本実施例においては、市販の細胞培養用384ウェルプレートを用い、各ウェルを空洞部を有する構造体として使用して、本発明の方法を使用して、マウスES細胞から胚様体を作製することにより、プレート単位面積あたりに作製できることの細胞塊数および一群の細胞塊の作成における効率性を調べることを目的として行った。
本実施例においては、市販の細胞培養用1536ウェルプレートを用い、各ウェルを有するプレートを空洞部を有する構造体として使用して、本発明の方法を使用して、マウスES細胞から胚様体を作製することにより、プレート単位面積あたりに作製できることの細胞塊数および一群の細胞塊の作成における効率性を調べることを目的として行った。
本実施例おいては、市販品である各種の細胞培養用プレートにおいて、細胞塊を作製するために培養液が突出部分を形成し、かつプレートを上下反転しても培養液が落下することのない、突出部分の形成に必要な適正溶液量と各種プレートの開口部内径の関係について検討した。
Claims (6)
- 少なくとも下側の一端が開口している内径が1.83〜3.00 mmである空洞部を有する構造体の前記空洞部内に、細胞を含む培養液を注入し、
前記培養液の一部を前記開口端より下側に突出させ、
前記培養液の前記突出部分で細胞を培養し、細胞塊を形成することを特徴とする、細胞塊の製造方法。 - 前記空洞部の他端が閉鎖しており、前記空洞部内への前記培養液の注入を、前記開口端を上にしかつ前記培養液の一部が前記開口端より上に突出させることによりおこない、前記開口端を下側に向けることを特徴とする、請求項1に記載の細胞塊の製造方法。
- 前記空洞部が前記構造体に複数形成されており、それによって複数の細胞の培養を同時にできることを特徴とする、請求項1または2のいずれかに記載の細胞塊の製造方法。
- 請求項1〜3のいずれかに記載の細胞の培養方法により胚性幹細胞を培養して胚様体を形成することを特徴とする、細胞塊の製造方法。
- 前記一端側の開口端より下側に突出した前記培養液の一部を別の培養液の液面に接触させることにより、細胞塊を前記別の培養液中に回収することを特徴とする、請求項1〜4のいずれか1項に記載の細胞塊の製造方法。
- 前記開口端の内径が1.83〜1.98 mmである、請求項1〜5のいずれか1項に記載の細胞塊の製造方法。
