Description
1)光合成微細藻類バイオマスを光束に暴露する工程であって、上記光束は、実質的に10μmol・m−2・s−1以上であって実質的に13μmol・m−2・s−1以下である光強度を有する、工程、
2)誘導培地中のNaNO3濃度が2.5g/Lを超えるように窒素源を添加する工程
を含む。
・a)微細藻類バイオマスを回収する工程
・b)上記バイオマスの増殖を阻害する工程
・c)フィコビリタンパク質の合成を誘導する工程
・d)上記フィコビリタンパク質が豊富なバイオマスを回収する工程
本発明の方法は以下の2つの必須工程に分けられる。
1)市販グレードのサリチル酸粉末をフィコビリタンパク質抽出物に添加する。溶液(サリチル酸+抽出物)を15分間撹拌する。この工程によって、溶液を均質化し、溶液中のフィコビリタンパク質分子と酸との接触を促進することができる。
サリチル酸(g)=0.007×フィコビリタンパク質(mg)/0.52
Log10Y=4−0.08Temp−0.013×Lum+0.0005×Temp2+0.0002×temp×Lum
サリチル酸を用いた選択的沈殿により抽出したフィコビリタンパク質の清澄化を試みる。抽出は、乾燥又は新鮮なスピルリナバイオマスから行うことができる。
抽出物(様々な希釈及び遠心分離条件により得られたもの)中のフィコビリタンパク質及び断片の様々な初期濃度、並びに、サリチル酸の様々な濃度において、沈殿したフィコビリタンパク質の清澄度の変化を調べる。
下記組成を有するシワ防止クリームが得られる。
水:59%
甘扁桃油:18%
グリセリンに溶解したフィコビリタンパク質が豊富な安定した沈殿物:7%
パーム油:5%、ヒマシ油:3%
藻類抽出物:3%(β−カロチン)
蜜蝋:2%
キサンタンガム:1.7%
香料:1%
エチルパラベン:0.1%、メチルパラベン:0.1%、ベンジルパラベン:0.1%
下記組成を有するシミ消しクリームが得られる。

Claims (14)
- フィコビリタンパク質が豊富な安定した沈殿物を得る方法であって、
フィコビリタンパク質の水性抽出物にサリチル酸を添加する沈殿工程を少なくとも1つ含み、
上記沈殿物のフィコビリタンパク質含量が上記水性抽出物中の初期含量の少なくとも50%に等しく、
上記沈殿物の清澄度が少なくとも70%に等しい、方法。 - 上記沈殿物のフィコビリタンパク質含量は上記水性抽出物中の初期含量の少なくとも50%に等しく、上記沈殿物の清澄度は少なくとも80%、好ましくは90%又は100%に等しい、請求項1に記載の方法。
- 上記沈殿物のフィコビリタンパク質含量は上記水性抽出物中の初期含量の少なくとも70%に等しく、上記沈殿物は膜断片を含まない、請求項1に記載の方法。
- 上記フィコビリタンパク質の水性抽出物は上記フィコビリタンパク質を少なくとも1.5g/L含有する、請求項1〜3のいずれか1項に記載の方法。
- 上記フィコビリタンパク質の水性抽出物に上記サリチル酸を4g/L〜20g/Lの範囲で添加する請求項1に記載の方法。
- 上記フィコビリタンパク質の水性抽出物に上記サリチル酸を4g/L〜13g/Lの範囲で添加する請求項1又は3に記載の方法。
- フィコビリタンパク質の水性抽出物に上記サリチル酸を13〜14g/Lの範囲で添加する請求項1又は2に記載の方法。
- サリチル酸による沈殿工程の前に、デカンテーションにより清澄化する工程を追加する請求項1〜7のいずれか1項に記載の方法。
- 上記沈殿物を溶解する工程を追加する請求項1〜8のいずれか1項に記載の方法。
- グリセリン、グリセロール、又は、多価アルコールであるプロパン−1,2,3−トリオール(又は1,2,3−プロパントリオール)の任意の誘導体若しくは任意の形態から選択される天然ポリオールに上記沈殿物を溶解する請求項9に記載の方法。
- 事前に、光合成微細藻類バイオマスのフィコビリタンパク質を富化する工程を含む請求項1に記載の方法。
- 上記フィコビリタンパク質を富化する事前工程は、増殖が阻害されたバイオマスにおいてフィコビリタンパク質合成を誘導することを含む、請求項11に記載の方法。
- a.バイオマスの増殖を阻害する工程、
b.フィコビリタンパク質の合成を誘導する工程、
c.サリチル酸で沈殿させる工程、
d.工程cで得られた沈殿物を溶解する工程
を含む請求項1、11又は12のいずれか1項に記載の方法。 - 上記フィコビリタンパク質の水性抽出物は、アルスロスピラ・プラテンシス(Arthrospira platensis)である光合成微細藻類から得られる、請求項1〜13のいずれか1項に記載の方法。
