本発明の化合物の治療剤としての有効性は、適切な細胞結合剤の慎重な選択によって変わる。細胞結合剤は、現在公知である、または公知になるいずれの種類でもよく、ペプチドおよび非ペプチドを含む。細胞結合剤は、特異的または非特異的な様式のどちらかで細胞を結合することができるいずれの化合物でもよい。一般にこれらは、抗体(とりわけモノクローナル抗体)、リンフォカイン、ホルモン、成長因子、ビタミン、栄養素輸送分子(トランスフェリンなど)、または他のいずれかの細胞結合分子もしくは物質であることができる。

細胞傷害性コンジュゲートを形成するために本発明に使用され得る細胞傷害性剤には、メイタンシノイドおよびメイタンシノイド類似体がある。好適なメイタンシノイドの例は、メイタンシノールおよびメイタンシノール類似体を含む。メイタンシノイドは、微小管形成を阻害し、哺乳動物細胞に対して高い毒性である薬物である。

メトトレキサート、ダウノルビシン、ドキソルビシン、ビンクリスチン、ビンブラスチン、メルファラン、レプトマイシン誘導体、マイトマイシンC、クロラムブシル、カリケアマイシン、チューブリシンおよびチューブリシン類似体、デュオカルマイシンおよびデュオカルマイシン類似体、ドラスタチンおよびアウリスタチンなどのドラスタチン類似体などの薬物も、本発明のコンジュゲートの調製に好適である。薬物分子は、血清アルブミンなどの中間担体分子を通じて抗体分子に連結することもできる。例えば米国特許第6,630,579号に記載されたドキソルビシンおよびダウノルビシン化合物も、有用な細胞傷害性剤であり得る。

有効量の本発明の抗体、抗体フラグメントまたは抗体コンジュゲートおよび
不活性であり得るまたは生理学的に活性であり得る医薬的に許容される担体を備えた医薬組成物を提供する。
別の実施形態において,本発明は、EphA2受容体活性を阻害する方法であって、前記EphA2受容体と拮抗する抗体を、これが必要な患者に投与することによる方法を提供する。本発明のいずれの種類の抗体、抗体フラグメント、または細胞傷害性コンジュゲートも治療的に使用され得る。本発明はこのため、アンタゴニスト抗EphA2抗体、これのフラグメント、またはこれの細胞傷害性コンジュゲートの医薬品としての使用を含む。好ましい実施形態において、アンタゴニスト抗EphA2抗体は、2H11R35R74抗体またはこれのヒト化バリアントである。

RTにて磁気撹拌しながら、hu2H11R35R74 4ml(14.36mg/緩衝液A 1ml)、次に緩衝液A 7.5ml、HEPES 1M 1.45ml、DMA 1.14ml、続いてL−DM4−アリル−PEG4−CONHS活性化エステルの10mM DMA溶液0.3mlを添加する。RTにて30分後、L−DM4−アリル−PEG4−CONHS活性化エステルの10mM DMA溶液0.125mlをさらに添加する。RTにて1時間25分後、粗コンジュゲーション媒体をHGS緩衝液65mlで希釈して、TFFによりPellicon 3カセットで精製する。試料を、試料の体積の約10倍のHGS緩衝液でダイアフィルトレーションして、次に収集する。TFFタンクおよびラインをさらに添加のHGS緩衝液10mlで洗浄する。2つの溶液を混合して、Amicon 15で濃縮し、PVDF 0.22μmでフィルタ滅菌する。このためhu2H11R35R74−PEG4−アリル−DM4コンジュゲート8.0ml(c=5.22mg/ml)が得られた。次にコンジュゲートを最終薬物負荷およびモノマー純度について分析する。SEC分析(H):DAR(SEC)=5.3;RT=16.767分;モノマー純度=99.4%;HRMSデータ:図16を参照のこと。

hu2H11R35R74によるEphA2自己リン酸化活性の阻害
物質および方法
株化細胞および抗体
