Description
本発明は、(i)メイタンシノイド(maytansinoid)と化学的に結合した抗体を含有する治療に効果的な量の複合体、(ii)緩衝剤、(iii)等張化させる(tonicifying)量の塩化ナトリウム、(iv)水、および、必要に応じて(v)界面活性剤を含有しており、pHが約5〜6である組成物を提供する。本発明はまた、(i)メイタンシノイドと化学的に結合した抗体を含有する治療に効果的な量の複合体、(ii)緩衝剤、(iii)抗凍結剤、(iv)充填剤、および、必要に応じて(v)界面活性剤を含有しており、再構成した(水にもどした)(reconstituted)ときのpHが約5〜6である、凍結乾燥された組成物を提供する。さらに本発明は、ヒトにおいて細胞を死滅させる方法であって、上述の組成物の何れかをヒトに投与する工程を含み、その結果抗体が細胞の表面に結合し、メイタンシノイドの細胞毒性が活性化され、これによって細胞を死滅させるという方法を提供する。
本発明は、(i)メイタンシノイドと化学的に結合した抗体を含有する治療に効果的な量の複合体、(ii)緩衝剤、(iii)必要に応じて界面活性剤、(iv)等張化させる量の塩化ナトリウム、および(v)水を含有しており、pHが約5〜6である組成物を提供する。
本実施例では、メイタンシノイドと化学的に結合した抗体を含有する複合体、緩衝剤、界面活性剤、等張化させる量の塩化ナトリウム、および水を含んでなる組成物の製造を実証する。
本実施例では、メイタンシノイドと化学的に結合した抗体を含有した複合体、緩衝剤、界面活性剤あるいはスクロース、等張化させる量の塩化ナトリウム、および水を含んでなる組成物の製造を実証する。
本実施例では、メイタンシノイドと化学的に結合した抗体を含有する複合体、緩衝剤、界面活性剤、等張化させる量の塩化ナトリウム、酸化防止剤、および水を含んでなる組成物の製造を実証する。
本実施例では、メイタンシノイドDM1と化学的に結合したモノクローナル抗体huMy9−6を含有する複合体、緩衝剤、等張化させる量の塩化ナトリウム、および水を含む組成物であって、界面活性剤およびスクロースを含むものおよび界面活性剤およびスクロースを含まないものの製造を実証する。
本実施例では、メイタンシノイドDM4と化学的に結合したモノクローナル抗体huMy9−6を含有する複合体、緩衝剤、等張化させる量の塩化ナトリウム、および水を含む組成物であって、界面活性剤を含むものおよび界面活性剤を含まないものの製造を実証する。
本実施例では、メイタンシノイドDM1と化学的に結合したモノクローナル抗体huN901を含有する複合体を含む凍結乾燥された組成物の製造を実証する。
本実施例では、メイタンシノイドDM1と化学的に結合したモノクローナル抗体huN901を含有する複合体を含む凍結乾燥された組成物の安定性を実証する。
Claims (66)
- (i)メイタンシノイドと化学的に結合した抗体を含有する0.1mg/ml〜5mg/mlの複合体、(ii)クエン酸塩バッファー、酢酸塩バッファー、コハク酸塩バッファー、およびヒスチジンバッファーからなる群から選択される2mM〜50mMの緩衝剤、(iii)50mM〜500mMの塩化ナトリウム、および(iv)水、を含有しており、pHが5〜6である、組成物。
- 上記組成物が、該組成物の全体積の0.002%〜0.1%の界面活性剤をさらに含む、請求項1記載の組成物。
- 上記界面活性剤がポリソルベート80である、請求項2記載の組成物。
- 上記界面活性剤がポリソルベート20である、請求項2記載の組成物。
- 上記組成物中のポリソルベート20の濃度が、該組成物の全体積の0.01%である、請求項4記載の組成物。
- 上記組成物中の上記複合体の濃度が1mg/mlである、請求項1〜5の何れか1項に記載の組成物。
- 上記組成物中の上記複合体の濃度が5mg/mlである、請求項1〜5の何れか1項に記載の組成物。
- 上記組成物が、クエン酸ナトリウムを10mMの濃度で含有し、そのpHが5.5である、請求項1〜7の何れか1項に記載の組成物。
- 上記組成物中の塩化ナトリウムの濃度が100mM〜200mMである、請求項1〜8の何れか1項に記載の組成物。
- 上記組成物中の塩化ナトリウムの濃度が120mMである、請求項9記載の組成物。
- 上記組成物がさらに酸化防止剤を含有する、請求項1〜10の何れか1項に記載の組成物。
- 上記組成物中の上記酸化防止剤の濃度が100μM〜100mMである、請求項11記載の組成物。
