- Separation Using Semi-Permeable Membranes (AREA)
Description
6.66%の固形分含有量を有するホエイパルミエートの流れが、アニオン交換樹脂(XA3112(Applexion製)、塩化物型)上を、そしてカチオン交換樹脂(XA 2033(Applexion製)、ナトリウム型)上を続けて流された。この工程において、適用される搭載量に依存して、ある量のホスフェート及びカルシウムが、それぞれ、塩素及びナトリウムイオンに交換されることが見出された。約25床量(BV)のホエイパルミエートが使用されるとき、存在するカルシウムイオンの約90%及び存在するホスフェートの約50〜60%が除去されることが見出された。典型的なパルミエート軟化の実験データが表1にまとめられる。
実施例1で得られた生成物が、ナノろ過を使用して濃縮された。ナノろ過は、NF−2540膜(ダウ)で実行された。ろ過工程の間、20〜25バールの範囲の膜間圧力が使用された。濃縮物は、膜の上で出発物質の連続再循環により得られた。その結果、濃縮物の固形分が増加した。このようにして、濃縮物の固形分が>15%に設定されたが、20%の固形分が到達されたとき、ろ過を留めることも可能であった。このようにして、存在する鉱物の大部分が除去された。典型的なナノろ過工程の実験結果が表2にまとめられる。
実施例2で得られた生成物の中で、約20%の固形分含有量を有する200リットルが以下のカラム上を連続的に通過された。
カラム1は25リットルのカチオン交換体(Applexion 748H)を含んでいた。
カラム2は25リットルのアニオン交換体(DIAION SA20)を含んでいた。
Claims (15)
- 脱脂された乳の流れから、シアル酸結合オリゴ糖を単離する方法において、該乳の流れが、30〜48%の水分含有量を有する遊離の塩基型の第一のアニオン交換樹脂を使用する第一のアニオン交換工程に付され、続いて、該第一のアニオン交換工程において得られた透過液が、該シアル酸結合オリゴ糖を吸着するために第二のアニオン交換樹脂を使用する第二のアニオン交換工程に付される、前記方法。
- 該乳の流れがたんぱく質を含まない、請求項1に記載の方法。
- 該シアル酸結合オリゴ糖がシアリルラクトースである、請求項1又は2に記載の方法。
- 該第二のアニオン交換樹脂がマクロ孔性又はゲルタイプのアニオン交換樹脂である、請求項1〜3のいずれか1項に記載の方法。
- 該第二のアニオン交換樹脂が45%超の水分含有量を有する、請求項1〜4のいずれか1項に記載の方法。
- 該第二のアニオン交換樹脂が50〜70%の水分含有量を有する、請求項1〜5のいずれか1項に記載の方法。
- 該乳の流れとしてホエイの流れが使用される、請求項1〜6のいずれか1項に記載の方法。
- 該乳の流れとしてホエイパーミエートの流れが使用される、請求項1〜7のいずれか1項に記載の方法。
- 該乳の流れが、該第一又は該第二のアニオン交換工程のいずれかに付される前に軟化される、請求項1〜8のいずれか1項に記載の方法。
- 該軟化がナトリウム/塩素型の複数のイオン交換樹脂を使用して行われる、請求項9に記載の方法。
- 該軟化が、カルシウム及びホスフェートイオンを沈殿させることにより実行される、請求項9に記載の方法。
- 該乳の流れが、該第一又は該第二のアニオン交換工程のいずれかに付される前に、ナノろ過に付されて、絶対量で、より少ない数の単独イオン及び非たんぱく質の窒素化合物を有する濃縮物が形成される、請求項9〜11のいずれか1項に記載の方法。
- シアル酸結合オリゴ糖を含む組成物において、少なくとも4重量%の、固形分に基づくシアリルラクトースの含有量を有し、かつ6より大きいシアリルラクトース/リンの比を有し、且つ、請求項1〜12のいずれか1項に記載の方法を使用して得られたものである、前記組成物。
- 該シアル酸結合オリゴ糖がシアリルラクトースである、請求項13に記載の組成物。
- 幼児用食品、臨床食品又は経腸食品において、請求項13又は14に記載の組成物を使用する方法。
