過敏性腸症候群は慢性的な機能性胃腸障害である。世界中で約4〜30%の人間はIBSに苦しむ。IBSは腹痛(慢性的疼痛)と排便習慣の変化との二つの主要症状が特徴である。前記主症状によれば、IBSは、下痢に伴うIBS(IBS−D)、便秘に伴うIBS(IBS−C)、混合形IBS(IBS−M)と分類不能型IBS(IBS−U)の四つのサブグループに分類することが可能である。IBS患者の中には、約30%の患者は胃腸炎の後にIBSに苦しむ。また、それらは感染/炎症後IBSに属する。さらに、患者が健康管理を探し求めるようにモチベーションを持たせ、生活の品質を著しく低下させることの主な要因は、内臓過敏(VH)と関連する腹痛である。内臓過敏は、機能性胃腸障害を起こす主要メカニズムの一つだと考えられる。また、最近の研究は、内臓過敏はIBSに対して高特異性であることを示す。
内臓過敏は、患者にセンセーションの強度増加と内臓痛に対する閾値の低下とで定義される。持続するVHは、ニューロン興奮性の増加に示されるニューロン感作性(neuronal sensitization)に関連する。神経伝達物質、たとえば、セロトニン(5−ヒドロキシトリプタミン、5−HT)は、ニューロン感作性において重要な役割を果たす。セロトニンはモノアミンニューロン伝達物質である。従来の研究では、すでに覚醒ラットに5−ヒドロキシトリプトファン(5−HTP、セロトニンの前駆体)を皮下注射すると、VHが誘発されたことを示している。セロトニンを合成と貯蔵する重要な場所は腸粘膜のクロム親和性細胞である。クロム親和性細胞から放出されたセロトニンは腸の分泌、運動性、及びセンセーションと関連する神経反射を活性化させる。臨床研究によると、IBS患者は通常セロトニン代謝の異常が伴っている。さらに、セロトニン受容体拮抗薬は既に治療薬剤として広く使用されている。これは、IBSの病状はセロトニンと関連することを示している。
内臓過敏を示す直腸膨満試験の痛覚閾値の低下は、IBS患者の顕著な特徴である。動物試験によると、結腸直腸の痛覚閾値はバロスタット膨張に反応する筋電図(EMG)信号から監視でき、EMG信号が高ければ高いほど、痛みの感受性が高いことを意味する。VHに加えて、脊髄にP物質の表現の増加もすでにIBSラットモデルの結腸直腸膨張時に内臓痛覚を示すバイオマーカーとして実証されている。
IBS症状を改善する治療方法はさまざまあり、セロトニン受容体拮抗薬、抗うつ薬、運動促進薬、鎮痙薬とプロバイオティクスを含む。プロバイオティクス(有益菌)は生きている微生物であり、適切な量で投与したとき、宿主に健康利益を与える。薬理的な副作用がないので、プロバイオティクスはIBSの治療にとって有望な方法だと思われる。一方、特定なプロバイオティクスは便秘症状に有利な効果を与えることもすでに記載されている。
便秘は、ワールドワイドの機能性胃腸障害であり、便の低頻度または通過困難と定義されている。便秘の原因は多くあり、化合物、食生活、腸内細菌の組成、妊娠、及び心理的ストレスを含む。多くの種類の下剤はすでに確認されているが、これらの薬の大部分は潜在的な不利な副作用があり、たとえば薬物の耐性、大腸メラノーゼ、または下剤性結腸を含む。便秘患者の腸内細菌は腸内毒素があり、プロバイオティクスの消費により改善できる。さらに、プロバイオティックバクテリア、特に乳酸桿菌とビフィズス菌は、結腸のpHを低下させることができ、乳酸、酢酸、及びその他のものを製造できる。低下したpHは大腸の蠕動運動を向上させる傾向があり、その結果、結腸の輸送時間を減少させ、便秘症状の治療にとって有利である。
プロバイオティクスは腸管内菌叢のバランスを維持すること、粘膜の障壁機能を強化させることと、免疫システムを調節することにより、宿主をより健康にし得る。数多い証拠は、プロバイオティクスは感染症、アレルギー、ガンと壊死性腸炎を含む疾患の治療法になることを示している。プロバイオティクスが機能性胃腸障害の治療に関する用途をさらに研究する価値がある。
それらのなかで、プロバイオティクスとして使用される最も重要な微生物種である乳酸菌(lactic acid bacteria)(本明細書では、これ以後LABと時々称される)は、宿主腸細菌叢を調節できるため、消化管(gut)の健康を予防または治療するための選択肢として認識されている。さらに、近年、LABが宿主の心理的ストレスに対する心理的及び生理的反応を変えることができることを明らかに示す証拠が増えている。
