Description
また、発明者らはマイクロ流体回路内でレーザートラップ、誘電泳動、マイクロキャピラリーフローを複合的に利用して分離を行う非接触式マニピレーション手法を用いた高速分離システムを考案した(特許文献1〜4)。しかし、直接レーザートラップで微生物を搬送するため、微生物に損傷を与える危惧があり、これを避けるために、マイクロツールをレーザートラップして、微生物を押したり引いたりすることで、間接的に搬送する方法も考案したが、実際は操作が難しい等の問題があった。
1.ランダムに分散した大量のサンプルから光学顕微鏡で観察しながら必要なサンプルを選択する。これは一般のセルソーターでは不可能である。
2.ランダムに分散したサンプルから任意のサンプルを選択する。必要なときに、必要なものだけをとりだせる。
3.高速分離を可能とする。
4.多量のサンプル(例えば、5万サンプル)から数サンプルを選ぶ場合に、簡単かつ精度よく分離する。
5.レーザーによる分離ターゲットへの直接照射を避け、分離物の損傷を回避する。特に微生物を分離する場合には、レーザー光が直接あたると、温度上昇や紫外線により微生物が死滅してしまう場合がある。
6.蛍光観察と画像処理を組み合わせて,選択のプロセスを自動化する。蛍光観察を行う場合は、観察面の垂直方向に存在する物体からの蛍光発光により、バックグラウンドノイズが発生し、蛍光観察の画像の分解能が低下し、像がぼやけるというの問題がある。このため、ノイズの原因となる不要な物体を除去できれば、バックグラウンドノイズを大幅に削減でき、コントラストの高い観察が可能となり、近接場のような高額な光学系を備える必要がない等のメリットがある。
7.微生物の培養チップとあわせて、集積化を可能とし、分離から培養までをワンチップ上で行う。
本発明の方法においては、このような多数の微小物を分散したゲルとゾルの相転移を可逆的に引き起こす物質又はそれを含む溶液を微小流路内に流し込む。ある特定の観察領域に存在する微小物体を、例えば顕微鏡により観察して、選択する。特に支持体(容器の底面等)付近に存在しているものの中から目標とするものを選択して、その周辺を局所的に目標物とともにゲル化すると、目標物を支持体(容器の底面等)に固定することができる。目標物をゲルとともに支持体に固定したあとに、微小流路内に流れをおこして、固定されていないものを除去する。十分に除去した後に媒体をゾル化することで粘度を下げる。再び、目標物を回収するための流れをおこすことで、目標物を回収することができる。
1.電極によりゲル化開始温度直前のT2に加熱する。このとき、溶液中はすべてゾル化している。
2.微生物や細胞やDNAなどサンプルが混入した溶液を流し入れる。
3.レーザーにより対象周辺のみをゲル化開始温度を超えT4まで加熱し、ゲル化する。その後すぐにレーザーを切る。しかし一度ゲル化温度T4を超えているため、対象周辺はゲル化したままになっており、底面に保持されたままとなる。また、対象物が微生物のように、直接レーザーを照射したりその他の加熱手段のより直接対象物を加熱することが適当でない場合には、対象物の周辺を局所加熱して対象物を加熱しないまま支持体に固定してもよい。その例を図2に示す。
4.工程3を繰り返すことで、複数の対象を加熱電極上に保持することが可能である。なお、微生物や細胞を対象とする場合には、通電によりこれらを死滅させないように、加熱電極を絶縁体でカバーする。
5.非対象物を除去するためのクリーンな流れを流す。このとき対象はゲル化した部分により底面に固定されており、流されることは無い。よって非対象のもののみが除去される。
6.工程2〜5を繰り返し、対象物を任意数保持することが可能となる。
7.電極加熱を切ることで温度は瞬時にT1になり、対象周辺のゲル化部分が瞬時にゾル化して底面に付着することなく流れに乗せることが可能な状態となる。
8.回収用の流れを流し、対象を回収ポートより回収する。
支持体は目標物を固定し、不要物を除去する際に目標物を固定しておくためのものであるので、このような目的にかなうのもであれば特に制限はない。通常は微小物体と媒体を入れておく容器であるが、容器とは別に、棒状、板状、網状あるいは格子状などの形状をした支持体を用いてもよい。支持体の材質には特に制限はない。但し、支持体が透明であると、その支持体を通して光を当てたり、目標物を観察したりすることが可能となり、更に、透過照明法により観察するタイプの顕微鏡を利用することができるため、好ましい。また、媒体が、温度によりゾルとゲルの相転移を起こすものである場合には、この媒体が加熱出来るものであれば好都合である。例えば、支持体が透明電極(ITO)であれば、これに赤外レーザー光を照射して局部加熱することも出来るし、通電により全体又は部分を加熱することも出来るため特に好ましい。
