2.1.市販のmAb
一次ウサギ抗ヒトビメンチン(クローンEPR3776)、ウサギ抗ヒトPAD2(クローンpab0197)、ウサギ抗ヒトシトルリン(クローンab6464)を全て、Abcamから購入した。一次ウサギ抗ヒトPAD−4(クローンpab0199)をCovalabから入手し、抗ヒトHLA−DR PE−Cy7コンジュゲート化抗体(クローンL243)をeBioscienceから入手した。抗CD25抗体(クローンPC61)、抗IFNγ抗体(クローンXMG1.2)、抗IL−17(クローン17F3)抗体、および抗CD4(クローンGK1.5)抗体をBioXcellから購入した。抗CTLA4抗体を、HB304ハイブリドーマ細胞培養上清(ATCC、USA)からセファロースプロテインGアフィニティークロマトグラフィーにより精製した。抗HLA−DR抗体(クローンL243)を、HB−55ハイブリドーマ細胞(ATCC、USA)培養上清からセファロースプロテインGアフィニティークロマトグラフィーにより精製した。ウサギmAb抗HMGB1(クローンD3E5)をCell Signaling Technologyから購入した。
機能的クラスII DR4(DRB1*0401;T2 DR4)またはDR1(DRB1*0101;T2DR1)を安定的にトランスフェクションされたT細胞/B細胞ハイブリッド細胞株T2[80]は、記載されており[82、83]、親切にも、Janice Blum博士およびLawrence Stern教授によって提供された。マウスメラノーマB16F1およびB16F10細胞株を、ATCCから入手した。全ての細胞株を、他に記述がない限り、10%FCS、L−グルタミン(2mM)を補充し、かつ重炭酸ナトリウム緩衝のRPMI培地1640(GIBCO/BRL)中で培養した。HB304ハイブリドーマ細胞を、Hybridoma SFM(Invitrogen、UK)において培養した。インビトロアッセイのために、シトルリン化エピトープを提示する腫瘍標的を作製するために、1%BSAを含有する0.1Mクエン酸(pH3.0)で、4℃、2分間、細胞を処理した。続いて、細胞を、培地で洗浄し、血清の非存在下、37℃で20時間、培養した。オートファジー阻害剤およびPAD阻害剤である、3−メチルアデニン(Sigma)およびCI−アミジン(Calbiochem)を、無血清培地中、20時間の培養の間、それぞれ、10mMおよび50μg/mlの最終濃度で加えた。
2.3.1.ペプチド
90%より高い純度のペプチドは、Peptide Synthetics(Fareham、UK)により合成された。0.2mgアリコートとして、−80℃で、凍結乾燥した状態で貯蔵した。使用する当日、それらを、10%ジメチルホルムアミド中、適切な濃度に再構成した。
抗体DNA構築物の作製は、他の所で詳細に記載されている[84]。簡単に述べれば、抗体DNA構築物を作製するために、標準分子生物学的技術を用いて、抗体の重鎖および軽鎖可変領域の相補性決定領域へエピトープを組み入れた。エピトープチロシナーゼ448〜462(DYSYLQDSDPDSFQD)、マウスおよびヒトgp100 44〜59エピトープ(WNRQLYPEWTEVQGSN/WNRQLYPEWTEAQRLD)由来のHLA−DR4拘束性ヘルパーCD4エピトープ、ならびにHepB核タンパク質128〜140(TPPAYRPPNAPIL)由来のI−Ab拘束性エピトープを、κ鎖のCDRL1と置きかえて挿入した。同様に、エピトープ MMP7 247〜262(SQDDIKGQKLYGKRS)、SSX2 33〜48(KEEWEKMKASEKIFY)、NYESO−1 87〜111(LLEFYLAMPFATPMEAELARRSLAQ)、および119〜143(PGVLLKEFTVSGNILTIRLTAADHR)由来のHLA−DR1拘束性ヘルパーCD4エピトープを、CDRL1へ組み入れ、同時に、エピトープトリオースリン酸イソメラーゼ23〜37(GELIGILNAAKVPAD)由来の修飾型エピトープをκ鎖のCDRL3と置きかえて挿入した。HLA−DR4拘束性CD4ビメンチン415〜433(LPNFSSLNLRETNLDSLPL)およびHLA−DR1拘束性エピトープ28〜49(RSYVTTSTRTYSLGSALRPSTS)をどちらもCDRH3へ組み入れた。DR1 CD4 NYESO−1 87〜111(LLEFYLAMPFATPMEAELARRSLAQ)はまた、抗体DNA二重発現ベクターのCDRH3部位へクローニングされた、拡張された配列83〜111内にコードされた。3つ全部のビメンチンエピトープを含有するヒトIgG1およびマウスIgG2a ImmunoBodyベクターも作製した。HLA−DR1 28〜49(RSYVTTSTRTYSLGSALRPSTS)、65〜77(SAVRLRSSVPGVR)、およびHLA−DR4拘束性ヒトCD4ビメンチン415〜433(LPNFSSLNLRETNLDSLPL)エピトープをそれぞれ、CDRH1、CDRH2、およびCDRH3部位へ組み入れた。