乳酸菌は宿主の腸‐脳軸への影響を有することが示されているため、従来の精神疾患用薬だけでなく、乳酸菌もストレス誘発失調を治療する代用品として使用できる。無菌マウスの活動力増加と不安減少が不足することは、正常菌群の脳部発育における重要な役割を強調することが開示されている。従来の研究には、ラクトバチルス・ラムノサス(Lactobacillus rhamnosus)が迷走神経により健康マウスにおける中枢神経系機能を変化させていることが開示され、また、ビフィドバクテリウム・インファンティス(Bifidobacterium infantis)は、強制水泳試験(forced swimming test、単にFSTと称する)の行動欠陥を逆転させ、脳幹におけるアドレナリン濃度を還元することが開示されている。上記研究は、プロバイオティクスが精神失調の治療及び神経伝達物質として開発される得ることを支持している。
数多い研究により、生産及び製造方法並びにフードキャリアはプロバイオティック株の特性に影響を及ぼし得ること、および臨床的介入研究の結果に影響を与えることが示されている。ここで、発明者らは、精神障害及び機能性胃腸障害の予防または治療の可能性を示す乳酸菌株を同定した。
本発明の一形態においては、ラクトバチルス・プランタルム亜種プランタルムPS128及び担体を含む組成物を提供する。
本発明の一態様においては、前記担体は生理学的に許容される賦形剤または希釈剤である。生理学的に許容される賦形剤または希釈剤の例としては、乳糖、澱粉、デキストリン、シクロデキストリン、カルボキシメチルスターチナトリウム、プロピオン酸カルボキシル化スターチ、微結晶セルロース、カルボキシメチルセルロース、マルトデキストリン、およびステアリン酸マグネシウムを含むが、これらに限定されない。
さらに具体的な態様においては、ラクトバチルス・プランタルム亜種プランタルムPS128を投与した後、血清コルチコステロン濃度は個体において統計学的に有意に低下する。また、ラクトバチルス・プランタルム亜種プランタルムPS128を投与した後、神経伝達物質濃度は個体において統計学的に有意に上昇する。前記神経伝達物質はドーパミンであることが好ましい。
本発明の一形態においては、有効量のラクトバチルス・プランタルム亜種プランタルムPS128を個体に投与することを含む、機能性胃腸障害を予防または治療するための方法を提供する。
本発明の別の形態においては、有効量のラクトバチルス・プランタルム亜種プランタルムPS128を個体に投与することを含む、個体の内臓過敏を予防または治療するための方法を提供する。好ましくは、前記内臓過敏は機能性胃腸障害に関連する。
本発明の一態様においては、前記機能性胃腸障害は、機能性腹痛、過敏性腸症候群、便秘、機能性下痢及び機能性消化不良からなる群から選ばれる。好ましい態様においては、前記機能性胃腸障害は、機能性腹痛、過敏性腸症候群または便秘である。
本発明のさらに具体的な実施例においては、ラクトバチルス・プランタルム亜種プランタルムPS128を投与した後、痛覚のバイオマーカーレベルは前記個体において統計学的に有意に低下する。好ましくは、前記バイオマーカーはP物質である。また、前記ラクトバチルス・プランタルム亜種プランタルムPS128投与した後、前記個体において筋電図信号が統計学的に有意に低下する。また、ラクトバチルス・プランタルム亜種プランタルムPS128を投与した後、筋電図信号は統計学的に有意に低下する。
好ましい実施例においては、前記単離乳酸菌を前記個体に経口投与する。
多数の実施例によって本発明を説明する。以下の実施例は本発明の範囲を限定するものと解してはならない。
ラクトバチルス・プランタルム亜種プランタルムPS128(以下、単にPS128と称する)は、台湾伝統のカラシナの発酵食品(中国表記「福菜」)から単離された。
ERIC−PCRを行い、さらに高い配列類似性を有する細菌亜種を識別した。
表1及び表2が示す条件でPS128のERIC−PCRプロフィールを行った。その菌株から抽出したDNAはテンプレートとして16S rDNAの増幅を行った。得られた増幅産物を電気泳動して、図1に示すようにそのパターンを比較した。プライマーは、SEQ ID NO:1及びSEQ ID NO:2が示すプライマーを使用した。

