本発明の一般的な実施形態は、p40を認識するためのモノクローナル抗体、それらの調製のための方法、免疫組織化学における使用などを含み得る。実施形態において、抗p40抗体クローンBC28などの抗p40抗体クローンは、ヒトp40タンパク質アミノ酸1−17のサブセットに対応する1つ以上のペプチドを用いてBalb/Cマウスを免役することによって得ることができる。p40ペプチドは、腹腔内注射を介して、アジュバントとともに、BALB/cマウスに注射されてもよく、おそらく、約3週間の間隔で約5回である。p40に対する免疫反応は、組換えp40タンパク質への直接ELISAによって評価されてもよい。最高の力価を有するマウスは、細胞融合によるハイブリドーマの発生のために選択されてもよい。ヒト組織でのp40に対する最良の反応を実証するハイブリドーマクローンが選択されてもよく、BC28と名付けられてもよい。BC28クローンはアイソタイプについて試験されてもよく、マウスIgG1/kappaとして同定されてもよい。BC28抗体は、ハイブリドーマ細胞の大規模組織培養によって、およびBALB/cマウスにおける腹水によって、産生されてもよい。上清および抗体の腹水が収集されてもよく、抗体はプロテインAアフィニティーカラムによって精製されてもよい。BC28は、ELISA、ウェスタンブロッティング、およびさらにはヒト組織によって、ヒトp40タンパク質に対する特異的反応を実証してもよい。

p40タンパク質へのBC28の結合はウェスタンブロットによって実証され得る(図31、左のパネル、レーン2)。BC28のTAp63タンパク質への同様の結合の非存在もまた、ウェスタンブロットによって示され得る(図48および図98、左のパネル、レーン3)。逆に、抗p63抗体4A4は、p40タンパク質とTAp63タンパク質の両方を結合し得る(図48および図98、右のパネル、レーン2およびレーン3)。BC28のより高い特異性(p40の結合であって、TAp63ではない)は、その優れた特性に寄与する利点であり得る。
Claims (13)
- アメリカンタイプカルチャーコレクション(ATCC)にATCC特許寄託番号PTA−120163のもとで寄託されたハイブリドーマにより産生される単離された抗体、または前記単離された抗体の単離された抗原結合フラグメントであって、前記抗体またはその抗原結合フラグメントは、配列番号4に記載のアミノ酸配列を含むp40に特異的に結合する、単離された抗体またはその単離された抗原結合フラグメント。
- 請求項1に記載の単離された抗体またはその単離された抗原結合フラグメントと、少なくとも1つのさらなる単離された抗体またはその単離された抗原結合フラグメントとを含む組成物であって、少なくとも1つのさらなる単離された抗体またはその単離された抗原結合フラグメントは、DSG−3、CK4、CK5、ナプシンA、HMWCK、p504S、CK5/14、CK7/18、CK8/18、ウロプラキンII、ウロプラキンIIIおよびGATA−3からなる群より選択される抗原に特異的に結合する、組成物。
- 配列番号2からなる核酸配列によりコードされるアミノ酸配列を含む軽鎖可変領域と配列番号1からなる核酸配列によりコードされるアミノ酸配列を含む重鎖可変領域とを含む、請求項1に記載の単離された抗体またはその単離された抗原結合フラグメント。
- 前記単離された抗体またはその単離された抗原結合フラグメントと、前記少なくとも1つのさらなる単離された抗体またはその単離された抗原結合フラグメントとが、少なくとも2つの異なる種に由来する、請求項2に記載の組成物。
- 前記少なくとも2つの異なる種が、マウス、ウサギ、ヤギ、ウマ、ニワトリおよびヒトからなる群より選択される、請求項4に記載の組成物。
- 前記単離された抗体またはその単離された抗原結合フラグメント、および前記少なくとも1つのさらなる単離された抗体またはその単離された抗原結合フラグメントはそれぞれ、p40およびDSG−3およびCK5およびナプシンA;p40およびDSG−3およびCK5;p40およびナプシンA;p40およびHMWCKおよびp504s;p40およびp504sおよびCK5/14;p40およびCK5/14およびCK7/18;p40およびCK5/14およびCK8/18;p40およびウロプラキンIIおよびウロプラキンIII;ならびにp40およびウロプラキンIIおよびウロプラキンIIIおよびGATA−3からなる群より選択されるタンパク質に特異的に結合する、請求項2に記載の組成物。
- 前記単離された抗体またはその単離された抗原結合フラグメント、および前記少なくとも1つのさらなる単離された抗体またはその単離された抗原結合フラグメントはそれぞれ、p40およびDSG−3およびナプシンA;p40およびDSG−3;ならびにp40およびCK5からなる群より選択されるタンパク質に特異的に結合する、請求項2に記載の組成物。
- 請求項1に記載の単離された抗体またはその単離された抗原結合フラグメントと;p40抗原に結合したときの請求項1に記載の単離された抗体またはその単離された抗原結合フラグメントの抗体検出試薬とを含むキット。
- 標識と結合体化された、請求項1に記載の単離された抗体またはその単離された抗原結合フラグメント。
- 前記標識が、放射性元素、磁気粒子、ラジオアイソトープ、蛍光色素、酵素、毒素、シグナル、染料、検出酵素、西洋ワサビペルオキシダーゼ(HRP)、アルカリホスファターゼ(AP)、β−ガラクトシダーゼ、色素原、ファストレッド、3,3’−ジアミノベンジジン、3−アミノ−9−エチルカルバゾール、5−ブロモ−4−クロロ−3−インドリルホスフェート、3,3’,5,5’−テトラメチルベンジジン、5−ブロモ−4−クロロ−3−インドリル−β−D−グルクロニド、およびそれらの任意の組み合わせからなる群より選択される、請求項9に記載の単離された抗体またはその単離された抗原結合フラグメント。
- 生物学的サンプル中のp40を検出するための方法であって、
(a)請求項8に記載のキットを提供する工程;および
(b)免疫アッセイにより前記単離された抗体またはその単離された抗原結合フラグメントの前記p40との結合を検出する工程であって、(i)前記生物学的サンプルと、アメリカンタイプカルチャーコレクション(ATCC)にATCC特許寄託番号PTA−120163のもとで寄託されたハイブリドーマにより産生される単離された抗体、またはその単離された抗原結合フラグメントであって、配列番号4に記載のアミノ酸配列を含むp40に特異的に結合する、単離された抗体、またはその単離された抗原結合フラグメントとを接触させ、複合体を形成する工程、および(ii)前記キットにおいて提供される前記抗体検出試薬により複合体を検出する工程を含み、前記免疫アッセイが、免疫組織化学(IHC)、FFPEのIHC、凍結組織切片のIHC、免疫細胞化学、およびELISAからなる群より選択される、工程
を包含する方法。 - 前記生物学的サンプルが、肺組織、膀胱組織、乳房組織、および前立腺組織を含む、請求項11に記載の生物学的サンプル中のp40を検出するための方法。
- 前記生物学的サンプルが、正常組織、新生物組織、膀胱組織、腎臓組織、卵巣組織、甲状腺組織、子宮内膜組織、腎組織、扁桃腺組織、膵臓組織、結腸組織、リンパ節組織、新生物膵臓組織、胃組織、前立腺組織、肺組織、および乳房組織からなる群より選択される、請求項11に記載の生物学的サンプル中のp40を検出するための方法。
