Description
本出願は米国仮出願第09/854,786号(2001年5月14日出願)の一部継続出願であってその内容に基づきまたその全体がこの参照により開示に含まれ、また35U.S.C.§120に基づく優先権の利益をここに主張する。
(i)脂質単層の吸着を支持する疎水性表面(たとえば、末端メチル基を示している自己組織化単層膜);これらは膜貫通性Gタンパク質を組み込むのに用いることができないため、有用性は限られている(非特許文献5);(ii)脂質二重膜を結合する親水性表面(たとえば、ガラス表面);これらもまた、吸着された水の層(〜1nm)より厚みの少ない膜外領域を有する膜貫通性Gタンパク質を組み込むのに用いることができるだけであるため、有用性が限られている(非特許文献3;および非特許文献7); そして(iii)疎水性と親水性の部分を含み脂質二重膜を結合する両親媒性表面;これらは多様な膜貫通性タンパク質を組み込むための可能性を与える(非特許文献4;非特許文献6;および非特許文献8)。
(i)誘導体化された単層と反応できる末端官能基;

を含む、部分直鎖または分枝鎖C10〜C25アルキル、アルキニル、アルケニル、アリール、アラルキル、ヘテロアルキル、ヘテロアルキニル、ヘテロアルケニル、ヘテロアリール、またはヘテロアラルキル分子を含むことができる。
図1で説明するように、本発明のアレイ(10)は表面(14)を覆う複数の生体膜プローブマイクロスポット(16)を有する表面(14)を有する基材(12)を含む。アレイ上の個々のプローブマイクロスポットは既知または未知の組成の生体膜を含み、また好ましい実施形態では、膜結合タンパク質を含む。マイクロスポットは複数の異なるタンパク質を含みうる。たとえば、ヘテロダイマーの組に関与する2種類の異なるタンパク質を1つのマイクロスポットに含むことができる。アレイ上のプローブマイクロスポットは任意の都合の良い形状であることができるが、典型的には円形、楕円形、卵形、輪形、またはある種の他の類似の曲線状の形であり、ここでその形は、ある実施形態では、アレイを製造するために使用した特定の方法の結果でありうる。基材の表面上のマイクロスポットすべて、すなわちプローブスポットおよびたとえば、校正スポット、対照スポット、などといった非プローブスポットの両方の密度は、少なくとも約1/cm2であり通常は少なくとも約10/cm2であるが約1000/cm2を超えず、多くの実施形態では約500/cm2を超えず、ここで一部の好ましい実施形態では、その密度は約400/cm2を超えず、通常は約300/cm2を超えず、そしてより普通には約60/cm2を超えない。マイクロスポットはアレイの表面をわたってまたは覆って、たとえば格子を形成するように行と列に、また円形に、など、任意の便利なパターンに配列させることができ、ここで一般的にスポットのパターンは固体支持体の表面をわたる格子状に存在する。
本主題アレイの基材はプローブスポットのパターンが存在する少なくとも1つの表面を含み、ここでその表面は平滑または本質的に平面、またはたとえば窪みまたは隆起といった不規則性を有することができる。スポットのパターンが存在する表面は、表面の性質を望ましい方法で修飾し以下により詳細に記載する1またはそれ以上の異なる層の化合物で修飾されうる。表面はまた多孔質でありうる。
本発明のアレイは選択的にさらにプローブマイクロスポットを含む被覆材料を基材の全体または一部の上に含みうる。好ましくは被覆材料は生体膜マイクロスポットの基材に対する親和性を向上させる。非常に好ましくは、被覆材料は約15°から80°までの範囲の接触角を与える。
本発明に基づいて、本発明で言う「生体膜」はたとえば、小胞、リポソ−ム、脂質単層膜、脂質二重膜、受容体を組み込んだ膜、細胞膜の全体または一部、または再折りたたみタンパク質を含むリポソ−ム、または再折りたたみタンパク質を含む界面活性剤ミセル、などを含むがこれらに限定されない、合成または天然に存在する膜を含む。本発明での使用に適した膜はたとえば、リン脂質、スフィンゴミエリン、コレステロールまたはその誘導体を含むがこれらに限定されない、両親媒性分子である。好ましい実施形態では、膜は膜タンパク質を含む。