・ インターロイキン(例えばIL-1、IL-2、IL-4、IL-5、IL-6、IL-7、IL-12などのサイトカイン[83、84]、インターフェロン(例えばインターフェロン−γ)、マクロファージコロニー刺激因子、および腫瘍壊死因子を含む、ヒト免疫調節薬、
・ キトサンおよびその誘導体などの生体付着性物質および粘膜付着性物質、エステル化されたヒアルロン酸細粒 [85]またはポリ(アクリル酸)、ポリビニルアルコール、ポリビニールピロリドン、多糖類およびルボキシメチルセルロースの交差結合した誘導体などの粘膜付着性物質[86]、
・ 生物分解性のおよび非毒性である材料から形成した微小粒子(すなわち、直径が最大100nm〜150μm、より望ましくは直径が最大200nm〜30μm、および最も望ましくは直径が最大500nm〜10μmの粒子)(例えばポリ(α-ヒドロキシ酸)、ポリヒドロキシ酪酸、ポリオルトエステル、酸無水物、ポリカプロラクトンなど)、
・ リポソーム [[49、87-89]の13章および14章、);
・ ポリオキシエチレンエーテルおよびポリオキシエチレン エステル[90]、
・ PCPP製剤[91および92]、

以上の同一性を持つアミノ酸配列、および/または(b)少なくとも7個の連続的な配列ID番号:18または配列ID番号:60のアミノ酸か、または配列ID番号:18または配列ID番号:60に対して80%以上の同一性を持つポリペプチドかの断片であり、かつ配列ID番号:18ま
たは配列ID番号:60の抗原決定基を含むアミノ酸配列、を含むかまたはそれにより構成されるポリペプチドであり、
前記ToxA-GT抗原が、以下:(a)配列ID番号:4または配列ID番号:56に対して80%以上の同一性を持つアミノ酸配列、および/または(b)少なくとも7個の連続的な配列ID番号:4または配列ID番号:56のアミノ酸か、または配列ID番号:18または配列ID番号:56
に対して80%以上の同一性を持つポリペプチドかの断片であり、かつ配列ID番号:4または配列ID番号:56の抗原決定基を含むアミノ酸配列、を含むかまたはそれにより構成されるポリペプチドであり、
前記TcdA抗原が、以下:(a)配列ID番号:1に対して80%以上の同一性を持つアミノ
酸配列、および/または(b)少なくとも7個の連続的な配列ID番号:1のアミノ酸か、ま
たは配列ID番号:1に対して80%以上の同一性を持つポリペプチドかの断片であり、かつ配列ID番号:1の抗原決定基を含むアミノ酸配列、を含むかまたはそれにより構成されるポ

前記TcdB抗原が、以下:(a)配列ID番号:2に対して80%以上の同一性を持つアミノ

号:1)から任意に選択される1、2、3、4、5、6、7、8、9、10、11、12、13、14、15個もしくはそれを上回るTcdA抗原を含む、項目1〜3のうちどれか一つの免疫原性組成物。

(1)ToxB-ED抗原(配列ID番号:17)、(2)ToxB-GT抗原(配列ID番号:18)、(3)ToxB-CP抗原(配列ID番号:19)(4)ToxB-T抗原(配列ID番号:20)、(5)ToxB-B抗原(配列ID番号:21)、(6)ToxB-B2抗原(配列ID番号:22)(7)ToxB-B7(配列ID番号:23)または(8)完全長TcdB抗原(配列ID番号:2)から任意に選択される1、2、3、4、5、6、7、8個もしくはそれを上回る追加的TcdB抗原をさらに含む、項目4の免疫原性組成物。
(項目6)
ToxA-GT抗原および(1)ToxB-ED抗原(配列ID番号:17)、(2)ToxB-GT抗原(配列ID