完全長NY−ESO−1およびマウスTap2、HHDII、ならびにマウスMHCクラスIIおよびマウスβ2ミクログロブリンの発現をノックダウンするためのsiRNAをコードする発現ベクターを、リポフェクタミントランスフェクション試薬を用いて、B16F10細胞に連続してトランスフェクションした。トランスフェクションされた細胞を、それぞれ、Zeocin(300μg/ml)、G418(500μg/ml)、およびブラストサイジン(4μg/ml)の存在下における成長により選択した。株を限界希釈によりクローニングし、発現をフローサイトメトリーにより確認した。
簡単に述べれば、関心対象となるペプチドを、増加性濃度における既定濃度のビオチン化HA306-318参照ペプチドと混合し、プレート結合化HLA DR0401に加えた。HLA DR0401に結合するビオチン化参照ペプチドの量を、ストレプトアビジン連結型酵素を用いて定量化し、続いて、発色基質で検出した。最大結合を、ビオチン化HA306-318ペプチド単独により達成された値として採用する。陽性対照として、非標識HA306-318ペプチドを用いて、ビオチン化バージョンと競合させた。
2.7.1.免疫化プロトコール
週齢8〜12週間の、C57BL/6マウス(Charles River、UK)、HLA−DR4マウス(Taconic、USA)、HHDIIマウス(Pasteur institute、France)、およびHHDII/DR1マウス(Pasteur institute、France)が用いられ、Nottingham Trent Universityのスタッフにより世話された。全ての作業は、Home Officeプロジェクトライセンスの下、実行された。ペプチドを、10%ジメチルホルムアミド中に1mg/mlまで溶解し、その後、異なるアジュバント:CpGおよびMPLA、各6μg/マウス(Invivogen、UK)、不完全フロイント50μl/マウス(Sigma、UK)、およびGMCSF 10μg/マウス(Peprotech、UK)で乳化した(一連の希釈物)。ペプチド(25μg/マウス)を尾の付け根に皮下注射した。DNA(1μg/マウス)を、製造会社の使用説明書を用いて1.0μm金粒子(BioRad、Hemel Hempstead、UK)上にコーティングし、遺伝子銃(BioRad)により皮内に投与した。Homspera(10nM/マウス)(PeptideSynthetics、UK)を、遺伝子銃免疫化と共に皮内注射した。マウスを、ペプチド免疫化について0日目か、またはペプチドおよび遺伝子銃免疫化について0日目、7日目、および14日目のいずれかにおいて免疫化した。脾臓を分析のために、他に記述がない限り、ペプチドについては14日目、ペプチドまたは遺伝子銃免疫化については20日目に取り出した。400μgの抗CD25抗体(PC61)を、免疫化の3日前に、食塩水において、腹腔内に投与した。200μgの抗CTLA−4抗体(UC10−4F10−11)を、遺伝子銃免疫化またはペプチド免疫化のいずれかで、食塩水において、7日目および14日目に腹腔内に投与した。
2.8.1.エクスビボElispotアッセイ
Elispotアッセイを、マウスIFNγ、IL−17、およびIL−10捕獲および検出試薬を製造会社の使用説明書(Mabtech、Sweden)により用いて、実施した。簡単に述べれば、抗IFNγ、IL−17、およびIL−10抗体で、96ウェルImmobilin−Pプレートのウェル上をコーティングした。(様々な濃度での)合成ペプチドおよびウェルあたり5×105個の脾細胞を、プレートのウェルに3連で加えた。腫瘍標的細胞を、関連した所に、5×104個/ウェルで、3連で加え、プレートを37℃で40時間、インキュベートした。インキュベーション後、捕獲されたIFNγ、IL−2、IL−17、およびIL−10を、ビオチン化抗IFNγ、IL−17、およびIL−10抗体によって検出し、ストレプトアビジンアルカリホスファターゼおよび発色基質で発色させた。スポットを分析し、自動プレートリーダー(Cellular Technologies Ltd)を用いてカウントした。機能的結合活性を、ペプチド濃度に対するエフェクター機能のグラフを用いて、50%最大エフェクター機能を媒介する濃度として計算した。
