また、フィコシアニン調製の原料として、生の藍藻類を使用することもできるし、乾燥処理した藍藻類を使用してもよい。藍藻類の乾燥品は、生の藍藻類を常法に従い乾燥品としてもよく、市販の乾燥品を使用することもできる。
さらに好ましい実施態様としては、C-フィコシアニンとアロフィコシアニンとを含有する美肌用飲食品が挙げられる。例えば、スピルリナ属の藍藻類から抽出することにより得られるC-フィコシアニンとアロフィコシアニンからなるフィコシアニンの混合物を美肌用飲食品に含有させることができる。
フィコシアニンを抽出する方法としては、特に制限は無く、一般に知られている方法を用いることができる。
抽出方法の好ましい実施態様としては、例えば、特開2006-230272号公報に記載の抽出方法を挙げることができる。具体的には、下記抽出方法(i)で記載する抽出方法が挙げられる。係る抽出方法(i)により、高純度であざやかな色調のフィコシアニンを得ることができる。
抽出方法(i)は、
藍藻類中のフィコシアニンを水懸濁液中に抽出させた抽出液を得る第一工程と、
該抽出液中でカルシウム塩とリン酸塩とを反応させてリン酸カルシウムを生成させると共に該リン酸カルシウムにフィコシアニンの夾雑物を吸着させ吸着物を得る第二工程と、
該抽出液から藍藻類の残渣及び吸着物を除去する第三工程と、を有する。
<抽出方法(ii)>
抽出方法(ii)は、
藍藻類中のフィコシアニンを水懸濁液中に抽出させた抽出液を得る第一工程と、
該抽出液中でカルシウム塩とリン酸塩とを反応させてリン酸カルシウムを生成させると共に該リン酸カルシウムにフィコシアニンの夾雑物を吸着させ吸着物を得る第二工程と、
該抽出液から藍藻類の残渣及び吸着物を除去する第三工程と、
第三工程より前に、抽出液にキレート剤を含有させる工程と、を有する。
特にスピルリナ属の藍藻類に対して、上記抽出方法(i)又は(ii)を用いることにより、C-フィコシアニンとアロフィコシアニンとの混合比が良好な品質のよいフィコシアニンを抽出することができる。
尚、上記抽出方法において、抽出条件を適宜選択することにより、C-フィコシアニンとアロフィコシアニンとの混合比を所望の範囲とするよう調整するとよい。
あるいは、アロフィコシアニンを含有し、C-フィコシアニンとアロフィコシアニンとの混合物を飲食品に含有させてもよい。
C-フィコシアニンとアロフィコシアニンとの混合比は、例えば、質量比で3~9.5:0.5~7が好ましく、6~9.5:0.5~4がより好ましく、7~8:2~3がさらに好ましい。
フィコシアニンにより、肌の保湿機能や弾力機能などを高めることができ、肌を美しくする効果(美肌効果)を奏する。
ここで、肌を美しくする具体例としては、例えば、水分蒸散量(TEWL)の増加を抑制したり、角質水分量を増加したり、皮膚粘弾性を増加したりすることなどが挙げられる。また、シワを改善したり、肌のキメをよくしたり、シミを改善したりするなども挙げられる。さらに、肌の乾燥感を防ぐ、肌のはりやつやを向上させる、あるいは肌のしっとり感を向上させるといったことも挙げられる。
このように、本発明に係るフィコシアニンは、肌の保湿機能や弾力機能を高めたり、皮膚の粘弾性を向上させることで、肌を美しくすることができる。したがって、本発明のフィコシアニンを含有する飲食品は、経口摂取によって美肌効果を満足させる飲食品として有効に使用することができる。
また、本発明のフィコシアニンを含有する飲食品は、肌の保湿機能や弾力機能が不調である場合にこれを改善して通常の状態に戻す効果のみならず、肌の保湿機能や弾力機能を通常の状態からさらに優れた状態にする効果をも有する。すなわち、本発明の美肌用飲食品は、肌の保湿機能や弾力機能修復用飲食品としてのみならず、肌の保湿機能や弾力機能亢進用飲食品としても用いることができる。またさらに、本発明の美肌用飲食品は、肌の保湿機能や弾力機能低下を予防する効果も奏し得る。つまり、肌の保湿機能や弾力機能が低下はしていないものの、低下する可能性がある場合に予め摂取しておくことで肌の保湿機能や弾力機能の低下を抑制することができる。
例えば、健康食品、機能性表示食品、特別用途食品、栄養補助食品、サプリメント又は特定保健用食品等に使用することができる。これらの食品の一部は機能表示が許可された食品であるため、一般の食品と区別することができる。
