Description

[0044] 膜は、混合物からのガスの分子分離、および液体からの化学物質の分子分離を含めた、多くのさまざまなタイプの分子分離に対して有用である。有望な需要先は、木質バイオマスを糖類、有機酸、およびアルコールに転換するバイオリファイナリーである。最新の膜技術によれば、糖類を酢酸やフルフラールから分離することができる。しかしながら、フルフラール化合物を酢酸から分離する新たな膜技術が求められている。蒸留ではなく膜による分子分離の大きな利点は、主としてエネルギー節約の面でコストがより低いことである。こうした膜を使用することができる他の工業としては、石油産業、石油化学工業、石炭ガス化工業、紙・パルプ工業、及び天然ガス製造業などがある。
[0053] 分子分離用セラミック膜(以下これを「分子分離膜」と呼ぶ)は下記のように製造される。管状のセラミックナノ濾過膜(以下これを「ナノ濾過膜」と呼ぶ)を、分子分離膜を形成するための基材として使用する。DNA分子を含有するゾル−ゲル中にナノ濾過膜を浸漬することで、ナノ濾過膜上にコーティングが形成される。次いで、ゾル−ゲルで被覆されたナノ濾過膜を強い磁場の中に置く。この磁場により、DNA分子がナノ濾過膜の表面に対して垂直に整列する一方で、ゾルゲルが重合してセラミック膜を形成する。ゾル−ゲルが凝固した後に、DNA分子をカ焼によって除去すると、セラミック膜中に細孔が残り、これによって分子分離膜が得られる。

[0057] i.基本原理: 液晶状態は、結晶固体のように整えられているが、液体のように流動する物質相である。DNAのようなポリアニオンを高密度で充填することは、ホスフェート基の電荷−電荷反発が、対イオンを加えることによって最小限に抑えられる場合にのみ達成することができる。二本鎖DNAの150塩基対長さを有するヘキサゴナル液晶相の構造は、小角中性子散乱(Dai2007)によって研究されている。この研究では、ヘキサゴナル液晶相に対し、一価のテトラメチルアンモニウム(TMA+)イオンが、液晶状態の形成を容易にするための対イオンとして使用された。この状態におけるDNAの長軸間の間隔は4nmと測定された。最大で100持続長(〜5μm)のDNAのセグメントは、局所的なヘキサゴナル構造を表わすことが示された。
[0063] DNAテンプレート化膜の合成
[0064] DNAをテンプレートとして使用する新しい手法の特徴は、DNAを巧みに処理して、膜表面に対して垂直な細孔をつくり出せる可能性にある。膜細孔の整列は、カ焼前のDNAの整列状態に依存する。DNA整列のための幾つかの方法が検討されている。最適の方法は、技術のスケールアップに対し、コスト効率の良い方法で実施する上で最も簡単な方法である。整列方法が水性環境にていったん確立されたら、該整列方法を、ゾル−ゲル出発物質の存在下で確認する。成功は、これはクロス偏光顕微鏡(cross−polarized microscopy)によって確認される、ゾル−ゲル中での配向液晶DNAの凝固によって決定される。

[0080] 細孔整列の代替法としては、最初は一本鎖DNAをアルミナに付着させるために開発された方法によって、魚の精子の二本鎖DNAの末端を表面に化学的に結合するという方法がある(Saprigin 2004)。アミノシランN−(2−アミノメチル)−3−アミノプロピルトリメトキシシラン(AEAPS)を使用して、アルミナ基材の表面をシラン化することができる。AEAPSの末端第一アミンは、カルボジイミド架橋メカニズムによってDNAの末端ホスフェートに共有結合する。結果として、約100塩基対の長さを有する魚の精子の二本鎖DNA(一端が表面に結合している)の単一層が得られる。中性pHにおいて第一アミンは正に帯電し、DNAは負に帯電するので、DNAは表面(第一アミンを含有する)上に横たわりやすい。この影響は、DNAを正に帯電したアミノシランで前処理することによって最小限に抑えることができる。さらに、アミノシランアミノプロピルトリエトキシシラン(APTES)も、DNA分子に近接してゾル−ゲルの形成を容易にする。DNAの結合単分子層は、薄すぎるために効果的な分離膜として機能することができないが、その代わり、後続する液晶DNAテンプレート化材料の層を整列させるための指向剤として作用する。