脾細胞を、抗体コーティング化磁気ビーズ(Miltenyi Biotech)を製造会社の使用説明書に従って用いて、CD4またはCD8細胞のポジティブ単離に供した。MHCクラスII遮断研究について、20μg/mlの抗HLA−DR(クローンL243)抗体をelispotアッセイに加えた。
脾細胞におけるエクスビボのIFNγ elispotアッセイからの上清を、40時間後、取り出し、elisaアッセイ(R&D systems)により製造会社の使用説明書に従って、グランザイムBについて評価した。
IL−10、IL−17、IFNу、TNFα、IL−2、およびIL−4に対するIgG抗体についての多重Luminexアッセイ(Invitrogen)のために3工程間接手順を用いた。標準、対照、および未知の血清を、50%アッセイ希釈緩衝液(Invitrogen)および50%無血清RPMI中に1:2に希釈した。標準の段階希釈物は各アッセイに含まれた。標準、対照、および未知の血清の各希釈物を、96ウェル濾過プレート(Millipore Multiscreen;Millipore Corporation、Bedford、Mass.)内で1セットの結合したLuminexミクロスフェアと混合し、振盪させながら室温で2時間、インキュベートした。ミクロスフェアを減圧濾過により収集し、PBSTで洗浄した。ビオチン化検出抗体を、振盪させながら室温で1時間、各ウェルへ加えた。ミクロスフェアを減圧濾過により収集し、PBSTで洗浄した。ストレプトアビジンコンジュゲート化R−フィコエリトリンを各ウェルに加えた。30分間のインキュベーションおよび洗浄工程後、ミクロスフェアをPBST中に再懸濁させ、XYプラットフォームを備えたBiorad BioPlex Luminexアナライザーにおいて読み取った。データ獲得および解析を、Luminexソフトウェア(BioPlex Systems)を用いて実施した。
Ficol−Hypaque(Sigma)勾配遠心分離により、新鮮に引き出されたヘパリン化血液からPBMCを単離した。2mlの最終体積における、5%プールされた自己ヒト血清、2mMグルタミン、20mM HEPES、およびペニシリン−ストレプトマイシン(1%)を含有するRPMI中、単一のペプチド(最終濃度10μg/ml)でPBMC(1.5×106細胞/ウェル)を刺激した。精製されたタンパク質誘導体、PPD(最終濃度10μg/ml)での刺激は、PBMCの増殖能力についての陽性対照としての役割を果たす。陰性対照として、PBMCを培地単独とインキュベートした。PBMCを、37℃で5%CO2の大気中、4日間、7日間、および11日間、培養した。これらの時点における増殖を評価するために、各培養物からの3連での100μlを96ウェルプレートの丸底ウェルへ等分し、3H−チミジンを加え(0.0185MBq/ウェル)、さらに8時間、37℃でインキュベートした。培養物を、unifilterプレート上に回収し、3H−チミジンの取り込みをβ−シンチレーションカウンティングにより決定した。エピトープ刺激された培養物の1分間あたりのカウントの平均(cpm)対刺激されていない培養物の平均の比率として刺激指数(SI)を計算することにより結果を評価した。増殖アッセイは、SI>2.5である場合、陽性とみなされた。
標的細胞を、10μg/mlのペプチド有り、または無しで、1.85MBq(51Cr)クロム酸ナトリウム(Amersham、Essex、UK)で1時間、標識した。インキュベーション後、それらをRPMI中、3回洗浄した。標的5×103個/96ウェルV底プレートのウェルを配置し、200μlの最終体積のRPMI、10% FCS(Sigma)、20mM HEPES緩衝液、2mM L−グルタミン、100ユニット/ml ペリシリン、および100μg/ml ストレプトマイシン中、異なる密度のエフェクター細胞と共インキュベートした。37℃での20時間後、50μlの上清を各ウェルから取り出し、Lumaplate(Packard、Rigaweg、the Netherlands)へ移した。プレートを、Topcountマイクロプレートシンチレーションカウンター(Packard)上で読み取った。パーセンテージ特異的溶解を、以下の式を用いて計算した:特異的溶解=100×[(実験的放出−自発的放出)/(最大放出−自発的放出)]
2.9.免疫組織化学的分析