(1)ToxA-ED抗原(配列ID番号:3)、(2)ToxA-GT抗原(配列ID番号:4)、(3)ToxA-CP抗原(配列ID番号:5)、(4)ToxA-T抗原(配列ID番号:6)、(5)ToxA-T4抗原(
配列ID番号:7)、(6)ToxA-B抗原(配列ID番号:8)、(7)ToxA-PTA2抗原(配列ID番
号:9)、(8)ToxA-P5-7抗原(配列ID番号:10)、(9)ToxA-P5-6抗原(配列ID番号:11)、(10)ToxA-P9-10抗原(配列ID番号:12)、(11)ToxA-B2抗原(配列ID番号:13)、(12)ToxA-B3抗原(配列ID番号:14)、(13)ToxA-B5抗原(配列ID番号:15)、(14)ToxA-B6抗原(配列ID番号:16)または完全長TcdA抗原(配列ID番号:1)から任意に選択される1、2、3、4、5、6、7、8、9、10、11、12、13、14、15個もしくはそれを上回る
追加的TcdA抗原をさらに含む、項目6の免疫原性組成物。
(項目8)
ToxB-GT抗原およびTcdA抗原を含む、前述の項目のうちどれかによる免疫原性組成物
であって、前記TcdA抗原が、ToxA-P5-6およびToxA-B2から成る群から選択される、免疫原性組成物。
(項目9)
i)ToxA-GT抗原またはToxB-GT抗原、およびii)ToxA-B、ToxA-PTA2、ToxA-P5-7、ToxA-P5-6、ToxA-P9-10、ToxA-B2、ToxA-B3、ToxA-B5および/またはToxA-B6から選択される少なくとも一つのTcdA抗原、およびToxB-B、ToxB-B2抗原、および/またはToxA-B7から選択される少なくとも一つのTcdB抗原を含む、免疫原性組成物。
(項目10)
i)ToxA-GT抗原およびToxB-GT抗原、およびii)ToxA-B、ToxA-PTA2、ToxA-P5-7、ToxA-P5-6、ToxA-P9-10、ToxA-B2、ToxA-B3、ToxA-B5および/またはToxA-B6から選択される少なくとも一つのTcdA抗原、およびToxB-B、ToxB-B2抗原、および/またはToxA-B7から選択される少なくとも一つのTcdB抗原を含む、免疫原性組成物。
(項目11)
ToxB-GT抗原、TcdA抗原およびさらなるTcdB抗原を含む前述の項目のうちどれかによ
る免疫原性組成物であって、前記組成物が任意に(a)ToxB-GT+ToxA-B2+ToxB-B、また
は(b)ToxB-GT+ToxB-B+ToxA-P5-6を含む、免疫原性組成物。
(項目12)
ToxB-GT抗原、ToxA-GT抗原、さらなるTcdA抗原およびさらなるTcdB抗原を含む前述の項目のうちどれかによる免疫原性組成物であって、前記組み合わせが任意にToxB-GT+ToxA-GT+ToxA-B2+ToxB-Bを含む、免疫原性組成物。
(項目13)
前述の項目のうちどれか一つによる免疫原性組成物であって、前記組成物中の前記抗原のうち少なくとも二つが混成ポリペプチドの形態である、免疫原性組成物。
(項目14)
項目1〜12のうちどれか一つによる免疫原性組成物であって、前記抗原のいずれもが
混成ポリペプチドの形態ではない、免疫原性組成物。
(項目15)
前述の項目のうちどれか一つによる免疫原性組成物であって、前記組成物がC. difficile毒素Aおよび毒素Bに対する中和価を生じさせる、免疫原性組成物。
(項目16)
少なくとも一つのさらなるC. difficile抗原を含む、前述の項目のうちどれか一つによる免疫原性組成物であって、前記のさらなるC. difficile抗原が任意に糖類抗原である、免疫原性組成物。
(項目17)
ワクチン組成物である、前述の項目のうちどれか一つによる免疫原性組成物。
(項目18)
アジュバントをさらに含む、項目17のワクチン組成物。
(項目19)
製薬としての用途の項目17または項目18によるワクチン組成物。