[0085] フェーズI/IBプロジェクトにより、DNAテンプレート化膜を形成するのに必要なテンプレート法と浸漬被覆法が開発された。この開発作業は、スライドガラスを膜用基材として使用して行われた。このプロトタイプ膜は、平面状であり;セラミック材料で構成され;並びにマクロポーラスゾーン、メソポーラスゾーン、およびマイクロポーラスゾーンで多層化されている;という特徴を有する多孔性材料によって支持される。プロトタイプの開発に対し、ゼオマトリックスは、Inocermic GmbHから供給されるγ−アルミナ基材、及びフェーズI/IBプロジェクト時に開発された被覆法を使用する。基材が多孔性であることにより、細孔配列と流量の測定が可能となる。単一ガスのパーミアンス、細孔のサイズ分布、気孔率、及び選択的な層厚さ等の膜特性も調べる。多孔性基材を浸漬被覆するための条件は、表面接着特性が異なることから非多孔性基材と同じではないことがある、ということが考えられる。多孔性基材は、Inocermic GmbHから購入することができる。複合アルミナ基材は、直径が2cm、厚さが3mmで、10nmの平均細孔径を有する。これらの基材は、10nmの多孔性γ−アルミナがより高多孔性のα−アルミナ層上に存在する形で構成される。Z−SEP(商標)プロトタイプ膜は、カプセル封入DNAゾル−ゲル前駆体溶液(フェーズI/IBに関して開発された方法に従って調製されている)中に多孔性基材を浸漬して浸漬被覆することによって形成される。これらの膜を乾燥し、次いでフェーズI/IBに関して開発された方法に従ってカ焼する。下側の基材に応じたZ−SEP(商標)層の焼結を含む最終プロセスのために、さらなる他の方法を開発する必要がある。こうした方法の開発は、別のタスク(タスク3)に分類される。

[0088] この研究フェーズにおいて、ゼオマトリックスは、コンサルタントであるWilliam DeSisto教授とともに作業する。DeSisto教授は、支持テンプレート化膜の浸漬被覆と特性決定において9年の経験を有する(Higgins 2006,Kennard 2008)。ゼオマトリックスが使用する方法は、教授が自分の研究において使用している手順を基にして適合させた方法であり、液晶DNAゾル−ゲル材料の溶液中に基材を浸漬することからなる。フェーズI/IB時の初期浸漬被覆作業は、メーヌ大学のDeSisto研究室の装置を使用して行った。フェーズIIの作業は、ゾル−ゲル溶液を通しての、より正確でより再現性の高い基材の「引き出し(draw)」を可能にする特注装置を使用して、ゼオマトリックスの設備で行う。基材が数秒間にわたって材料中に置かれ、これにより表面に対するゾル−ゲルの結合が可能となる。次いで基材を所定の一定速度で引き出すと、均一な厚さの膜層が得られる。厚さは、ゾル−ゲルの粘度だけでなく引き出し速度にも影響される(Brinke and Hurd 1994)。乾燥工程時に、膜の欠陥が取り込まれることがある。したがって、温度と湿度が制御されたチャンバー〔エレクトロ−テックシステムズ社から購入〕内にて浸漬被覆プロセスを行うことによって蒸発速度を調整する。浸漬被覆の速度と角度に対する正確で且つ再現性のある制御を可能にするために、環境チャンバー内に被遠隔制御モーターを据え付ける。
[0091] 膜厚は、SEMを使用して得られる断面画像によって測定する。分岐ヘキサンの吸着ポロシメトリーまたは吸着パームポロメトリー(Clark 2003,CaO 1993,Cuperus 1992)を使用して、欠陥を定量化し、平均細孔径と細孔径分布を測定する。この方法は、凝縮性蒸気による制御された細孔ブロッキングを測定することによって、そして膜を通過する別の非凝縮性ガスのフラックスを測定することによってアクティブな細孔(膜を横切っている細孔)を測定する。ブロッキング剤として作用するよう、ヘキサンの蒸気分圧は広い範囲にわたって変動する。ヘリウムガスの流量を、ヘキサンの活動度(hexane activity)の関数として測定する(ヘキサンの蒸気圧をヘキサンの飽和圧力に応じて標準化する)。ヘキサンの活動度は、ケルビン式(式中、σはヘキサンの表面張力であり、Vmはヘキサンのモル体積であり、Rはガス定数であり、Tは温度であり、aはヘキサンの活動度であり、tは吸着された単分子層の厚さである)を使用して細孔径Rpに関係づけることができる(Cuperus 1992)。