乳腺癌株化細胞MDA−MB−231をECAAC(参照# 92020424)から入手した。非小細胞肺癌株化細胞NCI−H1299をATCC(参照# CRL−5803)から入手した。両方の株化細胞を、10%熱不活性化ウシ胎仔血清および2mM L−Glnを添加したダルベッコ変法イーグル培地(DMEM)中で維持した。組換えマウスエフリン−A1細胞外ドメイン/Fcキメラ(エフリンA1/Fc)をSigma(参照# E9902)またはR & D Systems(参照# 602−A1)から入手した。抗Eck/EphA2クローンD7抗体(Ab)および抗ホスホチロシンAb4G10をMilliporeから入手した(それぞれ参照# 05−480および05−321)。chKTIアイソタイプ対照Abは、Immunogen Inc.にて生成された。これはIg重鎖および軽鎖定常領域がヒトκ軽鎖およびヒトγ1重鎖によって置換された、キメラ型の抗Kunitz SoybeanトリプシンインヒビタAb(KTI,ATCC 参照# HB9515)に相当する。組換えAbは、ImmunoGen(chKTI ロット# 2539−91)にて、重鎖および軽鎖について発現プラスミドを一過性にトランスフェクトしたHEK293T細胞の培養物上清から精製した。ヒト化抗EphA2 Abs hu2H11(ロット# LP08191)およびhu2H11R35R74(ロット# LP09077)は、Sanofi−Aventisにて安定的にトランスフェクトされたロンザ・チャイニーズ・ハムスター卵巣(CHO)/GS株化細胞から産生した。どちらのAbも標準手順に従って、タンパク質A sepharoseでの親和性クロマトグラフィーと、続くアニオン交換ロマトグラフィーにより培養物上清から精製した。追加のクロマトグラフィーステップは、セラミックのヒドロキシアパタイトで行った。すべての調製物を4℃の1×PBS中で貯蔵して、動態LAL法を使用して低い内毒素レベルについて試験した。抗アクチン抗体クローンC4は、Millipore(参照# MAB1501)から得た。ペルオキシダーゼ(PO)コンジュゲートヤギ抗マウスIgG Abは、Jackson Immunoresearch(参照# 115−035−003)から得た。
MDA−MB−231細胞をP100ペトリ皿(試料当り5皿)の完全培地中で、1皿当り3×106細胞にて播種して、CO2インキュベータ内で37℃にて48時間インキュベートした。細胞は18時間にわたって血清を欠乏させ、続いて10g/mL hu2H11またはhu2H11−R35/R74 または2μg/mLエフリンA1/Fcによって37℃にて10分間から2時間処置した。
血清が欠乏したMDA−MB−231またはNCI−H1299細胞をchKTI、hu2H11またはhu2H11R35R74(10μg/mL)によって37℃にて1時間インキュベートした。次に1μg/mLエフリンA1/Fcを添加して、細胞を37℃にてさらに10分から30分間インキュベートした。
細胞試料を掻爬によって氷上で収集して、15mL円錐管に移し、1300rpmにて5分間遠心分離した。1×リン酸緩衝食塩水(PBS;Invitrogen# 14190)で1回洗浄した後、細胞を1mMフェニルメチルスルホニルフルオリド(PMSF)、1×プロテアーゼ・インヒビタ・カクテル(Sigma参照# P2714)および1×Haltホスファターゼインヒビタ(Pierce参照# 78420)を即時添加した溶解緩衝液300μL(Biosource参照# FNN0011)に再懸濁させた。細胞抽出物をときどきボルテックス処理しながら氷上で45分間維持して、15000rpmにて10分間遠心分離し、さらに使用するまで−80℃にて維持した。タンパク質濃度は、Pierceによるキット(参照# 23227)を使用して、ビシンコニン酸(BCA)によって決定した。
