Description
〔1〕特定の抗原受容体遺伝子について遺伝子再構成後の遺伝子型を有する機能的な分化細胞の製造方法であって、
該遺伝子型を有する幹細胞を培地中で培養して、該幹細胞由来の分化細胞を得ることを含む、方法;
〔2〕該遺伝子型を有する幹細胞が、該遺伝子型を有する細胞の核の移植により樹立された幹細胞である、上記〔1〕の方法;
〔3〕幹細胞がES細胞である、上記〔1〕又は〔2〕の方法;
〔4〕抗原受容体がT細胞受容体である、上記〔1〕〜〔3〕のいずれかの方法;
〔5〕T細胞受容体がNKT細胞特異的T細胞受容体である、上記〔4〕の方法;
〔6〕NKT細胞の核を除核卵母細胞に移植することにより樹立されたES細胞を培地中で培養して、NKT細胞系譜の細胞を得ることを含む方法である、上記〔1〕の方法;
〔7〕NKT細胞系譜の細胞がNKT細胞である、上記〔6〕の方法;

ntES細胞
末梢Tリンパ球の一種であるNKT細胞由来の核移植(nuclear transfer:nt)ES細胞としては、NKT細胞より直接核移植法によって先に樹立したES細胞(WO2006/018998参照)のうちクローン7の細胞を用いた。
細胞培養に用いたOP9細胞培地は、MilliQ水1リットルにつき10gのαMEM(Gibco社)、2.2gの重炭酸ナトリウム(和光純薬)、ペニシリン、ストレプトマイシン(Gibco BRL)、20%(v/v)の牛胎児血清(FCS)(Equitech Bio Inc,Kerrville,TX)を含むように調製した培地を、Corning社のボトルフィルター(0.45μm)に通して滅菌することにより作製した。
OP9細胞は、独立行政法人理化学研究所 バイオリソースセンター Riken Cell Bankより購入した細胞(RCB1124)を使用した。notchのリガンドであるdelta−like 1を強制発現させたOP9細胞(OP9−dlk細胞)は、既報(Schmitt et al., Nature Immunology 5: 410-417 (2004))に従い調製した。マウスdelta−like 1は、RT−PCR法によりマウス胸腺のcDNAライブラリから取得した。IRES−ヒトNGFRの構造を持つレトロウイルスのIRESの上流にdlkを挿入し、OP9細胞に形質導入した後、抗ヒトNGFRに対するモノクローナル抗体を用いてその発現を確認した。
NKT細胞由来のntES細胞(クローン7)は、20%FCS、ペニシリン、ストレプトマイシン(Gibco BRL)、LIF(Chemicon社,2×103U/ml)を含むDMEM培地(Sigma社)中において、マウス胎児線維芽細胞上で培養維持された。またOP9細胞及びOP9−dlk細胞は、90%コンフルエントで1/4に分割し継代維持された。ntES細胞との共培養に供されるときは完全にコンフルエントになった後さらに2日以上おいたものを使用した(以下、分化誘導用OP9細胞及びOP9−dlk細胞と称する)。
NKT細胞はFACS(Fluorescence activated cell sorting)法によって確認した。具体的には、FITC標識された抗マウスTCRVβ(PharMingen社)並びにPE標識されたα−ガラクトシルセラミド(αGalCer)がのせられたCD1dダイマー分子(PharMingen社)の双方で染色されるもの(αGalCer−CD1dダイマー+)をNKT細胞と規定した。またネガティブコントロールとしてビヒクル分子がのせられたPE標識CD1dダイマー分子を用いた。
LIFの存在下クローン7を3.5cm培養皿で20−30%コンフルエントまで培養したものをリン酸バッファー(PBS)で2回洗浄後、TE(トリプシン/EDTA,Sigma社)を加えて37℃のインキュベーター中で5分静置後、電動ピペットでよく攪拌し500×gで5分間遠心分離して上清を除去後、細胞数を計測した。これらの細胞をOP9細胞培養液で懸濁し、分化誘導用のOP9細胞もしくはOP9−dlk細胞上に撒いた(各1.0×105/10cm培養皿)。その後、37℃、5%CO2のインキュベーター中に3日静置させ培養液を交換した。さらに2日間培養を継続し、mesoderm(中胚葉)群の細胞を誘導した。これらの中胚葉細胞群はTE処理によってOP9細胞もしくはOP9−dlk細胞と分離された。共培養細胞はPBS、4mlで二度洗浄した後、培養皿一枚当たり4mlのTEを入れ37℃、5%CO2のインキュベーター中に5分間静置後8mlのOP9細胞培養液で中和し電動ピペットでよく攪拌し再び37℃、5%CO2のインキュベーター中に30分間静置した。