従来法のエバネッセント照明を利用する方法(例えば、オリンパス光学工業製の全反射蛍光顕微鏡システム、TIRFM)は、エバネッセント光を励起光としてガラスのごく近傍の蛍光分子のみを光らせる方法であるが、エバネッセント照明のための高価な光学系が必要となる。本発明の方法(熱ゲル化)を用いれば、図3に示す支持体近傍の微小物体だけを一時固定し、洗浄によって観察ノイズになる蛍光分子や観察障害物を除去できるため、一般的に利用されている蛍光顕微鏡で感度の高い(バックグラウンドノイズの低い)観察が可能になる。
本発明において、ゲルとゾルの相転移を可逆的に引き起こす手段は、ゲルとゾルの相転移を可逆的に引き起こす物質の性質によって適宜選択すればよいが、温度によりゲルとゾルの相転移を可逆的に引き起こす物質を用いるのが簡便である。
局所加熱の方法としては特に制限はないが、微小電極を微細加工により形成し、抵抗線加熱を行ったり、赤外線照射装置により局所的に加熱してもよい。
加熱電極等の抵抗線加熱を行う場合であって、対象物が微生物等の場合には、抵抗線を絶縁体で被覆して、これらが通電により死滅しないような配慮をすることが好ましい。絶縁体としては、酸化シリコン、窒化シリコン、ポリイミド樹脂等が挙げられる。
また、加熱方法として、注入ポートから湯を注いでもよい。
以下、実施例にて本発明を例証するが本発明を限定することを意図するものではない。
イースト菌(サイズ:約6μm)0.01gをメチルセルロース(信越化学工業社製、メトローズSM−4000)の2%水溶液8mlに混合したものをスピンコートにより基板(表面に電極(ITO)を成膜したガラス)上に平面状に広げる。
目標物をゲルとともに基板底面に固定したあとに、それ以外の不要物を水で洗浄し除去した。十分に除去した後に目標物の加熱をとめて、ゾル化することで固定を解除し、マイクロマニピュレータに取り付けた回収用のピペットを用いて目標物を回収した。
これらのマイクロチップは交差する2つの流路を備えている。これら2つの流路は微細加工により型をつくり、PDMS(ポリジメチルシロキサン)により型どりして微小流体チップを製作した。図5のものは2つの流路が十文字型に交差しており、図6のものは2つの流路がT型に交差しており、一つの排出ポートがサンプルと洗浄液の排出を兼ねている。
一方の流路は分離したい目標物を含むゾル/ゲル相転移物質を投入し、その後固定した目標物以外の不要物を洗浄して除去するための水の流路であり、もう一方の流路は固定した目標物を回収するための流路である。
このマイクロチップをこの光学顕微鏡のステージの上にセットした。このステージは可動であり、観察領域を移動させながら、顕微鏡による観察とレーザー照射が出来る。
その後、透明電極を通電して暖め、透明電極の表面上をゲル化し、表面付近に存在する試料や不純物を透明電極上に一旦固定する。その後、洗浄水注入ポートより水を注入して、固定されていないメチルセルロース等を除去する。
以後、再び抽出ポートのバルブを閉めて、投入ポートから試料を含んだ溶液を投入し、同様の手順を連続的に繰り返すことで、目標物を必要数回収した。
図5のものに比べて図6のものの方が洗浄液や回収液の滞留が少ないため、回収物の純度が高かった。
このマイクロチップに実施例1と同様の試料とメチルセルロースの混合物を投入した。
両方の透明電極を通電して暖め、試料や不純物を全て一旦固定する。その後、実施例1と同様の手順で目標物付近にレーザーを照射する。その様子を図8に示す。その後、透明電極の通電を止め、洗浄水注入ポートより水を注入して、不要物を除去する。その後、残った目標物を回収した。
この方法によれば、下部と上部の電極の局所加熱により固定を行うため、底面付近にない試料であっても環境中に固定できるメリットがある。