細胞抽出物(試料当り0.3から0.5mg)のプレクリーニングステップは、溶解緩衝液中で先に平衡させた(ローテータ上で4℃にて1時間)、タンパク質G Sepharose 4 fast flow(GE Healthcare Life Sciences参照# 17−0618−01)を添加することによって行った。インキュベーションは、ローテータ上で4℃にて30分間行った。試料を1500rpmにて2分間遠心分離して、上清を収集し、抗EphA2 AbクローンD7(試料当り4μL)と共にローテータ上で一晩インキュベーションした。免疫沈降は、タンパク質G Sepharoseを用いてローテータ上で4℃にて4時間行った。試料を1500rpmで4℃にて2分間遠心分離して、溶解緩衝液100μLによって5分間、3回洗浄した。免疫沈降したビーズは、NuPAGE還元剤(Invitrogen参照# NP0009)を添加した4×NuPAGE LDS試料緩衝液50μL(Invitrogen参照# NP0007)に再懸濁させて、95℃にて5分間加熱し、1500rpmで5分間遠心分離して、−20℃にて維持するか、電気泳動を行った。
免疫沈降物または細胞抽出物を基準分子量マーカー(GE healthcare Life Sciences参照# RPN800)と共に4から12% Bis−Tris Midiゲル(Invitrogen# NP0322BOX)に投入して、電気泳動をMOPS−SDS緩衝液1×(Invitrogen参照# NP0001)中で150Vにて3時間行った。電気ブロットは、PVDF膜(Invitrogen参照# LC2007)上で、i−blot(商標)装置(Invitrogen)によりプログラム3を使用して行った。膜のブロッキングは、5%ウシ血清アルブミンを添加したTBST 1×(即ちトリス緩衝食塩水 Sigma参照# T5912、0.1%ツイン20 Sigma参照# P1379)中で行った。抗EphA2 AbクローンD7または抗ホスホチロシン4G10 Abによる標識を、同じ緩衝液中で4℃にて一晩行った。抗アクチンAbクローンC4による標識を室温にて1時間行った。続いての1×TBSTによる膜の洗浄後に、PO−コンジュゲートヤギ抗マウスAbおよびPerkin ElmerによるECLキット(参照# NEL 104001EA)を使用して、免疫ブロットの展開を行った。発光はFuji4000装置で読み取った。
結果
組換えエフリンA1/Fcを正の対照として使用して、MDA−MB231細胞におけるhu2H11またはhu2H11R35R74 AbによるEphA2のリン酸化の誘発について調査した。結果を図4に示す。リン酸化の誘起は、2つのAbのいずれによっても10分間から2時間で検知することができなかったが、組換えエフリンA1/Fcは、10分にてEphA2受容体の強いリン酸化を誘発し、シグナルの低下は1時間後に開始し、受容体の分解が2時間後に見られ始めた。
コンジュゲート抗EphA2抗体、hu2H11R35R74およびhu2H11の結合キャラクタリゼーション
裸であるかまたはPEG4−NHAcリンカーによってDM4にコンジュゲートされているかのどちらかの、抗EphA2抗体hu2H11およびhu2H11R35R74の固定されたEphA2−Fcとの相互作用を、表面プラスモン共鳴検出によりBIAcore 3000装置(GE healthcare,N°CH321)を使用して監視した。EDC(N−エチル−N’−[ジメチル−アミノプロピル]カルボジイミド)/NHS(N−ヒドロキシスクシンイミド)による標準アミン・カップリング・プロトコルを使用して、EphA2−Fc(10μg/ml;酢酸塩緩衝液pH=4.5)をC1センサチップのマトリクス(GE healthcare BR−1005−40)に10μl/分にてカップリングした。動的結合実験では、密度は応答レベルを約100応答単位(RU)上昇させて制御した。IgGは、0.15M NaCl、3mM EDTAおよび0.005% P20を含有する0.01M Hepes、pH7.4で希釈した。続いての希釈物はすべて同じ緩衝液で作製した。すべての結合実験は、25℃にて通例50から0.2nMの範囲に及ぶIgG濃縮物を流速50μl/分で用いて行った。