上清を回収して細胞数を計測した後、1.0×106の中胚葉細胞群を新たに分化誘導用のOP9細胞もしくはOP9−dlk細胞上(10cm培養皿)に撒き直した。この時から、OP9細胞培養液中にはマウスflt3−リガンド(flt3L)(R&D社)を5ng/mlの濃度で添加した。その後2日おきに培養液並びにflt3Lを交換した。また共培養開始後7日目からはマウスIL−7(R&D社)、ヒトIL−15(R&D社、OP9−dlk細胞との共培養時のみ)も同時に各5ng/mlの濃度で培養液中に添加し、培養液とともに2日ごとに交換した。この状態で血液・リンパ球系の幹細胞(hematoblast,lymphoblast)を出現させた。その後これらの細胞数が数百万になるまで培地交換とサイトカインの交換を続けた。その後分化誘導用のOP9細胞もしくはOP9−dlk細胞の6ウェルプレートに1ウェル当たり1×106の濃度で細胞を撒き直し、cobble stone(未熟なT、B細胞のコロニー)を形成させた(共培養開始後13−16日で出現)。cobble stoneがOP9細胞もしくはOP9−dlk細胞を全て覆い尽くすまで上記のように培地交換とサイトカインの交換を続けた。その後はこれらのcobble stoneの細胞を電動ピペットでよく攪拌しOP9細胞もしくはOP9−dlk細胞から分離し、新たに分化誘導用OP9細胞もしくはOP9−dlk細胞上(10cm培養皿)に撒き直した。その後も培地、サイトカイン交換を2日ごとに行い培養を継続した。さらに細胞数が10cm培養皿あたり1×108を超えた時点での別の分化誘導用OP9細胞もしくはOP9−dlk細胞(10cm培養皿)上に分割し、増殖させた。
本実験では核移植技術によってできたntES細胞(クローン7)と並行して通常の受精卵より樹立したES細胞を用いて実験が正常に進行しているか否かを確認した。即ち、後者の細胞を用いるとOP9−dlk細胞上ではCD4+CD8+の二重陽性T細胞(DP細胞)が産生され、OP9細胞上ではB細胞が生ずるはずである。
これらの細胞は抗マウスCD19、及び抗マウスGr−1モノクローナル抗体(PharMingen社)によって確認した。OP9細胞上ではクローン7からも通常のES細胞からもCD19+(B細胞マーカー)、Gr−1+(顆粒球マーカー)の細胞が出現するのに対してOP9−dlk細胞を用いた場合ではこれらの細胞は生成しなかった。
先の報告(Schmitt et al., Nature Immunology 5: 410-417 (2004))で示されているように通常のES細胞(M1)をOP9−dlk細胞上で培養・分化させた場合はCD4+CD8+のDP細胞産生が見られた。一方、クローン7をOP9−dlk細胞上で培養・分化させた場合はこれらDP細胞に加えてCD8+SP細胞、CD4−CD8−細胞などの細胞の出現が見られた。この時細胞を抗マウスTCRVβとαGalCer−CD1dダイマーで染色するとクローン7由来のリンパ球の80%以上がNKT細胞(TCRVβ+/αGalCer−CD1dダイマー+)であった。一方、ネガティブコントロールとしてビヒクル分子がのせられたCD1dダイマー分子を用いるとこれらの細胞は染色されなかった。また通常のES細胞からできたDP細胞中にはこのような細胞は出現してこなかった。以上の結果より、末梢の遺伝子再構成の済んだNKT細胞の核の核移植により作製したES細胞(ntES細胞)を用いると非常に高い効率でTCRVβ+αGalCer−CD1dダイマー+で規定されるNKT細胞が産生できることが判明した。
マウス胸腺中で見られるTCRVβ+/αGalCer−CD1dダイマー+で規定されるNKT細胞はその殆どがNK1.1+、CD44high、CD69+、CD4+であり、CD8+の集団は殆どいない。ところがクローン7より産生されたTCRVβ+/αGalCer−CD1dダイマー+で規定されるNKT細胞表面上ではNK1.1やCD69の発現は殆ど観察されず、CD44を発現している細胞もごく僅かであった。さらに同細胞中のCD4を発現している細胞は殆ど存在せず約半数がCD8単一陽性であり、残りはCD4−CD8−であった。また使用されているTCRVβ鎖のレパートリーはVβ8であった。