本発明はAPI50CHLキット(bioMerieux、フランス)でPS128の糖利用能を評価し、その結果を表3で示す。その発酵試験は、PS128がラクトバチルス・プランタルム亜種プランタルムと類似する生化学的特性を有することを示している。

(1)PS128の準備
PS128を培養液(10%脱脂粉乳、1%酵母粉、0.1%tween80、及び2%グルコース)に播種し、37℃で18時間培養した後、遠心分離して回収した。PS128は保護剤(脱脂粉乳1%、砂糖2%、オリゴフルクトース2%、マルトデキストリン3%、及びグリセロール2%)に埋め込んで凍結乾燥して最終濃度が1×1011コロニー形成単位(colony formation unit、以下単にCFUと称する)/g粉末に調整し、使用するまで−20℃冷蔵庫に貯蔵した。
(2)実験動物及び飼育
8週齢のC57BL/6JNarl雄マウスまたは定期的に妊娠するC57BL/6Jnarl雌マウスは、いずれも台湾国家実験動物センターから購入し、出産の約一週間前に、台湾国立陽明大学の病原体フリー動物センターの特定無菌室で、フィルターキャップ付き飼育ケージの中に、標準条件(温度22℃、湿度50〜60%に保持し、12時間の明暗周期)で飼育し、標準実験室食物(LabDiet Autoclavable Rodent Diet 5010, PMI Nutrition International, Brentwood, USA)と水を無制限に提供した。全部の動物実験工程は台湾国立陽明大学動物管理委員会により審査され、許可された。
従来研究を修正し、低温環境下の母性剥奪を早期生活ストレス(Millstein and Holmes 2007、Desbonnet et al., 2010)とした。即ち、新生仔の生後2〜14日目、室温下(〜22℃)で、毎日新生仔を親と兄弟から3時間(11:00〜14:00)分離させていた。それから2週間、毎週の床敷きの交換以外は邪魔しなかった。その後、ストレスを受けた幼若マウスを離乳させ、ケージあたり5〜6匹で無作為に割り当てた。雄の幼若マウスのみを実験に使用した(ストレス群は12匹で、ストレス群に109のPS128を与えた)。それら早期生活ストレスを受けていないマウスはコントロール群とした(コントロール群は10匹)。
(4)実験設計及びサンプリング
離乳(生後28日目)から8週齡までのマウスには、4週間連続で生理食塩水、またはPS128(109 CFU/匹/日)を投与した。これらのマウスに対し、照明期で最も軽微のストレスから最大ストレスへとオープンフィールド試験及び強制水泳試験などを含む一連の行動試験を行った。屠殺する当日、前記マウスから後眼窩採血して、ストレスの原因として頸椎脱臼の30分間前に短い強制水泳を行わせ、これらはいずれも11:00〜14:00の間で行ったため、日周リズムへの影響は減少した。前記マウスの脳を速やかに取り出し、特定領域の前頭葉前部皮質を氷上で切片にし、液体窒素で冷凍して、または0.6%過塩素酸緩衝液に保存した後、使用するまで−80℃に貯蔵していた。
オープンフィールド試験は、ラット/マウスの探索行動及び基本活動能力を測定する一般測定方法であり、さらに定性及び定量も同時にできる。
プレキシグラス壁を囲む二つの光束センサーリングを含む競技場(25.4×25.4×38 cm)を備えるオープンフィールド活動システム(Tru Scan activity system, Coulbourn Instruments, PA, USA)で、運動能力を監視して計算した。各マウスは同じコーナーから競技場に10分間置いて、全移動距離などを含むいくつかの活動を測定した。毎回試験終了後、前記競技場は水及び70%エタノールで洗浄し、前記マウスは元の飼育ケージに戻した。前記活動はTru Scan 2.2ソフトウェア(Coulbourn Instruments)で自動的に記録して定量した。
強制水泳は齧歯動物の行動試験であり、これらの憂うつ様状態を起こす或いは予防する抗憂うつ藥物、新規化合物の抗憂うつの治療効果、または実験操作を評価するためのものである。