Lovo腫瘍細胞の阻害
Lovo腫瘍細胞(ウェル当り2000)を、完全血清含有培地中の96ウェル組織培養プレートに播種した。コンジュゲートを10−7から10−12Mの間の範囲に及ぶ濃度で段階希釈して3組のウェルに添加した。細胞を37℃/5% CO2にて抗体−細胞傷害性化合物コンジュゲートの存在下で5日間培養して、この時間の後、4時間のWST8アッセイを製造者の説明書(Dojindo Cell Counting Kit−8,カタログ#CK04)に従って行い、細胞の生存および成長を評価した。無細胞試薬ブランクを試験ウェル読み取り値から引いて、コンジュゲート処置細胞の読み取り値をビヒクル処置細胞の対照ウェルからの読み取り値の平均で割ることにより得られたデータを、生存画分としてプロットした。
ヒト化2H11R35R74−PEG4−NHAc−DM4による腫瘍細胞を発現するEphA2の成長の阻害
MDA−MB231およびSKMEL28の成長の阻害
対数成長期にある細胞をトリプシン処理して、これのそれぞれの培養培地に再懸濁させた(MDA−MB231細胞にはDMEM/F12 Gibco #21331;10% SVF Gibco# 10500−056;2nMグルタミンGibco# 25030;SKMEL−28細胞にはDMEM(Gibco# 11960)10%SVF Gibco# 10500−056;2nMグルタミンGibco# 25030)。完全血清含有培地中の96ウェルCytostar培養プレート(GE Healthcare Europe,#RPNQ0163)に、細胞懸濁物を5000細胞/ウェルの密度で(MDA−MB231、SKMEL−28)分散させた。4時間コーティングした後、コンジュゲートの10−7から10−12Mの間の範囲に及ぶ濃度の段階希釈物を3組のウェルに添加した。細胞を37℃/5%CO2にて抗体−細胞傷害性化合物コンジュゲートの存在下で3日間培養した。第4日に、14Cチミジンの溶液10μl(0.1μCi/ウェル(Perkin Elmer# NEC56825000)を各ウェルに添加する。実験を開始して96時間後に、14Cチミジンの吸収をmicrobeta放射線カウンタ(Perkin Elmer)で測定した。無細胞試薬ブランクを試験ウェル読み取り値から引いて、コンジュゲート処置細胞の読み取り値をビヒクル処置細胞の対照ウェルからの読み取り値の平均で割ることにより得られたデータを、生存画分としてプロットした。多くの実験において、裸の抗体(2H11または2H11R35R74)を実験開始時に1μMの濃度でウェルに添加して増殖の阻害を上記のように測定した。
表IIIに報告した結果は、2H11R35R74−PEG4−NHAc−DM4ならびに2H11R35R74−PEG4−Mal−DM4コンジュゲートが以前のコンジュゲートよりも、MDA−MB231細胞に対するインビトロ増殖阻害および抗原マイナス細胞(SKMEL−28)に対する選択性が良好であることを示唆する。
薬物動態研究
本研究は、hu2H11R35R74−PEG4−NHAc−DM4コンジュゲート(DAR=5.5)の薬物動態挙動を、CD−1マウス中のhu2H11−SPDB−DM4コンジュゲート(DAR=3.9)と比較して評価するために設計された。動物に各コンジュゲート20mg/kgを静脈内経路により投与して、血液を注射の0、8、24、48、72、120、168、240、336、504、672時間後に収集した。抗体薬物コンジュゲートの血漿レベルを測定して、標準条件下で基本単回用量薬物動態パラメータを制定した。コンジュゲートおよびこれの抗体構成要素(全抗体、コンジュゲート抗体および任意の脱コンジュゲート抗体の和)の血漿濃度を特異的ELISA技法によって測定した。
