Description
本発明の第一の形態によれば、バイオフラボノイド組成物で含浸させた材料が提供され、ここで該組成物のバイオフラボノイド分は、少なくともナリンギンおよびネオヘスペリジンを含む。
好ましい選択肢は、R2が水素であり、R1が3位または4位にある場合である。他の選択肢は、R1が3−ヒドロキシであり、R2が4−メトキシルである場合である。好ましくは、Xは、Hである。より好ましくは、Xは、糖である。
サリチル酸は、ヤナギ樹皮抽出物から得ることができる。あるいは、サリチル酸の合成方法が当業者公知である。
同様に、追加の有機酸が存在する場合、その有機酸も同様に、その塩ではなく酸の形態である。好適な追加の有機酸としては、一塩基性(すなわち1つのCO2H基)、二塩基性または三塩基性酸であり、場合により1、2または3個のヒドロキシル基を含有する最大8個の炭素原子の酸が挙げられる。このような追加の有機酸は、クエン酸、リンゴ酸、乳酸(latic acid)、酒石酸、フマル酸などのうち1種またはそれより多くであってもよい。
現時点では、本組成物においてサリチル酸およびクエン酸を採用することが好ましい。
本明細書で説明される組成物は、極めて有利な安全性および環境プロファイルを有する。極めて有効な抗菌活性を示すのと同様に、本組成物または、非毒性、非腐食性、再生可能、および完全に生分解性でもある。WO2012/017186(参照により本明細書に組み入れられる)で開示された組成物は、本発明の好ましい組成物である。
タケ材料は、天然のタケから抽出または加工されたセルロース繊維であるタケ繊維で形成されていてもよい。タケは、環境を破壊せずに利用できる農産物であり、完全に植物性セルロースから誘導される天然産物であるため、タケ繊維は土壌中の微生物によって生分解可能であり、さらには日光によっても生分解可能である。本発明のタケ材料は100%タケ繊維で形成されることが好ましいが、他のセルロース繊維との混合物も考慮される。
好ましくは、本材料に含浸させるバイオフラボノイドコーティングの量は、本材料全体にわたり均一である。
本明細書で説明されるバイオフラボノイド組成物は、グラム陽性細菌、グラム陰性細菌、菌類、ウイルス、原生動物、および寄生虫などの多様な生物に対して活性を示す。本組成物は、メチシリン耐性黄色ブドウ球菌(MRSA:methicillin resistant Staphylococcus aureus)、クロストリジウム・ディフィシレ(clostridium difficile、C. diff)、ヘリコバクター・ピロリ(helicobacter pyroli、H. pylori)、およびバンコマイシン耐性腸内細菌などの困難な細菌に対して採用される可能性がある。また本組成物は、ノロウイルスや空気との接触によって伝染する他の病原体に対しても使用できる。特に、本明細書で説明される本組成物は、大腸菌、黄色ブドウ球菌、サルモネラ属、枯草菌、および緑膿菌に対して活性を示す。
好ましくは、上述した方法により、セルロース系繊維材料全体にわたり均一なバイオフラボノイドの含浸が達成される。0.005〜0.75%、好ましくは0.005〜0.5%、より好ましくは0.025%〜0.5%、さらにより好ましくは0.025〜0.1%の濃度のバイオフラボノイド組成物が使用される。本明細書で説明される組成物は水溶性であり、バイオフラボノイド組成物を望ましい濃度に希釈するのに水が使用できる。
(2) 前記ナリンギンおよびネオヘスペリジンが共に、バイオフラボノイド分の少なくとも50%を構成する、(1)に記載の材料。
(3) 前記ナリンギンおよびネオヘスペリジンが共に、バイオフラボノイド分の少なくとも70%を構成する、(2)に記載の材料。
(4) 前記ナリンギンおよびネオヘスペリジンが共に、バイオフラボノイド分の少なくとも75%を構成する、(3)に記載の材料。
(5) 前記ナリンギンおよびネオヘスペリジンが共に、バイオフラボノイド分の75%〜80%を構成する、(4)に記載の材料。
(6) 前記組成物のバイオフラボノイド分が、ネオエリオシトリン、イソナリンギン、ヘスペリジン、ネオジオスミン、ナリンゲニン、ポンシリン、およびロイフォリンの群より選択される1種またはそれより多くの化合物をさらに含む、(1)〜(5)のいずれか一項に記載の材料。

(17) 表面上の細菌量を低下させる方法であって:a)(9)、(10)または(11)のいずれか一項に記載の材料を水分で湿潤させる工程;および、b)表面上に該材料をを置く工程、および/または該材料を表面全体に塗り付ける工程、を含む、上記方法。
(18) 前記細菌量が、黄色ブドウ球菌(S. aureus)、大腸菌(E. coli)、緑膿菌(P. aeruginosa)、枯草菌(B. subtilis)、サルモネラ属、MRSA、クロストリジウム・ディフィシレ(C.diff)、およびヘリコバクター・ピロリ(H.pylori)の群から選択される細菌で構成される、(17)に記載の方法。