強制水泳試験の評価方法は前記(Cryan et al., 2001)を修正したものである。即ち、不動程度(immobility)はビデオカメラで紀録し、マウスは透明なアクリルシリンダー(高さ30cm×内径10cm、深さ15cmの水を含み、水温は23±1℃に調整する)中に放り込み、さらにビデオ追跡ソフトウェアEthoVision(Noldus Information Technology, Wageningen, Netherlands)で、単回の6分間の強制水泳の内4分間(1〜5分)の不動程度を分析した。試験終了後、マウスはティッシュペーパーで乾燥させて元の飼育ケージに戻した。透明なアクリルシリンダー(高さ30cm×内径10cm)には深さ15cmの水を含み、水温は23〜25℃に調整し、単回の6分間試験に使用した。ビデオカメラでその行動を記録した。ビデオ追跡ソフトウェアEthoVision (Noldus Information Technology, Wageningen, Netherlands)で分析した。1〜5分の不動時間はいずれも同じパラメータで計算して人的エラーを避ける。試験終了後、前記マウスはティッシュペーパーで乾燥させて元の飼育ケージに戻した。
図2A及び2Bに示すように、オープンフィールド試験において、ストレスを受けたマウスはコントロール群と似ている全移動距離を有したが(図2A)、強制水泳試験においては、より多くの不動時間を有している(図2B)。ストレスを受けるマウスにPS128を投与すると、コントロール群マウスと同様な程度に、オープンフィールド試験における全移動距離が有意に上昇し、強制水泳試験における不動程度が有意に低下した(図2A及び2B)。*はコントロール群と比較してp<0.05であることを表し、$はストレス群と比較してp<0.05であることを表す。
血漿コルチコステロン濃度は、視床下部‐下垂体‐副腎系活性化の指標であり、ストレスまたは恐怖下での体−脳相互作用の基本反応である。
犠牲にする前にマウスから採取した血液サンプルを4℃、3000×gで10分間遠心分離して血清を得た。血清を適宜に希釈して市販コルチコステロン酵素免疫測定キット(CORT EIA kit,Cayman, USA)に加えて、そのキットが提供する標準品から内挿で濃度を算出した。
図3に示すように、血清濃度を測定することにより、本願発明者は、ストレスを受けるマウスにおいて、コルチコステロンは有意に上昇し、PS128(ストレス群には109を与える)を投与した後で逆転することを見出した。*はコントロール群と比較してp<0.05であることを表し、$はストレス群と比較してp<0.05であることを表す。
HPLC-ECDは、生物または環境サンプル、例えばカテコールアミン(ドーパミン、ノルアドレナリン、及びアドレナリン)、モノアミン(例えば、セロトニン、ドーパミン、ノルアドレナリン、及びアドレナリン)アセチルコリン、グルタミン酸、アルギニン、γ−アミノ酪酸、及び他の物質のルーチン測定に対して高性能を有する(Cheng et al., 2000)。
測定しようとする神経伝達物質は、ドーパミン(dopamine、以下単にDAと称する)、ジヒドロキシフェニル酢酸(dihydroxyphenylacetic acid、以下単にDCと称する)、及びホモバニリン酸(homo−vanillic acid、以下単にHVAと称する)を含む。前記高速液体クロマトグラフィー−電気化学検出器(HPLC-ECD)システムは前記(Cheng et al. 2000)のように、マイクロポンプ(CMA−100,CMA,Stockholm,Sweden)、オンライン注射器(CMA−160)、Microtech液体クロマトグラフィーポンプ(Micro−tech Scientific, Sunnyvale, CA)、及びBAS−4C電気化学検出器(Bioanalytical Systems, Inc., West Lafeyette, IN)を含む。逆相クロマトグラフィーカラム(Kinetex C18、2.6μm、100×内径2.1mm、Phenomenex、USA)で分析した。室温下(25℃)で、Ag/AgClの参照電極に対し、グラッシーカーボン作用電極の電位を+650mVに設定した。