本発明はまた本明細書に記載のプロテアーゼ変異体をコードする分離されたポリヌクレオチドを提供する。更に、本発明は本明細書に記載のポリヌクレオチドを含む、プロモータと動作可能に組み合わされた発現ベクターを提供する。
本明細書で用いる「プロテアーゼ」(protease)及び「タンパク質分解活性」(proteolytic activity)はペプチド又はペプチド結合を持つ基質を加水分解する能力を示すタンパク質又はペプチドを言う。タンパク質分解活性を測定するための多くの知られた手順が存在する(例えば、Kalisz,「微生物プロチナーゼ」(Microbial Proteinases)Fiechter (編纂), Advances in Biochemical Engineering/Biotechnology, [1988]を参照)。例えば、タンパク質分解活性は比較アッセイにより確認しても良く、それにより市販の基質を加水分解するプロテアーゼの各能力を分析する。その様なプロテアーゼ及びタンパク質分解活性の分析で有用な代表的な基質はジメチル カゼイン(Sigma C-9801), 牛のコラーゲン (Sigma C-9879), 牛のエラスチン(Sigma E- 1625)及び牛のケラチン (ICN Biomedical 902111)を含むがこれに限定されるものではない。これらの基質を使用する熱量アッセイは技術分野で良く知られている(例えば、WO 99/34011及び米国特許第6,376,450号を参照願いたい。)pNAアッセイ(例えば、Del Mar他、Anal Biochem, 99:316-320 [1979]を参照願いたい)はまた、勾配溶離中に回収される部分の活性酵素濃度を決定するのに用いることが出来る。このアッセイは、酵素が可溶合成基質、スクシニルーアラニンーアラニンープロリンーフェニルアラニンーp−ニトロアニリド(sAAPF-pNA)を加水分解するに連れてp−ニトロアニリンが放出される速度を測定する。加水分解反応から黄色の生成される速度が分光光度計において410nm波長で測定され、これは活性酵素濃度に比例する。さらに280nmでの吸光測定は全タンパク質濃度の決定に用いることができる。活性酵素/全タンパク質の比率は酵素純度を示す。
アクチノバクテリア網(Class Actinobacteria)のメンバーとして分類されるグラム陽性細菌であるセルロモナス属内の全ての種を指す。セルロモナス属は分類状の再編成を受けていると認められる。この様に、属は再分類された種を含むことが予定されている。
Qβ レプリカーゼの場合, MDV-I RNAがレプリカーゼの特異鋳型である(例えば、 Kacian 他、Proc. Natl. Acad. Sci. USA 69:3038 [1972]を参照)。他の核酸はこの増幅酵素によっては複製されない。同様に、T7 RNAポリメラーゼの場合、この増幅酵素はそれ自身のプロモータに厳格な特異性を持つ(Chamberlin他、Nature 228:227 [1970]を参照)。T4 DNAリガーゼの場合、この酵素は、結紮接合点でオリゴヌクレオチド又はポリヌクレオチド基質及び鋳型間にミスマッチのある場合、2つのオリゴヌクレオチド又はポリヌクレオチドを結紮しない(Wu 及びWallace, Genomics 4:560 [1989]を参照)。最後に、Taq 及びPfuポリメラーゼは、高温で機能するという能力のため、プライマにより画され(bounded)、それにより規定される配列に高い特異性を示すことが判明しており、高温はプライマが標的配列とハイブリッドするのに都合が良いが、非標的配列とハイブリッドするにはそうはならない熱力学条件を作り出す。
「分離された」(又は、単離された)(isolated)又は「精製された」(purified)はその元の環境(例えば、もし天然(自然)に発生する場合は自然環境)から取出された材料を言う。例えば、材料は、それが特別の組成物中で、天然に発生する又は野生型有機体に存在する、又は天然に発生する又は野生型有機体から発現して通常存在しない組成物と組み合わされたものよりも、より高い又は低い濃度で存在する場合「精製されている」と言う。例えば、生きている動物中で天然に発生するポリヌクレオチド又はポリペプチドは分離されていないが、天然のシステム中の共存する材料の一部又は全部から分離されている同じポリヌクレオチド又はポリペプチドは分離されている。ある実施の態様においては、その様なポリヌクレオチドはベクターの一部であり、及び/又はその様なポリヌクレオチド又はポリペプチドは組成物の一部であり、及びさらにその様なベクター又は組成物はその自然環境の一部でないと言う意味で分離されている。ある好ましい実施の態様において、例えば、もしそれが電気泳動ゲル又はブロットで本質的に一つのバンドを生成する場合は核酸又はタンパク質は精製されていると言われる。