そのような膜タンパク質は、たとえば、膜貫通タンパク質、膜周辺タンパク質および受容体(たとえば、Gタンパク質−結合受容体、イオンチャンネル受容体、チロシンキナーゼ結合受容体、受容体チロシンキナーゼ、サイトカイン受容体、および固有の酵素活性を有する受容体)を含む。別の実施形態では、膜はイオノフォア(たとえば、バリノマイシン、ノナクチン、メチルモネンシン、クラウンエーテル、クリプタンドまたはその誘導体でありうるがこれらに限定されない)、イオンチャンネル(たとえばタンパク質イオノフォアなどであるがこれに限定されない)、またはたとえば、抗体、酵素、レクチン、色素、キレート剤などであるがこれらに限定されない合成または天然に存在する分析対象、などであるがこれらに限定されないものを組み込んだ脂質二重膜でありうる。
アレイに組み込まれたタンパク質は当該分野の通常の技能を有する者に知られているさまざまな方法のうち任意のものにより製造することができる。本発明のアレイへの組み込みの準備として、タンパク質は天然の起源から得ることができ、あるいは選択的に組み換えDNA法を用いて過剰発現させることができる。タンパク質は、たとえば、GPCR(たとえば、アドレナリン受容体、アンギオテンシン受容体、コレシストキニン受容体、ムスカリン性アセチルコリン受容体、ニューロテンシン受容体、ガラニン受容体、ドーパミン受容体、オピオイド受容体、セロトニン受容体、ソマトスタチン受容体、など)、Gタンパク質、イオンチャンネル(ニコチン性アセチルコリン受容体、ナトリウムおよびカリウムチャンネル、など)、受容体チロシンキナーゼ(たとえば、上皮成長因子(EGF)受容体)、および他の膜結合タンパク質を含む。そのようなタンパク質の突然変異または修飾もまた用いることができる。たとえば、1つまたは複数の点突然変異を有する一部のGPCRは生物学的機能を保持し、疾患に関わっている可能性がある。(非特許文献14を参照。)
さらに、タンパク質はまた(あるいは独立に)N末端に結合した作用剤(またはペプチド)を持つように修飾することができる。そのような方法で修飾されたGPCRは構造的に活性化されている(非特許文献15)。
薬物候補化合物の、同じ器官、組織または単一の細胞さえの中の他のGPCRに対する標的GPCRへの選択性は、医薬開発の間に考慮および観察すべき極めて重要な因子である。現在、ほとんどすべてのHTS技術は一度に1つの標的スクリーニングに関係している。GPCRアレイはさまざまな受容体への複数の目的化合物の選択性を評価するのに用いることができる。本発明の1つの実施形態では、アレイはGPCRテーマアレイを作り、そこで表面上に配列した受容体はGPCRの単一またはいくつかの関連したサブファミリーの成員を含む(実施例3を参照)。
本発明のアレイはマイクロパターン技術を用いて作製する。そのような技術は当該分野でよく知られている。作製の好ましい方法においては、プローブの先端(「ピン」ともいう)を生体膜の溶液に浸す。先端を溶液から取り出し、先端に付着した溶液を得る。溶液を基材の表面に接触させ、それによって溶液を先端から表面へ移す。
本発明はまた生体膜アレイを使用する方法も提供する。本発明のアレイは特に医薬開発、医療診断学、プロテオミクスおよびバイオセンサーにおける使用に適している。アレイに送達される試料は典型的には液体である。
本発明のアレイは、GPCRの活性化およびコエフェクターを同定するためのマイクロアレイに基づく不均一性分析に用いることができる。この用途のためには、分析では、標識された加水分解されないGTP(たとえば、放射性[35S]GTPγSまたはその蛍光類縁物質(たとえば、BODIPY−FL−GTPγS))を用いてGTPγSのi)過剰発現したGPCRおよびGタンパク質を含む細胞膜調製物;またはii)目的の受容体およびそのコエフェクターを含む再構成された小胞/ミセル、のアレイへのリガンド刺激結合を観察する。この手法はGPCRに対する作用剤の高処理量でのスクリーニングを可能にするだけでなく、GPCRのコエフェクター(たとえば、結合Gαタンパク質)の同定も可能にする。
本発明の生体膜アレイはマイクロプレートを用いて製作することができる。受容体がその生物学的機能を保持するように受容体をマイクロプレート表面上に固定化するため、マイクロプレートの少なくとも底面は修飾されている。ある実施形態ではウェル全体の表面を修飾するのが望ましい。修飾に応じて、マイクロプレートの底はガラス、高分子、または金を基礎としたものとすることができる。いくつかの異なる表面化学を修飾に用いることができ、たとえば下記が含まれる:
(1)シラン化表面。たとえば、ガラス底を有するマイクロプレートは3−アミノプロピルトリエトキシシランといったアミン末端を有するシランの気相蒸着法または溶液相析出法を用いて修飾することができる。金で被覆したマイクロプレートはアルカンチオラートのSAMのシラン化を用いて修飾することができる。ポリスチレン底を有するマイクロプレートは、これらの表面をガンマ線で活性化後に気相蒸着法または溶液相析出法を用いて、またはこれらの表面へのシランの架橋を用いてシラン化することができる。
材料
ヒトβ−アドレナリン受容体サブタイプ1(β1)およびドーパミン受容体サブタイプ1(D1)の膜調製物はBiosignal Packard(Montreal, Canada)より購入した。これらの受容体結合膜は10mM Tris−HCl、pH7.4および10%グリセロールを含む緩衝溶液中に懸濁されて販売されていた。ヒトクローン化ニューロテンシン受容体サブタイプ1(NT1R)およびBODIPY−TMR−ニューロテンシン(BT−NT)はPerkin Elmer Life Science (Boston, MA)から購入し、10mM Tris−HCl(pH7.4)および10%スクロースを含む緩衝溶液中の膜結合懸濁液として受領した。BODIPY−TMR−CGP12177(BT−CGP)およびBODIPY−FL−SCH23390(BF−SCH)はMolecular Probes(Eugene, OR)から購入した。CGP12177およびSCH23390はTocris Cookson, Inc (Ballwin, MO)から購入した。ニューロテンシンはSigma Chemical Co.(St. Louis, MO)から購入した。Corning CMT−GAPSスライドは受領時の状態で使用した。蛍光標識リガンドおよびニューロテンシンはDMSOに溶解し−20℃にて保存した。使用前に、リガンド溶液は50mM Tris−HCl、2mM EDTA、1mM MgCl2、pH7.4および0.1%ウシ血清アルブミン(BSA)から成る結合緩衝液を用いて希釈した。
管状ピン(Telechem)を装備した自動ピンプリンター(Model PS 5000, Cartesian Technologies Inc.)を用いて、GPCRまたは脂質の複数アレイを各スライド(Corning CMT−GAPSスライド)にプリントした。各3X3または5X5要素のアレイは隣接するアレイから少なくとも6mm離した。GPCRを含む膜調製物は、メーカーから受領したままでさらに精製または希釈を行わずにプリントに使用した。プリント後アレイを湿潤室内で室温にて1時間インキュベートし、その後リガンド結合実験に使用した。より長期の保存には、アレイはデシケータ内で4℃にて保存した。
任意のスライド状の各アレイはリガンドを含む緩衝溶液(50mM Tris−HCl、2mM EDTA、1mM MgCl2、pH7.4、0.1%BSA)10μLと共に1時間インキュベートした。インキュベート後、真空ポンプに接続したピペットチップを用いて溶液を注意深く除去した。スライドを水で簡単に洗浄し窒素気流下で乾燥した。スライドをGenePix4000Aスキャナ(Axon Instruments, Foster City, CA)でイメージングした。
2%(mol%)テキサスレッドDHPEと混合した1,2−ジラウロイル−sn−グリセロ−2−ホスホコリン(DLPC)の小さな1枚膜リポソーム(SUV)を、緩衝液中の脂質(1mg/mL)懸濁液を超音波処理して生成した;これらの小胞を次いで基材と共にインキュベートした。注意深く徹底的に洗浄後、支持された脂質膜がこれらの表面上に形成された。FRAP実験は裸のガラスおよびGAPSスライド上のこれらの支持された脂質膜上で、CCD検出器(Princeton Instruments)を装備したOlympus AX70エピ蛍光顕微鏡を用いて実施した。
GPCRアレイの製作と保存