(項目20)
哺乳動物での免疫応答を高める用途の、項目17〜19のうちどれか一つのワクチン組成物。
(項目21)
前記哺乳動物がヒトである、項目20のワクチン組成物。
(項目22)
C. difficileに関連する病気の治療または予防の用途の、項目18〜21のうちどれか一つによるワクチン組成物。
(項目23)
前述の項目のうちどれかの有効量の免疫原性組成物またはワクチンを哺乳動物に投与する手順を含む、前記哺乳動物での免疫応答を高めるための方法。
Claims (16)
- Clostridium difficile抗原の組み合わせを含む免疫原性組成物であって、前記組み合わせが、以下:
a)ToxB-GT抗原およびTcdA抗原、
を含み、前記TcdA抗原が、配列ID番号:11に対して80%以上の同一性を持つアミノ酸配列により構成されるToxA-P5-6抗原である、免疫原性組成物。 - 前記ToxB-GT抗原が解毒される、請求項1の免疫原性組成物。
- 前記ToxB-GT抗原が、以下:(a)配列ID番号:18または配列ID番号:60に対して80%以上の同一性を持つアミノ酸配列、および/または(b)少なくとも7個の連続的な配列ID番号:18または配列ID番号:60のアミノ酸か、または配列ID番号:18または配列ID番号:60に対して80%以上の同一性を持つポリペプチドかの断片であり、かつ配列ID番号:18または配列ID番号:60の抗原決定基を含むアミノ酸配列、を含むかまたはそれにより構成されるポリペプチドである、
請求項1または請求項2の免疫原性組成物。 - (1)ToxB-ED抗原(配列ID番号:17)、(2)ToxB-GT抗原(配列ID番号:18)、(3)ToxB-CP抗原(配列ID番号:19)(4)ToxB-T抗原(配列ID番号:20)、(5)ToxB-B抗原(配列ID番号:21)、(6)ToxB-B2抗原(配列ID番号:22)(7)ToxB-B7(配列ID番号:23)または(8)完全長TcdB抗原(配列ID番号:2)から任意に選択される1、2、3、4、5、6、7、8個もしくはそれを上回る追加的TcdB抗原をさらに含む、請求項1〜3のうちどれか一つの免疫原性組成物。
- ToxB-GT抗原、TcdA抗原およびさらなるTcdB抗原を含む、請求項1〜4のうちどれかによる免疫原性組成物であって、前記組成物が任意にToxB-GT+ToxB-B+ToxA-P5-6を含む、免疫原性組成物。
- 請求項1〜5のうちどれか一つによる免疫原性組成物であって、前記組成物中の前記抗原のうち少なくとも二つが混成ポリペプチドの形態である、免疫原性組成物。
- 請求項1〜5のうちどれか一つによる免疫原性組成物であって、前記抗原のいずれもが混成ポリペプチドの形態ではない、免疫原性組成物。
- 請求項1〜7のうちどれか一つによる免疫原性組成物であって、前記組成物がC. difficile毒素Aおよび毒素Bに対する中和価を生じさせる、免疫原性組成物。
- 少なくとも一つのさらなるC. difficile抗原を含む、請求項1〜8のうちどれか一つによる免疫原性組成物であって、前記のさらなるC. difficile抗原が任意に糖類抗原である、免疫原性組成物。
- ワクチン組成物である、請求項1〜9のうちどれか一つによる免疫原性組成物。
- アジュバントをさらに含む、請求項10のワクチン組成物。
- 製薬としての用途の請求項10または請求項11によるワクチン組成物。
- 哺乳動物での免疫応答を高める用途の、請求項10〜12のうちどれか一つのワクチン組成物。
- 前記哺乳動物がヒトである、請求項13のワクチン組成物。
- C. difficileに関連する病気の治療または予防の用途の、請求項11〜14のうちどれか一つによるワクチン組成物。
- 請求項1〜10のうちどれかの有効量の免疫原性組成物または請求項11〜15のうちのどれかの有効量のワクチン組成物を含む、哺乳動物での免疫応答を高めるための組成物。