組織アレイ切片をまず、キシレンで脱パラフィンし、段階的アルコールを通して再水和し、内因性ペルオキシダーゼ活性を遮断するために、0.3%過酸化水素を含有するメタノール中に20分間、浸漬した。抗原性を賦活するために、切片を、500mlのpH6.0クエン酸緩衝液中に浸漬し、マイクロ波の第6センス設定で20分間、加熱した。内因性アビジン/ビオチン結合を、アビジン/ビオチン遮断キット(Vector Labs)を用いて遮断した。一次抗体の非特異的結合を遮断するために、その後、全ての切片を、PBS中、100μlの1/5正常ウマ血清(NHS)で15分間、処理した。試験切片を、PBS中に希釈された100μlの一次抗体と、22℃で1時間、または4℃で一晩、インキュベートした。陽性対照組織は、1/1000希釈でのβ2−ミクログロブリン(PBS中;Dako)で染色された結腸直腸癌組織の切片全体を含んだ。一次抗体を陰性対照から取り除き、それをNHS中、インキュベートの状態にしておいた。PBSでの洗浄後、全ての切片を、NHS中1:100に希釈された100μlのビオチン化ヤギ抗マウス/ウサギ免疫グロブリン(Dako)と30分間、インキュベートした。切片をPBS中で再び洗浄し、100μlの前もって形成されたストレプトアビジン−ビオチン/HRP複合体(Dako)と室温(RT)で60分間、インキュベートした。その後、エピトープ発現の可視化を、DABを用いて達成した。最後に、切片を、ヘマトキシリン(Dako)で軽く対比染色し、アルコール中で脱水し、キシレン(GentaMedica、York、UK)中で透徹し、ジスチレン、可塑剤、およびキシレン(DPX;BDH)でマウントした。
抗血清を、胃癌TMAにおいて腫瘍結合についてスクリーニングした。患者研究および設計:研究集団は、University Hospital、Nottingham、UKにおける組織学的に証明された散発性原発性結腸直腸癌の待機的外科的切除を受けた一連の462人の継続患者を含んだ(表1)。これらの患者は、1994年1月1日〜2000年12月31日の間、処置された;この期間は、研究される予後マーカーの意味ある評価を可能にした。この時間枠の間に処置された全ての患者は、本研究への算入について適格とみなされた。腫瘍体積の50%より多くがムチンからなる場合、腫瘍を粘液性癌腫として分類した。
抗血清を、卵巣癌TMAにおいて腫瘍結合についてスクリーニングした。卵巣癌TMAは、2000年〜2007年の間にNottingham University Hospitalsで処置された原発性卵巣癌を有する362人の患者のコホートを表す。癌の病期分類を、International Federation of Obstetrics and Gynaecology(FIGO)判定基準を用いて実施した。この研究に含まれる全患者を、現在の標準化学療法レジメンに従って処置し、65人の患者(41.4%)において単一の薬剤カルボプラチン、または89人の患者(56.7%)において白金に基づいた併用化学療法のいずれかであり、3人の患者は化学療法を拒否した。白金抵抗性症例を、処置の間、初回白金化学療法において進行した患者、または処置後6カ月以内に再発した患者として定義した。全ての患者は手術を受けた;症例(n=69)の44%より多くが、最初の手術後、最適以下に減量した(残存腫瘍<1cm)とみなされた。患者を物理学的検査、コンピュータ断層撮影、およびCA−125レベルにより追跡調査した。これらの患者の腫瘍からのヘマトキシリンおよびエオシン染色切片は、臨床データおよび病理診断を知らされていない婦人科病理学者により再検査された。各腫瘍について、その型および分化の再検査もまた、SDによって行われた。各症例に関連した臨床データを、患者のメモから、または病院の電子記録(NotIS)を介して収集および記録した。そのような情報は以下を含んだ:診断時の患者の年齢、FIGOステージ、外科的腫瘍縮小の程度、ならびに化学療法の型、持続期間、および応答。アジュバント処置、疾患特異的生存率(DSS)、および全生存(OS)の詳細を、全ての患者について文書化した。生存期間は、手術日から、いかなる残っている生存者も打ち切られる2008年5月30日までで計算された。追跡期間中央値は36カ月間であった。本研究のために試料および関連データを収集することへの倫理的承認は、Nottinghamshire Local Research Ethics Committeeによって与えられた。
正常組織TMAは、38個の正常器官を表す59個のコアを含有した。各コアを、以下の試料の起源に従って分類した;非癌患者由来の正常組織、癌患者由来の正常組織であるが、その癌は非関連器官に関わり、正常組織がその癌に隣接している。組織型およびカテゴリーは表2に詳細が示されている。
自己および外来エピトープに対する野生型マウスにおけるCD4応答
腫瘍関連エピトープに対するT細胞応答は、寛容および胸腺内のT細胞除去のせいで弱く、または存在しない場合が多い。それにも関わらず、本発明者らは、様々な自己および外来CD4エピトープをそれらのヘルパー応答を刺激する能力についてスクリーニングした。以前の研究により抗体DNA構築物が最も強い免疫応答を与えたことが示されているため[84]、様々なCD4外来および自己エピトープを、別々の構築物中へ組み入れ、野生型マウスにおいてスクリーニングした。表3は、全てのエピトープの配列、および必要に応じて、それらのマウス相同体を列挙している。