多数の部位評価ライブラリー(Site Evaluation Library)は成熟タンパク質の各アミノ酸が殆んどの他のアミノ酸により置換されているNprEのために構築された(米国特許出願第10/576,331号及びWO 2005/052146を参照願いたい)。
本明細書に記載の様に、BMIミクロ材料見本アッセイでの洗濯能力及び酵素の表面の全体の電荷の間の関係が決定された。本発明の方法は種々の酵素及びタンパク質(例えば、アミラーゼ、セルラーゼ、オキシダーゼ、クチナーゼ、マンナーゼ、ペクチナーゼ、リパーゼ、プロテアーゼ及び他の酵素)の能力を向上させるために使用することができる。
AmyS-S242Qについて、成熟酵素の4つの位置に組み合わせ置換を導入することにより組み合わせ電荷ライブラリーが構築された。このライブラリーはBODIPYデンプン加水分解、 米デンプンミクロ材料見本洗浄能力、及び酵素発現のためにスクリーニングされた。その後スクリーニングデータは変異により起こった電荷の変化の効果が分析された。操作されたAmyS-S242Q変異体においてはタンパク質発現及び良好な酵素の能力の増加が望ましい。しかしこれらの特性は逆相関することが判明した。本発明は良好な 米デンプン洗浄能力を示す、より高度に発現された変異体を生成する手段を提供する。この様に本発明は過度に他のパラメータを犠牲にすることなく発現又は酵素活性を増大させる変異を同定する手段を提供する。

実施例1
アッセイ
以下に記載のアッセイが以下に述べる実施例で実施された。以下に記載の手続きからの逸脱部分については実施例に記載する。これらの実験では反応の終了後に形成された製品の吸光度の測定に分光光度計を用いた。材料見本の反射率の測定には反射率計を使用した。
1.タンパク質含有量決定のためのBCA(ビシンコニン酸)アッセイ
これらのアッセイでは、マイクロタイタープレート(MTP)スケール上でプロテアーゼサンプル中のタンパク質濃度の決定にBCA (Pierce)アッセイが用いられた。このアッセイシステムでは使用された化学品及び試薬溶液は:
BCA タンパク質アッセイ試薬及びPierce Dilution 緩衝液 (50 mM MES, pH 6.5, 2mM CaCl2, 0.005% TWEEN(登録商標)-80)である。
このアッセイでは、Bradford染料試薬(Quick Start)アッセイがMTPスケールでプロテアーゼサンプル中のタンパク質濃度を決定するために用いられた。
使用した機器はEppendorf Thermomixer及びSpectraMAX (type 340:分子装置) MTPリーダーを含む。MTPはCostar (type 9017)製であった。
Milli-Q水が6 gpgの硬度(Ca/Mg=3/1)に調製され、0.78 g/1 TIDE(登録商標)2X Ultra, Clean Breeze洗剤が加えられた。洗剤は製剤中に存在する全ての酵素を不活性にするために1時間95℃で事前に予備加熱された。洗剤溶液は15分攪拌された。その後5 mM HEPES(遊離酸)が加えられ、pHが8.2に調整された。
0.25インチ円状直径のミクロ材料見本はCFT Vlaardingenから得た。材料見本を切断する前に、織物(EMPA 116)が水で洗濯された。一つのミクロ材料見本が各96ウエルマイクロタイタープレートの各ウエルに置かれた。
所望の洗剤溶液が上に述べた様に生成された。Thermomixerを25°Cで平衡させた後、190 μlの洗剤溶液がMTPのミクロ材料を含む各ウエルに加えられた。この混合物に10 μlの希釈酵素溶液が添加され、最終酵素濃度がlμg/ml(BCAアッセイにより決定)とした。MTPはテープでシールされ培養器に30分間置かれ、1400rpmで攪拌された。適当な条件下で培養するのに続いて、各ウエルから100 μlの溶液が採られ新たなMTPに移された。各ウエル100 μlの溶液を含む新しいMTPはMTP SpectraMaxリーダーを用いて405nmで読み取られた。ミクロ材料見本及び洗剤を含むが酵素を含まない実験対照(コントロール)及び何も含まない対照(ブランクコントロール)も含んでいた。