GPCRのアレイを管状ピンプリンターを用いて従来の自動ピンプリンターによって、実験の項に記載した通り製作した。Boxerと共同研究者は膜を固体支持体上へ水中で移すことの重要性を記載している;我々は、しかし、ピン上を濡らす脂質溶液が水中で一部がピンから解離しプリント中にクロスコンタミネーションの原因となることを危惧した。さらに、市販プリンターのスライドラックは水中でのプリント用には設定されていない。既製のプリント装置を膜タンパク質アレイの製作に用いることができることは、生物分析用途にこれらのアレイが幅広く製作および開発されることに向けた重要な一歩である。
我々は膜タンパク質アレイのための頑健な結合分析の開発に関心があった。Boxerおよび共同研究者は、裸のガラス基材上に吸着された脂質は、空気−水界面を通って引き出したとき自発的に脱離したことを報告している(非特許文献19)バイオアッセイでは手順でしばしばスライドを溶液から引き出す必要があり(たとえば、連続的浸漬による洗浄の間)、我々は、この挙動がバイオアッセイのための膜タンパク質アレイの使用を制限すると考えた。我々はしたがって、機械的に安定な支持された膜の吸着を支持する表面を調べた;安定性についての我々の判定基準は、空気−水界面を通って引き出したとき、支持された膜が吸着されたまま残ることであった。試験した数種類の表面のうち、CMT−GAPS表面が最も安定して支持された脂質を与えた。図3Aは空気−水界面を通って引き出し、水に浸漬し、乾燥し、再び水中に浸漬した、緩衝液に浸漬したフルオレセイン−DHPEを添加したDPPC/DMPCから成る支持された膜のアレイの蛍光画像を示す。我々はこれらの連続した浸漬と引き出しを通じて、これらの脂質マイクロスポットの蛍光強度に何ら低下を認めなかった;これらの結果は、結合した脂質は安定であることを示す。図3Bは卵PCから成る脂質についてのデータを示す;連続した浸漬と引き出しに供された場合、これらの脂質のアレイもまた安定である。室温で、DMPC/DPPC脂質はゲル相にあり、一方で卵PCは液相にある。これらの実験は、支持された脂質アレイが、それらがゲル相にあるか液相にあるかにかかわらず、GAPS被覆基材上で機械的に頑健であることを実証する。
GPCRのアレイは上記の通り、管状ピンプリンターを用いることによって製作された。アレイは次いで直接分析または競合分析で、蛍光標識同族リガンドとインキュベートした。図4は1枚のCMT−GAPSスライド上にプリントした5つの別々のアレイの彩色蛍光画像を示す;個々のアレイそれぞれが5連のスポットを含む3列を含む;各列は異なるGPCRタンパク質を表す。これらのタンパク質は、左から右へ、それぞれアドレナリン受容体(β1)、ニューロテンシン受容体(NTR1)およびドーパミン(D1)受容体である。第1のアレイ(図4A)は結合緩衝液だけとインキュベートした。予想される通り、蛍光は観察されなかった。第2のアレイ(図4B)は蛍光標識ニューロテンシン(BT−NT、2nM)を含む緩衝液とインキュベートした。画像はNTR1に対応するアレイだけが強い蛍光シグナルを見せることを示す;この観察は、BT−NTのNTR1への結合は選択的であることを示唆する。その相互作用の特異性はさらに、BT−NT(2nM)、CGP12177(1nM)(図4C)、SCH23390(1nM)(図4D)、またはニューロテンシン(1nM)(図4E)のいずれかとを含む溶液とアレイをインキュベートすることによって実証された。図4Bと比較して、NTR1に対応するスポットの強度に図4Cおよび4Dでは目立った減少は見られない。CGP12177およびSCH23390はNTR1と結合しない;ゆえに、それらを結合溶液に添加しても、我々の観察と合致して、BT−NTのNTR1との相互作用を阻害しない。ニューロテンシンはNTR1の同族リガンドであり、ゆえに、それはNTR1アレイ上の結合部位を奪い合う。図4Eでは、ニューロテンシンをBT−NTに対して500倍過剰に含む溶液とアレイをインキュベートした;この比率では、ニューロテンシンはBT−NTのNTR1への結合を完全に阻害すると予想される。我々はNTR1アレイに対応するシグナルを観察しなかった;ゆえに、ニューロテンシンはNTR1への結合を特異的に阻害することができる。これらの実験は、リガンドと阻害剤の結合を試験する分析がGPCRアレイを用いて実現可能かどうかを実証する。
