Description
(1)内径1mm、外形2mm以下、長さ50〜80mm、両端が開いている毛細管の一端を完全に融解した試料につけて10mmの高さまで試料をみたし、速やかに氷片等で固化させる。
(2)10℃以下で24時間、あるいは氷上で1時間放置した後試験に供する。
(3)上昇融点測定器(ELEX SCIENTIFIC社製 EX−871A)に毛細管をセットする。
(4)予想される融点よりも約20℃低い温度の水を満たした容器に毛細管を浸し、温度計の下端を水面下30mmの深さに置く。
(5)容器内の水を適当な方法でかき混ぜながら、最初は2℃/分ずつ水温が上昇するように加熱する。
(6)予想される融点の約10℃低い温度に達した後は0.5℃/分ずつ水温が上昇するように加熱する。
(7)試料が毛細管中で上昇を始める温度を上昇融点とする。
表2に示す原料、及び配合により、乳化物を製造した。以下に乳化物の製造手順を示す。脂質類及び水を量りとり、60℃に加熱後、水にリン酸塩類、及びタンパク質類を溶解させる。溶解後、脂質類と混合し、ホモミキサー(SILVERSON社製、LABORATORY MIXER EMULSIFIER)を用いて攪拌数2500rpmにて予備乳化を行った。
表2の実施例5又は11の乳化物を炭素源として使用し、微生物の培養を行った。微生物としてE.coli HB101株(タカラバイオ社製)を使用した。
炭素源として、微生物培養用乳化物に替えて乳化処理していないPFAD又はPKFADを使用した以外は、実施例14〜15と同様の操作を行った。結果を表3に示す。
表2の実施例5、11、12、又は13の乳化物を炭素源として使用し、PHAの生産を行った。微生物としてCandida maltosa AHU−71 pARR−149/171NSx2−phbB株(国際公開公報第2005/085415号パンフレット参照)を使用した。
炭素源として、微生物培養用乳化物に替えて乳化処理していないPFAD、PKFAD、CPO、又はCPKOを使用した以外は実施例16〜19と同様の操作を行った。結果を表4に示す。
表2の実施例5、11、12、又は13の乳化物を炭素源として使用し、PHAの生産を行った。微生物としてCupriavidus necator KNK−005株(米国特許7,384,766号公報参照)を使用した。
炭素源として、微生物培養用乳化物に替えて乳化処理していないPFAD、PKFAD、CPO、又はCPKOを使用した以外は実施例20〜23と同様の操作を行った。結果を表5に示す。
Claims (12)
- 脂質類、リン酸塩類、並びに、カゼイン、カゼインNa、及びホエーからなる群から選択される1以上の乳タンパク質を含み、乳タンパク質の濃度は、脂質類に対して0.01〜10重量%であることを特徴とする、微生物培養用乳化物。
- 前記脂質類の25℃での水への溶解度が10g/L以下であることを特徴とする、請求項1に記載の微生物培養用乳化物。
- 前記脂質類に遊離脂肪酸が5%以上含まれていることを特徴とする、請求項1又は2に記載の微生物培養用乳化物。
- 前記微生物培養用乳化物中の前記脂質類の含量が3〜70%であることを特徴とする、請求項1から3のいずれかに記載の微生物培養用乳化物。
- 請求項1から4のいずれかに記載の微生物培養用乳化物を培地中に添加することを特徴とする、微生物の培養方法。
- 前記微生物培養用乳化物に含まれる脂質類の上昇融点が、微生物の培養温度より2℃以上高いことを特徴とする、請求項5に記載の培養方法。
- 前記微生物が細菌であることを特徴とする、請求項5又は6に記載の培養方法。
- 前記細菌がCupriavidus属に属する細菌であることを特徴とする、請求項7に記載の培養方法。
- 前記Cupriavidus属に属する細菌がCupriavidus necator、又は、その遺伝子組み換え体であることを特徴とする、請求項8に記載の培養方法。
- 請求項5から9のいずれかに記載の方法で微生物を培養することを特徴とする、微生物代謝産物の製造方法。
- 前記微生物代謝産物がポリヒドロキシアルカン酸であることを特徴とする、請求項10に記載の製造方法。
- 前記ポリヒドロキシアルカン酸がポリ(3−ヒドロキシブチレート−co−3−ヒドロキシヘキサノエート)であることを特徴とする、請求項11に記載の製造方法。