0.1Mリン酸二水素ナトリウム(NaH2PO4)、8%メタノール、0.74mMオクチル硫酸ナトリウム(SOS)、0.03 mMエチレンジアミン四酢酸(EDTA)、及び2mM塩化カリウム(KCl)を含む移動相緩衝溶液をリン酸でpH3.74に調整した。マウス頸椎脱臼後に直ちに全脳を収集して、ドライアイスで冷凍した。前頭葉前部皮質を解剖して0.6%過塩素酸緩衝液に保存した。超音波で均質化し、かつ12000×gで10分間遠心分離するまで−80℃に貯蔵した。分析する前、上清を0.22μmPVDF膜(4mmシリンジフィルター、Millex−GV,Millipore,USA)でろ過した。適宜に希釈した上清(20μl)を、流速0.2ml/分間でクロマトグラフィー系に注入した。以下の標準品:ドーパミン、ジヒドロキシフェニル酢酸、及びホモバニリン酸(Sigma−Aldrich, St. Louis, MO, USA)から、範囲1〜100ng/mlで、内挿でモノアミンの濃度を算出した。
図4A〜Cに示すように、早期生活ストレスプロトコルを適用することにより前頭葉前部皮質内の神経伝達物質が変化した。ドーパミン関連経路において、ドーパミン自体は低下し、かつその代謝物であるジヒドロキシフェニル酢酸はコントロール群と似ており(図4A及びB)、同時に、もう一つの代謝物であるホモバニリン酸は有意に低下した(図4C)。PS128を投与することで、ストレスを受けたマウスのドーパミン、ジヒドロキシフェニル酢酸、及びホモバニリン酸濃度は有意に上昇した(図4A〜C)。*はコントロール群と比較してp<0.05であることを表し、$はストレス群と比較してp<0.05であることを表す。
従来の研究では、覚醒ラットに5−ヒドロキシトリプトファン(5−HTP)(セロトニン前駆体)を皮下注射すると、VHを誘発したことが示されている。この研究では、5−HTP誘発VHモデルは、プロバイオティック株であるラクトバチルス・プランタルム亜種プランタルムPS128のラットに対する改善効果を評価するのに使用されていた。
PS128を培養液(10%脱脂粉乳、1%酵母粉、0.1%tween80、及び2%グルコース)に播種し、37℃で18時間培養した後、遠心分離して回収した。PS128は保護剤(脱脂粉乳1%、砂糖2%、オリゴフルクトース2%、マルトデキストリン3%、及びグリセロール2%)に取り込んで凍結乾燥して最終濃度が1×1011コロニー形成単位(CFU)/g粉末になるように調整した。PS128粉末を−20℃で貯蔵し、動物治療の前、食塩水に溶解して5×109CFU/mLとした。
(2)実験動物及び飼育
国家実験動物センター(NLAC、台北、台湾)から6〜8週齢のSprague-Dawley(SD)雄ラット(220〜330g)を購入した。これらのラットを恒温、恒湿、12時間の明暗周期で飼育し、水と餌を無制限に提供した。全部の動物実験工程は台湾国立陽明大学動物管理委員会により審査され、許可された。
(3)実験手順
図5Aを参照し、まず、電極植え込みのためにラットをイソフルランで麻酔した。電極植え込み手術の後、ラットを単独で飼育した。手術後約7〜10日目、ベースラインと5−HTP注射誘発の内臓過敏検知のため、ラットに結腸直腸膨張(CRD)試験を行った。この試験は、細菌を投与していない覚醒雄ラットにおける5−HTP−内臓過敏を検証するために設計された。10日目、CRD試験後の二週間にラットにPS128(一日あたり約109CFUの0.2mL食塩水)または食塩水(一日あたり200μl)を経口投与した。その後またCRD試験を行い、VHにおけるPS128の効果を測定した。CRD手順と筋電図(EMG)記録を同時に行った。EMG曲線下面積(AUC)はパラメーターとして計算した。脊髄におけるP物質のタンパク質濃度をウエスタンブロット法で検出するため、ラットをペントバルビタールナトリウム(65mg/kg、腹腔内注射)で麻酔して生理食塩水を経心的に(transcardially)灌流した。ラットの腹部感覚を担当するL6−S1脊髄を単離した。
ラットをイソフルランで麻酔した。少なくとも実験の7日前、テフロン(登録商標)被覆ステンレス鋼線(A−M systems inc.)の電極をラットの腹部外斜筋に埋め込んだ。頸部の背に電極を露出させた。
(5)結腸直腸膨張(CRD)試験