我々は、プリントしたGPCRアレイの、異なる濃度の同族リガンドへの反応を調べている。図5AはBT−NTで処理したニューロテンシン受容体のアレイの蛍光画像を示す;データは、より高い濃度の蛍光標識リガンドで処理された場合、アレイの蛍光強度が増加することを示す。BT−NTのNTR1アレイへの結合については、検出限界は0.1−0.2nMBT−NTであった。これらの結果は、この系については蛍光標識リガンドを利用したGPCRアレイのダイナミックレンジは〜2logsであることを示唆する。図SBはcy5−標識アンタゴニストDのガラニン受容体アレイへの結合についてのデータを示す;画像はマイクロスポットの蛍光強度はリガンドの濃度に依存することを示す。
小麦胚芽凝集素表面修飾
スライドガラス(Corning)は使用前にピラニアエッチング液(濃硫酸:30%過酸化水素、7:3)に30分間浸漬後蒸留水ですすいで洗浄した。スライドガラスは3−イソシアナートプロピルトリエトキシシラン5%を含むエタノール溶液に1時間浸漬し、次いでエタノールと水ですすぎ、最後に窒素気流下で乾燥した。
ファミリー特異的アレイ
アドレナリン受容体という同一のファミリーに属する3種類の受容体を同じ格子の中に配列し、この格子の複製を1つのスライド上に配列した。受容体はそれぞれβ1、β2およびα2Aであった。CGP12177はβ1/β2アンタゴニストで、またβ3部分作用剤であることが報告されている。アレイは5nM BODIPY−TMR−CGP12177と共にさまざまな濃度のICI118551の存在下および非存在下でインキュベートした。図9A、9Bおよび9Cに図解する通り、β1およびβ2受容体と対応するスポットだけが明るい。10nM ICI118551(選択的β2作用剤)の存在下で、β2受容体の蛍光強度は減少し、一方でβ1受容体の強度はほぼ同じレベルに留まる。しかし、ICI118551の濃度が500nMに上昇すると、β1の蛍光強度は劇的に低下し、一方でβ2の強度は10nM ICI118551の存在下で観察された強度と同様である。これらの結果は、ICI118551はβ1およびβ2受容体の両方との結合で、しかしβ2とのほうがより高い親和性でBODIPY−TMR−CGP12177と競合することができること、およびGPCRテーマアレイを用いてβ1およびβ2受容体に対するICI118551の選択性を識別できることを説明する。
GPCR固定化のための金表面上のアルカンチオラートのアミノシラン化SAM
実験の詳細
表面の調製
顕微鏡用金被覆スライド(10nM Cr接着層、100nmAu)はEvaporated Metal Filmsから購入し、使用前にピラニアエッチング液(濃硫酸:30%過酸化水素、7:3)に30秒間浸漬後蒸留水で徹底的にすすいで洗浄した。11−メルカプトウンデカン酸(MUA)または11−メルカプトウンデカノール(MUD)のSAMは金スライドをチオールの1mMエタノール溶液に少なくとも1時間浸漬することによって形成した。MUA SAMのカルボン酸基は、試料を30分間、75mMの1−エチル−3−(3−ジメチルアミノプロピル)カルボジイミド(EDC)および15mMのN−ヒドロキシスクシニミド(NHS)を含む蒸留水溶液中で反応させることによって、活性エステルに変換した。この活性化工程は、アミノ末端を有するシランの結合に、必要ではないが好ましいことが分かった。MUDおよびNHS−エステル修飾MUAのSAMのシラン化は、スライドを一時間、3−アミノプロピルトリエトキシシラン(APTES)の5%(v/v)トルエン溶液に浸漬することによって達成された。試料はその後、トルエンに5分間浸漬し、エタノールですすぎ、そして乾燥した。
GPCRのアレイは、管状ピン(Telechem)を装備したCartesianプリンターを用いてAPTESシラン化金表面上にプリントした。ヒトβ−アドレナリン受容体サブタイプ1(β1)の膜調製物はBiosignal Packard (Montreal, Canada)から購入した。これらの受容体結合膜は10mMTris−HCl、pH7.4および10%グリセロールを含む緩衝液に懸濁されて販売されており、プリントに直接用いられた。プリント後、アレイを20μM非標識CGP12177、プロプラノロール、またはベタキソロールの非存在下または存在下で、5nM BODIPY−テトラメチルローダミン(BT)標識CGP12177を含む溶液10μLとインキュベートした。これらの反応は50mM Tris−HCl(pH7.4)、10mM MgCl2、および0.1%BSAを含む結合緩衝液中で実施した。

