Description
本発明は、水溶性生理活性物質、有機イオンおよびポリマーを含む徐放組成物に関連し、その生理活性物質の含有量は、有機イオンの非存在下より約2倍〜4倍高い。
現在、種々の生理活性物質(例えば、GnRH類似物、ヒト成長ホルモン、リスペリドン、および酢酸オクトレオチドを例とするソマトスタチン類似物)を含む市場において、多くの徐放処方物が存在する。そのような処方物は、医師および獣医師、ならびに彼らの患者に好まれる。なぜなら、そのような処方物は、複数回の注射の必要性を減少するからである。不幸にも、多くの一般的な生理活性物質がその実質的な水溶性に起因して、徐放組成物に適した候補ではないという、徐放組成物についての処方物に関連する多くの問題が存在する。高い溶解性の生理活性物質の使用は、低薬物含有量、および水溶液との接触(例えば、患者への投与によってか、または生理的媒体への導入による)における生理活性物質の好ましくない「破裂」を生じ得、多くの場合、そのことは、実施において達成可能な生理活性物質の使用可能な含有量を制限する。
ここに、活性物質を長時間放出する、濃縮した低用量容積形態で投与可能な徐放組成物が、最初に生理活性物質を水溶性形態に変換しない水溶性生理活性物質から生じ得ることが見出された。この発見は、水溶性生理活性物質が、持続性放出組成物を形成する前に何らかの方法で操作される(例えば、その生理活性物質に水溶性を与える)必要があることを示唆した、この分野における先の研究と、正反対である。さらに、先の組成物において必要とされる変換プロセスは、高い割合の上記生理活性物質の分解を生じるのに対し、本発明の組成物は、分解を減少する方法の理解によって調製される。生理的に関連する形態にある上記生理活性物質の存在は、先に調製された組成物に関して、増加した核負荷(core load)を引き起こし、このことは生理活性物質の顕著な分解を生じやすい。減少した分解は、増加した非改変生理活性物質の放出に形を変え、従って、より小さい注射容積で、同じ用量の生理活性物質の送達を可能とする。本発明の組成物は、減少した用量容積を使用し、長時間にわたって比較的分解していない生理活性物質を送達する方法を提供することにおいて、先の発明を超える大きな改良点を提供する。
(定義)
本発明の意図に関して、以下の用語は、以下の意味を有する。
本発明の1つの実施形態において、上記生理活性物質は、タンパク質、核酸、炭水化物、ペプチドまたは低分子の薬学的物質からなる群より選択される。本発明において使用するタンパク質としては、抗体、治療的タンパク質、ヒト成長ホルモン、インターフェロンα、インターフェロンβ、インターフェロンγ、インスリン、カルシトニン、インターロイキン−1、インターロイキン−2などが挙げられるが、これらに限定されない。本発明において使用する核酸としては、DNA、RNA、化学的に改変されたDNAおよび化学的に改変されたRNA、アプタマー、アンチセンス、RNA干渉、および低分子RNA干渉が挙げられる。炭水化物としては、ヘパリン、低分子量ヘパリンなどが挙げられる。ペプチドとしては、LHRHアゴニストおよびそれらの合成類似物、ソマトスタチンおよびそれらの合成類似物、ホルモン、オクトレオチド、グルカゴン様ペプチド、オキシトシンなどが挙げられる。低分子の薬学的物質としては、抗感染薬、細胞傷害薬、高血圧治療薬、抗真菌薬、抗精神病薬、抗糖尿病薬、免疫賦活薬、免疫抑制薬、抗生物質、抗ウイルス薬、抗痙攣薬、抗ヒスタミン薬、心血管作動薬、抗凝血薬、ホルモン、抗マラリア薬、鎮痛薬、麻酔薬、ステロイド剤、非ステロイド性抗炎症薬、制吐薬が挙げられるが、これらに限定されない。
本発明において使用する有機イオンとしては、アニオン性物質およびカチオン性物質が挙げられる。アニオン性物質としては、以下の有機酸:パモ酸の、ドデシル硫酸の、コール酸の、トリフルオロメチル−p−トルエン酸の、2−ナフタレン硫酸の、2,3−ナフタレンジカルボン酸の、1−ヒドロキシ−2−ナフト酸の、3−ヒドロキシ−2−ナフト酸の、2−ナフト酸の、およびサリチルサリチル酸の、酸ならびにそれらの塩が挙げられるが、これらに限定されない。アニオン性物質の塩形態としては、ナトリウム、アンモニウム、マグネシウム、カルシウムなどが挙げられ得る。
特定の実施形態において、上記徐放組成物は、マイクロ粒子である。
特定の実施形態において、徐放組成物は、ナノ粒子である。