ここで、本発明をより詳細に理解するために、単なる一例として、以下の図を参照しながら本発明を説明する。
手法
適切な液体培地中で単一種の微生物の純粋培養物を増殖させた。培養物を標準的な微生物学的技術を使用して標準化して、1ミリリットルあたり100万個の細胞に極めて近い濃度にした。微生物の培養物が基準に近ければ近いほど、テスト結果はの再現性はより高くなる。滅菌希釈剤を使用して抗菌剤を数回1:1希釈した。抗菌剤を希釈した後、各希釈容器に、希釈した抗菌剤の体積に等しい標準化した接種材料の体積を添加して、微生物濃度を1ミリリットルあたりおよそ500,000個の細胞にした。菌が植え付けられた抗菌剤の連続希釈液を、予め決められた期間(通常18時間)、テスト生物にとって適切な温度でインキュベートした。インキュベート後、通常、混濁および/または容器底部への微生物の沈殿によって示される微生物の増殖に関して、一連の希釈容器を観察した。連続希釈において増殖を示さなかった最後のチューブが、抗菌剤の最小阻止濃度(MIC)に相当する。
MICテストは、液体抗菌剤の生物抑制(および場合によってはさらに殺菌)活性の確立された「選別法」である。これは、さらなる有効性テストに使用するための抗菌活性の適切な濃度を発見するのに使用されることが多い。MICテストとMBCテストの両方を行うことにより、殺菌性または生物抑制性の作用機序を区別することが可能になると予想される。使用される活性物質の濃度と接触時間に応じて、活性物質は、生物抑制性の作用機序と殺菌性の作用機序の両方を有すると実証されることがある。
手法
標準的な生菌数計測手法を使用して、全ての時間殺菌テストを行った。NBX34689を参考にした。

時間殺菌テストは、所定の微生物集団を殺菌するのにかかる時間を推定する。シトロックスBC活性物質で多種多様の微生物を殺菌した。抗菌性タオルで使用するためのこの活性物質を特徴付ける際における重要な第一の工程は、殺菌性の要件を検証することであった。有効性の要件は、所定時間枠内で殺菌された細菌の数に基づく。最も厳格な要件は、食品との接触に関してなされる要件であり、これは、活性物質は、30秒で攻撃生物の5の対数減少値を達成しなければならないというものである。
実施例2(a):時間殺菌テスト:10分の接触時間
殺菌の速度論は、細菌数、使用される活性物質の濃度、および接触時間の影響を受ける。シトロックスBC活性物質の最も有効な範囲を決定するために、10分の殺菌テストで黄色ブドウ球菌を使用して、シトロックスBC活性物質の様々な濃度の有効性を推定した。テストされたシトロックスBC活性物質の全ての濃度(0.45〜0.65%)で6.56より大きい対数減少値が観察された(表3)。
大腸菌、緑膿菌、および黄色ブドウ球菌を使用した時間殺菌研究を、0.5%のシトロックスBC活性物質を使用して30秒の接触時間で行った。全ての生物で6.4より大きい対数減少値が観察された(表5)。これにより、この活性物質は、食品と接触する状況で使用するための基準を満たすと予想されることが確認された。
上述したように、あらゆる抗菌活性物質にとって最も重要なテストは、殺胞子能力である。定義によれば、細菌の胞子を殺す化学物質または方法はいずれも、滅菌剤である。シトロックスBC活性物質が殺胞子性かどうかを推定するために、実際の胞子懸濁液で殺菌テストを行った。シトロックスBCを0.5%〜1.5%の範囲にわたり1時間かけてテストした。このテストにはいくつか制限があった。このテストにおける胞子懸濁液(枯草菌、ATCC6633、6.4×104CFU/ペレット、マイクロバイオロジー(Microbiologies))はわずか約2×104CFU/mlであり、対数減少値の計算が制限された。凍結乾燥ペレットには、シトロックスBC活性物質のバイオフラボノイド成分を中和することが知られている物質である炭が含有されていることが見出された。これらの制限下で、胞子においておよそ2の対数減少値が実証された。これは、シトロックスBC活性物質は、胞子に対して明確な活性を有することを示す。この活性をさらに調査するためには、炭添加剤を含まないより高い力価の胞子懸濁液を使用すべきである。
1)手法:ペーパータオルへのシトロックスBCの添加
バウンティ(登録商標)(「バウンティ」は、プロクター&ギャンブル(Procter & Gamble)の登録商標である)ブランドのペーパータオルを使用して、乾燥した抗菌性タオルを作製した。バウンティ(登録商標)ペーパータオルは、一般的な市販のペーパータオル製品である。シトロックスBC活性物質の濃縮物を望ましい濃度に希釈した。1枚のペーパータオルを希釈した活性物質に完全に浸漬し、手で絞った。タオルを一晩乾燥させた。
54℃のオーブン中で、バウンティ(登録商標)ブランドのペーパータオルを一定の重量になるまで乾燥させた。シトロックスBC活性物質の様々な希釈液を、上述したようにバウンティ(登録商標)ブランドのペーパータオル上で乾燥させた。タオルを室温で一晩乾燥させた。次いで、処理済みタオルを一定の重量になるまで54℃で乾燥させた。未処理のタオルと処理済みタオルとの重量差をシトロックスBC活性物質の重量と想定した。
代表的な硬質の非多孔質表面として実験用ベンチトップを使用して、テープで格子状の区切をつくった。攻撃生物の液体培地培養物を染み込ませた綿棒を使用して表面に植菌し、空気乾燥した。シトロックスBC活性物質の希釈液で処理したペーパータオルを湿潤させ、次いで植菌されたベンチトップを清浄化するのに使用した。ベンチトップは見た目で3分湿潤した状態にし(接触時間)、次いでそのまま完全に空気乾燥させた。RODAC(Replicate Organism Detection and Counting)プレートを使用して、清浄化された表面を、生存細菌のためにサンプリングした。攻撃生物に適した温度でプレートを一晩インキュベートした。コロニーを数え、その数値を使用してCFU/cm2を計算した。3インチ×3インチの「格子で仕切られた正方形」5つからのカウント数の平均を出すことにより結果を計算した。
RODACプレートを使用して、サンプリングされる表面に接触し、その後プレートを適切な温度でインキュベートした。様々な微生物の増殖を促進する培地中にはが栄養素が含まれる。寒天中にレシチンおよびポリソルベート80が取り入れられており、これらは消毒剤/殺菌剤の中和剤として機能する。寒天培地の表面上のコロニーの外観から、微生物のタイプと数を検出した。消毒剤での清浄および処理の前後に同じ領域からサンプルを収集することにより、衛生手法の評価を行った。
水で湿潤させたシトロックスBC活性物質非含有のペーパータオルを、汚染された硬質の表面から細菌を除去する能力に関して推定した。図1および2において、この対照(すなわち含浸されていないペーパータオル)の結果は「0」と標識されたバーで示される。図1および2は、黄色ブドウ球菌および大腸菌の両方の結果を示す。シトロックスBC活性物質の1:200より大きい希釈液を含有するペーパータオルは、黄色ブドウ球菌を50CFU/cm2を超えるレベルから1CFU/cm2未満のレベルに低下させることができた。シトロックスBC活性物質の1:200より大きい希釈液を含有するペーパータオルは、大腸菌を7CFU/cm2を超えるレベルから1CFU/cm2未満のレベルに低下させることができた。
結果の考察
シトロックスBC活性物質の様々な希釈液を、バウンティ(登録商標)ブランドのペーパータオル上で乾燥させた。これらの処理済みタオルを、細菌が大量に植え付けられた実験用ベンチを浄化するのに使用した。RODACプレートを使用して、実験用ベンチ上での汚染物質の減少に影響を与える処理済みタオルの能力を評価した。