[00101] i.基本原理 亀裂や基材からの層間剥離などのZ−SEP(商標)膜中の欠陥は、プロジェクトの次の段階を始める前に解決しておかなければならない。欠陥の形成は、浸漬被覆/乾燥工程またはカ焼/焼結工程時に起こりうる。パーベーパレーション試験(タスク4)時にZ−SEP膜を評価するために、欠陥は最小限でなければならない。より高い流量とより良好な分離係数との組み合わせは、分子流が主として、膜の細孔を通って、そしてより大きくてより選択性の低い欠陥を通らないで起こるときに達成される。
[00103] 選択的シリカ膜層を基材に結合させなければならない。焼結はこの結合を達成するための一般的な方法であるが、細孔サイズを維持しようとすると、焼結温度が限定される。多孔性シリカ膜中の細孔の構造が変わり始める温度は約600℃からである(Tatsu 2005)。細孔の崩壊を避けるために、DNAの除去は可能な最も低い温度で行わなければならない。熱重量分析(TGA)により、加熱したときのサンプルの塊の変化をモニタリングすることが可能である。いったんDNAが完全に除去されたら、DNAを除去するのに必要な最低温度を設定する。減衰全反射フーリエ変換赤外分光法(ATR−FTIR)により、表面上の固体物質の化学組成の分析が可能であり、ATR−FTIRは膜の組成をモニターするのに使用される。膜を焼結するにはより高い温度が使用される。膜の細孔特性は、焼結温度の関数としてモニターされる。これらの実験は、メーヌ大学のDeSisto教授の研究室において行うことができる。これらの温度により、我々には、Z−SEP(商標)生成物に対する最適焼結温度を決定するための操作範囲がわかる。欠陥の除去がこの工程の目的である。選択層の亀裂、浮き、及び離層は実際に起こりうることである。
[00108] ATR−FTIRスペクトルは、DNAテンプレート材料の存在もしくは除去に対する証拠を与える。タスク2において略述した細孔特性、パーミアンス、及び欠陥に関するデータの分析と解釈がそのままタスク3に当てはまる。このタスクにおける異なったアプローチは、欠陥を最小限に抑えるべく手順を系統的に適用することである。圧力が増大するにつれてパーミアンスの増大が観察されることがあるが、これは欠陥を通過するガス流量によるものである。パーミアンスは、膜の両端間の圧力低下(ΔP)の関数としてグラフ化される。右上がり勾配は欠陥が存在していることを示す。パーミアンスvs.ΔPの勾配がゼロということは欠陥が存在していないこと示す。欠陥の数と程度が低下すると、パーミアンスvs.ΔP線の勾配が減少する。
[00110] 問題: 欠陥の除去は、膜のパーミアンスの大幅な減少を引き起こす。これは、多くの要素からなる浸漬被覆法によるものと考えられる。DNAテンプレートを除去するために膜層をカ焼した後に、残された細孔が次のゾル−ゲル浸漬で詰まってしまう、ということが起こりうる。
