形成された構造を分析した。配列番号37の効能を試験するために、以下の条件をアッセイした。

(1)アッセイの設定
24時間にわたり欠乏状態となした細胞を、96ウエルプレート中に3000個の細胞/ウエルの密度で蒔き、種々の条件下で培養した。5日後に、増殖をMTTアッセイにより評価した。使用した条件を、以下に示す:2%のFCS及び種々の濃度のVEGF(20~30~50ng/ml)。2%のFCSは、細胞が基礎増殖速度を保ち、培養物が細胞停止に入るのを回避するのに必要である。図21Aは、細胞の増殖の3倍の誘発が、VEGFに応答して、試験したその濃度にかかわらずもたらされたことを示している。抗VEGF剤の阻害剤としての作用を分析するために、細胞を、30ng/mlのVEGFの存在下で、ベバシズマブ(Avastin)の濃度を減少させて(12nMから開始し、1:2で希釈した)培養した。この培養は、IC50を決定するためであった。図21Bに示すように、ベバシズマブのIC50は、0.85nMであることが見出された。
細胞に電気穿孔を行い、細胞を、1%のゼラチンでコートした96ウエルプレート中に5000個の細胞/ウエルの密度で蒔き、その後、細胞を24時間の期間にわたり回復させた。細胞を、回復させてから、10%のFCS及び100ng/mlのVEGFを補充した基本培地中で培養した。平板培養の3日後に、細胞の増殖をMTTにより分析した。試験した条件を、以下に示す。

これらの研究を、8μmの不透明なメンブランフィルターを有する24トランスウエルシステムプレート(Transwell HTS FluroBlok(商標)Multiwell Insert System、Becton Dickinson社製)中で実施した。細胞の吸収を促進するために、メンブランの両方の表面を、I型コラーゲンを15μg/mlの濃度で用いて、37℃で2時間かけて被覆した。細胞を50,000個の細胞/ウエルの密度で蒔き、4時間の期間にわたり血清が欠乏する状態となし、その後、遊走刺激剤を添加した。刺激剤をトランスウエルの下の部分に添加してから24時間後に、上のコンパートメントまで遊走するに至った細胞を、Calcein-AMを用いて染色した後に、その数を分析した。
細胞に電気穿孔を行い、細胞を24時間の期間にわたり回復させ、その後、細胞を、15μg/mlのコラーゲンでコートした8μmの不透明なメンブランフィルターを有する96トランスウエルプレート上に、10,000個の細胞/ウエルの密度で蒔いた。試験条件を全て、基本培地の存在下及び(10%のFCS及び100ng/mlのVEGFを補充した)完全培地の存在下でアッセイした。試験条件を、以下に示し、平板培養してから24時間後に、Calcein-AMを用いて細胞を染色することによって、遊走を分析した。

細胞に電気穿孔を行い、細胞を24時間の期間にわたり回復させ、その後、細胞を、1%のゼラチンでコートした96ウエルプレート中に、70,000個の細胞/ウエルの密度で蒔いた。平板培養の24時間後に、細胞の狭い領域を削り取ることによって、病変を誘発し、病変を誘発してから16時間後に、培養物を分析した。10μΜのcalceine-AMを用いて細胞を染色し、削った領域、及び細胞により再び覆われた領域のパーセントを分析することによって、分析を実施した。試験条件を全て、基本培地の存在下及び(10%のFCS及び100ng/mlのVEGFを補充した)完全培地の存在下でアッセイした。試験条件を、以下に示し、平板培養してから24時間後に、遊走を分析した。