非相同残基はイタリック体で示されている
図5は、外来CD4エピトープの大部分に対して良いCD4応答であるが、自己エピトープに対して良い応答ではないことを示す。CD4 I−Abヘルパーエピトープである、B型肝炎核タンパク質128〜140は、C57Blマウスにおいて50〜1,000/脾細胞100万個(平均382/脾細胞100万個)のELISPOTによる応答を示した。癌精巣エピトープNYESO−1およびSSX2は、マウスにおいて外来エピトープであり、DR1エピトープNYESO−1(87〜111)およびSSX2(34〜48)は、HLA−DR1トランスジェニックマウスにおいて良い応答を刺激した;NYESO−1(87〜111)について250〜900/脾細胞100万個(平均567/脾細胞100万個)であり、SSX2(34〜48)については500〜1200/脾細胞100万個(平均765/脾細胞100万個)であった。野生型と1アミノ酸、異なるだけであるHLA−DR1突然変異型TPIエピトープは、300〜1300/脾細胞100万個の応答を示す。ヒトgp100 HLA−DR4エピトープ44〜59は、相同性マウスエピトープからの1つのアミノ酸変化を有し、263〜1521/脾細胞100万個(平均745/脾細胞100万個)の応答を示す。対照的に、相同性マウス自己エピトープは、HLA−DR4トランスジェニックマウスにおいて応答を刺激することができなかった。ヒトHLA−DR4チロシナーゼエピトープは、相同性マウスエピトープから6つのアミノ酸変化を有し、405〜832/脾細胞100万個(平均555/脾細胞100万個)の応答を示す。HLA−DR1 MMP7 247〜262エピトープは、マウスとヒトとの間で4つのアミノ酸変化を有し、そのうちの1つは、コアMHC結合/TCR認識領域内にあると予測される(表3)。このエピトープは、HLA−DR1トランスジェニックマウスにおいてバックグラウンドより高い応答を刺激することができなかった。HLA−DR4ビメンチン415〜433エピトープは、ヒトとマウスとの間で2つのアミノ酸相違を有するが、コアMHC結合/TCR認識領域内にあるとは予測されていない(表3)。それは、野生型HLA−DR4トランスジェニックマウスにおいて応答を刺激することができなかった。対照的に、ビメンチン28〜49は、HLA−DR4トランスジェニックマウスにおいてマウスとヒトにおいて相同であり、200〜500/脾細胞100万個(平均390/脾細胞100万個)の応答を刺激する真の自己エピトープである。この応答は、興味をそそられ、本発明者らを、なぜこのエピトープに対して除去/寛容がないのかを説明してみたいと思わせた。
DNA免疫化は、シトルリン化vim 28、vim 415、MMP7、およびNY−ESO−1に対する応答を生じる
RA患者は、シトルリン化ビメンチンエピトープに対してT細胞応答を起こすことが示されている。APCはエピトープを恒常的にシトルリン化することができるので、抗体DNA構築物がシトルリン化され、これが応答を刺激するだろうことは起こり得た。したがって、HLA−DR4トランスジェニックマウスを、自己vim 28エピトープをコードする抗体−DNA構築物で免疫化した。これらのマウス由来の刺激されたT細胞を、インビトロで、シトルリン化vim 28ペプチドと非シトルリン化vim 28ペプチドの両方に対するIFNγ、IL−17、およびIL−2応答についてスクリーニングした。図6は、マウスが、ELISPOTによりアッセイされた場合、3つ全てのサイトカインの産生(平均:IFNγ 400、IL−17 120、およびIL−2 150/脾細胞100万個)により野生型ペプチドに対して応答したが、シトルリン化ペプチドに対する応答は、IFNγおよびIL−17について有意に高かったが、IL−2についてはそうではなかった(平均:IFNγ 1250/脾細胞100万個;p=0.0046、IL−17 250/脾細胞100万個;p=0.0392、IL−2 250/脾細胞100万個)ことを示している。同様の応答がvim 415エピトープに対して発生するかどうかをみるために、抗体DNA構築物を用いてマウスを免疫化した(図7)。vim 28〜49とは対照的に、かつ本発明者らの最初の観察と一致して、野生型vim 415〜433に対する応答はなかったが、そのシトルリン化ペプチドに対する強い応答があり、IL−17への応答は、IFNγ応答と同じくらい強かった(平均:IFNγ 400/脾細胞100万個;p=0.0067、IL−17 350/脾細胞100万個;p=0.002、およびIL−2 250/脾細胞100万個;p=0.0056)。これにより、抗体DNAが翻訳される時、それがシトルリン化されることが確認される。