本発明の徐放組成物は、当該分野で公知のエマルジョンプロセスのいずれかによって作製され得る。
血管内注射または筋肉内注射などの非経口投与を目的として処方された化合物に加えて、本発明の他の代替的な投与方法もまた使用され得、その投与方法としては、皮内投与、肺投与、舌下投与、経皮的投与および経粘膜的投与(transmucosal administration)が挙げられるが、これらに限定されない。経粘膜的投与としては、眼投与、膣投与、直腸投与、および鼻腔内投与が挙げられ得るが、これらに限定されない。そのような投与方法の全ては、当該分野で周知である。
本発明の徐放組成物は、生物系または生物学的存在が許容し得る任意の賦形剤中に処方され得る。そのような賦形剤の例としては、水、生理食塩水、Ringer溶液、デキストロース溶液、Hank溶液、および他の水性の生理的に平衡化された塩溶液が挙げられる。非水性ビヒクル(例えば、不揮発性油、ポリエチレングリコール、およびオレイン酸エチルのような注射可能な有機エステル)もまた、使用され得る。他の有用な処方物としては、粘度増強剤(例えば、カルボキシメチルセルロースナトリウム、ソルビトールまたはデキストラン)を含む懸濁液が挙げられる。
(これまでに使用された方法による共溶媒を使用する、ポリ(ラクチド−コ−グリコリド)(PLGA)マイクロ粒子中にカプセル化された酢酸オクトレオチドの従来の調製)
酢酸オクトレオチドマイクロ粒子処方物を、有機相中の異なる共溶媒の効果を調査するために調製した。水中油型のエマルジョン/溶媒抽出技術を使用して調製した、処方物A〜処方物Fを、表1に要約する。PLGAポリマー(50:50 ラクチド/グリコリド、分子量24,000、180mg)を、酢酸エチル(EtOAc、900μL)中に溶解し、そして共溶媒(表1)中に溶解した酢酸オクトレオチド(20mg)を、そのポリマー溶液に添加した。生じた均一な有機相を、1%ポリビニルアルコール(PVA)を含む水相(2mL)に添加し、そしてその混合物を、15秒間〜30秒間ボルテックスした。上記エマルジョンを、溶媒抽出溶液(10mMリン酸ナトリウム、pH8.0、150mL)中に注ぎ、そしてそれを、4時間攪拌してEtOAcを抽出した。粒子を濾過によって単離し、水で洗浄し、そして一晩風乾した。上記処方物を、粒子サイズ、走査型電子顕微鏡(SEM)、形態学、オクトレオチド核負荷、およびインビトロ放出プロフィールによって特徴付けした。
(オクトレオチドの非水溶性有機酸塩(複合体)の生成、およびこれまでに使用された方法によるPLGAマイクロ粒子中へのカプセル化)
有機イオン物質を調査し、その有機イオンを、最初に酢酸オクトレオチドと複合体化して非水溶性塩を形成し、後にPLGAマイクロ粒子中にカプセル化した。
(本発明による水性エマルジョン相における、有機酸塩を使用するPLGAマイクロスフェア中の酢酸オクトレオチドカプセル化)
意外にも、上記エマルジョンプロセスの水相における有機酸塩の使用が、水溶性ペプチドの使用を可能にし、そして上記処方物を調製する前に、独立の工程における複合体化した化学種の調製の必要性を排除することを見出した。本発明は、別の利点(例えば、増加した薬物核負荷、一貫したオクトレオチド/有機イオン比率、およびインビトロ放出間の減少したペプチドの分解)を提供した。
(本発明による水性エマルジョン相中の有機酸塩を使用するPLGAマイクロスフェア中のさらなるペプチドのカプセル化)
酢酸オキシトシンおよび酢酸ロイプロリドを、以下の実施例に記載されるように、本発明に従ってPLGAマイクロ粒子中に処方した。これらの調査の結果は、従来の方法論に対して、上昇した核負荷および上昇したカプセル化効率(処方物BI対処方物BJ〜処方物BK、および処方物BL対処方物BM)に対する本発明の利用を、実証する(表9)。
(水性エマルジョン相における有機酸塩を使用するPLGAマイクロ粒子中のインスリンカプセル化)