Lesker Inc.)の熱蒸発によって調製した。ヒトガラニン受容体サブタイプ1(NEN Life Sciences)およびβ−アドレナリン受容体サブタイプ1で、APTESで修飾したMUD SAM上にアレイを形成した。試料はSF−10等辺プリズムに屈折率マッチング液(n=1.730、Cargille Labs)を用いて結合し、次いでSPR Imager装置(GWC Instruments)に取り付けた。要約すると、この機器は平行白色光源、帯域通過干渉フィルター(794nm±1nm)、回転ステージ、試料台、フローセル、および8ビットCCDカメラから成る。CCDカメラレンズは6.3X顕微鏡対物レンズ(Melles Griot)と交換した。アレイの最初の画像は結合緩衝液中で得られた。アレイを次いでガラニン(5nM)の結合緩衝液溶液に曝露し、60分間反応させ、その後、緩衝液で洗浄した。アレイの最後の画像を撮り、そして最後のSPR画像から最初のSPR画像を差し引いて差分SPR画像を生成した。画像はScion Image(Scion Image for Windows(登録商標)は米国国立衛生研究所にて開発された公有画像取り込みおよび解析プログラムであるNIH Imageの移植である;www.scioncorp.comを参照)を用いて解析した。
金表面上へのGPCRの結合のために2つの異なる表面化学が開発された(図10Aおよび10Bを参照)。図式1(図10A)は3−アミノプロピルトリメトキシシラン(APTES)と1工程で反応する開始層として11−メルカプトウンデカノール(MUD)のヒドロキシル末端を有するSAMを利用する。この反応はクロロ−およびアルコキシシランと、シリコンの表面水酸基とのよく知られた反応と類似している。シランの化学はアルカンチオラートのSAMの修飾にも用いられてきている。たとえば、Itoh et al.は銅表面上のMUDのSAMを修飾するのにアルキルトリクロロシランを用い、銅の腐食耐性を向上させた。Sun et alは、金表面上のチオフェノールのヒドロキシルおよびアミン末端を有するSAMと、ジメチルオクチルクロロシランとの気相反応を実証した。図式2(図10B)は11−メルカプトウンデカン酸(MUA)の開始層を使用する。APTESとの反応の前に、このSAMのカルボン酸基は最初に、1−エチル−3−(3−ジメチルアミノプロピル)−カルボジイミド(EDC)とN−ヒドロキシスクシニミド(NHS)を用いて反応性のNHSエステルに変換される。APTES分子のネットワークはアミド結合形成によって共有結合で表面に固定される。ここで記載する作業については、予備的な偏光測定はMUDおよびMUA表面上でそれぞれ〜43Åおよび〜96ÅのAPTESの厚みを示す。
{1−(S阻害/S対照)}×100
ここでS阻害は標識CGPおよび非標識リガンドと共にインキュベートしたアレイの正味の蛍光シグナルで、S対照は標識CGPだけと共にインキュベートしたアレイの正味の蛍光シグナルである。図12はMUAおよびMUDのSAMに結合したAPTESについて実施したこれらの実験の結果を示す。標識リガンドのβ1受容体への結合の強く特異的な阻害が3つすべての阻害剤について観察された;わずかに良好な阻害レベルがMUAについて得られた。同様なレベルの阻害が、CMT−GAPSスライド上にプリントしたアレイについて観察された(データ記載せず)。
我々は生物学的に活性な形でGPCRの固定化を可能にする金表面上の表面化学を開発した。金基材の使用の結果、ガラス基材上にプリントされたアレイと比較してシグナル対バックグラウンド比が向上した。これらの表面のもう一つの利点は、たとえばSPRといった非標識技術による直接分析方式においてリガンドを検出する可能性である。本発明は金表面へのGPCRの固定化のためのAPTESの特定用途を記載するが、いろいろなシランによるアルカンチオールの化学修飾はさまざまなタンパク質の固定化のために広く有用であると予想される。
機能分析
実験
ヒトオピオイド受容体サブタイプμおよびδ2はPerkin Elmer Life Science (Boston, MA)から購入した。受容体結合膜は最初は10mM Tris−HCl、5mM MgCl2および10%スクロース、pH7.4に懸濁されており、それ以上の処理無しでプリントに直接用いられた。