本発明の美肌用飲食品は、美肌を目的にした飲食品(特に肌の保湿機能や弾力機能向上を目的にした飲食品)として、係る飲食品の製品に、「美肌を維持する」、「肌の潤いを保つ」、「肌の水分を保持する」、「肌のバリア性向上」、「肌の弾力性を維持する」、「肌の乾燥防止」、「シワを防止する」、「シミを防止する」、「シワを改善する」、「シミを改善する」、「肌のはり低下を防止する」、「肌のつや低下を防止する」、「肌のはりを改善する」、「肌のつやを改善する」等の表示が付されていてもよい。
賦形剤としては、でんぷん及びその誘導体(デキストリン、カルボキシメチルスターチ等)、セルロース及びその誘導体(メチルセルロース、ヒドロキシプロピルメチルセルロース等)、糖類(乳糖、白糖、ブドウ糖、トレハロース等)等、クエン酸または同塩類、リンゴ酸または同塩類、エチレンジアミン四酢酸または同塩類が挙げられる。
結合剤としては、でんぷん及びその誘導体(アルファー化デンプン、デキストリン等)、セルロース及びその誘導体(エチルセルロース、カルボキシメチルセルロースナトリウム、ヒドロキシプロピルメチルセルロース等)、アラビアゴム、トラガント、ゼラチン、糖類(ブドウ糖、白糖等)、エタノール等が挙げられる。
滑沢剤としては、ステアリン酸、ステアリン酸カルシウム、ステアリン酸マグネシウム、タルク、酸化チタン、リン酸水素カルシウム、乾燥水酸化アルミニウムゲル、ショ糖脂肪酸エステル、食用油脂等が挙げられる。
保存剤としては、パラオキシ安息香酸エステル類、亜硫酸塩類(亜硫酸ナトリウム、ピロ亜硫酸ナトリウム等)、リン酸塩類(リン酸ナトリウム、ポリリン酸カルシウム、ポリリン酸ナトリウム、メタリン酸ナトリウム等)、デヒドロ酢酸、デヒドロ酢酸ナトリウム、ソルビン酸グリセリン、糖類等が挙げられる。
酸化防止剤としては、亜硫酸塩類(亜硫酸ナトリウム、亜硫酸水素ナトリウム等)、エリソルビン酸、L-アスコルビン酸、システイン、チオグリセロール、ブチルヒドロキシアニゾール、ジブチルヒドロキシトルエン、没食子酸プロピル、アスコルビン酸パルミテート、dl-α-トコフェロール等が挙げられる。
等張化剤としては、塩化ナトリウム、硝酸ナトリウム、硝酸カリウム、デキストリン、グリセリン、ブドウ糖等が挙げられる。
コーティング剤としては、セルロース誘導体(ヒドロキシプロピルセルロース、酢酸フタル酸セルロース、ヒドロキシプロピルメチルセルロースフタレート等)、セラック、ポリビニルピロリドン、ポリビニルピリジン類(ポリ-2-ビニルピリジン、ポリ-2-ビニル-5-エチルピリジン等)、ポリビニルアセチルジエチルアミノアセテート、ポリビニルアルコールフタレート、メタアクリレート・メタアクリル酸共重合体等が挙げられる。
矯味剤としては、糖類(ブドウ糖、白糖、乳糖等)、サッカリンナトリウム、糖アルコール類等が挙げられる。
溶解補助剤としては、エチレンジアミン、ニコチン酸アミド、サッカリンナトリウム、クエン酸、クエン酸塩類、安息香酸ナトリウム、ポリビニルピロリドン、ポリソルベート類、ソルビタン脂肪酸エステル類、グリセリン、ポリプレングリコール、ベンジルアルコール等が挙げられる。
基剤としては、脂肪類(豚脂等)、植物油(オリーブ油、ゴマ油等)、動物油、ラノリン酸、ワセリン、パラフィン、樹脂、ベントナイト、グリセリン、グリコール油、等が挙げられる。
分散剤として、アラビアゴム、トラガント、セルロース誘導体(メチルセルロース等)、アルギン酸ナトリウム、ポリソルベート類、ソルビタン脂肪酸エステル類等が挙げられる。
安定化剤としては、亜硫酸塩類(亜硫酸水素ナトリウム等)、窒素、二酸化炭素等が挙げられる。
例えば、飲料としては、水、清涼飲料水、果汁飲料、乳飲料、アルコール飲料、ノンアルコール飲料、スポーツドリンク、栄養ドリンク等が挙げられる。
食品としては、パン類、麺類、米類、豆腐、乳製品、醤油、味噌、菓子類等が挙げられる。
このような各飲食品は、常法によって調製することができる。
