Description
ビタミンは、人の健康にとって重要な物質である。ビタミンは様々な体内のプロセスを調節しており、多くの生命機能に対して好ましい作用を有する。一般論として、ビタミンは補酵素またはプロホルモンとして作用するものの、抗発癌物質または抗酸化作用のようなその他の機能の多くはまだ研究されていない。ビタミンの大部分は人体で合成することができないので、食物とともに摂取しなければならない。ビタミンは、構造および機能が様々であることから、最初の取り組みは、ビタミン類を水溶性群と脂肪性群とに分類することである。水溶性ビタミンは全身に分布し、ほとんど全ての器官に作用するが、一方で、脂肪性ビタミンは主に膜中に存在する。これらビタミンの血流中の輸送および消化は、十分な量の油脂質およびミセルが存在している場合にのみ行われる。
近年、超臨界二酸化炭素による天然材用の抽出に関する研究が増えつつある。二酸化炭素の臨界パラメータは、7.4MPa、304.21Kである(例えば、非特許文献6参照。)。これら低臨界パラメータのため、超臨界二酸化炭素を用いることにより、装置の運転に起因するコストを低コスト化でき、また、低臨界温度のため、超臨界二酸化炭素を用いることにより温度感受性材料であっても抽出できる。また、有機溶媒とは逆に、二酸化炭素は、無毒、不燃性および不活性である。さらに、二酸化炭素は、容易に入手可能であり、所望の産物から完全に取り除くことができ、残留物が発生しない。また、二酸化炭素の可溶性は密度で調節することができるので、供給材料によって調節して適用させることができる。これらが超臨界二酸化炭素抽出の分野における膨大な研究の理由である。例えば、イレス外(例えば、非特許文献7参照。)、およびグナイフィード外(例えば、非特許文献8参照。)は、トコフェロールおよびカロテン含量に関して、超臨界CO2および近臨界プロパンによる赤トウガラシおよびコショウ油の抽出を研究している。超臨界二酸化炭素の溶解性はカロテンを溶解するには不十分であることがわかった。両方の溶媒へのトコフェロールの溶解性は良好であり、CO2およびプロパン両方にトコフェロールを溶解することができた。また、ナジシャ外(例えば、非特許文献9参照。)およびシシクラ外(例えば、非特許文献10参照。)は、大豆脱臭剤の蒸留液からの超臨界流体抽出によるトコフェロールの富化を研究している。さらに、オリーブオイル産物の中間物からのスクアレン富化(例えば、非特許文献11参照。)および酢酸トコフェロールおよびトコフェロール富化、フレック外(例えば、非特許文献12参照。)に関する更なる研究が行われている。
特開2002−226723号公報は、パーム油からのカロテン富化方法を示している。パーム油は、最初にエステル交換される。次いで、このエステルを濃縮エステル層に分離するために、メタノールなどの親水性の溶媒で抽出する。次いで、この濃縮物は、カロテンを吸着するためにシリカゲルを充填したカラムに供される。次いで、カロテンは、約4000ppmのアセトンを含むヘキサンを用いて脱着される(例えば、特許文献3参照。)。
パーム油由来混合物からのトコトリエノールを主に含む高度富化トコクロマノール画分製造のための新しい方法は、カラム上部のより軽い揮発性物質、(即ち、超臨界化合物(二酸化炭素)中にてより溶解性の高い化合物)を除去するために、超臨界流体、好ましくは二酸化炭素を用いる連続的な向流多段階分離を兼ね備える。カラム下部は、ステロールおよびその他のやや極性がある化合物を比較的高含量で含むトコトリエノール富化画分である。この下部画分は、適切な吸着剤に通される。シリカゲルが適用可能な吸着剤であることが分かった。吸着された混合物は、超臨界流体、好ましくは二酸化炭素によって、2工程の溶出または抽出による脱着工程により脱着される。第1脱着工程は、約140barすなわち約14MPaの圧力にて行われ、揮発性がより高い化合物、または超臨界二酸化炭素により可溶な化合物を除去する。第2脱着工程は、約250barすなわち約25MPaの圧力にて行われ、主にトコトリエノールを含む高度富化トコクロマノール画分が抽出される。抽出された画分は、別の容器に採取することによって更に分割することもできる。画分の分け方によって、トコトリエノール濃度は原材料混合物の約50%の収率の、85%から略100%までの範囲に及ぶ。吸着剤はエタノールなどの液体溶剤によって再生され、吸着工程に再利用される。
