Description
発明の分野
本発明は、創傷ドレッシング(wound dressing)、詳細には、銀メッキした(silvered)ゲル形成繊維をベースとする抗細菌性創傷ドレッシング、並びにかかる創傷ドレッシングを製造するための改良された方法に関連する。
銀は、ほ乳類組織との適合性と併せて幅広い活性を有する有用な抗細菌剤であることが長年に渡り知られており、そして創傷ドレッシング中での銀の有利な殺菌特性を得るために、銀を創傷ドレッシング中へと組み込むための多くの提案がなされてきた。加えて、銀は、従来から創傷ドレッシングのための目的ではなく、通常は繊維性材料に対して電気伝導度を高める目的で適用されてきた。例えば、UK−A−927,115、WO−A−92/16,589、DE−C263,9287、US−A−5,302,415、US−A−5186984、US−A−4752536、US−A−4,643,918、JP−010,207,473、及びJP−020,153,076などを参照のこと。銀は、一般にゲル形成性ではないかかる繊維に対して、それら参考文献中の様々な方法で適用されてきた。そのある方法とは、繊維を銀溶液中に浸すことを伴うが、使用されている手順の詳細は往々にして十分に分かってはいない。
我々は、高品質な創傷ドレッシングは、銀メッキしたゲル形成繊維であって、銀メッキをするために用いられる溶液中の有効な銀イオンよりも銀イオンを吸収することができる部位を多く有し、従って、銀メッキ作業中、吸収される銀イオンがそれらを吸収できる部位に対して実質上均一に分配されることによって全部の部位が銀イオンを吸収しないような銀メッキしたゲル形成繊維から獲得することができることを発見した。
(i)銀イオンを前記ゲル形成繊維に対して結合せしめるために、銀イオンとの交換をすることができる交換可能部位をその上に有する前駆体ゲル形成繊維の集成体を形成し、そして
(ii)前記前駆体ゲル形成繊維の集成体と銀イオンを含有する溶液とを、当該繊維の不可逆的ゲル化が生じない条件下で接触せしめることによって、銀イオンを当該繊維に対して交換可能部位にて結合せしめる、
段階が含まれ、そして、
創傷ドレッシングを形成するための当該前駆体ゲル形成繊維の集成体の全体が本質的に同時に銀イオンを含有する溶液全体と接触しており、当該銀イオンを含有する溶液は、当該前駆体ゲル形成繊維の交換可能部位の全てではなくいくつかと、存在する銀イオンを結合せしめるために十分な量でのみ使用されていることを特徴とする。
(i)銀メッキされたトウ(tow)を形成するために、銀イオンとのイオン交換をすることができる部位をその上に有する前駆体繊維を形成し、そして
(ii)当該繊維の不可逆的ゲル化が生じない銀イオン含有溶液を用いて当該繊維に銀メッキを施す、
段階を伴い、そして
銀の吸収が均一になるようにするために、当該繊維を直接当該銀溶液中へと沈めてすぐに当該繊維を当該溶液の水面下に押出すことによって、当該繊維を当該溶液中へと沈めることを特徴とする。次いで、生成される銀メッキ繊維は、創傷ドレッシングを形成するために、常用の段階などで処理されうる。
上記のキヤー方法及び本発明の方法を個別に、置換の程度が約0.3のカルボキシメチル化されたリオセルに対して行った。前記キヤー方法を、「発明の背景」に記載したようにして行った。本発明の方法は、2.7lのIMS、1.8lの水及び25gのAgNO3を含んで成る溶液を用いてカルボキシメチル化されたリオセルのハンクに対して行った。ハンクが前記溶液を吸収するように、ハンクを約5秒に渡り全体的に当該溶液に沈めて浸し、従って、当該溶液の全てがカルボキシメチル化されたリオセルと接触し且つ本質的に全てを吸収した。そのような方法で獲得した銀メッキされたハンクは、キヤー方法によって処理したハンクと平均銀含有量が本質的に同じであった。それぞれの場合において、反応したトウを縦に広げて、暗闇ではなく光にさらして乾燥せしめた。キヤーを用いて作製した材料は、トウの着色されていない大部分、典型的に75%の部分と、灰色又は桃色の範囲が極端に異なった。本発明のこの方法によって作製した材料は、着色されていないトウは多分ほんの僅か約5〜10%であり、大部分が灰色/桃色に着色されていた。