種々の形態の飲食品を調製するには、本発明に係るフィコシアニンを任意の食品材料、もしくは他の有効成分、又は食品に許容される添加物(例えば溶剤、軟化剤、油、乳化剤、防腐剤、酸味料、甘味料、苦味料、香味剤、pH調整剤、界面活性剤、安定剤、着色剤、紫外線吸収剤、酸化防止剤、保湿剤、増粘剤、固着剤、分散剤、流動性改善剤、発泡剤、湿潤剤、香科、調味料、風味調整剤、等)等と適宜組み合わせて、常法に従って調製することができる。
上記他の有効成分(生物活性成分)としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、本発明に係るフィコシアニンと共通する保湿性を目的とする保湿性成分であっても、肌の美白を目的とする美白性成分などであっても構わない。例えば、アミノ酸、糖、グリセリン等の保湿性成分や、ビタミンA、B、C(アスコルビン酸)、D、E等のビタミン類、コラーゲン、コラーゲンペプチド、ヒアルロン酸、ヒアルロン酸塩、セラミド、プラセンタ、アスタキサンチン等の美白性成分などを本発明の美肌用飲食品に含有させることができる。
また例えば、食品におけるフィコシアニンの含有量は、その全質量中、該フィコシアニンの固形分換算で、0.008質量%以上であると好ましく、0.06質量%以上であるとより好ましい。また、食品におけるフィコシアニンの含有量は、その全質量中、該フィコシアニンの固形分換算で、99質量%以下であると好ましく、70質量%以下であるとより好ましい。
また特に、美肌用飲食品の形態が錠剤である場合は、例えば、錠剤におけるフィコシアニンの含有量は、その全質量中、該フィコシアニンの固形分換算で、2.5質量%以上であると好ましく、20質量%以上であるとより好ましい。また、錠剤におけるフィコシアニンの含有量は、その全質量中、該フィコシアニンの固形分換算で、98質量%以下であると好ましく、70質量%以下であるとより好ましい。
尚、本発明において、フィコシアニンの含有量とは、飲食品中にC-フィコシアニン、R-フィコシアニン、及びアロフィコシアニンから選ばれる2種類以上のフィコシアニンが含有されている場合、これらフィコシアニンの合計量とする。
尚、本発明において、フィコシアニンの摂取量とは、飲食品中にC-フィコシアニン、R-フィコシアニン、及びアロフィコシアニンから選ばれる2種類以上のフィコシアニンが含有されている場合、これらフィコシアニンの合計量とする。
本発明の美肌用飲食品が、C-フィコシアニンとアロフィコシアニンとを含有する場合には、例えば、C-フィコシアニンの摂取量は、成人1人に対して1日当たり、19mg以上が好ましく、225mg以上がより好ましく、また、1,000mg以下が好ましく、500mg以下がより好ましい。また、アロフィコシアニンの摂取量は、成人1人に対して1日当たり、6mg以上が好ましく、75mg以上がより好ましく、また、333mg以下が好ましく、167mg以下がより好ましい。
<フィコシアニンの抽出>
1%塩化カルシウム(無水)溶液1300Lに屋外培養槽で生産したスピルリナ乾燥藻体(噴霧乾燥品)65kgを加え、15分間の攪拌により均一懸濁液とした後、20℃15時間、静置条件下で藍藻類中のフィコシアニンを溶液中に抽出し抽出液を得た。
この抽出液にリン酸二水素ナトリウム32kgを添加し、0.5時間攪拌した後、20℃、静置下で2.5時間反応させ、リン酸カルシウムを生成させると共に該リン酸カルシウムにフィコシアニンの夾雑物を吸着させて吸着物を得た。この後抽出液を遠心分離機に導き、重力加速度が10,000Gで、15分間の遠心分離を行い、藍藻類の残渣及び吸着物を抽出液から除去した。得られたフィコシアニンの抽出液は、分画分子量10,000の分離膜を使用した限外濾過により低分子成分及び塩類を除去した後、トレハロース、クエン酸三ナトリウムを加えて混合し、噴霧乾燥を行い、フィコシアニン色素乾燥物15kgを得た。これを、フィコシアニン1とする。尚、フィコシアニン1の色素粉末100質量%中、フィコシアニン含量は約30質量%(C-フィコシアニン約22質量%、アロフィコシアニン約8質量%であった。
フィコシアニンの定量は、Journal of AOAC International Vol.91、No.3、2008、p524~529に従って、100mMリン酸緩衝液(pH6.0)に試料を溶解し、620nm及び650nmの吸光度を測定した。更に以下の計算式に従ってC-フィコシアニン(CPC)及びアロフィコシアニン(APC)量を算出した。