結腸直腸膨張(CRD)は、内臓敏感度の評価に広く使用され、かつ再現できる方法である。筋電図信号の測定はCRDにおける評価項目のひとつであり、かつ最も正確な定量試験項目である。
図5Bを参照し、CRD試験は5−HTP皮下注射(5mg/Kg)の前と後に行ない、5−HTP誘発内臓過敏を測定した。
CRD試験のためラットをプラスチックトンネル(直径6cm、長さ25cm)内に置いた。試験に先立つ3日間、実験条件に慣らすためにラットを一日あたり3時間単独にトンネル置いて訓練した。結腸直腸膨張(CRD)バルーンは直腸カテーテル(Medtronic Inc.)付着ラテックス手袋の指(長さ7cm)より構成される。バルーンを膨張して一晩放置することで、壁面の張力を平衡した。膨張装置を肛門管によって意識があるラットの直腸に完全に導入し、尾の基部に固定した。当該カテーテルをバロスタット(Medtronic Inc.)に接続し、バルーンを所望の圧力(20、40、または60mmHg)になるように30秒間の間隔を挟んで10秒間膨張させて結腸を膨張させた。各シリーズの間に5分間の間隔をあけ、各実験プロトコルについて膨張を4回繰り返した。CRDの反応をEMG装置で記録した。EMG信号分析は内臓運動反射(面積/秒間)としてSpike2(Cambridge Electronic Design Limited)で行われた。
図6Aに示すように、内臓運動は生のEMG信号から算出され、各群において圧力依存の方式で徐々に増加する。ラットにおけるベースライン信号に対し、5−HTP皮下注射により、CRDに反応するEMG信号が有意に増加している。
図6Bに示すように、二週間PS128投与のラットは5−HTP誘発EMG信号の上昇を抑制できるものの、5−HTPを注射した後、二週間食塩水を投与したら、EMG信号は依然として有意に上昇した。
さらに、PS128の投与がEMG信号の上昇を抑制する効果を確認するために、我々は5−HTP注射後10日目と24日目のEMG上昇の差を計算した。EMG差の数式は次のように示す。

図6Cに示すように、食塩水投与後、全部の膨張圧力刺激において、10日目のEMG信号の上昇と比べて、24日目のEMG信号の上昇がより大きい。それに対して、PS128群では、全部の膨張圧力刺激において、10日目のEMG信号の上昇と比べて、24日目のEMG信号の上昇が低い。これらのデータは、PS128が5−HTP注射誘発VHを改善する可能性を有することを示唆している。
ウエスタンブロット法でPS128の痛覚関連の脊髄神経細胞ペプチドに対する効果を確認するため、市販のタンパク質抽出キット(EC21 Inc.)によりL6−S1脊髄組織の全部のタンパク質を抽出した。抽出物を10%ポリアクリルアミドゲルで分別して電気泳動でフッ化ポリビニリデン膜(Roche Ltd.)に移し、次に、ブロッキングバッファ(5%脱脂粉乳を含むTBST)で1時間ブロッケージし、ブロッキングバッファの中の一次抗血清P物質(1:1000、GeneTex inc.)と一緒に4℃で一晩インキュベートした。TBSTで二回洗浄した後、膜をブロッキングバッファの中のヤギ抗ラットIgG−HRP(1:5000、Santa Cruz Biotechnology inc.)と一緒にインキュベートした。抗体−タンパク質複合体をイモビロンTMウェスタン化学発光HRP基質(Millipore inc.)で可視化してルミノ・イメージアナライザー(富士フイルムホールディングス株式会社)で検出した。
P物質はラット脊髄の痛覚のバイオマーカーである。これは、P物質の異常な発現レベルがIBSの病因に関与することを示唆する。なお、P物質含有神経経路は胃腸機能調節の役割を果たす神経経路の一つであると考えられる。図7に示すように5−HTPを注射していないコントロールラットは基礎レベルの生成を有しているものの、食塩水を二週間経口投与したラットは、CRD試験を経てから5−HTPを注射した後、L6−S1脊髄において高濃度のP物質の生成が発見された。二週間のPS128投与は5−HTP注射誘発P物質産生の上昇を抑制した。すなわち、PS128投与で、P物質のレベルはほぼ5−HTPを注射していないラットにおけるレベルにまで下がった。