カロテノイド濃縮物(1〜10重量%)は、5段階高圧ミキサ・セトラとして組み立てられた連続運転の抽出装置に供給される。この装置による三つの分離工程で、20MPa〜30MPaの圧力および323K〜358Kの温度において、最大30重量%までのカロテノイドを含む生成物を製造することが可能だった。この生成物を特別なシリカゲル、好ましくはZeofree5170である吸着剤に通し、この吸着剤によってカロテノイド、ステロイド、およびその他の物質、ならびに脂肪酸エステルの混合物が吸着される。混合比は4から10の範囲である。続いて、充填された吸着剤を例えば5MPaと323K、好ましくは2〜7MPaと313〜343Kの範囲の条件にて、最初に超臨界二酸化炭素と接触させ、次に近臨界プロパンと接触させる。二酸化炭素によって主に非カロテノイドが脱着されて溶出され、近臨界プロパンでカロテノイドが脱着される。原材料に対するプロパンの溶媒比は、20から100の範囲、好ましくは約30である。吸着工程において吸着剤の画分のみが原材料に充填された場合には、溶出したカロテノイドの濃度が高められた。
およそ10重量%のカロテノイドを含むカロテノイド濃縮物は、50mlの内部体積を有する高圧力パイプ中の吸着剤に通された。165m2/gの最少表面積のシリカゲルが原材料に対して4:1(12g:3g)の比で吸着剤として用いられた。次に、最初は150barおよび60℃の超臨界二酸化炭素で、次いで250barおよび60℃の超臨界二酸化炭素で抽出を行った。第三工程では、近臨界プロパンが7MPa、60℃で用いられた。画分は回収された。最終画分は約60%のカロテノイドからなる。

揮発性が前記天然化合物より高く超臨界流体または近臨界流体によって可溶性である上部の揮発性物質を取り除き、前記天然化合物を富化した下部画分をもたらす超臨界流体および近臨界流体の少なくともいずれかによる第1の向流多段階分離と、上部の前記標的天然化合物を取り除き、さらには低揮発性不純物から標的天然化合物を洗浄する超臨界流体および近臨界流体の少なくともいずれかによる前記第1の向流多段階分離とは別の第2の向流多段階分離とを有する第1の分離工程と、
吸着剤にて超臨界流体および近臨界流体の少なくともいずれかを含む前記第1の分離工程からの画分を直接的に吸着する吸着工程と、この吸着工程の後に同一または別の超臨界流体および近臨界流体の少なくともいずれかによって前記吸着剤から前記混合物を溶出する溶出工程とによる選択的吸着および脱着、ならびに様々な画分回収を含む第2の分離工程とを備え、
この第2の分離工程では、同一の圧力および温度でまたは減少した密度で、吸着剤に、超臨界流体および近臨界流体である二酸化炭素およびプロパンの少なくともいずれかを含む向流分離からの画分を4から10の割合で混合し、流入する原材料が吸着剤の一部のみに充填されるように直接的に吸着することによる選択的吸着および脱着を含み、少なくとも一つの工程にて、同一または別の超臨界流体および近臨界流体の少なくともいずれかで溶出または抽出による脱着を行い、
第1の脱着工程では、吸着された前記混合物中にまだ残っている揮発性がより高い天然化合物を12〜15MPaの低い圧力で除去し、
第2の脱着工程では、揮発性がより低い天然化合物を22〜35MPaの圧力でかつ313〜373Kの温度で抽出し、
第3の脱着工程では、3〜10MPaの条件での近臨界プロパンの使用を含む
ことを特徴とするパーム油またはパーム油派生物から天然化合物の高度富化した画分を製造する方法。 - 超臨界二酸化炭素および近臨界プロパンの少なくともいずれかを、313〜383Kの温度、5〜45MPaの圧力、溶媒対原材料比が1〜500の範囲の操作条件で用い、カラム上部の抽出物の還流比を0.1〜50の範囲として用いる
ことを特徴とする請求項1記載のパーム油またはパーム油派生物から天然化合物の高度富化した画分を製造する方法。 - 製造する天然化合物は、トコクロマノール画分、カロテノイド画分、ステロール画分およびコエンザイムQ10の少なくともいずれかである
ことを特徴とする請求項1または2記載のパーム油またはパーム油派生物から天然化合物の高度富化した画分を製造する方法。