BMIの能力の計算
得られた吸光度は空試験値(すなわち、酵素の存在しない場合のミクロ材料見本を培養した後に得た)に対比し修正された。得られた吸光度は試験された酵素の加水分解活性の測定値を与えるものであった。
野生型及び変異プロテアーゼの安定性が25%の熱処理されたTIDA(登録商標)2x Ultra Clean Breeze液体洗濯洗剤中でので培養ステップを経た後に測定された。当初及び残留活性が以下に述べるAGLAアッセイ、蛍光96ウエルプレートリーダー、培養器/振とう器(iEMS; Thermoelectron)及び培養器/振とう器(Innova; New Brunswick (type 4230))を用いて決定された。MTPはCostar (type 9017)及びGreiner (black plates, type 655076)製であった。
このアッセイシステムで使用された化学品及び試薬溶液は以下の通りである:
TIDE(登録商標) 2x Ultra Clean Breeze
125 g のTIDE(登録商標)2x Ultra Clean Breeze (上と同じく95°Cで熱処理)を50 gの50 mM HEPES pH 8.2及び275 ml 水の混合したものに溶解させた;TIDE(登録商標)の濃度は上澄み25 % (以下に"TIDE(登録商標)"と呼ぶ)で希釈した後27.7%であった。

AGLA基質

AGLA基質溶液
16 ml N,N ジメチルフォルムアミド(dimethylformamide)に溶解させた451 mgのAGLA;この溶液は304 mlのMES緩衝液 (52.6 mM MES/NaOH, 2.6 mM CaCl2, 0.005% TWEEN(登録商標)-80, pH 6.5)に攪拌しつつ注いだ。
ストレスのない条件
まず、20 μl ろ過培養液が180 μl MES 希釈緩衝液により希釈された。その後20 μl のこの希釈培養駅が180 μl MES希釈緩衝液により希釈された。その後に 10 μl のこの希釈培養駅が25℃で事前に暖められたプレートで190 μl AGLA-基質溶液で希釈された。全ての空気泡が取り払われそしてプレートがAGLA プロテアーゼアッセイ手順に従い測定された。
まず20 μl ろ過培養液が180 μl TIDE(登録商標)洗剤溶液で希釈されiEMS振とう器中で5分事前混合された後にさらにInnova 振とう器で培養した。プレートは32℃で、200rpmで回転振動しつつ全60分の間培養された。さらに20 μl ろ過培養液が180 μl TIDE(登録商標)洗剤溶液で希釈されiEMS振とう器中で5分事前混合された後にさらにInnova 振とう器で培養した。プレートは20℃で、200rpmで回転振動しつつ全40分の間培養した。その後20 μlのこれらの溶液のいずれかが180 μl MES希釈緩衝液で希釈され、そして10 μlのこの希釈液は25℃で事前に暖められたプレートで190 μl AGLA-基質溶液で希釈された。全ての空気泡が取り払われそしてプレートがAGLA プロテアーゼアッセイ手順に従い測定された。
蛍光測定が励起350 nm及び放射415 nmで行われた。蛍光分光光度計用ソフトが相対蛍光単位(RFU)ミリ/分の線形回帰線に従って各ウエルの蛍光の増加の反応速度を計算した。
D. アミノベンゾイル−L−アラニングリシル−L−ロイシン−アラニノー4−ニトロベンジルアミド プロテアーゼアッセイ(Abz-AGLA-Nba)
以下に述べる方法は、時間と場所に係わりなく再現可能なプロテアーゼアッセイデータを与えるあるレベルの技術の詳細を提供する。このアッセイは与えられた実験室条件に適合させることができるが、変更された手順により得られたデータは全てもとの方法により得られる結果と調整させねばならない。
52.6 mM MES/NaOH, 2.6 mM CaCl2, pH 6.5 − MES緩衝液
MES酸 (10.28 g) 及び292 mg 無水CaCl2が約900mLの精製水に溶解された。