OP9細胞並びにOP9−dlk細胞上で分化させたES細胞並びにntES細胞であるクローン7の分化誘導に伴う変化を遺伝子レベルで追跡するため、半定量的RT−PCR法によって、T、B細胞分化に重要な因子のmRNAの発現を検討した。TCRVβ+/αGalCer−CD1dダイマー+で規定されるNKT細胞特異的なmRNA(vα14−jα281)はOP9細胞上では観察されなかったが、OP9−dlk細胞上では培養15日目で出現が確認された(図1)。またB細胞受容体、T細胞受容体(TCR)のレパートリーの生成に必要なragの発現はOP9細胞上では培養19日で出現したのに対してOP9−dlk細胞上では培養12日目から確認された。さらにT細胞分化のプログラムに必須のgata3の発現も同条件下で観察された。これに対してB細胞の分化・成熟に必要と考えられるIgα、λ5の発現は通常のES細胞、ntES細胞(クローン7)の場合ともOP9細胞上でのみ観察された。以上の事実より以前の報告(Schmitt et al., Nature Immunology vol.5, p410-417 (2004))で示されているような胚性幹細胞からのT細胞の分化にはdlkが必須であることが確認され、TCRVβ+/αGalCer−CD1dダイマー+で規定されるNKT細胞はT細胞分化の場合とほぼ同一条件下で発生・分化してくることが判明した。
1)インビトロでのサイトカイン産生機能
TCRVβ+/αGalCer−CD1dダイマー+で規定されるこれらのNKT細胞が実際に機能を有するか否かを検証した。マウスより調製したこれらの細胞はαGalCerが提示された樹状細胞(DC)と相互作用しIFN−γ、IL−4などのサイトカインを産生する。そこでクローン7から誘導したTCRVβ+/αGalCer−CD1dダイマー+細胞をαGalCer提示DCと共培養し、培養液中に放出される各サイトカインの濃度をELISA法にて定量した。マウスIL−10はOptEIA mIL−10 ELISAキット(日本ベクトン・ディッキンソン社)、マウスIFN−γ、IL−4、IL−13はR&D systems社、Duoset(各DY485,DY404,DY413)を用いた。DCは、NKT細胞欠損マウス(C57BL/6バックグランド TCR Jα281鎖欠損マウス)の脾細胞からマウスCD11c−ビーズ〔CD11c(N418)マイクロビーズ,Miltenyi社〕を用いて純化した。このビーズを用いて磁気カラムを通した後の純度は96%以上であった。これらのCD11c+細胞はコントロールとして通常のES細胞より分化させたDP細胞とαGalCer提示DCとを共培養して上清中のサイトカインの定量にも使用した。実験の結果、これらのDP細胞を用いた培養ではIFN−γもIL−4も産生されなかったのに対してTCRVβ+/αGalCer−CD1dダイマー+細胞を用いると両サイトカインの産生が観察された(図2、3)。さらにαGalCer提示DC単独の培養からはこれらのサイトカイン産生が見られなかったのでIFN−γ並びにIL−4はTCRVβ+/αGalCer−CD1dダイマー+細胞より放出されたものと結論付けられる。
上記で示されたように、クローン7から誘導したTCRVβ+/αGalCer−CD1dダイマー+細胞は適切な刺激に呼応してインビトロでIFN−γ、IL−4などのサイトカイン産生能を示す。また、NKT細胞はそのアゴニストであるαGalCerにより活性化する際に樹状細胞を成熟化させ、インビボで免疫アジュバント効果を発揮し、抗原特異的なT細胞を誘導することが知られている(Fujiiら2003, J. Exp. Med. 198, p267-279)。そこでC57BL/6由来(自己)やBALB/c由来(アロ)のNKT細胞を、NKT細胞欠損マウス(C57BL/6バックグランド TCR Jα281鎖欠損マウス)に輸注した後、OVAタンパク質抗原とαGalCer(2μg/マウス)を同時投与して、免疫アジュバント能を発揮するか否か検討することにより、獲得免疫誘導を可能にするためのNKT細胞数の閾値を検討した。その後、我々が樹立したntES細胞由来NKT細胞によるアジュバント効果が誘導できるか否か検討し、自己又はアロNKT細胞輸注の場合と比較した。