メスSCIDマウスに移植された原発性結腸腫瘍CR−LRB−004Pに対する、hu2H11R35R74−PEG4−NHAc−DM4およびhu2H11−PEG4−NHAc−DM4コンジュゲートの抗腫瘍効果
物質および方法

hu2H11−PEG4−NHAc−DM4−コンジュゲートおよびhu2H11R35R74−PEG4−NHAc−DM4の抗腫瘍効果を、メスSCIDマウスに皮下移植された、標的を強力に発現する測定可能な原発性結腸腫瘍CR−LRB−004Pに対する2つの用量レベルにて評価した。対照群は未処置のままであった。用量を1kg当りのタンパク質のミリグラムで表した。第15日に静脈内(IV)ボーラス注射によって、40および10mg/kgのhu2H11R35R74−PEG4−NHAc−DM4を投与した。DM4と等用量を与えるために、hu2H11−PEG4−NHAc−DM4を44および11mg/kgで投与した。

抗体薬物コンジュゲートの抗腫瘍活性に対するDARの効果
hu2H11R35R74−PEG4−NHAc−DM4を、メスSCIDに皮下移植された前立腺PC−3腫瘍に対して、6つの異なる薬物抗体比(DAR)で2つの低い有効用量を比較して評価した。対照群は未処置のままであった。用量を1kg当りのタンパク質のミリグラムで表した。DARをUV法によって決定した。第16日に、10および5mg/kgのhu2H11R35R74−PEG4−NHAc−DM4をそれぞれ3.4、4.4、5.9、6.2、7.4および8.4のDARにて、静脈内(IV)ボーラス注射により投与した。

2.1Å分解能でのhu2H11−R35−R74からのFabフラグメントと複合したEphA2受容体の細胞外ドメインの結晶構造の構造決定による、エピトープのマッピングおよびパラトープの同定
物質および方法
hu2H11R35−74のFabと複合したEphA2受容体−Fcのグリコシル化細胞外ドメインに対して、所期結晶化の試験を行った。トリプシンの存在下のみで結晶が得られた。これらの結晶を分析し、ペプチドマッピングはこれらがFabを含有し、EphA2およびFcの細胞外ドメインのN末端ドメインのある部分を含有していることを示した。複合体の別のバッチを産生し、今回はhu2H11R34−R74−Hisからの組換えFabフラグメントと複合した末端Hisタグを有するEphA2受容体のグリコシル化細胞外ドメインを使用した。EphA2受容体のどちらの構築物も、EphA2受容体とhu2H11R34−R75との間の複合体の同じ構造を提供する。
エピトープマッピング
図10:hu2H11R34−R75のEphA2受容体エピトープの細胞外ドメインのマッピングを示す(エピトープ残基は、hu2H11R34−R74 FabフラグメントのCDR残基のいずれかの原子から4Å以内に存在する原子を含有する残基として定義されている。)。
R74はインタフェースの近くに存在して、EphA2受容体の細胞外ドメインの外方を向き、最も近い細胞外ドメインから約10Å離れている。R35はパラトープ残基の1つであり、これはEphA2受容体の細胞外ドメインからのAsp53とH結合を作製する。リジン残基は抗原との同じ相互作用を非常に生じやすく、これのコンジュゲーションは抗体結合にとって明らかに有害である。
EphA2受容体の細胞外ドメインとhu2H11R35−R74との間の界面は、CDRループをすべて含むわけではない。図12からわかるように、主に重鎖CDRが結合に関与している。このことは、hu2H11のCDRにおける、特に軽鎖における変化が、ただし排他的ではないが、EphA2受容体への結合に悪影響を及ぼさずに導入できることを示唆している。軽鎖のループ3はインタフェースに全く関与していないが、L1末端の1残基(Arg35)のみがEphA2の細胞外ドメインと相互作用する。このことは、ループL3の変化がEphA2への結合に影響するはずがないことと、アミノ酸残基の変異/挿入がArg35残基の立体配座および向きを不安定化させない限り、ループL1がアミノ酸残基の変異/挿入に耐えるはずであることを暗示している。