水中(1l)硝酸銀(1.58g)の溶液二つを研究室にて室温で作製した。第1番目には、アルギン酸塩トウ(100g)を非常にゆっくりとした態様で、即ち全てを浸すのに約30秒を要し、添加した。第2番目には、類似量のアルギン酸塩トウを加えたが、この時間は5秒未満であった。
Claims (20)
- 銀イオンが、ゲル形成繊維上の交換可能部位の20%以下に対して結合しているゲル形成繊維に由来する、該ゲル形成繊維を銀含有溶液に沈めることによって得られる抗細菌性創傷ドレッシング(wound dressing)であって、該ドレッシングの均一な着色によって示されるように、当該交換可能部位に対する銀イオンの分配が実質的に均一であることを特徴とする抗細菌性創傷ドレッシング。
- 前記ゲル形成繊維がカルボキシメチル化されたセルロース繊維である、請求項1に記載の創傷ドレッシング。
- 前記ゲル形成繊維がアルギン酸カルシウム繊維又はアルギン酸ナトリウム/カルシウム繊維である、請求項1に記載の創傷ドレッシング。
- 抗細菌性創傷ドレッシングにおいて用いるための銀メッキした(silvered)繊維を生産する方法であって:
(i)銀イオンを前記ゲル形成繊維に対して結合せしめるために、銀イオンとの交換をできる交換可能部位をその上に有する前駆体ゲル形成繊維の集成体を形成し、そして
(ii)前記前駆体ゲル形成繊維の集成体と銀イオンを含有する溶液とを、当該繊維の不可逆的ゲル化が生じない条件下で接触せしめることによって、銀イオンを交換可能部位の全てではなくいくつかにて結合せしめる、
段階を伴い、そして
前駆体ゲル形成繊維の集成体の全体が本質的に同時に銀イオンを含有する溶液全体と接触しており、当該銀イオンを含有する溶液は、当該前駆体ゲル形成繊維上の交換可能部位の全てではなくいくつかと、存在する銀イオンを結合せしめるために十分な量でのみ使用されていることを特徴とする方法。 - 前記銀イオンを含有する溶液は、前記前駆体ゲル形成繊維上の交換可能部位の20%以下に対して、存在する銀イオンを結合せしめるために十分な量で使用されている、請求項4に記載の方法。
- 前記銀イオンを含有する溶液は、前記前駆体ゲル形成繊維上の交換可能部位の10%以下に対して、存在する銀イオンを結合せしめるために十分な量で使用されている、請求項4に記載の方法。
- 前記方法が、前駆体ゲル形成繊維の集成体を、前記銀イオンを含有する溶液溶中へと急速且つ全体的に沈めることによって当該溶液全体と接触せしめているバッチ方法である、請求項4〜6のいずれか1項に記載の方法。

- 前記時間間隔が5秒以下である、請求項9に記載の方法。
- 沈められる前記前駆体ゲル形成繊維の集成体が乾燥している、請求項7〜10のいずれか1項に記載の方法。
- 前記銀イオンを含有する溶液が、閉鎖された容器中に入れられている請求項8〜11のいずれか1項に記載の方法。
- 前記銀イオンを含有する溶液が、開放された容器中に入れられている請求項8〜11のいずれか1項に記載の方法。
- 前記前駆体ゲル形成繊維がトウ(tow)形態かあるいはステープル繊維形態である、請求項4〜13のいずれか1項に記載の方法。
- 前記前駆体ゲル形成繊維の集成体がハンクの形態である、請求項4〜14のいずれか1項に記載の方法。
- 前記前駆体ゲル形成繊維がカルボキシメチルセルロース繊維である、請求項4〜15のいずれか1項に記載の方法。
- 前記カルボキシメチルセルロース繊維がカルボキシメチル化されたリオセルである、請求項16に記載の方法。
- 前記カルボキシメチル化されたリオセルの置換の程度が0.2〜0.4である、請求項17に記載の方法。
- 前記前駆体ゲル形成繊維がアルギン酸カルシウム繊維又はアルギン酸ナトリウム−カルシウム繊維である、請求項4〜15のいずれか1項に記載の方法。
- 前記銀メッキされたゲル形成繊維の集成体が創傷ドレッシングを形成するために加工されている、請求項4〜19のいずれか1項に記載の方法。