約28 mgのAbz-AGLA-Nbaが小さな管に入れられ、DMF(容量は集められたAbz-AGLA-Nbaにより変わる)で溶解され数分間攪拌された。溶液は光から遮断され室温で保存された。
アルファ溶液
1mL Abz-AGLA-Nba原液が19 mL MES 緩衝液に加えられ攪拌された。溶液は光から遮断され室温で保存された。
この緩衝液は5 mL精製水を95 mL MES緩衝液に加えて生成された。
5mL の純粋なDMFが95 mL MES 緩衝液に加えられた。この緩衝液は反応速度パラメータを決定するために用いられた。
酵素原料溶液が酵素希釈緩衝液により1 ppm (1 mg/mL)濃度に希釈された。
まず全ての緩衝液、原液及び希釈標準溶液が準備された。特に断らない限り各酵素希釈液剤は3回アッセイされた。まったく一杯というわけではないが、酵素希釈標準溶液原料マイクロプレートがプレートの左から(プレートリーダーを収めるために)垂直解カラムを満たす様に準備された。対応するアッセイプレートは同様jに設定された。マイクロプレート蛍光光度計が先に述べた様に設定された。
バチルス スブチリスにおけるNprEの生成
この実施例では、バチルス スブチリスでNprEプロテアーゼを生成する実験について述べる。特にバチルス スブチリスにpUBnprEプラスミドを形質転換するために用いられる方法について述べる。形質転換は技術分野で知られた方法で実施された(例えば、WO 02/14490及び米国特許出願第11/581,102号を参照)。以下に記載するDNA配列(バチルス アミロリケファシエンス由来のnprE リーダー, nprE プロ(pro)及びnprE成熟DNA配列)はNprE前駆体タンパク質をコードする:
PCR手順:
1) 30 秒98°C;
2) 10秒98°C;
3) 20秒55°C;
4) 1 分 72°C;
5) 2 から4のステップを25サイクル;
6) 5 分72°C.;
この結果1.9 kb DNA断片が生成され、このDNA断片はBgIII及びBcII DNA制限酵素を用いて消化された。複数コピーバチルスベクターpUBl10 (例えば、Gryczan, J. Bacterid., 134:318-329 [1978]を参照)はBamHIにより消化された。PCR断片xBgIIIxBcII はその後pUB110 x BamΗIベクター中で結紮されpUBnprE発現ベクターを形成した。
部位評価ライブラリー(SEL)の生成
この実施例においてはnprE SELsの構築で使用される方法が記述される。
pUB-Bglll-RV GTCTCGAAGATCTGATTGCTTAACTGCTTC (配列番号7).
各SELの構築は、pUB-BgIII-FWプライマー及び特異nprE逆突然変異誘発プライマーを用いて2つの主PCR増幅により開始された。第2のPCRではpUB-BgIII−Rvプライマー及び特異nprE前進突然変異誘発プライマー(前進及び逆突然変異プライマーのための同等nprE 成熟コドンの位置が使用された。
主PCR1:

PCR融合プログラムは以下の通り:
30秒98℃, 30x (10 秒 98℃, 20 秒 55℃, 2.40分 72℃)及び5 分72℃、PTC-200 Peltier 熱サイクル(MJ Research)で実施された。
(Qiagen, Cat. No. 28106)を用いて精製され、融合断片の両端に付着端を生成するためにBgIII制限部位で消化された:
- 35 μL 精製線状DNA 断片
- 4 μL REACT(登録商標)3緩衝液 (Invitrogen)
- 1 μLBglII, 10 単位/ml (Invitrogen)
反応条件:1時間、30℃
BgIIIによる消化及びQiaquick(登録商標)PCR精製キット(Qiagen, Cat. No
. 28106)を用いて精製された断片の結紮により、所望の突然変異を含む環状及び多重結合DNAが生成された:
- 30μL 精製BglII消化DNA 断片
- 8 μL T4 DNAリガーゼ緩衝液 (Invitrogen(登録商標)Cat. no. 46300-018)
- 1 μL T4 DNA リガーゼ, 1 単位/μL (Invitrogen(登録商標)Cat. No.15224-017)
反応条件:16-20時間、16℃