ペプチド免疫化は、シトルリン化vim 28、vim 415、およびvim 65に対する応答を生じる。
自己エピトープに対するCD4応答はナイーブ応答か、メモリー応答か、またはTreg応答かの決定
マウスは、ヒトvim 415 citの単一免疫化に対して強力なIFNγおよびIL−17応答を起こし、それが、メモリー応答またはTreg応答をブーストしていることが示唆された。これが、IFNγ/IL−17応答へ変換されるだろう内在性Treg応答であるかどうかを決定するために、抗CD25 mAbを用いてマウスから内在性Tregを枯渇し、マウスをマウスvim 415 citで免疫化した。内在性Tregの枯渇は、応答の頻度または結合活性に影響を及ぼさず、内在性Tregが応答集団ではないことが示唆された(図17)。この応答がまた他のアジュバントで誘導されるかどうかを決定するために、マウスを、アラム、不完全フロイントアジュバント(IFA)、GMCSF、MPLA、TMX201、MPLA/TMX201、またはCpG/MPLAのいずれかにおいてvim 415〜433 citおよび28〜49 citで免疫化し、IFNγ、IL−17、またはIL−10の産生についてスクリーニングした(図18)。vim 415〜433 citおよび28〜49 citエピトープに対する強力なIFNγ/IL−17応答は、CpG/MPLA、GMCSF、およびTMX201アジュバントで誘導されたが、そのペプチドがアラムまたはIFAにおいて投与された場合、応答は見られなかった。IFAにおけるペプチドでの免疫化は、そのシトルリン化ペプチドに対して高頻度のIL−10応答を誘導した。
癌患者は自己ペプチドに応答する
9人のメラノーマ患者を、一連の自己ペプチドに対する彼らの応答についてスクリーニングした(図20)。8人の患者のうち5人は、4日目において(1人)、7日目において(1人)、または11日目より後(3人)において、vim 415 citに対する応答を示した。11人の患者のうちの6人は、10日目より後に、非修飾型vim 415に対して応答した。これらの患者の4人だけが、修飾型ペプチドと非修飾型ペプチドの両方に対して応答した。8人の患者のうちの2人は、11日目にvim 28に対する応答を示したが、5人の患者は、vim 28 citに対して応答した。非修飾型ペプチドに対して応答したその2人の患者はまた、修飾型ペプチドを認識した。8人の患者のうちの4人は、vim 65に対して応答し、8人のうちの3人はvim 65 citに対して応答した。これらの患者のうちの2人だけが、修飾型ペプチドと非修飾型ペプチドの両方に対して応答した。8人の患者はまた、シトルリン化NYESO−1 119〜143に対する応答を示した;これらの応答のうちの6つは、4〜7日目にピークに達し、強いまたはメモリー応答を示唆した。8人の患者は非修飾型NYESO−1 119〜143に対する応答を示した。これらの患者は、様々なHLA型を有する(表4)。1人だけが、HLA−DR4であり、そのことは、他のHLAハプロタイプがこれらのペプチドに応答できることを示唆した。
PAD酵素、ビメンチン、およびシトルリンの発現
シトルリン化は、PAD酵素、特にPAD2およびPAD4酵素によって実行される。これらは、高レベルのカルシウムを必要とし、通常、死細胞または瀕死の細胞において活性化される。したがって、健康な腫瘍細胞がシトルリン化タンパク質を発現するだろうとは可能性が低いように思われる。したがって、結腸直腸および卵巣腫瘍、ならびに正常組織を、ビメンチン、シトルリン化、ならびにPAD2およびPAD4酵素の発現について染色した。
ビメンチン、PAD2、PAD4、およびシトルリンの発現は、正常組織について表5に示されている。
卵巣腫瘍は、間葉系起源であり、したがって、ビメンチンを発現することが予想される。PAD4は、正常卵巣により弱く発現している。219個の卵巣腫瘍を、ビメンチン特異的mAbで染色した。腫瘍の9個/219個(4%)だけが染まることができず、さらなる16個/219個(7%)が弱く染まったが、194個/219個(89%)は強く染まった。カプラン・マイヤー生存分析により、ビメンチン発現と生存の相関はないことが示された。ビメンチンの発現と、ストレス関連タンパク質ULBP1(p=0.017)、PAD4(p=0.018)、およびCEA−CAM4(p=0.033)との間に弱い相関があった。
結腸直腸腫瘍は、上皮起源であり、それらが、上皮間葉転換を受けるだろうことがない限り、ビメンチンを発現するとは予想されない。PAD2およびPAD4の発現は正常結腸において見られた。