hu2H11抗体は機能活性を示す:これはEphA2のエフリン−A1結合およびエフリンA1誘発リン酸化を阻害する。エフリン−A1と複合したEphA2受容体のLBDおよびCRDドメインの構造は公開されている(PDBコード3MBW)。本構造をhu2H11R35R74のFabフラグメントと複合したEphA2の構造に重ね合せるとき、2H11R35R74のFabフラグメントの軽鎖がEphA2上のエフリン−A1の結合範囲と重複し、このためhu2H11抗体の競合的性質が確認されることが明確に考えられる。

対数成長期にあるMDA−MB231細胞をトリプシン処理して、培養培地に再懸濁させた(DMEM/F12 Gibco #21331;10% SVF Gibco# 10500−056;2nMグルタミン Gibco#)。完全血清含有培地中の96ウェルCytostar培養プレート(GE Healthcare Europe,#RPNQ0163)に、細胞懸濁物を5000細胞/ウェルの密度で分散させた。4時間コーティングした後、コンジュゲートの10−7から10−12Mの間の範囲に及ぶ濃度の段階希釈物を3組のウェルに添加した。細胞を37℃/5%CO2にて抗体−細胞傷害性化合物コンジュゲートの存在下で3日間培養した。第4日に、14Cチミジンの溶液10μl(0.1μCi/ウェル(Perkin Elmer # NEC56825000)を各ウェルに添加する。実験を開始して96時間後に、14Cチミジンの吸収をmicrobeta放射線カウンタ(Perkin Elmer)で測定した。無細胞試薬ブランクを試験ウェル読み取り値から引いてコンジュゲート処置細胞の読み取り値をビヒクル処置細胞の対照ウェルからの読み取り値の平均で割ることにより得られたデータを、生存画分としてプロットした。多くの実験において、裸の抗体(2H11または2H11R35R74)を実験開始時に1μMの濃度でウェルに添加して増殖の阻害を上記のように測定した。
表IXに報告された結果は、試験した2H11R35R74DM4コンジュゲートはすべて、MDA−MB231細胞の成長を阻害するのに、hu2H11R35R74−Peg4−AcNH−DM4と同じくらい強力であることを示唆した。
SCIDメスマウスにおける結腸腺癌Lovoに対する2h11−DM4コンジュゲートの抗腫瘍活性についての異なるリンカーの評価
物質および方法
コンジュゲートの抗腫瘍活性の評価のために、動物の体重を毎日測定し、腫瘍をカリパスにより週2回測定した。腫瘍の重量は、塊体(mg)=[長さ(mm)×幅(mm)2]/2の式を使用して計算した。抗腫瘍活性の評価は、最大非毒性用量(HNTD)にて行った。
%腫瘍回帰は:規定の観察日の処置群における、第1の処置の第1日の腫瘍体積と比較した腫瘍体積減少%として定義する。特定の時点および各動物について、%回帰を計算する。次に群の中央値%回帰を計算する。
異なる切断不能なリンカーhu2H11−R35R74−PEG4−AcNH−DM4、hu2H11−R35R74−PEG8−AcNH−DM4、hu2H11−R35R74−PEG4−AcNMe−DM4、hu2H11−R35R74−PEG4−アリル−DM4およびhu2H11−R35R74−アセチル−DM4を有する抗体薬物2h11−DM4コンジュゲートの抗腫瘍活性は、メスSCIDに皮下移植された結腸Lovo腫瘍に対する同じ用量のDM4/kg 600μgを比較して評価した。対照群は未処置のままであった。用量を1kg当りのDM4のマイクログラムで表した。コンジュゲートを静脈内(IV)注射により第14日に投与した。
Claims (27)
- EphA2受容体に特異的に結合して、少なくとも1つの重鎖および少なくとも1つの軽鎖を備える抗体またはそのエピトープ結合フラグメントであって、