その後、結紮された混合物はバチルス スブチリス(ΔaprE, ΔnprE, oppA, ΔspoIIE, degUHy32, ΔamyE::(xylR,pxylA-comK)株に形質転換された。バチルス スブチリスへの形質転換は、WO 02/14490に記載の様に実施された。各ライブラリーについて、96単一コロニーが採取され、配列分析(BaseClear)及びスクリーニングの目的のために、ネオマイシン及び1.25 g/L イーストエキスを含むMOPS媒体中で成長させた。各ライブラリーは最大19 nprE部位特異変異体を含んでいた。

本明細書に記載のこれらの方法は関心の対象となる他の酵素のSELの生産に適している(例えば、Asp)がこの実施例では、nprE SELを生成する他の方法について述べる。

方法
一つの突然変異(上の実施例4及び米国特許出願第11/581,102号に記載の様に、nprE SELにより同定される)を含むpUBnprEプラスミドを得る為に、関心の対象となる各バチルス株単一コロニーが5ml LB + l0ppm ネオマイシン管(例えば、開始培養液)に植菌するために用いられた。培養液は37℃で、225rpmで6時間振とうさせて成長させた。その後、100 mlの新しいLB + l0ppmネオマイシンが1mlの開始培養液により植菌された。この培養液は、225rpmで振とうさせつつ37℃で一夜成長させた。この培養に続いて、細胞ペレットを得るために十分な遠心分離を行うことにより細胞ペレットが収集された。細胞ペレットは10 ml 緩衝液Pl (Qiagen Plasmid Midi Kit)中で再けん濁された。その後l0ulのReady-Lyse Lysozymeが再けん濁細胞ペレットに添加され、37℃で30分間培養された。細胞培養液の増加させるためである10ml の緩衝液P2及びP3を使用してQiagen Plasmid Midi Kitの手順が続けられた。単一nprE変異を含む各pUBnprEプラスミドがバチルス(Bacillus)から分離された後各プラスミドの濃度が決定された。プラスミドはその後製造者の指示に従いdam Methylase Kit (New England Biolabs)を用いてdamメチル化され一つの管当り約2 ug のpUBnprEプラスミドがメチル化された。チモクリーン ゲルDNA(Zymoclean Gel DNA)回収キットがdam メチル化されたpUBnprE プラスミドを精製し及び濃縮するために用いられた。dam メチル化されたpUBnprEプラスミドは数値化され、その後各50ng/ulの作業濃度に希釈された。混合された部位特異突然変異誘発プライマが各反応のため別個に準備された。例えば、pUBnprE T14R プラスミドを鋳型ソースとして用いて、混合された部位特異突然変異誘発プライマ管は10 μl のnprE-S23R, 10 μl nprE-G24R, 10 μl nprE-N46K, 及び10 μl nprE-T54R (全てのプライマは各10 μM)を含む。QuikChange Multi Site-Directed Mutagenesis Kit(多部位特異突然変異誘発キット)(Stratagene)を用いたPCR反応が以下の製造者指示に従い実施された(例えば、一つの突然変異を含む1 μl damメチル化されたpUBnprE プラスミド(50 ng/μl), 2 μl nprE 部位特異突然変異誘発プライマ(10 μM), 2.5 μl 10x QuikChange Multi Reaction 緩衝液, 1 μl dNTP Mix, 1 μl QuikChange Multi 酵素ブレンド(2.5U/μl),及び17.5 μl 蒸留加圧滅菌水で全反応混合物量で25 μlとなる)。
変異プロテアーゼの発現、発酵、精製及び特性評価
この実施例では、先行する実施例の形質転換されたバチルス スブチリスで発現されたプロテアーゼを発現、発酵及び精製するために用いられる方法について述べる。
プロテアーゼ活性及び安定性に対する突然変異の効果の平衡
この実施例では2つの競合する酵素特性を最適化するために操作された複数置換プロテアーゼ変異体について記載する。
アミラーゼ活性及び発現に対する変異効果の平衡
この実施例では2つの競合する酵素特性が複数のアミノ酸置換を導入することにより同時に最適化することができることを示す。