シトルリン化ペプチドに対する抗腫瘍応答
マウスとヒトのどちらも、シトルリン化自己ペプチドおよびDNAワクチンに対する応答を示す。しかしながら、これらのエピトープは、腫瘍においてシトルリン化されず、それゆえに、T細胞は抗腫瘍活性を生じないだろう。
異なる種間のビメンチンの相同性
ビメンチンは、ニワトリ、マウス、イヌ、ヒツジ、ウシ、ウマ、ブタ、およびヒトの間で高度に保存されている(図27)。ワクチンは、ヒトおよびマウスにおいてT細胞応答を、ならびにマウスにおいて抗腫瘍応答を誘導するため、類似した応答が他の種において見られるだろうと想定することができる。
他のHLAハプロタイプにより拘束されるビメンチン応答
HLA−DR4マウスは、ヒトvim 415 citペプチドの単一免疫化に対して強力なIFNγおよびIL−17応答を起こした。このまたは他のシトルリン化ビメンチンエピトープが他のハプロタイプにおいて免疫応答を誘導することができるかどうかを決定するために、ビメンチンの全スパンを網羅し、かつあらゆるアルギニンをシトルリン残基で置きかえて組み入れた、様々な20merペプチド(表8)を、HLA−DR1およびC57/BlマウスにおいてIFNγ/IL−17応答についてスクリーニングした(図28)。CpGおよびMPLAアジュバントと組み合わせて、3つのシトルリン化vimペプチドの組み合わせでマウスを免疫化し、その後、組み合わせにおけるそれぞれ個々のペプチドに対するIFNγおよびIL−17応答についてスクリーニングした。図28aは、HLA−DR1トランスジェニックマウスにおいて、シトルリン化vimペプチド9、10、14、15、および16は有意なIFNγ応答(200〜350スポット/脾細胞100万個、p<0.02)を示し、ペプチド10はIL−17応答(350スポット/脾細胞100万個)を示したことを示す。図28bは、C57Bl/6マウスにおいて、シトルリン化vimペプチド16、17、18、19、および20は有意なIFNγ応答(300〜500スポット/脾細胞100万個、p<0.02)を示し、ペプチド19および20は、IL−17応答(約400スポット/脾細胞100万個、p<0.05)を示したことを示す。表7は、これらのペプチドの配列、シトルリンアミノ酸の位置、およびビメンチンタンパク質内の位置を示す。
シトルリン化T細胞エピトープについてスクリーニングする方法
文献に記載されているいかなるシトルリン化タンパク質もT細胞の潜在的な標的であり得る。しかしながら、それは、まず、MHCクラスIおよび/またはクラスII MHC抗原上に提示される能力を有しなければならず、かつそれは、T細胞受容体によって認識されなければならない。抗原提示細胞は、恒常的に、オートファジーを起こし、それはこれらの二重膜オートファゴソーム内であり、そのオートファゴソームは十分な細胞内Ca2+を蓄積して、PAD酵素を活性化することができ、その酵素はエピトープをシトルリン化し、その後、そのエピトープはMHC抗原上に提示される。最後に、抗腫瘍標的であるために、腫瘍細胞もまた、オートファゴソーム内でシトルリン化を誘導し、MHC抗原上に同じ修飾型エピトープを提示しなければならない。したがって、なお抗腫瘍免疫を刺激することができるシトルリン化エピトープを同定するために、T細胞応答の誘導および腫瘍認識について標的タンパク質をスクリーニングすることが必要である。
ヒト末梢血は、例5において概要が示されているように、インビトロで刺激することができる。タンパク質全体に及ぶシトルリン化20merペプチドを、T細胞増殖についてスクリーニングすることができる。CD4およびCD8 T細胞のソーティングは、CD4およびCD8エピトープを同定することができ、HLA拘束性は、ドナーのHLAタイピングによって同定することができる。
シトルリン化サイトケラチン−8エピトープが免疫応答を誘導することができるかどうかを決定するために、サイトケラチン−8の全スパンを網羅し、かつ推定のコア結合領域内のアルギニンをシトルリン残基で置きかえて組み入れている、様々な20merペプチド(表8)を、HLA−DR1およびC57 BIマウスにおいてIFNγ/IL−17応答についてスクリーニングした(図29)。CpGおよびMPLAアジュバントと組み合わせて、3つのサイトケラチン8ペプチドの組み合わせでマウスを免疫化し、その後、組み合わせにおけるそれぞれ個々のペプチドに対するIFNγおよびIL−17応答についてスクリーニングした。図28aは、HLA−DR1トランスジェニックマウスにおいて、シトルリン化サイトケラチン8ペプチド1、2、3、13、16、および17は有意なIFNγ応答(200〜300スポット/脾細胞100万個、p<0.02)を示し、ペプチド1、2、3、13、および14はIL−17応答(250〜350スポット/脾細胞100万個;p<0.02)を示したことを示す。図29bは、C57Blマウスにおいて、サイトケラチン8ペプチドは応答を刺激しなかったことを示す。
