・大腸菌(Escherichia coli)株RR1ΔM15(Ruether, Nucleic Acids Res 10:5765-5772, 1982)。
・大腸菌(Escherichia coli)株XL1-Blue(Agilent Technologies,カタログ番号200268)。
・ヒト補体タンパク質C5(hC5)。完全な1676個のアミノ酸プロタンパク質は、GenBank受入番号:NP_001726(配列番号:760)を有し、ここで、アミノ酸19〜673がベータ鎖であり、アミノ酸678〜1676がアルファ鎖である。本明細書に用いたヒトC5は、Quidel(カタログ番号A403)から購入した。
・カニクイザル(Cynomolgus)補体タンパク質C5(cC5;配列番号:762)。cC5配列は、カニクイザル(Cynomolgus)(オナガザル科(Macaca fascicularis))ゲノム配列(www.ebi.ac.uk/ena; Ebeling et al. (2001) Genome Res. 21(10):1746-1756)から誘導した。ヒトC5のコード領域をwww.ensembl.orgから検索し、C5遺伝子を染色体15に限局化した。この遺伝子を含む領域は、約110000塩基長であり、コンティグCAEC01154150〜CAEC01154178に含まれる。コンティグを手作業で単一ファイルにつなぎ、カニクイザルゲノム原物質に対してヒトC5のコード領域を整列させるsim4ソフトウェアにゲノムコンテキスト(genomic context)として用いた。本明細書に用いるカニクイザルC5は、3ステップ手順;PEG6000沈降、イオン交換およびOmCIアフィニティークロマトグラフィーを用いて血清から組織内で精製した。
・ラット補体タンパク質C5(rC5;GenBank受入番号:XP_001079130)。本明細書に用いるラットC5は、3ステップ手順;PEG6000沈降、イオン交換およびOmCIアフィニティークロマトグラフィーを用いている血清から組織内で精製した。
・フリースタイルHEK293細胞中に組織内で産生したヒトC5のアミノ酸残基822〜931(配列番号:760;Fredslund et al. (2008) Nature Immunology 9: 753-760)に対応する補体タンパク質C5のヒトMG7(hMG7)ドメイン。
・hMG7結合タンパク質。
・C末端にHis6タグを有するヒメダニOrnithodoros moubata OmCIからのOmCI(AF2999、前出のNunn, M. A.ら)(配列番号:761)を、E. coli株Origami(DE3)中で組織内で産生し、HisTrap1カラムで精製した。
物質および方法
標的タンパク質hC5のビオチン化:No-Weigh EZ-Link Sulfo-NHS-LC-Biotin(Pierce, カタログ番号21327)を10倍(10×)モル過剰で用いて室温(RT)で40分間、hC5を製造元の推奨に従ってビオチン標識した。その後、緩衝液をPBS(10mMリン酸塩、137mM NaCl、2.68mM KCl、pH7.4)に交換するのは、Protein Desalting Spin Columns(Pierce, カタログ番号89849)を、製造元の説明書に従って用いて実施した。
補体タンパク質C5結合ポリペプチドのファージディスプレイ選択:ビオチン化hC5に対するファージディスプレイ選択の2〜4サイクル後に個々のクローンを得た。
物質および方法
Z変異体のサブクローニング、タンパク質発現および精製:5つの補体タンパク質C5結合Z変異体(Z05363(配列番号:510);Z05477(配列番号:509);Z05483(配列番号:511);Z05538(配列番号:512)およびZ05692(配列番号:513)を、pAffi1/pAY00065/pAY02947ライブラリーベクターから増幅した。N末端His6タグを有する二量体Affibody分子の構築のためのサブクローニング戦略は、標準分子生物学的手法を用いて、WO2009/077175に詳細に記述されたとおり適用した。Z遺伝子フラグメントを、コードされた配列MGSSHHHHHHLQ−[Z#####][Z#####]−VDが得られる発現ベクターpAY01448にサブクローニングした。
Z変異体のサブクローニング:選択した5つ独特なクローン(Z05477(配列番号:509)、Z05363(配列番号:510)、Z05483(配列番号:511)、Z05538(配列番号:512)およびZ05692(配列番号:513))を、発現ベクターpAY01448中で二量体としてサブクローニングするために選択し、その後、シークエンシングによって確認した。