ドデシル硫酸ナトリウム:マイクロ粒子処方物を、水中油型のエマルジョン/溶媒抽出法を使用して調製した。PLGAポリマー(分子量11,800、150mg)およびPEG化インスリン(50mg)からなる有機相を、CH2Cl2(2mL)中に溶解した。水相を、1% PVAおよび14mM SDSから構成した。上記均一な有機相および均一な水相を、1:5の割合で混合し、水相エマルジョン中に有機相エマルジョンを生成した。そのエマルジョンを、0.3% PVA溶媒抽出溶液(100mL)中に直接収集し、そしてそれを、100mL 2% IPAを添加する前に10分間攪拌した。その後、その溶媒抽出溶液を、さらに3時間攪拌してCH2Cl2を抽出した。硬化したマイクロ粒子を、濾過によって収集し、水で洗浄し、風乾し、そして−20℃で保存した。生じたマイクロ粒子は、21%の核負荷(カプセル化効率84%)を有した。これらのマイクロ粒子を、37℃のPBS中の24時間での、50%の大きいインビトロ破裂によって特徴付けた。
(Sprague Dawleyラットへの投与後のPLGAマイクロ粒子中のオクトレオチドの薬物動態の評価)
血液血清レベルを、ラットの皮下に注射した、PLGAマイクロ粒子処方物から放出されたオクトレオチドについて測定した。動物(n=6/群)を、6つの異なるオクトレオチドPLGAマイクロ粒子処方物の、単一用量レベル(約8mg/kg〜10mg/kg)の皮下注射によって、1回処置した。1時間および6時間、ならびに1日、4日、7日、11日、14日、20日、28日、42日、および54日において、血清サンプルを、それぞれの動物から得てオクトレオチド薬物動態を評価した。血清濃度を、市販の非抽出型放射免疫アッセイキット(extraction−free radioimmunoassay kit)(#S−2211)(Peninsula Labs)によって測定した。そのアッセイの定量化の限界(LOQ)は、0.1ng/mLであった。それぞれの時点における平均オクトレオチド血清濃度を、表10に示す。試験したオクトレオチドPLGA処方物の調製は、以下に記載する。
(処方物BC)
PLGAポリマー(分子量24,000、720mg)を、EtOAc(4000μL)中に溶解した。酢酸オクトレオチド(80mg)を、BnOH(4000μL)中に溶解し、そしてそれを、均一な有機相を生成する上記ポリマー溶液に添加した。生じた有機相を、10mMパモ酸二ナトリウムを含む1% PVA水相と混合してエマルジョンを得た。そのエマルジョンを、0.3% PVA溶媒抽出溶液(600mL)中に直接収集し、そしてそれを、4時間攪拌してEtOAcを抽出した。硬化したマイクロ粒子を、濾過によって収集し、水で洗浄し、風乾し、そして4℃で保存した。このことは、55.0μmの粒子サイズの中央値を有する、処方物BC(754mg、収率94%)を提供した。その核負荷(8.5%)、そのカプセル化効率(85.0%)、およびそのインビトロ破裂(7.4%)を、RP−HPLCアッセイによって決定した。
PLGAポリマー(分子量24,000、680mg)を、EtOAc(4000μL)中に溶解した。酢酸オクトレオチド(120mg)を、BnOH(4000μL)中に溶解し、そしてそれを、均一な有機相を生成する上記ポリマー溶液に添加した。生じた有機相を、10mMパモ酸二ナトリウムを含む1% PVA水相と混合してエマルジョンを得た。そのエマルジョンを、0.3% PVA溶媒抽出溶液(600mL)中に直接収集し、そしてそれを、4時間攪拌してEtOAcを抽出した。硬化したマイクロ粒子を、濾過によって収集し、水で洗浄し、風乾し、そして4℃で保存した。これにより、58.7μmの粒子サイズの中央値を有する、処方物BD(694mg、収率94%)を提供した。その核負荷(11.8%)、そのカプセル化効率(78.7%)、およびそのインビトロ破裂(4.1%)を、RP−HPLCアッセイによって決定した。

Claims (24)
- 組成物であって、以下:
a)酢酸ロイプロリド、酢酸オクトレオチド、および酢酸オキシトシンからなる群より選択される生理活性物質;
b)ポリマーまたは生理活性物質を分解から保護する、有機イオンであって、該有機イオンが、トリフルオロメチル−p−トルアート、2−ナフタレンスルホナート、2,3−ナフタレンジカルボキシラート、1−ヒドロキシ−2−ナフトアート、3−ヒドロキシ−2−ナフトアート、2−ナフトアート、およびサリチルサリチラートからなる群より選択される、有機イオン;および
c)該生理活性物質および該有機イオンをカプセル化する、ポリ(ラクチド−コ−グリコリド)
を含有する組成物。 - 前記組成物が、マイクロ粒子およびナノ粒子からなる群より選択される、請求項1に記載の組成物。
- 前記マイクロ粒子および前記ナノ粒子が、生分解性である、請求項2に記載の組成物。
