Description
本発明の更年期障害予防・改善剤は、ラクトバチルス・アシドフィラスとともに培養することにより代謝・活性化されたリグナンを有効成分として含み、その効果は、代謝・活性化する前のリグナンを摂取するより高いものである。リグナンとしては、例えば、杜仲リグナン、亜麻仁リグナン、ゴマリグナン等が挙げられる。好ましくは、リグナンは杜仲リグナンである。リグナンは、ラクトバチルス・アシドフィラスによりまずリグナン配糖体からアグリコンに変換される。さらに、ピノレジノール、メディオレジノールはラクトバチルス・アシドフィラスにより代謝・活性化されて、種々のエストロゲン様活性中間体に変換される。本明細書において、エストロゲン様活性中間体とは、その物質自体がエストロゲン様活性をもつ物質、および摂取されたときに生体内でさらに代謝されてエストロゲン様活性をもつ物質に変換される物質の両方を含む。エストロゲン様活性中間体には、ラシリレジノールおよびセコイソラリシレジノール等が含まれる。本発明において好ましくは、エストロゲン様活性中間体はセコイソラリシレジノールである。
1.形態学的性質
1)細胞の形状:桿菌、2)運動性:なし、3)胞子の有無:なし、4)グラム染色性:陽性。
2.生理学的性質
1)カタラーゼ:陰性、2)インドール生成:陰性、3)硝酸塩の還元:陰性、4)酸素に対する態度:通性嫌気性菌、5)15℃で生育:なし、6)グルコースからホモ乳酸発酵によりDL(−)乳酸を生成し、ガスの産生はない。
7)各種炭水化物の分解性:グルコース;+、ラクトース;+、マンノース;+、フラクトース;+、ガラクトース;+、シュークロース;+、アラビノース;−、マルトース;+、キシロース;−、ラムノース;−、セルビオース;+、トレハロース;+、メルビオース;+、ラフィノース;+、マンニトール;−、ソルビトール;−、エスクリン;+、サリシン;+、N−アセチルグルコサミン;+、アミグダリン;+、ゲンチオビオース;+、メレチトース;−、デキストリン;+、スターチ;−。

各種微生物のリグナン代謝能を評価した。下記の表1−3に示される各種の微生物を、MRS培地若しくはGAM培地を用いて37℃で20−24時間前培養し、10mlのMRS培地若しくはGAM培地に1−3%で植菌した。培地中の最終濃度が0.5mMになるように杜仲リグナン(ピノレジノールおよびメディオレジノール)を添加し、37℃で培養を続けた。経時的にサンプリングを行い、添加したリグナンが代謝されてセコイソラリシレジノールに変わる様子をHPLCにより定量的に測定した。リグナンが50%以上変換されたものを○、ほとんど変換されなかったものを×として表した。結果を表1−3に示す。
1mMの試験試料(ピノレジノール・ジグルコシド、ピノレジノール、セコイソラリシレジノール)と0.5nMのトリチウムラベルしたエストラジオールを、組換え発現させたヒト型エストロゲンレセプターαまたはβを含むpH7.4の10mMトリスバッファー(0.1% BSA,10% Glycerol,1mM DTT含有)中で、25度にて2時間反応させた。反応終了後、レセプターを回収・洗浄して、定法にしたがって、レセプターに結合しているトリチウムラベルされたエストラジオール量を測定した。0.5nM [3H]Estradiolのレセプターへの結合を100%として、それぞれのリグナンまたは代謝中間体を添加したときの[3H]Estradiolの結合度を算出し、それを100から引いた値を結合阻害率とした。結果を下表に示す。
Claims (10)
- リグナンをラクトバチルス・アシドフィラスとともに培養することを特徴とするエストロゲン様活性中間体の製造方法。
- リグナンがピノレジノールまたはメディオレジノールである、請求項1記載の方法。
- リグナンが杜仲リグナンである、請求項1記載の方法。
- エストロゲン様活性中間体がセコイソラリシレジノールである、請求項1−3のいずれかに記載の方法。
- リグナンをラクトバチルス・アシドフィラスとともに培養した発酵物を有効成分として含有する更年期障害予防・改善剤。
- リグナンをラクトバチルス・アシドフィラスとともに培養した発酵物を有効成分として含有する更年期障害予防・改善用機能性飲食品。
- 杜仲リグナンをラクトバチルス・アシドフィラスとともに培養した発酵物を含む更年期障害予防・改善剤。
- 杜仲リグナンをラクトバチルス・アシドフィラスとともに培養した発酵物。
- ピノレジノールまたはメディオレジノールを含有する植物をラクトバチルス・アシドフィラスとともに培養した培養物を有効成分として含有する更年期障害予防・改善剤。
- ピノレジノールまたはメディオレジノールを含有する植物をラクトバチルス・アシドフィラスとともに培養した培養物。