本実施例では、成熟ライブラリーを構築した。ライブラリーは、hC5−結合ポリペプチドの選択に用いた。成熟ライブラリーからの選択は、通常、親和性が増加した結合剤になることが期待される(Orlova et al. Cancer Res 2006, 66(8):4339-48)。本研究では、その後に構築した二本鎖DNAの連結および制限を用いてトリヌクレオチド構成要素のランダム化セットの取り込みが可能となるSlonomics(R)技術によってランダム化二本鎖リンカーを生成した。
ライブラリーデザイン:ライブラリーは、実施例1および2に記述したhC5結合Z変異体の配列の選択に基づいた。新たなライブラリーでは、Z分子骨格中の13の可変位置を、配列番号:509〜513(Z05477、Z05363、Z05483、Z05538、Z05692)に定義されたZ変異体配列に基づく戦略に従って特定のアミノ酸残基の方へ偏らせた。制限部位XhoIおよびSacIに隣接するアミノ酸配列5'−AA ATA AAT CTC GAG GTA GAT GCC AAA TAC GCC AAA GAA/GAG NNN NNN NNN GCA/GCC NNN NNN GAG/GAA ATC/ATT NNN NNN TTA/CTG CCT AAC TTA ACC/ACT NNN NNN CAA/CAG TGG NNN GCC/GCG TTC ATC/ATT NNN AAA/AAG TTA/CTG NNN GAT/GAC GAC CCA AGC CAG AGC TCA TTA TTT A−3’(ランダム化コドンは、NNNとして示す)の147塩基対の部分ランダム化ヘリックス1および2を含む二本鎖DNAのSlonoMax(R)ライブラリーを、Sloning BioTechnology GmbH(Pucheim,ドイツ)に注文した。最終的に12個の可変Z位置を含む新たなライブラリーにおけるアミノ酸残基の理論分布を表4に示す。
AmpliTaq Goldポリメラーゼ(Applied Biosystems,カタログ番号4311816)を用いて、12サイクルのPCRの間、ライブラリーを増幅し、プールした生成物を、QIAquick PCR Purification Kit(QIAGEN,カタログ番号28106)を用いて、供給者の推奨に従って精製した。精製したランダム化ライブラリーフラグメントのプールを、制限酵素XhoIおよびSacI(New England Biolabs,カタログ番号R01460Lおよびカタログ番号R0156L)で消化し、PCR Purification Kitでさらに1回精製した。その後、1%アガロースゲル上で分取ゲル電気泳動を用いて生成物を精製した。
換細胞を37℃で50分間インキュベートした。滴定のため、そして形質転換体の数の測定のためサンプルを採取した。その後、細胞をプールし、2%グルコースおよび100μg/mlアンピシリンを補充したTSB−YE培地7l中37℃で一夜培養した。細胞を4,000gで15分間沈殿させ、PBS/グリセロール溶液(約40%グリセロール)中に再懸濁した。細胞を等分し、80Cで保存した。Z変異体のライブラリーからのクローンについて、含量を確認するため、そしてライブラリーデザインと比較して構築ライブラリーの結果を評価するため、シークエンシングした。実施例1に記述したとおり、シークエンシングを実施し、アミノ酸分布を確認した。
ライブラリー構築:確認された結合性(実施例1および2)を有するOmCIブロックトC5結合Z変異体のセットに基づいて新たなライブラリーを設計した。設計したライブラリーの理論サイズは、6.7×109のZ変異体であった。E. coli. RR1ΔM15細胞に形質転換後、滴定によって決定されたライブラリーの実測サイズは、1.4×109の形質転換体であった。
物質および方法
補体タンパク質C5結合ポリペプチドのファージディスプレイ選択:標的タンパク質hC5を、実施例1に記述したとおりビオチン化した。ファージディスプレイ選択は、実施例3に記述したZ変異体分子の新たなライブラリーを用いて、本質的に実施例1に記述したhC5に対して実施した。ファージ増幅には、E. coli XL1-Blueを用いた。選択は、2つの並行なトラックで最初に実施した。一方のトラックでは、選択の時間が2時間であったが、もう一方のトラックでは、より短い選択時間:第1のサイクルで20分およびその後のサイクル2〜4では10分を用いた。これらの2つのトラック(1および2)を第2のサイクルで更に分けて、全部で6トラック(1a−cおよび2a−c、標的濃度および洗浄条件が異なる)となった。選択は、合計4サイクルで実施した。選択のサイクル1では、25nM補体タンパク質C5を用い、0.1%PBSTにより5回の洗浄を実施した。その後の3つのサイクルでは、低下した標的濃度および増加した洗浄回数を用いて高めた厳密性を適用した。サイクル2、3および4では;10、5または2.5nM補体タンパク質C5、4、1または0.25nM補体タンパク質C5および1.6、0.2または0.05nM補体タンパク質C5を用いた。サイクル2、3および4では;0.1%PBSTを用いて10、15および、20回の洗浄を実施した。さらに、2番目の最後の洗浄は、2つのトラック(1cおよび2c)について洗浄溶液中の50×過剰の非ビオチン化hC5を用いて3時間に延長した。