具体的には、生体内DCに抗原を効果的に取り込ませるため、TAP遺伝子欠損マウスの脾細胞に高浸透圧(10mg/ml)で取り込ませた細胞結合(cell-associated)オボアルブミン(OVA)(2×107細胞/マウス)を同時に静脈内注入し、7日後マウスを頚椎脱臼により安楽死させ脾臓の単核細胞を調製した。これらの細胞を再びOVAペプチド存在下で6時間刺激した後、抗マウスCD8α抗体(PharMingen社)、並びに抗IFN−γ抗体を用いてCD8+細胞内におけるIFN−γの産生を調べた。その結果、αGalCer並びに細胞結合OVAを共投与したものではOVA再刺激によってCD8+細胞に抗原特異的なIFN−γの産生が確認されたがαGalCer又はOVA単独で免疫したマウスではOVA再刺激によっても同細胞内でIFN−γは確認されなかった。このOVA特異的なIFN−γ産生は、NKT細胞依存性であり(Fujiiら, 2003, J. Exp. Med. 198, p267-279)、前述したようにNKT細胞の欠損したマウスに抗原とαGalCerで免疫する前に予めNKT細胞を投与することによって回復できると考えられる。そこでNKT細胞をC57BL/6並びにBALB/cマウスから純化し、NKT細胞欠損マウスに養子移入した。戻すNKT細胞数が2×106で同系の場合(C57BL/6由来)にはαGalCer並びにOVAで免疫された脾細胞中のCD8+細胞はOVA再刺激によってIFN−γを産生するがその割合は0.38%であった。これに対して同数の同種異系NKT細胞(BALB/c由来)を戻した場合は0.13%の脾臓CD8+細胞中でIFN−γが確認された。このことは、同系に比べてアロNKT細胞が投与後にホストによって拒絶されている可能性を示す。我々の誘導したntES細胞(クローン7)由来のTCRVβ+/αGalCer−CD1dダイマー+細胞2×106個を同マウスに輸注した後に、OVAとαGalCerで免疫した場合は、OVA依存的に0.38%のIFN−γの産生CD8+T細胞が観察され、同系の場合とほぼ同様の力価を持った抗原特異的なCD8+T細胞が観察された。
上記2)の実験によりntES細胞由来のTCRVβ+/αGalCer−CD1dダイマー+細胞は、免疫アジュバント能を発揮することが明らかとなった。次に、この機能が、誘導された抗原特異的なT細胞による癌転移阻止機能に結びつくか否かを検討した。NKT細胞欠損マウス(C57BL/6バックグランド TCR Jα281鎖欠損マウス)にntES細胞由来のTCRVβ+/αGalCer−CD1dダイマー+細胞2×106個を静脈内注入によって戻し、一時間後にαGalCer並びにOVAで免疫した。その一週間後に、OVA抗原を発現している癌細胞であるEG7細胞(OVA抗原を発現しているEL4リンホーマ細胞)、又はこの抗原を発現していない別の癌細胞であるEL4リンホーマ細胞を2×106個ずつ皮下投与して腫瘍径を系時的に測定することにより腫瘍形成・増殖に及ぼす効果を比較検討した。この実験によりIFN−γ産生性の抗原特異的な獲得免疫の誘導が、抗腫瘍効果を示すかどうかを判定できる。結果を図4、5に示す。
15日後の評価により、ntES細胞由来のTCRVβ+/αGalCer−CD1dダイマー+細胞を投与したマウス5匹中4匹については、EG7細胞が拒絶され癌細胞の増殖を抑制した。1匹については腫瘍径の増大がみられたが、非常にわずかであった。
実験の結果、ntES細胞由来NKT細胞を輸注したものは抗原特異的に癌細胞の増殖を抑制したのに対してこれらの細胞を輸注していないマウス又は免疫抗原が異なる癌細胞を輸注したマウスでは癌の増殖を抑制することができなかった。以上の実験結果から、ntES細胞由来NKT細胞は抗原特異的な免疫アジュバント効果を発揮することにより癌細胞の増殖を抑止することが証明できた。
Claims (3)
- NKT細胞特異的T細胞受容体遺伝子について遺伝子再構成後の遺伝子型を有し、αGalCer提示樹状細胞と相互作用することによりIFN−γ及びIL−4を産生し得る、機能的なNKT細胞の製造方法であって、
該遺伝子型を有する幹細胞を、Dlk1を発現するOP9細胞の存在下で、Flt3リガンドを含む培地中で培養し、該OP9細胞との共培養開始後7日目にIL−7及びIL−15を培地に添加して更に培養して、該幹細胞由来の、該遺伝子型を有し、αGalCer提示樹状細胞と相互作用することによりIFN−γ及びIL−4を産生し得る、機能的なNKT細胞を得ることを含み、該幹細胞がES細胞又はES細胞になるように遺伝子操作された体細胞である、方法。 - 該遺伝子型を有する幹細胞が、該遺伝子型を有する細胞の核の移植により樹立されたES細胞である、請求項1記載の方法。
- 該ES細胞が、NKT細胞の核を除核卵母細胞に移植することにより樹立されたものである、請求項2記載の方法。
