Description
PGはカルボニル官能基を含有するN−保護基であり、かつこの官能基を介して窒素と結合している]のN−保護4−ケトプロリン誘導体を製造するための方法において達成され、この場合、この方法は、相当する4−ヒドロキシプロリン化合物を、酸化剤を用いて、触媒的に活性のルテニウム化合物の存在下で、酸化することにより製造され、この場合、酸化を水性の単相系中で実施し、かつ酸化生成物(I)が酸化剤の添加の間に晶出しうる。すでに記載したように、一般式(I)の化合物は、特にアルカリ条件下でさらに形質転換しうる(アルドール反応、開環反応)不安定な化合物である。さらに本発明による方法は、生じた酸化生成物(I)が、その製造後可能な限りすぐに晶出する場合にのみ優れて作用することを約束する。したがって、この系においてのさらなる酸化を回避することができる。この結果は、簡単な方法で、複雑な抽出工程なしに、酸化系から除去することができる極めて純粋な反応生成物の存在である。

69g(0.322モル)のNaIO4(Fluka)

(20.8mgからの全量を推定した):
乾燥粗生成物21gを、酢酸イソプロピル(250ml)および水(55ml)中で懸濁した。生成物を撹拌および加熱により溶解した。その後に、活性炭素(PWA)1gを2相混合物に添加し、かつ混合物を35℃で30分に亘って撹拌した。その後に、セライトを介して濾別し、かつ水相を除去した。有機相を真空下で150〜170mバール下で、浴温度40〜45℃で蒸留した。生成物は、蒸留中であっても沈澱し始めた。塔底中の量が80g(約25g%)になるまで蒸留した。得られた懸濁液(重結晶)を、その後に約2時間に亘って0℃に冷却し、濾過し、かつそれぞれ冷酢酸イソプロピル20mlで2回に亘って洗浄した。湿性の生成物28gが得られ、かつ水浴中で、45℃/15mバールで乾燥させた。乾燥生成物17.0gが得られた。

純度:HPLC(>99.5面積%)
旋光性:C=1、アセトン中20℃

Claims (4)
- 触媒的に活性のルテニウム化合物の存在下で、相当する4−ヒドロキシプロリン化合物を酸化剤で酸化させることによって、一般式(I)
PGがカルボニル官能基を有するN−保護基であり、かつこの官能基を介して窒素と結合している]のN−保護4−ケトプロリン誘導体を製造するための方法において、酸化を、水性の単相系中で実施し、かつ、酸化剤の添加の間に酸化生成物(I)を晶出させることを特徴とする、一般式(I)のN−保護4−ケトプロリン誘導体を製造するための方法。 - 酸化の間の温度を、≦30℃に維持する、請求項1に記載の方法。
- 次亜ハロゲン酸塩、ハロゲン酸塩および過ハロゲン酸塩を、酸化剤として使用する、請求項1または2に記載の方法。
- 50%の酸化剤の添加後に、種晶を反応混合物に添加する、請求項1から3までのいずれか1項に記載の方法。