CPC(mg/mL)=0.162×OD620-0.098×OD650
APC(mg/mL)=0.180×OD650-0.042×OD620
上記フィコシアニン1を用いて、下記表1に従い、被験食(フィコシアニン配合錠剤)を調製した。また、表1に従い、対照食(フィコシアニン非配合錠剤(プラセボ))を調製した。なお、対照食については、フィコシアニン成分をセルロースに置き換え、色を被験食に合わせるために合成着色料(青色2号)を添加し、調製した。
<被験者及び摂取方法>
20歳以上65歳未満の健常女性93名を2グループ(被験食摂取群47名、及び対照食摂取群46名)に分け、プラセボを対照としたランダム化二重盲検並行群間比較試験にて、8週間錠剤(一日一回5粒)を継続して経口摂取させた。
被験者のグループ割付けは以下の方法にて実施した。即ち、SAS(登録商標)9.4(SAS Institute Japan株式会社製)を用いて、発生させた乱数を各被験者に割り当て、乱数の若い順に2群に振り分ける。割付因子(年齢、スクリーニング検査時の水分蒸散量、スクリーニング検査時の角質水分量)において群間差がなくなるまで振り分け作業を行った。そのようにして振り分けた、被験食摂取群の被験者の平均年齢は、45.0歳であった。対照食摂取群の被験者の平均年齢は、45.4歳であった。
摂取開始時検査日、摂取開始4週間後検査日、及び摂取開始8週間後検査日に、経皮水分蒸散量測定計(Tewameter TM300、Courage+ Khazaka electronic GmbH社製)を用いて水分蒸散量、角層水分測定計(Corneometer CM825、Courage+ Khazaka electronic GmbH社製)を用いて角質水分量を測定した。
測定部位は、左目尻横1cmとした。
測定室の温度は21±1℃、湿度は50±10%とし、15分間以上環境に馴化させた後、測定を実施した。
水分蒸散量は、10回目以上測定し、連続する5回の測定データの標準偏差が0.2g/h/m2未満になった時点の5回の測定データの平均値を測定値として採択した。
角質水分量は、5回測定し中央値を測定値として採択した。
水分蒸散量(TEWL)の測定結果を下記表2及び図1に示す。表2及び図1は、摂取開始時検査日における水分蒸散量から、摂取経過後の検査日における水分蒸散量の変化量を示す。表2及び図1中の*はp<0.05を示す。
尚、表2及び図1は、摂取開始時検査日の水分蒸散量が、人の水分蒸散量の目安として一般的に知られている10g/m2/hr以上20g/m2/hr未満であった被験者に対して測定した結果を示す。
表2及び図1で示されるように、被験食摂取群、つまりフィコシアニンを摂取した群は、対照食摂取群(プラセボ摂取群)に比べ、水分蒸散量を抑制する効果が認められた。
この水分蒸散量の抑制効果は、摂取開始4週間後から摂取開始8週間後にかけて特に顕著に表れる。そこで、水分蒸散量の抑制効果をより発揮させるためには、フィコシアニンを含有する錠剤を4週間以上、継続して服用することがより好ましい。
また、被験食を摂取し続けることで、摂取開始4週間後から摂取開始8週間後にかけて肌の保湿性が顕著に向上されることは、角質水分量の結果においても水分蒸散量の結果と同様なことがいえる。
下記表3に、被験者における摂取開始4週間後の角質水分量から摂取開始8週間後の角質水分量の変化量の結果を示す。
<被験者及び摂取方法>
被験者及び摂取方法は、実施例2と同様である。
<<レプリカ法による評価>>
摂取開始時検査日、摂取開始4週間後検査日、及び摂取開始8週間後検査日に、レプリカ法によるシワの評価を実施した。評価部位は、左目尻とした。
被験者の評価部位に樹脂を当ててレプリカを作製し、機器分析によりシワを三次元解析した。
摂取開始時検査日、摂取開始4週間後検査日、及び摂取開始8週間後検査日に、コードレスデジタルマイクロスコープDG-3X(スカラ株式会社)により被験者のシワ画像を撮影した。その撮影した画像を、下記基準をもとに皮膚科医に判定してもらうことで、シワの評価を行った。評価部位は、左目尻とした。
シワの判定は、日本香粧品学会の新規効能取得のための抗シワ製品評価ガイドライン(Vol.30、No.4、pp316-332(2006))に従い、下記8段階の基準でおこなった。