GPCRμおよびδ2−オピオイド受容体のアレイを、固定(25nM)濃度のBODIPY−フルオレセイン標識GTPγSを含む溶液と、リガンドであるダイノルフィンAの存在下および非存在下でインキュベートした。図14A、14Bおよび14Cはこの実験の結果を示す。ダイノルフィンAの存在下でインキュベートしたアレイは顕著により高い蛍光を示す。具体的には、基底レベル蛍光の増加はμ受容体について〜2.5倍(図14Bを参照)、δ2受容体について〜10倍(図14Cを参照)であった。この結果は(i)ダイノルフィンAは細胞膜調製物中に存在するGαタンパク質へのGTPγSの結合を促進する;および(ii)アレイ中のGPCRは機能を有することを示唆する。
金表面上のアミノ末端を有するアルカンチオラートのSAM上の上皮成長因子受容体マイクロアレイ
実験
ヒト上皮成長因子受容体(EGFR)はPerkin Elmer Life Science(Boston, MA)から購入した。受容体膜調製物は、正常上皮細胞と比較してEGFRを〜100倍過剰に発現するヒトA431細胞(上皮腫瘍細胞)に由来した。受容体結合膜は最初は50mM HEPES−KOH、138mM NaCl、5mM KCl、および10%スクロース、pH7.7に懸濁されており、それ以上の処理無しでプリントに直接用いられた。上皮成長因子(EGF)およびテトラメチルローダミン標識上皮成長因子(TM−EGF)はMolecular Probes(Eugene, OR)から購入した。EGFおよびTM−EGFは2mMアジ化ナトリウムを含む蒸留水中で再構成し、−80℃での長期保存のために1回分の使用量ずつに分割した。
上皮成長因子受容体(EGFR)は170kDaの膜結合糖タンパク質で上皮細胞の表面に発現した受容体タンパク質チロシンキナーゼである。EGFRは細胞周期調節分子の一種であるタンパク質チロシンキナーゼの成長因子受容体ファミリーの一員である。受容体はそのリガンド(EGF、TGF−α)が細胞外領域に結合する場合、二量化、立体構造変化、内在化を受ける。EGFは、表皮細胞およびその他の細胞の分裂を刺激する、53アミノ酸のポリペプチドホルモンである。
Claims (9)
- 非共有的にまたは共有的に、ガラス基材の表面に安定に結合している、G−タンパク質共役受容体(GPCR)を含む複数の生体膜マイクロスポットを有するアレイであって、前記基材の表面がγ−アミノプロピルシランを含み、空気−液界面を通して引き上げたときに前記マイクロスポットの基材に対し相対的な位置が維持されることを特徴とするアレイ。
- 前記GPCRが、同様な組織分布または同様な生理的もしくは薬理的機能を有するGPCRサブファミリーの一員であるさまざまなGPCRを含むことを特徴とする請求項1記載のアレイ。
- 前記GPCRがa)細胞内領域またはb)細胞外領域を含み、さらに該細胞内領域または細胞外領域がそれぞれ基材の表面を向くようにGPCRが配向されていることを特徴とする請求項1または2記載のアレイ。
- 前記γ−アミノプロピルシランが3−アミノプロピルトリエトキシシランであることを特徴とする請求項1から3いずれか1項記載のアレイ。
- 前記基材の表面が多孔質であることを特徴とする請求項1から4いずれか1項記載のアレイ。
- GPCRプローブと標的化合物との間の結合事象を検出する方法であって、
前記標的化合物を含む溶液を、請求項1から5いずれか1項記載のアレイと接触させる工程、および
前記プローブと、前記標的化合物との間の結合事象を検出する工程、
を含む方法。 - 前記マイクロスポットのアレイを、標識同族標的および非標識標的化合物とインキュベートし、前記標識同族標的と前記非標識標的化合物間のプローブに対する競合による前記標識の信号の減少を測定することによって、前記非標識標的化合物と前記プローブとの間の前記結合事象が測定されることを特徴とする請求項6記載の方法。
- 前記標的化合物が非標識であって、標識された加水分解されないGTPの濃度を測定することによって前記標的化合物の結合を検出することを特徴とする請求項6記載の方法。
- 前記加水分解されないGTPが、放射性同位体または蛍光化合物で標識されているGTPγSであることを特徴とする請求項8記載の方法。
