Description
本発明は米国国立癌研究所(the National Cancer Institute)により資金援助を受けた研究過程において成されたものであった。米連邦政府は本発明における所定の権利を有する。
発明の背景
免疫応答の刺激化は宿主免疫系により外来物として認識される抗原の存在に依存する。癌特異的抗原の存在の発見により現在では宿主の免疫系を用いて腫瘍成長を妨害することの可能性が強まってきている。免疫系の両腕とも言うべき体液性免疫と細胞性免疫の両方を活用する様々なメカニズムが、癌の免疫療法について現在のところ探索されている。
細胞性免疫応答の因子は腫瘍細胞を特異的に認識かつ破壊することが可能である。腫瘍浸潤細胞集団からかもしくは末梢血からの細胞障害性T細胞(CTC)の単離により、このような細胞が癌に対する天然の免疫防御において重要な役割を担っていることが示唆されている(Cheever et al.、Annals N.Y. Acad.Sci. 1993 690:101−112)。サイトゾル(cytosol)中に存在する蛋白質に由来し、8〜10の残基からなり、主要組織適合遺伝子複合体(MHC)のクラスI分子により提示されるペプチドを認識するCD+8 T細胞(TCD8+)が特にこの応答においては重要な役割を担っていると考えられる。腫瘍細胞を特異的に認識し、かつ養子免疫細胞移入後に治療活性(幾つかの症例ではこの活性には完全緩解が含まれる)を示すマウスおよびヒトの両CD+8については、双方共現在多大な数の例が存在する。しかしながら、腫瘍を根絶するというT細胞についての可能性にも拘わらず、インビボではTCD8+による認識を多くの腫瘍が回避することが大半の癌の進行性成長から明らかである。インビボでは十分なT細胞の誘導が非常に有効という訳ではない。多種多様の腫瘍が免疫原性であることが見いだされてはいるものの、有効な抗腫瘍免疫応答の刺激化は未だに見いだされてはいない。
この現象についての一つの説明は、腫瘍はT細胞に抗原特異的シグナルを伝達することが可能であってよいが、ただしT細胞の完全な活性化にとって必要な共刺激シグナルを伝達することは可能ではないということである。T細胞の共刺激は抗原呈示細胞上の表面分子B7がCD28として知られるT細胞分子と相互作用を行う際に生じる。抗原特異的シグナル(ただしB7ではない)を受け取るT細胞が不応答性になることが観察されている。多くの腫瘍細胞はB7蛋白質を保持してはいないため、B7は癌細胞に添加されたのである(Travis,J.、Science 1993 259、310−311)。黒色腫細胞上での共刺激性リガンドB7の発現によりインビボでのマウス黒色腫の排除が誘導されることが示されている(Townsend,S.E. and Allison,J.P.、Science 1993、259、368−370)。この排除はCD8+ T細胞により媒介されることが見いだされており;CD4+ T細胞は必要とされなかった。これらの結果により、B7発現により腫瘍細胞は効果的な抗原呈示が可能となり、そのためインビボでのそれら腫瘍の根絶がもたらされてよいことが示唆される。
腫瘍進行の際のサイトカインの局在的分泌の効果も研究されている。レトロウイルスベクターを介して組込まれるヒトIL−2遺伝子でトランスフェクトさせたマウス線維肉腫細胞株における低レベルのインターロイキン−2(IL−2)の分泌は、これらの細胞の腫瘍形成性を排除し、かつ腫瘍形成性用量の親細胞での後続の攻撃誘発に対する長期持続性の防御免疫応答を誘導することが見いだされた(Gansbacher et al.、J.Exp.Med.1990、172、1217−1224)。もう一つの研究では、自然発症性マウス腎臓細胞腫瘍からの細胞を工学的に操作して局所的に大用量のインターロイキン−4(IL−4)を分泌させた(Golumbek et al.、Science 1991、254、713−716)。腫瘍細胞を注入した動物は、おおかたはT細胞に無関係な様式でIL−4でトランスフェクトさせた腫瘍を排除した。しかしながら、これらの動物はその親腫瘍に対するT細胞依存的全身性免疫を生じた。この全身性免疫は腫瘍特異的であり、かつCD8+ T細胞により媒介された。これらの実験により、親腫瘍を、遺伝子工学的に創製された腫瘍細胞の注入によって全身性免疫応答を生じさせることにより救済することが可能であってよいことが示唆される。
幾つかの腫瘍細胞がインビボおよびインビトロで低レベルのクラスI分子を発現することを示唆する証拠も存在する。細胞内抗原は、腫瘍組織適合遺伝子複合体(MHC)クラスI分子によるCD+8T細胞への呈示の以前にプロセシングを受けなければならない。26の異なるヒト腫瘍株の抗原プロセシング効率が研究されている(Restifo et al.、J.of Exp.Med.1993、177、265−272)。3つの異なる細胞株(全てヒト小細胞肺癌である)は一貫して、T細胞への呈示のための内因的に合成された蛋白質のプロセシングを行わない。パルス−チェース実験により、MHCクラスI分子はこれらの細胞株によっては小胞体から細胞表面には輸送されないことが示された。ノザン(Northern)ブロット分析により、これらの細胞はMHCをコードするプロテオソームおよび輸送体遺伝子をコードするmRNAはほとんど含まないか、あるいは全く含まないことが示された。インターフェロンγでの処置によりこれらmRNAの発現が亢進し、かつ観察された機能的および生化学的な欠失が逆行した。従って、MHCをコードするプロテアソームおよびトランスポーター遺伝子の転写のレベルで抗原プロセシングを亢進させることを含む可能な治療的適用法が示唆された。