ING4タンパク質は、それのNLS領域内の位置133においてPAD4によってシトルリン化される。これは、p53活性化に必須である、それのp53との会合を阻止する。シトルリン化ING4タンパク質は、迅速に分解され、それがMHCクラスII上に多量に発現され得ることを示唆している。ING4ペプチドAQKKLKLVRTSPEYGMPは、HLA−DR1トランスジェニックマウスにおいて少しの免疫応答も刺激することができなかったが、AQKKLKLVcitTSPEYGMPは、シトルリン化ペプチドに対するIFNγ/IL−17応答(平均200スポット/脾細胞100万個)を刺激したが、野生型ペプチドに対する応答を刺激しなかった。シトルリン化ペプチドに対する応答は、MHCクラスII遮断mabで遮断された(図31a)。これは、シトルリン化/腫瘍特異的CD4応答を刺激することができるタンパク質のさらなる例である。
本発明者らが例2において示しているように、抗原全体をコードするDNAで免疫化することは、結果として、シトルリン化ペプチドに対する応答を生じる。ここで、本発明者らが、抗原全体をコードするDNAで免疫化し、全ての可能なシトルリン化20merペプチドに対してスクリーニングした場合、HLA分子によって提示されたシトルリン化エピトープに対するT細胞応答だけが、免疫応答を刺激することを本発明者らは示す。シトルリン化ペプチドのパネルは表7に詳細を示している。図33aは、HLA−DR4トランスジェニックマウスにおけるビメンチンDNA免疫化から生じた応答を示し、図34bは、HLA−A2/DR1トランスジェニックマウスにおける応答を示す。HLA−DR4トランスジェニックマウスは、シトルリン化ビメンチン28〜49および415〜433ペプチドに特異的な高頻度の応答、加えて、ビメンチン19〜33、26〜44、および36〜54ペプチドに対するより低い頻度の応答を示す。HLA−A2/DR1トランスジェニックマウスは、シトルリン化ビメンチン65〜77ペプチドに特異的な高頻度応答を示す。これは、抗原全体をコードするDNAを用いて、シトルリン化されてT細胞応答を誘導することを例示し、さらなるシトルリン化T細胞エピトープについてスクリーニングするための優れた方法である。同様に、タンパク質は、高レベルのカルシウムの存在下でPAD酵素とインキュベートすることによりエクスビボでシトルリン化することができる。これらのタンパク質を、マウスを免疫化するために用いることができ、その後、全ての可能なシトルリン化20merペプチドに対してT細胞をスクリーニングする。
本発明者らは、既知のシトルリン化エピトープに対して応答を誘導することができることを示しているが、ここで、本発明者らはまた、ペプチドエピトープが、予測されるMHC結合スコアおよびコアMHC結合領域内のアルギニン残基の存在に基づいて選択し得ることを実証する。この例として、ペプチドを、SYFPEITHI予測アルゴリズム(www.syfpeithi.de)を用いてHLA−DR4への高いと予測される結合を有するBiP、HSP90、CXCL10、CXCL12、およびING4から選択し、その後、(IEDB予測アルゴリズム(www.iedb.org)を用いて決定される)コア結合領域内のアルギニンの存在を通してさらに限定した(表9)。全てのアルギニンをシトルリンへ変化させて含有するペプチドを試験した。
以前の研究により、ナイーブCD4細胞の、それらが刺激された時に存在するそれらのサイトカイン環境に依存して、異なるヘルパー表現型への分化を決定することは可能であることが示されている。対照的に、本発明者らは、ある特定のエピトープが、サイトカイン環境に関わらず、Tヘルパー細胞分化を決定することができることを図15において初めて示している。vim 415 citは、Th2アジュバント完全フロイントアジュバントの存在下で免疫化した場合でさえも、Th1/IL−17表現型を刺激した。これは、MHCペプチドとのCD4 T細胞受容体結合の強さがT細胞分化を決定することができることを示唆している。この関連において、本発明者らは、野生型vim 415が、Th1アジュバントCpG/MPLAの存在下でさえも、IL−10応答(図30;平均580スポット/脾細胞100万個、p=0.0249)を刺激することを示している。しかしながら、それは、有意なIFNγまたはIL−17応答を刺激することができなかった。

Claims (12)