補体タンパク質C5結合ポリペプチドのファージディスプレイ選択:選択は、それぞれ4サイクルを含む全部で6つの並行なトラックにおいて実施した。選択トラックが異なると、以下のように標的濃度および洗浄条件が異なる:1a)選択時間2時間、高い濃度、標準洗浄、1b)選択時間2時間、低い濃度、標準洗浄、1c)選択時間2時間、中程度の濃度、長い洗浄、2a)選択時間10分、高い濃度、標準洗浄、2b)選択時間10分、低い濃度、標準洗浄、および2c)選択時間10分、中程度の濃度、長い洗浄。各選択サイクルについては、標的濃度を低下させ、洗浄条件をより厳密にした。すべてのトラックから各ラウンドで溶出液中の十分な量のファージ粒子を得た。ほとんどのファージ粒子は、最も高い標的濃度および最も温和な洗浄条件を表すトラック1aおよび2aに見いだされた。
および発生頻度に基づいて選択した。43個の独特なZ変異体を、より低い濃度のhC5(0.05μg/ml)およびrC5(4μg/ml)について検定した。試験した40個のZ変異体について、rC5に対して陽性の結果が得られ、これは、陰性対照(0.4AU)についての2×シグナルとして定義された。hC5およびrC5のより低い濃度に対して試験したすべてのZ変異体についての結果を図3に示す。
物質および方法
発現ベクターへのZ変異体分子のサブクローニング:配列分析ならびにヒトおよびラット補体タンパク質C5に対するELISAにおける性能に基づいて、発現ベクターpAY01448へのサブクローニングのために、45個のクローンを選択した。単量体Z変異体フラグメントを、ファージミドベクターpAY02592から増幅し、pAY01448へのサブクローニングを、実施例2に記述したように実施して、タンパク質配列MGSSHHHHHHLQ−[Z#####]−VDをコードするベクターを得た。
タンパク質発現および精製:全45個のサブクローニングしたZ変異体は、発現しることができ、精製したすべての変異体についてin vitro溶解性は、良好であった。純度は、LC/MSによって、すべての変異体について90%を超えることが推定された。正確な分子量は、LC−MSによって確認した。
物質および方法
クローニングおよびタンパク質産生:C5結合Z変異体(配列番号:745〜757)のサブセットをコードするDNA、ここで、E. coliコドンを、GeneArt, GmbHによって最適化し、合成した。C5結合Z変異体を表す合成遺伝子をサブクローニングし、E. coli中で発現させた。場合によりアルブミン結合ドメイン(ABD094、配列番号:759)に結合されたZ変異体の単量体または二量体の構築物をコードする発現ベクターを、図4に概略的に説明する。

クローニングおよびタンパク質産生:「Z」が配列番号:745および配列番号:748〜757によって表すことができる、図4に概略的に記述した、産生されたタンパク質変異体を、MALDI−TOF MSによっておよびSDS−PAGEにおいて分析した(図5)。
たがって、試験したすべての第二世代のZ変異体(配列番号:748〜757)は、親のZ変異体Z05477a(配列番号:745)よりもヒトC5に対する高い結合親和性を示した。さらに、同じ方法を用いたヒトC5に対するOmCI結合のそれよりも親和性が高かった。
物質および方法
2つの異なる方法、サイズ排除クロマトグラフィーおよびBiacoreを用いて、C5結合Z変異体に融合したアルブミン結合ドメインABD094間の相互作用を研究した。
SECカラムでは、より大きな分子ほど、小さいものより速く溶出する。図8Aに見られるように、同時に注射されたHSA+Z06175a−GS−ABD094は、HSAを単独で注射したときよりも早く溶出しており、2つの分子がこれらの条件下で安定な複合体としてふるまうことを示唆している。より小さなZ06175a−GS−ABD094は、複合体またはHSA単独よりもゆっくり溶出し、これらのタンパク質単独では複合体より小さいことを示している。
物質および方法
