Description
分子量1.2MDaのヒアルロン酸ナトリウム(Hi−Aqua(登録商標)、(株)ジンウバイオ)を恒温恒湿器を用いて湿潤処理した。すなわち、相対湿度60%の条件下でヒアルロン酸ナトリウム10gを基準に1〜3時間放置してヒアルロン酸ナトリウム内の水分含有量を5〜30%に調整した。参考として、湿潤処理前のHA内の水分含有量は2%程度であった。
示差走査熱量計(DSC6100、Seiko、Japan)を用いて、30〜250℃の温度範囲、10℃/minの昇温速度条件で湿潤処理前のヒアルロン酸ナトリウムと実施例1で湿潤処理したヒアルロン酸ナトリウムの吸熱ピークをそれぞれ測定した。DSC測定の結果、湿潤処理が施されていないヒアルロン酸ナトリウム粉末の場合は、溶融に起因した吸熱ピークが全く感知されなかったが、これに対し、水分含有量が5%以上に調整されたヒアルロン酸ナトリウムの場合は、温度区間150〜200℃付近で溶融に起因した吸熱ピークが強く感知されることを確認することができた。よって、上記温度区間でヒアルロン酸ナトリウムの溶融紡糸が可能であることが分かった。
実施例1で湿潤処理したヒアルロン酸ナトリウムを溶融紡糸装置(Melt Spinning Apparatus)の溶融貯蔵部に入れ、これを150〜200℃で窒素圧処理して紡糸(spinning)させた後、常温で一般乾燥させてヒアルロン酸ナトリウムファイバー(HAFiber)を製造した。
実施例2と同様の方法で紡糸(spinning)させて製造したHAファイバー(Fiber)を50%エタノール、70%エタノール、最終95%エタノールに順次浸漬させて表面を硬化させた後、常温で一般乾燥させてヒアルロン酸ナトリウムファイバー(HA Fiber)を製造した。
実施例2と同様の方法で紡糸(spinning)させて製造したHAファイバー(Fiber)を95%エタノールに直ちに浸漬させて表面を硬化させた後、常温で一般乾燥させてヒアルロン酸ナトリウムファイバー(HA Fiber)を製造した。
実施例2と同様の方法で紡糸(spinning)させて製造したHAファイバー(Fiber)を50%エタノールに直ちに浸漬させて表面を硬化させた後、常温で一般乾燥させてヒアルロン酸ナトリウムファイバー(HA Fiber)を製造した。
実施例2〜5で得たヒアルロン酸ナトリウムファイバー(HA Fiber)を37℃、生理食塩水に浸漬させ、時間による膨潤度を測定した。その結果を表1に示す。
Claims (10)
- 下記の工程を含む、溶融紡糸によるヒアルロン酸塩ファイバーの製造方法。
(a)重量平均分子量500〜3,000kDaのヒアルロン酸塩の水分含有量を5〜20%に調節する工程、
(b)水分含有量が調節されたヒアルロン酸塩を溶融紡糸装置に入れ、150〜200℃で加熱した後、高圧紡糸してヒアルロン酸塩ファイバーを製造する工程、および
(c)ヒアルロン酸塩ファイバーをエタノール水溶液に浸漬させて表面を硬化させる工程。 - 前記ヒアルロン酸塩のpHは6〜8であることを特徴とする、請求項1に記載のヒアルロン酸塩ファイバーの製造方法。
- 前記表面を硬化させる工程は、エタノール水溶液に1回浸漬させるか、或いはエタノール水溶液のエタノール濃度を順次増加させながら2〜5回浸漬させることを特徴とする、請求項1に記載のヒアルロン酸塩ファイバーの製造方法。
- 前記エタノール水溶液の濃度は30〜99体積%であることを特徴とする、請求項1に記載のヒアルロン酸塩ファイバーの製造方法。
- 重量平均分子量500〜3,000kDaのヒアルロン酸塩が100%の純度で含有されたことを特徴とするヒアルロン酸塩ファイバー。
- 重量平均分子量500〜3,000kDaのヒアルロン酸塩が含有されており、37℃の食塩水に24時間浸漬したときは、下記式による膨潤度が141〜148%であり、ファイバーの形態が維持されることを特徴とするヒアルロン酸塩ファイバー。
膨潤度(%)=(一定時間経過後のHAファイバーの重量/初期ファイバーの重量)×100。 - 請求項5または6に記載のヒアルロン酸塩ファイバーを含む手術用縫合糸。
- 請求項5または6に記載のヒアルロン酸塩ファイバーを含む整形フィラー。
- 請求項5または6に記載のヒアルロン酸塩ファイバーを含むリフティング用糸。
- 請求項5または6に記載のヒアルロン酸塩ファイバーを含む組織工学用スキャフォールド。