- バイオフラボノイド組成物で含浸させた乾燥したセルロース系繊維材料であって、該組成物のバイオフラボノイド分は、少なくともナリンギンおよびネオヘスペリジンを含む、上記セルロース系繊維材料。
- 前記ナリンギンおよびネオヘスペリジンが共に、バイオフラボノイド分の少なくとも50%、少なくとも70%、少なくとも75%、または、75%〜80%を構成する、請求項1に記載の材料。
- 前記組成物のバイオフラボノイド分が、ネオエリオシトリン、イソナリンギン、ヘスペリジン、ネオジオスミン、ナリンゲニン、ポンシリン、およびロイフォリンの群より選択される1種またはそれより多くの化合物をさらに含む、請求項1または2に記載の材料。

- バイオフラボノイド組成物でセルロース系繊維材料を含浸させる方法であって:
a)該材料もしくは該材料の繊維を、請求項1〜4のいずれかに記載のバイオフラボノイド組成物に浸漬するか、または該材料もしくは該材料の繊維に該バイオフラボノイド組成物を噴霧する工程;
b)該材料を機械的に乾燥させる工程
を含む、上記方法。 - 表面上の細菌量を低下させる方法であって:
a)請求項6〜8のいずれかに記載の材料を水分で湿潤させる工程;および
b)表面上に該材料を置く工程、および/または該材料を表面全体に塗り付ける工程、
を含む、上記方法。 - 前記細菌量が、黄色ブドウ球菌(S. aureus)、大腸菌(E. coli)、緑膿菌(P. aeruginosa)、枯草菌(B. subtilis)、サルモネラ属、MRSA、クロストリジウム・ディフィシレ(C. diff)、およびヘリコバクター・ピロリ(H. pylori)の群から選択される細菌で構成される、請求項12に記載の方法。
- 請求項1〜5のいずれかに記載の乾燥したセルロース系繊維材料を含む、包装された製品。
- 前記セルロース系繊維材料が、請求項6、7または10のいずれかに記載のものである、請求項14に記載の製品。
- 前記製品が、タオル、クロス、食品用パッド、呼吸用マスク、または、ソックスなどの衣類の形態である、請求項14または15に記載の製品。