[シワの判定基準]
0 シワはない
1 不透明な浅いシワがわずかに認められる
2 明瞭な浅いシワがわずかに認められる
3 明瞭な浅いシワが認められる
4 明瞭な浅いシワの中に、やや深いシワがわずかに認められる
5 やや深いシワが認められる
6 明瞭な深いシワが認められる
7 著しく深いシワが認められる
<<レプリカ法による結果>>
レプリカ法による肌のシワの結果を下記表4に示す。
表4で示されるように、被験食摂取群、つまりフィコシアニンを摂取した群は、対照食摂取群(プラセボ摂取群)に比べ、シワを改善する効果が認められた。
マイクロスコープ撮影画像による肌のシワの結果では、すでに摂取開始4週間後検査日において、シワの改善効果が確認できた。
マイクロスコープ撮影画像による摂取開始4週間後検査日におけるシワの判定結果を、下記表5に示す。
表5で示されるように、被験食摂取群、つまりフィコシアニンを摂取した群は、対照食摂取群(プラセボ摂取群)に比べ、シワを改善する効果が認められた。
<被験者及び摂取方法>
被験者及び摂取方法は、実施例2と同様である。
<<マイクロスコープ撮影画像による評価>>
摂取開始時検査日、摂取開始4週間後検査日、及び摂取開始8週間後検査日に、コードレスデジタルマイクロスコープDG-3X(スカラ株式会社)により被験者のシミ画像を撮影した。その撮影した画像を、下記基準をもとに皮膚科医に判定してもらうことで、シミの評価を行った。評価部位は、左目尻とした。
シミの判定は、下記5段階の基準でおこなった。

マイクロスコープ撮影画像によるシミの判定結果を、下記表6に示す。
尚、表6は、被験者のうちシミの自覚症状のある被験者を対象としたため、45歳以上の被験者に対して測定した結果を示す。
<被験者及び摂取方法>
被験者及び摂取方法は、実施例2と同様である。
<<VAS(Visual Analogue Scale)による評価>>
摂取開始時検査日、摂取開始4週間後検査日、及び摂取開始8週間後検査日に、VAS法を用いて、被験者が「肌のはり」、「肌のつや」、「肌のしっとり感」の各項目について評価した。
具体的には、「肌のはり」、「肌のつや」、「肌のしっとり感」の各項目について、被験者が、図2で示すような両端に最良と最悪を記した黒線上に、肌の状態がどの程度の感覚かを記入してもらい、左側から被験者が記入した箇所までの長さをmm単位で、測定者が測定し、100-「測定値」を計測値として評価した。
<<VASによる結果>>
VASによる「肌のはり」、「肌のつや」、「肌のしっとり感」についての被験者の評価結果を下記表7及び図3~図5に示す。表7及び図3~図5は、摂取開始時検査日におけるVASの計測値から、摂取経過後の検査日におけるVAS計測値の変化量を示す。表7及び図3~図5中の**はp<0.01を、*はp<0.05を、†はp<0.1を示す。
表7で示されるように、被験食摂取群、つまりフィコシアニンを摂取した群は、対照食摂取群(プラセボ摂取群)に比べ、肌の状態(はり、つや、しっとり感)を改善させる効果が認められた。
したがって、本発明のフィコシアニンを含有する飲食品は、経口摂取によって美肌効果を満足させる飲食品として有効に使用することができる。

- フィコシアニンを有効成分として含有する美肌用飲食品。
- フィコシアニンが藍藻類由来のフィコシアニンである、請求項1に記載の美肌用飲食品。
- 藍藻類がスピルリナ属の藍藻類である、請求項2に記載の美肌用飲食品。
- フィコシアニンが、C-フィコシアニンを含有する、請求項1から3のいずれか一項に記載の美肌用飲食品。
- フィコシアニンが、C-フィコシアニンとアロフィコシアニンとを含有する、請求項4に記載の美肌用飲食品。
- 美肌用飲食品が肌の保湿性向上用飲食品である、請求項1から5のいずれか一項に記載の美肌用飲食品。
- 美肌用飲食品が肌の弾力性向上用飲食品である、請求項1から5のいずれか一項に記載の美肌用飲食品。
- 美肌用飲食品が肌のシワ改善用飲食品である、請求項1から5のいずれか一項に記載の美肌用飲食品。
- 美肌用飲食品が肌のシミ改善用飲食品である、請求項1から5のいずれか一項に記載の美肌用飲食品。