- 上記酸化防止剤がスーパーオキシド・ジスムターゼ、グルタチオンペルオキシダーゼ、トコトリエノール、ポリフェノール、亜鉛、マンガン、セレン、ビタミンC、ビタミンE、ベータカロチン、システイン、およびメチオニンからなる群から選択される、請求項12記載の組成物。
- 上記組成物が10mMの濃度でメチオニンを含有する、請求項13記載の組成物。
- 上記組成物がさらにスクロースを含有する、請求項1〜14の何れか1項に記載の組成物。
- 上記組成物中のスクロースの濃度が、該組成物の全体積の0.1%〜10%である、請求項15記載の組成物。
- 上記組成物中のスクロースの濃度が、該組成物の全体積の5%である、請求項16記載の組成物。
- 密封容器およびその中に分散された請求項1〜17の何れか1項に記載の組成物、ならびに不活性ガスオーバーレイを含有してなる、パッケージ化された組成物。
- 上記不活性ガスが窒素またはアルゴンである、請求項18記載のパッケージ化された組成物。
- 上記抗体が細胞の表面に結合し、メイタンシノイドの細胞毒性が活性化され、これによって細胞を死滅させる、ヒトにおいて細胞を死滅させるための、請求項1〜17の何れか1項に記載の組成物。
- 上記細胞が腫瘍細胞である、請求項20に記載の組成物。
- 上記抗体がモノクローナル抗体である、請求項20に記載の組成物。
- 上記抗体が、huN901、huMy9−6、huB4、huC242、トラスツズマブ、ビバツズマブ、シブロツズマブ、および、リツキシマブからなる群から選択される、請求項20記載の組成物。
- 上記メイタンシノイドがチオール基を有する、請求項20に記載の組成物。
- 上記メイタンシノイドがDM1またはDM4である、請求項24記載の組成物。
- 上記抗体が、ジスルフィド結合、酸に不安定な結合、光に不安定な結合、ペプチダーゼに不安定な結合、チオエーテル結合、およびエステラーゼに不安定な結合からなる群から選択される化学結合を介してメイタンシノイドと化学的に結合している、請求項20に記載の組成物。
- 上記抗体が細胞の表面に存在する抗原と結合する、請求項20に記載の組成物。
- 上記抗原が、NCAM/CD56、CD33、CD19、GD3、CanAg、PSMA、α葉酸受容体、Her2/neu、CD44v6、胎児膵腺房(FAP)抗原、Cripto−1、CA6、CD20、CA55.1、MN/CA IX、およびコンドロイチン硫酸プロテオグリカンからなる群から選択される、請求項27記載の組成物。
- 上記組成物が、ヒトの静脈内、腹腔内、あるいは腫瘍内に投与される、請求項20に記載の組成物。
- (i)メイタンシノイドと化学的に結合した抗体を含有する治療に効果的な量の複合体、(ii)クエン酸塩バッファー、酢酸塩バッファー、コハク酸塩バッファー、およびヒスチジンバッファーからなる群から選択される、複合体1mg当たり0.1mg〜2mgの緩衝剤、(iii)複合体1mg当たり0.5mg〜5mgの抗凍結剤、および(iv)複合体1mg当たり2mg〜20mgの充填剤、を含有しており、水で再構成したときのpHが5〜6である、凍結乾燥された組成物。
- 複合体1mg当たり0.3mgのコハク酸ナトリウムを含有する、請求項30記載の組成物。
- 上記抗凍結剤が、グリセロール、ジメチルスルホキシド、ポリエチレングリコール、デキストラン、グルコース、およびトレハロースからなる群から選択される、請求項30または31に記載の組成物。
- 上記抗凍結剤がスクロースである、請求項30または31に記載の組成物。
- 複合体1mg当たり1mgのスクロースを含有する、請求項33記載の組成物。
- 上記充填剤が、グリシン、マンニトール、およびデキストランからなる群から選択される、請求項30〜34の何れか1項に記載の組成物。
- 上記充填剤がグリシンである、請求項35記載の組成物。
- 複合体1mg当たり3.8mgのグリシンを含有する、請求項36記載の組成物。
- 複合体1mg当たり0.005mg〜0.1mgの界面活性剤をさらに含有する、請求項30〜37の何れか1項に記載の組成物。
- 上記界面活性剤がポリソルベート80である、請求項38記載の組成物。
- 上記界面活性剤がポリソルベート20である、請求項38記載の組成物。
- 複合体1mg当たり0.02mgのポリソルベート20を含有する、請求項40記載の組成物。
- 上記抗体がモノクローナル抗体である、請求項1〜17および請求項30〜41の何れか1項に記載の組成物。