生存相の齧歯動物:2つのC5結合構築物Z−ABD(Z06175a−GS−ABD094;GSリンカーによって配列番号:759に融合した配列番号:753、図4、構築物2)およびZ−ABD−Z(Z06175a−GS−ABD094−GSGGGGSGGGGS−Z06175a;(GSリンカーによって配列番号:759に融合し、続いてGS(G4)2リンカーによって配列番号:753の第2のモチーフに融合した配列番号:753、図4、構築物5)の薬物動態を、雄Sprague Dawley(SD)ラット(体重250〜300g)において研究した。各ラットに、Z−ABDまたはA−ABD−Zの単一用量をi.v.(250nmol/kg)またはs.c.(500nmol/kg)投与した(用量群当たりn=3)。血液サンプル(200L)を、i.v.群については投与後、5、20および45分、ならびに1.5、4、7、24、48、72、120、168、240および336時間で採血し、s.c.群については、投与後15および30分、1、2、4、7、24、48、72、120、168、240および336時間で採血した。血液をチューブに集め、冷蔵庫に20分間置いて凝固させた。続いて、4000rpmで10分間遠心分離した後、血清を得た。血清サンプルを−70℃に保ち、分析にかけた。

i.v.(250nmol/kg)およびs.c.(500nmol/kg)投与後のZ−ABDおよびZ−ABD−Zについての薬物動態データを表8にまとめる。Z−ABDは、i.v.群では投与後10〜14日まで、そしてs.c.群では14日まで血清中で定量化可能であったが、Z−ABD−Zは、両方の用量群において投与後10日まで血清中で定量化可能であった(図9)。14日は、最終的なサンプリング時点であった。
物質および方法
生存相における研究を、Charles River, Nevada(www.criver.com)で実施し、投与薬物の調製および血清サンプルの分析を組織内で実施した。Z−ABD変異体の薬物動態(GSリンカーによってABD094(配列番号:759)に融合したZ06175a(配列番号:753))を、雄カニクイザル(n=3)において、i.v.(静脈内)およびs.c.(皮下)投与後に調べた。薬物動態パラメータの評価は、実施例8に従って実施したが、しかし、静脈投与後、対数線形血清中濃度−時間曲線の最初の傾きに対応する初期血清半減期(T1/2α)、第二(中間)相に関連する、対数線形血清中濃度−時間曲線の傾きに対する対応する中間血清半減期(T1/2β)および対数線形血清中濃度−時間曲線の終末の傾きに対応する終末相血清半減期(T1/2γ)を決定した。T1/2は、ln2/λとして計算し、ここで、傾きλの見積りは、少なくとも4C=f(t)観察に基づく。sc投与について示した薬物動態データは、Z−ABDの投与前レベルについて補正したが、血清中濃度対sc投与後の時間を示すグラフは、決定された実際の血清中濃度を示す。サルは、2〜4歳で、体重2.3〜3kgであった。それぞれのサルは、1回のi.v.投与(540nmol/kg)、続いてi.v.投与の3週後に1回のs.c.投与(1635nmol/kg)を受けた。血液サンプルは、いずれの投与の後も、投与後10および30分ならびに1、2、4、8、24、48、72、120、168、240、336および504時間で採取した。血液サンプルを、室温で20〜40分間凝固させ、次いで、1500〜2200RCFで、2〜8℃で10〜15分間遠心分離した後、血清を回収し、凍結させた。分析まで、血清サンプルを−20℃未満の温度で保存した。
各用量群の平均(±標準偏差)薬物動態におけるデータを示す。Z−ABDの血清中濃度は、LC/LC/MS/MSによって、i.v.およびs.c.投与後のすべての時点で定量化可能であった(図14)。ELISAデータおよびLC/LC/MS/MSデータであるが、係数0.986によって線形的に補正されたLC/LC/MS/MSデータを計算に用いた。Z−ABDのi.v.投与後、初期血清半減期は9.1(0.8)時間であり、中間血清半減期は84(4)時間であり、そして終末相血清半減期は198(51)時間であった。平均滞留時間は246(62)時間であった。分布容積、VssおよびVzは、それぞれ110(23)ml/kgおよび127(27)ml/kgと計算され、クリアランスは0.45(0.02)mL/h*kgと推定された。
物質および方法
マウスへの投与:C57BL/6雌マウスは、ザイモサンで誘導する1時間前に腹腔内に(i.p.)、またはザイモサンで誘導する18時間前に皮下に(s.c.)、異なる濃度のZ−ABD融合分子(Z06175a−GS−ABD094、GSリンカーによって配列番号:759に融合した配列番号:753)または陽性対照OmCIを受けた。0.8mg/マウスのザイモサンをi.p.投与し、1時間後、眼窩血液サンプル(凝固活性化剤入りの血清バイアル中)をイソフルラン麻酔下で採取した。動物を頚椎脱臼により殺した。皮膚切開を行い、腹部筋肉壁を視覚化した。PBS溶液(2mM EDTAを含む)を、腹腔
