Description
本発明の一実施形態では、細胞分離モジュールまたは細胞分離装置は、脂肪組織を切除し、加熱し、消化し、分離するための、全ての必要な電子部品およびコンポーネントを収容する機器の独立型部品である。好ましい実施形態では、細胞分離コンポーネントは遠心分離機を備える。移植片ソッディングモジュール、細胞収集モジュール、または他のモジュールに接続される、細胞分離モジュールからの出口は、単離細胞の単一細胞懸濁物を供給する。図3は、人と機械のインタフェース、細胞処理デバイス(遠心分離機)、チューブカセット、媒質バッグ、シリンジポンプ、ピンチ弁、収集シリンジ、およびバーコードスキャナを含む細胞分離モジュール耐久性コンポーネントおよび使い捨てコンポーネントを示す。
本発明の細胞分離装置は、他のデバイスおよびシステムと共にコンポーネントが使用され、また、再使用されてもよいように、モジュール式であるようにデザインされる。一実施形態では、細胞分離装置は、細胞ソッディングデバイスまたは移植片ソッディングモジュールと共に使用するようになっている。移植片ソッディングモジュールは、細胞分離ユニットによって供給される細胞を、圧力ソッディング技法を使用して多孔質移植片骨格上に適用するために必要である、耐久性コンポーネントおよび使い捨てコンポーネントを指す。移植片ソッディングモジュール耐久性コンポーネントおよび使い捨てコンポーネントは、図6に示される。
本発明のある実施形態では、細胞分離装置はまた、収集モジュールまたは収集デバイスと共に機能するようデザインされる。収集モジュールまたは収集デバイスは、細胞治療で使用するために、細胞分離ユニットからシリンジ内に細胞を収集するのに必要である耐久性コンポーネントおよび使い捨てコンポーネントを指す。細胞収集モジュール耐久性コンポーネントおよび使い捨てコンポーネントは図7に示される。図8は、使い捨てコンポーネントが使用のために装填された状態での、細胞分離モジュールに接続された細胞収集モジュール耐久材を示す。ある実施形態では、収集モジュール耐久材は、細胞分離ユニットに物理的に嵌合して、電力および通信接続を提供する。収集耐久材は、リニアアクチュエータを収容し、リニアアクチュエータは、シリンジにインタフェースして、細胞分離ユニット内で生成される細胞産物を自動的に収集する。
本発明の臨床キットまたは手術室(OR)キットは、無菌流路を提供し、無菌流路を通して、脂肪組織が、消化され、分離され、多孔質血管移植片骨格上に圧力ソッディングされうる。システムはまた、圧力ソッディング動作のため、移植片骨格を調製するために移植片骨格を前処理することが可能である。一実施形態では、流路は、耐久性臨床(OR)キットシステム格納容器にインターロックする3つの使い捨てカートリッジを備える。使い捨てカートリッジは、流体リザーバを有する流路カートリッジ、本発明の細胞分離装置を備える使い捨て遠心分離機カートリッジ、および移植片骨格を事前装填される使い捨て移植片チャンバを含む。臨床(OR)キットシステムは、動作するために電力だけを必要とする内蔵式の独立型システムである。
臨床(OR)キットシステムは、移植片骨格を前処理し、移植片内にソッディングするために脂肪組織を調製し、移植片に細胞を適用し、移植片管腔をパージし、採取まで生存能を維持するために移植片にわたってM199E/血清溶液を再循環させるために必要な流れ通路を自動的に進む。図4は、臨床(OR)キット内のコンポーネント間の概念的な流路を説明する。
上述したデバイスによって調製される組織移植片および細胞懸濁物は、無数の治療的使用に使用されうる。たとえば、本発明の一実施形態では、上述したデバイスの任意のデバイスによって調製される少なくとも1つの組織移植片または細胞懸濁物を組織または器官内に埋め込むことによって、必要なときに、被検者の組織または器官の血管再生を行う方法が提供される。本明細書で使用される用語「血管再生を行う(revascularize)」、「血管再生を行うこと(revascularizing)」、「血管新生(neovascularization)」、または「血管再生(revascularization)」は、通常、疾患、先天性欠損、または傷害により、無血管性ゾーンまたは乏血管性ゾーンを有する組織または器官内で、既存の血管ネットワークを修正すること、または、新しい機能性のまたは実質的に機能性の血管ネットワークを確立することを指す。

Claims (13)
- 細胞分離装置であって、
媒質リザーバと、
少なくとも1つの入口および少なくとも1つの出口を介して前記媒質リザーバと流体連通した細胞処理デバイスであって、該細胞処理デバイスは垂直軸周りに回転可能な内部チャンバと、該内部チャンバと流体連通しており且つ垂直軸と直交する方向に横向きに突出した複数のローブと、を備えた細胞処理デバイスと、
少なくとも1つのローブと整列され、流体を噴射して細胞を前記ローブから排出する噴射ノズルと、
前記媒質リザーバおよび細胞処理デバイスと流体連通した少なくとも1つのポンプと、
前記細胞処理デバイスに連結された少なくとも1つの弁と、を備え、
前記少なくとも1つのローブはテーパを有するように形成されており、前記テーパは前記垂直軸からテーパ端部まで径方向に沿って縮小しており、前記内部チャンバが前記垂直軸周りに回転する間に、前記細胞処理デバイスは前記内部チャンバ内の細胞を前記複数のローブに向かって径方向外向きに移動させ、少なくとも1つの前記ローブは前記テーパ端部において収集し且つ維持し、
前記少なくとも1つの弁は前記細胞処理デバイスに流体を流すか、または前記細胞処理デバイスから流体を流すか、を選択的に制御する装置。 - 前記細胞処理デバイスは遠心分離機である請求項1に記載の装置。

- 前記細胞処理デバイスの出口と無菌細胞収集デバイスとの間にフィルタをさらに備える請求項1に記載の装置。
- 前記細胞処理デバイスは、互いに流体連通する、内側ボウルおよび外側ボウルをさらに備える請求項1に記載の装置。
- 前記内側ボウルは、消化のために構成されたエリアおよび分離のために構成されたエリアを備える請求項8に記載の装置。
- 前記内側ボウルは、前記外側ボウルと連通する少なくとも1つのアパーチャをさらに備える請求項8に記載の装置。
- 分離のために構成された前記エリアは、さらに、内皮細胞の収集を最適化し、非内皮細胞材料の収集を最小化するよう構成される請求項9に記載の装置。
- 前記媒質リザーバは、生理的pHのある生理的濃度の塩化ナトリウムを含有する溶液を含有する請求項1に記載の装置。
- 前記溶液は、M199、M199E、PBS、生理食塩水(saline)、およびジカチオンの無いDPBSからなる群から選択される請求項12に記載の装置。