前記重鎖が、配列番号12から成るアミノ酸配列と少なくとも95%の同一性を有する配列を含み、配列番号1、2および3によって表されるアミノ酸配列を有する3個の連続相補性決定領域を含み、74位にアルギニンを含み、
前記軽鎖が、配列番号14から成るアミノ酸配列と少なくとも95%の同一性を有する配列を含み、配列番号4、5および6によって表されるアミノ酸配列を有する3個の連続相補性決定領域を含み、
但し、細胞傷害性剤にコンジュゲートされたとき、コンジュゲーションは抗体の親和性に影響を与えない、前記抗体またはそのエピトープ結合フラグメント。 - 細胞傷害性剤にコンジュゲートされたとき、KDが、5.6から8.4の範囲に及ぶ薬物対抗体比で、裸の抗体と有意に異なっていない、請求項1に記載の抗体またはそのエピトープ結合フラグメント。
- 前記重鎖が、配列番号12から成るアミノ酸配列と少なくとも97%の同一性を有する配列を含み、前記軽鎖が、配列番号14から成るアミノ酸配列と少なくとも97%の同一性を有する配列を含む、請求項1に記載の抗体またはそのエピトープ結合フラグメント。
- 前記モノクローナル抗体またはそのエピトープ結合フラグメントがヒト化もしくは表面再処理抗体またはそのエピトープ結合フラグメントである、請求項1に記載の抗体またはそのエピトープ結合フラグメント。
- 前記重鎖が配列番号12から成るアミノ酸配列を含み、および前記軽鎖が配列番号14から成るアミノ酸配列を含む、請求項1に記載の抗体またはそのエピトープ結合フラグメント。
- 前記重鎖が配列番号18のアミノ酸配列に存し、および前記軽鎖が配列番号16のアミノ酸配列に存する、請求項1に記載の抗体またはそのエピトープ結合フラグメント。
- 前記細胞傷害性剤が抗体に共有結合されている、請求項7に記載のコンジュゲート。
- 細胞傷害性剤が式(XIII)のメイタンシノイドである、請求項7または8のいずれかに記載のコンジュゲート。
- nが4から7の間で構成される、請求項10に記載の抗体薬物コンジュゲート。
- 式(XV)の構造を有する、請求項11に記載の抗体薬物コンジュゲート。
- コンジュゲートを調製する方法であって:
(i)細胞結合剤の水溶液を細胞傷害性化合物の溶液と接触させるステップと;
(ii)次に、(i)で形成されたコンジュゲートを未反応試薬および溶液中に存在し得るいずれの凝集体からも分離するステップと;を備え

- 癌が膀胱、乳房、結腸、腎臓、肝臓、肺、卵巣、膵臓、胃、子宮頸、甲状腺および皮膚の癌腫を含む;扁平上皮細胞癌腫を含む癌腫;白血病、急性リンパ性白血病、急性リンパ芽球性白血病、B細胞リンパ腫、T細胞リンパ腫、バーキットリンパ腫を含む、リンパ系の造血器腫瘍;急性および慢性骨髄性白血病ならびに前骨髄球性白血病を含む、骨髄系の造血器腫瘍;線維肉腫および横紋筋肉腫を含む、間葉起源の腫瘍;黒色腫、精上皮腫、奇形癌(tetratocarcinoma)、神経芽細胞腫および神経膠腫を含む他の腫瘍;星状細胞腫、神経芽細胞腫,神経膠腫および神経鞘腫を含む、中枢および末梢神経系の腫瘍;線維肉腫、横紋筋肉腫(rhabdomyoscarama)および骨肉腫を含む、間葉起源の腫瘍;ならびに黒色腫、色素性乾皮症、角化棘細胞腫(keratoactanthoma)、精上皮腫、甲状腺濾胞癌および奇形癌を含む他の腫瘍の間から選ばれる、請求項16に記載の使用。
- 前記モノクローナル抗体またはそのエピトープ結合フラグメントのパラトープが軽鎖内に:ループ1のArg35、L2のTyr54、Arg58およびAsp60を備える、請求項1から6のいずれか一項に記載の抗体またはそのエピトープ結合フラグメント。

- 前記モノクローナル抗体またはそのエピトープ結合フラグメントのパラトープが位置:Thr H28に変異を備える、請求項1から6のいずれか一項に記載の抗体またはそのエピトープ結合フラグメント。