- 上記抗体が、huN901、huMy9−6、huB4、huC242、トラスツズマブ、ビバツズマブ、シブロツズマブ、および、リツキシマブからなる群から選択される、請求項42記載の組成物。
- 上記メイタンシノイドがチオール基を有する、請求項1〜17および請求項30〜43の何れか1項に記載の組成物。
- 上記メイタンシノイドがDM1またはDM4である、請求項44記載の組成物。
- 上記抗体が、ジスルフィド結合、酸に不安定な結合、光に不安定な結合、ペプチダーゼに不安定な結合、チオエーテル結合、およびエステラーゼに不安定な結合からなる群から選択される化学結合を介してメイタンシノイドと化学的に結合している、請求項1〜17および請求項30〜45の何れか1項に記載の組成物。
- 請求項30〜37の何れか1項に記載の凍結乾燥された組成物に水を加える工程を含む、再構成された組成物の調製のための方法。
- 請求項38〜41の何れか1項に記載の凍結乾燥された組成物に水を加える工程を含む、再構成された組成物の調製のための方法。
- 上記再構成された組成物中の上記界面活性剤の濃度が、該再構成された組成物の全体積の0.002%〜0.1%である、請求項48記載の方法。
- 上記再構成された組成物中の上記界面活性剤の濃度が、該再構成された組成物の全体積の0.01%である、請求項49記載の方法。
- 上記再構成された組成物中の上記複合体の濃度が0.1mg/ml〜5mg/mlである、請求項47〜50の何れか1項に記載の方法。
- 上記再構成された組成物中の上記緩衝剤の濃度が2mM〜50mMである、請求項47〜51の何れか1項に記載の方法。
- 上記再構成された組成物が、コハク酸ナトリウムを10mMの濃度で含有し、そのpHが5.5である、請求項52記載の方法。
- 上記再構成された組成物中の抗凍結剤の濃度が、該再構成された組成物の全体積の0.1%〜3%である、請求項47〜53の何れか1項に記載の方法。
- 上記再構成された組成物中の抗凍結剤の濃度が、該再構成された組成物の全体積の0.5%である、請求項54記載の方法。
- 上記再構成された組成物中の充填剤の濃度が、50mM〜500mMである、請求項47〜55の何れか1項に記載の方法。
- 上記再構成された組成物中の充填剤の濃度が、250mMである、請求項56記載の方法。
- 上記細胞が腫瘍細胞である、請求項47〜57の何れか1項に記載の方法。
- 上記抗体が細胞の表面に存在する抗原と結合する、請求項47〜58の何れか1項に記載の方法。
- 上記抗原が、NCAM/CD56、CD33、CD19、GD3、CanAg、PSMA、α葉酸受容体、Her2/neu、CD44v6、胎児膵腺房(FAP)抗原、Cripto−1、CA6、CD20、CA55.1、MN/CA IX、およびコンドロイチン硫酸プロテオグリカンからなる群から選択される、請求項59記載の方法。
- 上記再構成された組成物が、ヒトの静脈内、腹腔内、あるいは腫瘍内に投与される、請求項47〜60の何れか1項に記載の方法。
- 上記組成物が、(i)DM1と化学的に結合したhuN901を含有する5mg/mlの複合体、(ii)10mMのクエン酸ナトリウムバッファー、(iii)0.01%のポリソルベート20、(iv)120mMの塩化ナトリウム、および(v)水、を含有しており、pHが5.5である、請求項2記載の組成物。
- 上記組成物が、(i)DM1と化学的に結合したhuMy9−6を含有する1mg/ml以上の複合体、(ii)10mMのクエン酸ナトリウムバッファー、(iii)0.01%のポリソルベート20、(iv)135mMの塩化ナトリウム、および(v)水、を含有しており、pHが5.5である、請求項2記載の組成物。
- 上記組成物が、(i)DM4と化学的に結合したhuMy9−6を含有する1mg/ml以上の複合体、(ii)10mMのクエン酸ナトリウムバッファー、(iii)0.01%のポリソルベート20、(iv)135mMの塩化ナトリウム、および(v)水、を含有しており、pHが5.5である、請求項2記載の組成物。
- (i)DM1と化学的に結合したhuN901を含有する複合体、(ii)複合体1mg当たり0.3mgのコハク酸ナトリウムバッファー、(iii)複合体1mg当たり0.02mgのポリソルベート20、(iv)複合体1mg当たり1mgのスクロース、および(v)複合体1mg当たり3.8mgのグリシン、を含有する、請求項30記載の組成物。
- 上記充填剤がマンニトールである、請求項35に記載の組成物。