AmyS-S242QプラスミドDNAが形質転換されたバチルス スブチリス(Bacillus subtilis)株(遺伝子型:ΔaprE, ΔnprE, amyE::xylRPxylAcomK-phleo)から分離され、CCL構築の鋳型としてDNA2.0 Inc.に送られた。 DNA 2.0 Inc. (Mountain View, CA) に対してAmyS-S242Q (S242Q)アミラーゼの4つの部位の各々に、位置ライブラリーを生成する様要請された。変異体は96ウエルプレートでグリセロール原液として供給された。
AmyS, S242Q 又はAmyTS23t発現ベクターを含むバチルス スブチリス (Bacillus subtilis)クローンがグリセロール原料を150 μl of LB媒体 + 10 μg/ml ネオマイシンを含む96ウエル培養プレートに入れたものからスチール製96ウエル複製器により複製され、湿気を与えた密封した中で220rpm、37℃で一夜成長させた。一夜媒体から100 μlを採り5mlプラスチック管中に2000 μl の内容の明らかな媒体 + l0μg/ml ネオマイシンを接種するのに用いた。培養媒体はMOPS緩衝液に基づく濃縮された準定義済みの媒体であり、尿素を主要な窒素源とし、グルコースを主要な炭素源とし、強力に細胞を成長させるために1% ソイトーン(soytone)及び5 mMカルシウムで補強された。
本明細書に記載の様に、アルファ−アミラーゼの濃度及び比活性度が抑制多クローン抗体の滴定により決定された。バチルス ステアロテルモフィルス (Bacillus stearothermophilus)由来のアルファ−アミラーゼ(AmyS)対する多クローン抗体はAmyS及びバチルス種由来のアルファアミラーゼTS23の強い抑制作用を持つことが判明した(例えば、結合は活性ロスの線形滴定を作りだすほどに堅いものである)。したがって、この抗体は酵素の濃度を測定するために用いることができ、その濃度は更に比活性度を計算する点に使用される。端的に言えば、幾つかの知られている抗体濃度により生成される酵素抑制量が測定される。この情報から完全な抑制に必要な抗体濃度が推定される、それはサンプル中の酵素濃度に等しい。アルファ−アミラーゼ活性及び抑制は蛍光BODIPYデンプンアッセイを用いて測定された。緩衝液は0.005% Tween-80を含む50 mM MOPS, pH 7.0であった。
Bodipy(有機ホウ素色素)デンプンアッセイをEnzChek(登録商標)Ultra Amylase Assay Kit (E33651, Invitrogen)を使用して行った。DQ デンプン基質の1 mg/mL原液が100 μLの50mM、pH4.0の酢酸ナトリウム中の凍結乾燥された基質を含む小瓶の内容量を溶解することにより作られた。小瓶が約20秒撹拌され、暗室に、室温で置かれ、溶解するまで時々混合された。900 μLのアッセイ緩衝液 (2.6 mM CaCl2 pH 5.8を含む50 mM酢酸ナトリウム)が加えられそして小瓶は約20秒撹拌された。基質溶液は4℃で使用可能になるまで、暗室に、室温で保存された。アッセイのため、DQ基質の希釈標準溶液100 μg/mLがアッセイ緩衝液中の1 mg/mL基質溶液から作られた。190 μLの100 μg/mL基質溶液が96-ウエル平底マイクロタイタープレートの各ウエルに添加された。10 μLの酵素サンプルがウエルに加えられ、800 rpmでサーモミキサーを用いて30秒混合された。緩衝液及び基質のみを含む(酵素を含まない)ブランクサンプルがアッセイに含まれていた。蛍光強度の変化率が蛍光マイクロタイタープレートリーダーで25℃で5分間測定された(励起:485nm、蛍光:520nm)。
アミラーゼ変異体がCS−28 米デンプンミクロ材料見本と共に又はそれを使用せずに標準洗濯条件で30分培養された。遊離型酵素の量がBodipyデンプンアッセイにより測定された。ミクロ材料見本に結合している酵素の割合が以下の様に計算された:結合された割合=(材料見本が存在しない場合の酵素活性−材料見本が存在する場合の酵素活性)/(材料見本が存在しない場合の酵素活性)。
