常法により調整された、2‘−O−シアノエチルウリジンがサクシニルリンカーを介して導入されているロングアミノアルキルチェーンCPG(16μmol/g)を1μmol固相合成用ソケットに充填されたものを用い、2‘−O-シアノエチルウリジンホスホロアミダイトユニット(化合物7)を0.1Mの無水アセトニトリルの溶液としてDNA/RNA自動合成機(Applied Bio System)に縮合時間10分間の縮合時間であるRNA合成用プログラムを適用する。
合成終了後、アンモニア水1mlにて20分間処理して固相担体からオリゴヌクレオチドを切り出しを行う。その後、酢酸アンモニウムバッファーで適当量希釈し、逆相カラムカートリッジ並びに陰イオン交換HPLCを用いて精製する。逆相カラムカートリッジにて脱塩操作を行い得られたオリゴヌクレオチドを凍結乾燥して標題化合物を得る。(収率21%)。陰イオン交換高速液体クロマトグラフィーによる分析プロファイルを図1Aに示す。

合成終了後、アンモニア水-酢酸アンモニウム(10%wt/wt)1mlにて90分間処理して固相担体からのオリゴヌクレオチドの切り出し並びに核酸塩基部の保護基の脱保護を行う。その後、酢酸アンモニウムバッファーで適当量希釈し、逆相カラムカートリッジ並びに陰イオン交換HPLCを用いて精製する。逆相カラムカートリッジを用いて脱塩操作を行い得られたオリゴヌクレオチドを凍結乾燥して標題化合物を得る(収率 58%)。陰イオン交換高速液体クロマトグラフィーによる分析プロファイルを図1Bに示す。

Claims (16)
- 2’‐O‐シアノエチルウリジン。
- 2’‐O‐シアノエチルシチジン。
- 2’‐O‐シアノエチルアデノシン。
- 2’‐O‐シアノエチルグアノシン。
- N−4−ジメチルアミノメチレン−2’‐O‐シアノエチルシチジン。
- N6−ジメチルアミノメチレン−2’‐O‐シアノエチルアデノシン。
- N2‐ジメチルアミノメチレン‐6‐O‐トリイソプロピルベンゼンスルホニル‐2’‐O‐シアノエチルグアノシン。
- N4‐アセチル‐2‘‐O‐(2−シアノエチル)シチジン。
- N2‐ジメチルアミノメチレン‐2’‐O‐シアノエチルグアノシン。
- 請求項1〜10の何れか一項に記載のヌクレオシドの合成方法であって、炭酸セシウムとアクリロニトリルとヌクレオシド誘導体を合成原料として用い、t−ブチルアルコールの存在下又は非存在下で、2‘水酸基をシアノエチルエーテル化することを特徴とする、前記合成方法。
- 炭酸セシウムをヌクレオシド誘導体に対して0.1〜30equivの範囲で反応系に存在させ、アクリロニトリルに対して、0.05〜30equivの範囲のt−ブチルアルコールの存在下に反応を行う、請求項11記載の合成方法。
- 炭酸セシウムを用いる、請求項11又は12に記載の合成方法。
- 20℃〜30℃の温度範囲で反応させる、請求項11〜13の何れか一項に記載の合成方法。
- 反応時間が2時間〜3時間の範囲である、請求項11〜14の何れか一項に記載の合成方法。
- 請求項1ないし10のいずれか一項に記載のヌクレオシドを含むRNAオリゴマー。