上記で言及した研究から得られたHUVEC細胞を収集し、全RNAを抽出して、NRARP標的遺伝子のレベルをqPCRにより分析した。図29は、配列番号37のsiRNAの電気穿孔に応答した、NRARPのmRNAレベルの低下を示している。
2.1 レーザーにより誘発される脈絡膜新血管形成のラットモデルの網膜及び脈絡膜におけるNRARPの発現
2.1.1 目的
本研究の目的は、CNVがレーザーにより誘発されているNorway Brownラットの網膜及び脈絡膜におけるNRARPの発現を、種々の時点で評価することであった。この標的遺伝子の発現の分析は、i)新血管形成に関連する網膜疾患を治療するための新しい化合物を開発する目的で、この標的がサイレンシングを行うべき候補であるかどうかを研究するために、CNVに応答して、NRARPが上方制御されるかどうかを評価し、ii)標的遺伝子のサイレンシングを行うために、一時的なNRARPの発現を研究して、動物を治療するのに最もよい時間を決定するという、2つの目的を果たした。
CNVは、いくつかの脈絡網膜性疾患に共通する非特異的な病変である。これらの病変は、ブルッフ膜の切断又は破壊;網膜下の空間への及び/又は網膜下の色素上皮への、脈絡膜毛細血管内皮細胞、周皮細胞及び炎症細胞の侵入を伴う炎症及び血管新生の誘発を引き起こす一連の事象を特徴とする{Grossniklaus HEら、2010年}。網膜下の空間への脈絡膜毛細血管の貫通は、いくつかの網膜疾患に共通する顕著な特徴である。これらの疾患の一部の例として、AMD、PDR又はDREが挙げられる。
i)動物
レーザーを適用した直後に、3つのレーザー病変のうちの2つ以上において出血が明らかである全ての眼。
(l)RNAの単離及び逆転写
全RNAを、網膜及び脈絡膜から、RNeasy RNA抽出キット(Invitrogen社、CA、米国)を使用して単離した。4μgの全RNAを、High-Capacity cDNA Archiveキット(Applied Biosystems, Inc.社、Foster City、CA、米国)を使用して、製造元の使用説明に従って逆転写した。
qPCRを、Stepone plus検出システム(Applied Biosystems社)を使用して実施した。TaqMan 2× Universal Master Mix中で、500ナノグラムの各試料を、以下、すなわち、95℃、10分間;続いて、95℃、15秒間の40サイクル;及び60℃、1分間の条件下で増幅した。全てのqPCR増幅は、三つ組で実施し、少なくとも2回の独立した実験において繰り返し、これらの実験は常に、逆転写の対照を含むが、鋳型の対照は含まなかった。
CNVのレーザーによる誘発の後の種々の時点で、NRARPの発現を、網膜及びRPE/脈絡膜において分析した。得られた結果から、網膜において、レーザー病変の誘発から24時間後に、NRARPのmRNAレベルが若干減少したことが示されている。病変を誘発してから72時間後に、NRARPのmRNAレベルは、若干(時間=0と比して1.5倍に)増加している。研究の終わりに(t=504時間)、NRARPレベルは、病変を誘発する前に観察されたレベルと同等になった(図14)
この研究の結果を総合すると、NRARPは、網膜における新血管形成を制御するための治療を開発し得る有効な標的となると結論付けることができる。網膜及び脈絡膜の両方における発現のパターンから、この研究で使用したモデルにおいてNRARPの発現が顕著に誘発されたこと、及び血管新生におけるNRARPの役割を考慮すると、NRARPが、サイレンシングを行う良好な候補であり得るであろうことが示されている。
2.2.1 目的
レーザー誘発型の脈絡膜新血管形成(レーザーCNV)のラットモデルにおいて、1つの試験薬剤(配列番号37のsiRNA)の抗血管新生作用/血管を破壊する作用を、外用投与の後に決定する。
レーザー誘発型の脈絡膜新血管形成のモデルにおいて、配列番号37の外用点眼の抗血管新生作用/血管を破壊する作用を決定するために、雌のBrown Norwayラットを用いて、26日間の研究を実施した。
2.2.3.1 ラットにおけるレーザー誘発型の脈絡膜新血管形成(CNV)