ロペラミドはμ−オピオイド受容体のアゴニストであり、腸筋神経叢(myenteric plexus)におけるGタンパク質によりアデニルシクラーゼを抑制する結果、内在性アセチルコリンの放出を抑制する。ロペラミドは、腸神経系に直接に作用して便秘を起こすと推定され、便秘モデルマウスに頻用されている。
さらに、正常及びロペラミド誘発便秘マウスの両方の排泄におけるPS128の緩下効果が試験された。マウスにプラシーボ(0.2mL食塩水/マウス/日)またはPS128(109CFUの0.2mL食塩水/マウス/日)を経口投与した。二週間後、3時間における各マウスの糞便の湿質量を測定した。実験の便秘マウスに、塩酸ロペラミド(5mg/kg)を経口投与し、30分間後、3時間における各実験便秘マウスの糞便の湿質量の測定を開始した。図8に示すように、PS128群の糞便の湿質量は、プラシーボ群と比べて、有意に増加しているため、PS128の緩下効果を示唆している。これらの結果は、PS128は便秘を軽減する可能性があると示唆している。
各実験群の間の差異はPrism version 6(Prism、San Diego、CA)で分析した。ここに示す全部のデータは平均値±標準偏差(SD)で示されている。図2、3、4、及び7の統計的分析は一元配置分散分析(one−way ANOVA)で計算され、Bonferroni事後試験を行った。二標本t−検定は、EMG信号の差(図6)及びPS128の緩下効果(図8)の分析に使用された。p<0.05であれば、差異は統計的に有意と考えられる。各群の間の統計的有意差はp<0.05であれば、アスタリスク(*)またはドル記号($)で示し、p<0.01であれば、**で示し、p<0.0001であれば、***で示す。
本発明は、潜在株であるラクトバチルス・プランタルム亜種プランタルムPS128がストレスを受けるマウスに有益であることを見出し、乳酸桿菌株PS128を投与することにより、マウスの行動が改善されたと報告した。
なお、本発明は、PS128は内臓過敏及び便秘に有益であり、また、PS128は、5−HTP注射後、EMG信号の上昇を除去し、ラットの脊髄におけるP物質の生成の上昇を抑制することを見出した。
従って、本発明が提供する方法は、乳酸桿菌株PS128を投与することで、ストレス誘発失調または機能性胃腸障害を予防または治療することに有用なものである。
上記では本発明のいくつかの実施態様を詳細に述べてきたが、当業者は本発明の教示および利点を逸脱することなく示された特定の実施態様に種々の改変および変更を加えることができる。そのような改変および変更は特許請求の範囲に記載した本発明の主旨および範囲に含まれる。
Claims (14)
- 登録番号DSM 28632でドイツ微生物細胞培養コレクション(DSMZ)に寄託しているラクトバチルス・プランタルム亜種プランタルム(Lactobacillus plantarum subsp. plantarum)PS128である単離乳酸菌。
- 請求項1に記載の単離乳酸菌と、担体とを含む組成物。
- 請求項1に記載の単離乳酸菌を含む、個体の不安、憂うつまたは過敏性腸症候群を予防または治療するための組成物。
- 血清コルチコステロン濃度が個体において統計学的に有意(p値<0.05)に低下する、請求項3に記載の組成物。
- 神経伝達物質の濃度が個体において統計学的に有意(p値<0.05)に上昇する、請求項3に記載の組成物。
- 神経伝達物質がドーパミンである請求項5に記載の組成物。
- 請求項1に記載の単離乳酸菌を含む、個体の機能性胃腸障害を予防または治療するための組成物。
- 請求項1に記載の単離乳酸菌を含む、個体の内臓過敏を予防または治療するための組成物。
- 内臓過敏が機能性胃腸障害に関連する、請求項8に記載の組成物。
- 機能性胃腸障害が機能性腹痛、過敏性腸症候群または便秘である、請求項7または9に記載の組成物。
- 請求項1に記載の単離乳酸菌を含む、個体の機能性腹痛、過敏性腸症候群または便秘を予防または治療するための組成物。
- 痛覚のバイオマーカーとしてのP物質のレベルが個体において統計学的に有意(p値<0.05)に低下する、請求項7〜11のいずれか1項に記載の組成物。
- 個体の腹部外斜筋の筋電図信号が統計学的に有意(p値<0.05)に低下する、請求項7〜11のいずれか1項に記載の組成物。
- 経口投与用である、請求項3〜13のいずれか1項に記載の組成物。