に静かに注射した。腹部をマッサージし、液体サンプル(1〜2ml)を採取した。サンプルを試験管に移し、濡れた氷の上に保存した後、600gで10分間遠心分離した。上清中の総タンパク質量およびC5a濃度を分析した。
動物からの血清サンプルにおけるZ−ABDの血清中濃度および溶血活性の分析:Z−ABD融合分子(Z06175a−GS−ABD094、GSリンカーによって配列番号:759に融合した配列番号:753))の血清中濃度およびヒツジ赤血球における溶血に影響を及ぼす能力を、低用量(20nmol/kg)、中用量(100nmol/kg)および高用量(500nmol/kg)の投与後に評価した。血清中濃度は、比較的に用量と比例し、溶血の阻害からは、血清中の分子が活性であり、かつ溶血の阻害も実際に濃度依存性であることが確認された。
物質および方法
雌C57blマウスに気管内投与した後のC5結合構築物Z06175a−GS−ABD094(GSリンカーによって配列番号:759融合した配列番号:753)の薬物動態プロファイルを研究した。温度、相対湿度および照明を設定して22±1℃、55±5%および明12時間−暗12時間サイクルに維持し、食事および水は自由に摂らせた。動物にイソフルランで麻酔し、500nmol/kgのZ06175a−GS−ABD094入りのマイクロスプレー(provided ad libitum)を用いて肺に直接投与した。血清サンプルの調製のため、5分、30分、1時間、3時間、7時間、16時間、24時間、48時間および72時間(3匹の動物/時点)で、イソフルランによる麻酔下で大静脈からできるだけ多くの血液を回収した。チューブ中に血液を集めることによって血清サンプルを調製し、チューブを冷蔵庫に20分間置いた。その後、チューブを4000rpmで10分間遠心分離した。各血液サンプルから、少なくとも100μlの血清を調製した。血清サンプルを、分析前に−70℃に保った。各サンプル中のZ06175a−GS−ABD094の血清中濃度は、実施例10に記述したように、ECLによって決定し、ヒツジ赤血球中の溶血に影響を及ぼす血清サンプルの能力は、実施例6および8に記述したように決定した。
各サンプル中の血清中濃度およびヒツジ赤血球における溶血に影響を及ぼす対応する能力を、それぞれ、図16Aおよび図16Bに記述する。300〜1000nMの範囲の血清中濃度では30分以内にプラトーに達し、ここで、溶血はほとんど完全にブロックされる。投与後7時間より後の時点でサンプリングした血清では、溶血が徐々に再発生している。投与3日後の最終時点で、溶血は、対照の約70%であった(図16B)。これらのデータは、気管内投与後の体循環へのZ06175a−GS−ABD094の吸収および分子が溶血を機能的に阻害することを明らかに示している。
生存相のウサギ:Z変異体(Z06175a、配列番号:753、続いてGS(図4、構築物1))の薬物動態を、硝子体内投与後のウサギ眼において研究した。生存相研究および投薬した動物(有色ウサギ、2〜2.5kg)の眼の解剖を、Iris Pharma, La Gaude,フランス(www.iris-pharma.com)で実施した。動物は、20±2℃、相対湿度55±10%で個別に収容して食物および水は自由に摂らせた。
硝子体内投与後のZ変異体の濃度は、1日後、すべての区分で高く(6〜200μM)、驚くべきことに、投与4日後でも高いままであった(1.5〜78μM)。特に、硝子体中のZ分子の濃度は、注射1日後、118〜201μMの範囲(平均161μM、n=6眼)であり、注射4日後、26〜78μM(平均46μM、n=6)でとどまっており、数日のT1/2を示している。薬物のサイズと、ウサギ眼における硝子体内注射後の薬物の排出との間には相反関係があると考えられ、以下の例によって記述されている;モキシフロキサシン(MW<0.35kDa、T1/2=1.72時間、Mohan et al. Trans Am Ophthalmol Soc 2005, 103:76-83)、ESBA105(MW=26kDa、T1/2=25時間、Ottiger et al. Investigative Ophthalmology & Visual Science 2009, 50: 779-786)およびラニビズマブ(MW=48kDa、T1/2=2.88日、Bakri et al. American Academy of Ophthalmology 2007, 114:2179-2182)。ここで試験したZ変異体は、7.0kDaの分子量を有しており、Z分子の排除が、硝子体中でこのような小分子に期待されると考えられるものよりもゆっくりであったことを示唆している。