このマイクロチップを顕微鏡のステージ上に設置する。顕微鏡の観察倍率に応じて観察領域を変えることができ、ステージを移動することで、広範囲の観察が可能となる。電極の数は複数用意することで、大量の試料を処理することが可能となり作業効率が向上できる。電極への通電過熱による固定では、数ミクロン程度の目標物を一つだけ固定するためには電極の数を増やす必要がある。このため、数千から数万の微小物試料から目標物を一つだけ取り出すには多数の電極が必要になり、システムが複雑になる。そこで、一度に数千から数万の微小物試料を一度に固定できる電極を複数用意する。本実施例では電極A〜Dまで4つの独立した電極を用意した。
まず、微小物試料を投入後、全ての電極に通電加熱して、微小物試料を固定した。必要に応じて、洗浄水を流して固定されていない試料は捨てた。次に、電極Aの通電加熱をやめて、電極A上の微小物試料を評価して目標物を決め、レーザー照射による局所加熱により目標物を固定した。固定後は洗浄水を流して、電極A付近の目標物以外の資料を捨てた。その後、電極Aを通電加熱して先ほど選択した目標物を固定し、レーザー照射をやめた。次に電極B上の微小物試料を評価して先ほどと同じプロセスを繰りかえした。電極C、Dについても同様の手順を繰り返して、少なくとも4個の目標物を固定した。最後に洗浄後に欲しい目標物がのっている電極だけ通電加熱をやめ、回収用の流れによってその目標物だけを抽出した。
なお、ここでは電極が4つの例を示したが、電極の数を増やせば、大量の試料を高速に処理することが可能となる。顕微鏡のステージを自動化すれば、分離作業を自動化することができる。
Claims (22)
- ゾルとゲルの相転移を可逆的に引き起こす媒体、支持体、及び微小物体から成る系において所望の微小物体を回収する方法であって、回収しようとする微小物体の周辺の媒体を局所的にゲル化して該微小物体をこのゲル化した媒体と共に該支持体に固定する段階、該支持体上に固定されていない媒体及び微小物体を除去する段階、及びゲル化した媒体をゾル化して所望の微小物体を回収する段階から成る微小物体を回収する方法。
- 前記媒体が、ゾルとゲルの相転移温度を有し、該相転移温度以上に加熱されることによりゲル化し、該相転移温度以下に冷却されることによりゾル化する請求項1に記載の方法。
- 前記相転移温度が室温〜90℃である請求項2に記載の方法。
- 前記媒体が水溶性のセルロース誘導体又はその溶液である請求項2又は3のいずれか一項に記載の方法。
- 前記セルロース誘導体がメチルセルロースである請求項4に記載の方法。
- 前記支持体が透明である請求項1〜5のいずれか一項に記載の方法。
- 前記支持体が透明電極(ITO)である請求項6に記載の方法。
- 前記微小物体が識別のためのマーカーを有する請求項1〜7のいずれか一項に記載の方法。
- 前記微小物体がDNA分子、細胞又は微生物である請求項1〜7のいずれか一項に記載の方法。
- 前記媒体を相転移温度以上に加熱する手段が赤外線の照射によるものであり、相転移温度以下に冷却する手段がこの照射を止めることである請求項2〜9のいずれか一項に記載の方法。
- 前記加熱手段が赤外レーザーである請求項10に記載の方法。
- 前記赤外レーザーがYAGレーザー又はNd:YVO4レーザーである請求項11に記載の方法。
- 温度によりゾルとゲルの相転移を可逆的に引き起こす媒体及び支持体を保持する空間、該支持体全体を該媒体の相転移温度以上に加熱する手段、該支持体を局部的に該媒体の相転移温度以上に加熱する手段、該空間に該媒体を注入する流路、該空間に洗浄用液体を注入する流路、該空間から洗浄用液体を排出する流路、及び該空間から該媒体を排出する流路から成る、微小物体の回収装置。
- 前記媒体が水溶性のセルロース誘導体又はその溶液である請求項13に記載の装置。
- 前記セルロース誘導体がメチルセルロースである請求項14に記載の装置。
- 前記支持体が透明である請求項13〜15のいずれか一項に記載の装置。
- 前記支持体が透明電極(ITO)である請求項16に記載の装置。
- 更に、加熱手段として前記空間を照射する赤外線照射装置を備えた請求項13〜17のいずれか一項に記載の装置。
- 前記赤外線照射装置が赤外レーザーである請求項18に記載の装置。
- 前記赤外レーザーがYAGレーザー又はNd:YVO4レーザーである請求項19に記載の装置。
- 更に、赤外線照射装置の赤外線を前記空間に集光するレンズを備えた請求項20に記載の装置。
- 更に、前記空間を観察できる顕微鏡を備えた請求項13〜21のいずれか一項に記載の装置。