動物の眼を、1%のCyclogyl溶液を用いて拡張し、光から保護した。観察可能な拡張を得たら、ケタミン/キシラジンを用いて、動物を鎮静させた。Micron III小動物用眼底カメラ(Phoenix Research社)を使用して、鎮静させた動物の眼底を観察し、記録した。Micron III専用のレーザー用付属品を介して接続して、レーザーによる処置を、熱レーザーを使用して実施した。520nmの波長を使用して、それぞれの眼に、1眼当たり総数3つの病変を設けた。結果として得られた眼底の画像を、記録及び評価して、レーザーが、ブルッフ膜を通って気泡を首尾よく生成するに至ったことを確認した。
ケタミン/キシラジンを用いて、動物に麻酔を施し、Cyclogyl及び/又はトロピカミドを外用投与して、瞳孔を拡張した。鎮静及び拡張の後に、Hamilton社製シリンジ及び33ゲージ針を使用して、1眼当たり5μlの総体積を、硝子体内の毛様体扁平部に注射した。
レーザーの適用又は硝子体内への注射の後に出血の徴候を示す眼はいずれも、分析から除外した。
動物に、ケタミン/キシラジンを用いて麻酔を施し、次いで、10%のフルオレセインナトリウムを1μl/g体重でIP注射した。Micron III及び488nmの標的波長のための励起用フィルター/バリアフィルターを使用して、眼底の画像を、8ビットのTIFFファイルとして撮影した。それぞれの眼について、眼底の標準的なカラー写真もまた撮影した。
画像分析コンピュータ処理ソフトウエア(ImageJ、NIH、米国)を使用して、全てのTIFF画像を定量化した。面積をピクセルで示して定量化するために、個々の病変をフリーハンドでトレースし、眼底のカラー写真は、病変の場所を参照するのに使用した。病変の中心にある血管形成が生じていない領域は、面積の計算から除外した。出血がある場合又は2つの病変がオーバーラップする場合には、これらの病変は分析から除外した。
ケタミン/キシラジン(80/10mg/kg)を用いて、動物に麻酔を施し、次いで、Euthasol(ペントバルビタール)を200mg/kgでIP投与することによって、動物を安楽死させた。安楽死の後に、眼球を除去し、個々の眼を4%のパラホルムアルデヒド中に固定した。眼は、固定したら、全てを個別に、2mLのスクリューキャップ付きポリプロピレンチューブ中に保存した。
統計学的分析を、Graphpad Prismソフトウエア(4.0版)を用いて、両側マン-ホイットニーt検定を使用して実施した。<0.05のp値を示す変化のみが、統計学的に有意であると考えられる。
レーザー誘発型のCNVのラットモデルにおいて、外用点眼により投与した配列番号37の作用を評価した。レーザーによる処置の3週間後に、フルオレセイン血管造影を実施して、ラット眼中のCNV病変のサイズ(ピクセルで示す面積)を定量化した。
脈絡膜新血管形成のラットモデルにおいて、5mg/mlの配列番号37の外用点眼により、病変サイズは、ビヒクル単独の外用点眼と比べて有意に低下する。
Claims (14)
- siRNA分子を含む、網膜の新血管形成を予防及び/又は治療するための、点眼剤の形態の医薬組成物であって、
前記siRNAが、センス鎖と前記センス鎖に相補的であるアンチセンス鎖とを含むか、又はセンス鎖と前記センス鎖に相補的であるアンチセンス鎖とからなり、前記センス鎖が、配列番号37のヌクレオチド配列を含むか、又は配列番号37のヌクレオチド配列からなり、前記アンチセンス鎖が、配列番号38のヌクレオチド配列を含むか、配列番号38のヌクレオチド配列からなり、
前記siRNAが配列番号1のヌクレオチド配列を特異的に標的にする、医薬組成物。 - 前記siRNAが、19個のヌクレオチドの二本鎖領域を含む、請求項1に記載の医薬組成物。
- 前記siRNAが平滑末端を有する、請求項2に記載の医薬組成物。
- 少なくとも1つのヌクレオチドが、化学的改変を含む、請求項1から3のいずれか一項に記載の医薬組成物。
- ヌクレオチドの前記化学的改変が、2'-O-メチル改変、2'-フルオロ改変、ホスホロチオエート改変ヌクレオチドの導入、ウラシルの5-プロピニルウラシルによる置換、ウラシルの5'-メチルウリジンによる置換、ウラシルリボースヌクレオチドの4'-チオリボースによる置換及びウラシルリボースヌクレオチドのデオキシチミジンヌクレオチドによる置換、並びにそれらの組合せから選択される、請求項4に記載の医薬組成物。
- 前記化学的改変が、センス鎖上、アンチセンス鎖上又は両方にある、請求項4又は5に記載の医薬組成物。
- 緩衝生理食塩水をさらに含む、請求項1から6のいずれか一項に記載の医薬組成物。
- 約7.0~約7.4のpHを有する、請求項7に記載の医薬組成物。
- ゲルの形態である、請求項1から6のいずれか一項に記載の医薬組成物。
- ポリ(ラクチド-co-グリコリド)、カーボポール、ヒアルロン酸及びポリアクリル酸から選択される1つ又は複数のバイオポリマーをさらに含む、請求項1から9のいずれか一項に記載の医薬組成物。
- 前記siRNAは、細胞中への導入時にNRARP遺伝子の発現を低下させる、請求項1から10のいずれか一項に記載の医薬組成物。
- センス鎖が、配列番号37のヌクレオチド配列を含むか、又は配列番号37のヌクレオチド配列からなり、アンチセンス鎖が、前記センス鎖に相補的であるsiRNAを含む、網膜の新血管形成を予防及び/又は治療するための、医薬組成物。
- 配列番号37の配列を含むか、又は配列番号37のヌクレオチド配列からなるセンス鎖と、前記センス鎖に相補的であるアンチセンス鎖とを有する、siRNA。
- 前記アンチセンス鎖が配列番号38のヌクレオチド配列を含むか、配列番号38のヌクレオチド配列からなる、請求項13に記載のsiRNA。