本発明の開発中に確認された様に、AmyS-242Qの中央発現値はプラスの電荷が増えると低下した。しかし、特定のBodipyデンプン加水分解はプラスの電荷が増えると増大した。強化された組み換えアミラーゼの発現及びデンプンの加水分解は、例えば、燃料エタノール産業又は洗剤を使用する洗浄におけるデンプン液化にとり好適なAmyS-242Qの操作された変異体では望ましい。しかしこれらの特性は競合する特性である様に思われる。本発明の開発中に確認された様に、本発明の方法を用いて単一変異を選択的に組み合わせることによりデンプンの加水分解をひどく損なうことなくより高度に発現されたアミラーゼ変異体を生成することが可能である。本明細書に記載の戦略は、第一の特性(例えば、発現を主要特性として)が改良された複数置換されたAmyS-242Q変異体を生成しそして選択するために上手に用いることができ、他方第二の特性(デンプンの加水分解を第二の特性として)を改良し又は犠牲にすることがない。
Claims (17)
- 改良された酵素変異体の生成方法であって、前記方法は操作可能な順序に従い:
a) 複数の単一置換された酵素変異体を第一の特性の第一の試験及び第二の特性の第二の試験において試験を行い、その場合親酵素が各試験で1.0の値を持つとすると、有利な第一又は第二の特性は1.0よりも大きな値を持ち、過度に不利な第一又は第二の特性は0.8未満の値を持ち;
b) 有利な第一の特性に関連しているが、過度に不利な第二の特性に関連していない、単一置換された酵素変異体の少なくとも一つの置換を同定し;
c) 有利な第二の特性に関連しているが、過度に不利な第一の特性に関連していない、単一置換された酵素変異体の少なくとも一つの置換を同定し;及び
d) ステップb) からの置換及びステップc)からの置換を酵素に導入して複数置換の酵素変異体を得て、前記複数置換の酵素変異体は改良された酵素変異体である、各ステップを含み、
i)前記親酵素がプロテアーゼであり、前記第一の特性が安定性であり、第二の特性が洗浄能力であり;または
ii) 前記親酵素がアミラーゼであり、前記第一の特性がタンパク質の発現であり、第二の特性が酵素活性であり;及び
前記少なくとも一つの置換は親酵素に比べて0、−1、又は−2のネット電荷の変化を持ち、及び前記少なくとも一つの置換は親酵素に比べて+1又は+2のネット電荷の変化を持ち、及び改良された酵素変異体は第一及び第二の試験の両方において1.0より大きい値を、又は第一の試験において1.0より大きい値を、第二の試験において0.80から1.0の値を達成する変異体である、前記方法。 - 請求項1の方法であって、さらにe) 複数置換された酵素変異体を第一及び第二の試験において試験を行い、その場合改良された酵素変異体は第一及び第二の試験の両方において1.0より大きい値を又は第一の試験において1.0より大きい値を、第二の試験において0.80から1.0の値を達成するステップを含む前記方法。
- 更に、前記改良された酵素変異体を生成することを含む請求項2の方法。
- 前記第一及び第二の特性が逆相関する、請求項2の方法。
- 前記有利な第一又は第二の特性は1.2より大きい値を持つ、請求項2の方法。
- 前記過度に不利な第一及び第二の特性は0.60未満の値を持つ、請求項2の方法。
- 前記過度に不利な第一及び第二の特性は0.40未満の値を持つ、請求項6の方法。
- 前記安定性は洗剤組成物中の安定性を含み、洗濯能力は血液、ミルク、インク(BMI)洗濯能力を含む、請求項1の方法。
- 前記洗濯能力は5及び12の間のpHを持つ粉末又は液状洗剤組成物で試験される、請求項8の方法。
- 前記洗濯能力は塩基pHを持つ冷水液状洗剤で試験される、請求項8の方法。
- 前記プロテアーゼは中性金属プロテアーゼ及びセリンプロテアーゼから選択される、請求項1の方法。
- 前記セリンプロテアーゼはスブチリシンである、請求項11の方法。
- 前記中性金属プロテアーゼはバチルス科(Bacillaceae)のメンバーから得た中性金属プロテアーゼである、請求項11の方法。
- 前記アミラーゼはバチルス科(Bacillaceae)のメンバーから得たアルファアミラーゼである、請求項1の方法。
- 前記置換の位置は25%より大きい溶媒の接近可能な表面(SAS)を持つ親酵素中の位置である、請求項1の方法。
- 前記置換の位置は50%より大きい、又は65%より大きい溶媒の接近可能な表面(SAS)を持つ親酵素中の位置である、請求項1の方法。
- 前記親酵素は野生型酵素である、請求項1の方法。