- 前記有機イオンに対する前記生理活性物質のモル比が、1.0から1.5の範囲である、請求項1に記載の組成物。
- 前記生理活性物質が、酢酸オクトレオチドである、請求項1に記載の組成物。
- 前記有機イオンが、荷電した複合体または中性の複合体を形成するために、前記生理活性物質と相互作用する、請求項1に記載の組成物。
- プロセスを通じて生成されるポリマー中に生理活性物質を含有する徐放マイクロ粒子組成物であって、該プロセスが、以下:
a)ポリ(ラクチド−コ−グリコリド)と有機相とを混合する工程;
b)酢酸ロイプロリド、酢酸オクトレオチド、および酢酸オキシトシンからなる群より選択される生理活性物質と該有機相とを混合する工程;
c)トリフルオロメチル−p−トルアート、2−ナフタレンスルホナート、2,3−ナフタレンジカルボキシラート、1−ヒドロキシ−2−ナフトアート、3−ヒドロキシ−2−ナフトアート、2−ナフトアート、およびサリチルサリチラートからなる群より選択される有機イオンと水相とを混合する工程;
d)該有機相と該水相とを、エマルジョンプロセスの使用を通じて接触させる工程;および
e)徐放組成物を生成するために、該マイクロ粒子を回収する工程
を包含する、徐放マイクロ粒子組成物。 - 前記組成物が、マイクロ粒子およびナノ粒子からなる群より選択される、請求項7に記載の組成物。
- 前記マイクロ粒子および前記ナノ粒子が、生分解性である、請求項8に記載の組成物。
- 前記有機イオンに対する前記生理活性物質のモル比が、1.0から1.5の範囲である、請求項7に記載の組成物。
- 前記生理活性物質が、酢酸オクトレオチドである、請求項7に記載の組成物。
- 前記有機イオンが、荷電した複合体または中性の複合体を形成するために、前記生理活性物質と相互作用する、請求項7に記載の組成物。
- 組成物であって、以下:
a)酢酸ロイプロリド、酢酸オクトレオチド、および酢酸オキシトシンからなる群より選択される生理活性物質;
b)トリフルオロメチル−p−トルアート、2−ナフタレンスルホナート、2,3−ナフタレンジカルボキシラート、1−ヒドロキシ−2−ナフトアート、3−ヒドロキシ−2−ナフトアート、2−ナフトアート、およびサリチルサリチラートからなる群より選択される有機イオン;ならびに
c)該生理活性物質および該有機イオンをカプセル化するポリ(ラクチド−コ−グリコリド)であって、該生理活性物質の核負荷が9%を超える、ポリ(ラクチド−コ−グリコリド)
を含有する、組成物。 - 前記組成物が、マイクロ粒子およびナノ粒子からなる群より選択される、請求項13に記載の組成物。
- 前記マイクロ粒子および前記ナノ粒子が、生分解性である、請求項13に記載の組成物。
- 前記有機イオンに対する前記生理活性物質のモル比が、1.0から1.5の範囲である、請求項13に記載の組成物。
- 前記生理活性物質が、酢酸オクトレオチドである、請求項13に記載の組成物。
- 前記有機イオンが、荷電した複合体または中性の複合体を形成するために、前記生理活性物質と相互作用する、請求項13に記載の組成物。
- 組成物であって、以下:
a)酢酸ロイプロリド、酢酸オクトレオチド、および酢酸オキシトシンからなる群より選択される生理活性物質;
b)トリフルオロメチル−p−トルアート、2−ナフタレンスルホナート、2,3−ナフタレンジカルボキシラート、1−ヒドロキシ−2−ナフトアート、3−ヒドロキシ−2−ナフトアート、2−ナフトアート、およびサリチルサリチラートからなる群より選択される有機イオン;ならびに
c)該生理活性物質および該有機イオンをカプセル化するポリ(ラクチド−コ−グリコリド)であって、該生理活性物質の25%未満が、生理的媒体への該生理活性物質の放出の際に分解した形態である、ポリ(ラクチド−コ−グリコリド)
を含有する、組成物。 - 前記組成物が、マイクロ粒子およびナノ粒子からなる群より選択される、請求項19に記載の組成物。
- 前記マイクロ粒子および前記ナノ粒子が、生分解性である、請求項20に記載の組成物。
- 前記有機イオンに対する前記生理活性物質のモル比が、1.0から1.5の範囲である、請求項19に記載の組成物。
- 前記生理活性物質が、酢酸オクトレオチドである、請求項19に記載の組成物。
- 前記有機イオンが、荷電した複合体または中性の複合体を形成するために、前記生理活性物質と相互作用する、請求項19に記載の組成物。