抗原性ペプチドをコードする遺伝子を保持する組換え
もしくはサルモネラ(Salmonella)細菌で患者を免疫化することも経口腫瘍免疫療法として示唆されている(Boon et al.、Annu.Rev.Immunol.、1994、12、337−65)。経口投与された生の弱毒化サルモネラ(Salmonella)組換えワクチン(これは、全長のP.ペルグヘイ(P.perghei)の環種虫(circumsporozite)抗原を発現する)がマラリアからマウスを防御することが示されている。この免疫応答はCD8+ T細胞の誘導により媒介される(Aggarwal et al.、J.of Exp.Med.1990、172、1083−1090)。生の弱毒化サルモネラ(Salmonella)組換え体は、CTCにより媒介される免疫が重要であってよい他の疾患の研究において有用であってよいことが示唆されているが、他の実験は全く報告されていない。BCGは多種多様のウイルス性、細菌性、および単細胞動物性抗原に対する液性および細胞性の両免疫応答を刺激化するのに有効であることが判明してよい新規生ワクチン媒介体として作用することも示唆されている(Stover et al.、Nature 1991、351、456−460)。
今回我々は、腫瘍特異的抗原に対する免疫応答を、腫瘍特異的抗原もしくはその断片を発現する細胞内細菌リステリア モノサイトゲネス(Listeria monocytogenes)の組換え体(recombinant form)を含むワクチンベクターの投与により誘導することができることを見いだした。このワクチンベクターは、既存の腫瘍のサイズを縮小させ、かつ原発性腫瘍の形成を阻害するのに有効であることを見いだした。抗原呈示後の他の刺激化はこの応答を誘導するのには全く必要とされなかった。
発明の要約
本発明の目的は、癌を有する宿主内で腫瘍特異的抗原に対する免疫応答を誘導する方法を提供することであり、この方法は、腫瘍特異的抗原もしくはその断片を発現することが可能なリステリア モノサイトゲネス(Listeria monocytogenes)の組換え体を含むワクチンの有効量をその宿主に投与することを含む。
本発明のもう一つの目的は、腫瘍特異的抗原に対する免疫応答を誘導するための、腫瘍特異的抗原もしくはその断片を発現することが可能なリステリア モノサイトゲネス(Listeria monocytogenes)の組換え体を含むワクチンのための使用法を提供することである。

発明の詳細な説明
L.モノサイトゲネス(L.monocytogenes)に対する免疫応答はTH1、CD4+ T細胞、およびCD8+ T細胞の応答であり、液性応答は非常に微弱にのみ生じるに過ぎないことが示されている。外来性蛋白質であるβ−ガラクトシダーゼ(Schafer et al.、J.Immunol.、1992、149、53−59)、インフルエンザ核蛋白質、ならびにHIV gagおよびnef遺伝子産物を発現する野生型細菌の組換え形態が開発されている。リステリア(Listeria)の染色体内にこれらの蛋白質を、それらがその細菌により分泌される様式で安定に組み込ませるための組換え技術が開発されている。これら組換えベクターの全てが強力で抗原特異的なCTC応答をインビボで誘発する。従ってこの細菌は、腫瘍特異的蛋白質に対するCTC応答を増強させるための理想的ワクチンベクター、かつ癌に対するワクチンとして細胞性免疫応答を開始させるための独特な系として作用する。
例えばL.モノサイトゲネス(L.monocytogenes)のような生ワクチンの投与により、死滅化調製物もしくは可溶性蛋白質とアジュバントとでは誘導できないことがしばしばである長期持続性細胞性免疫がもたらされる。L.モノサイトゲネス(L.monocytogenes)のライフサイクルの独特な特徴は、L.モノサイトゲネス(L.monocytogenes)が宿主細胞に浸潤し、そしてファゴソーム内に取り込まれ、そしてそこから抜け出し、その後に細胞質内で生息および複製することである。Tilney,L.G.およびD.A.Portnoy、J.Cell Biol. 1989 109、1597。従ってこのL.モノサイトゲネス(L.monocytogenes)ベクターは、外来性蛋白質および蛋白質断片の標的をクラスI MHCに限定された回路に合わせる能力を提供する。一層効果的なベクターであることに加え、L.モノサイトゲネス(L.monocytogenes)(これはグラム陽性生物体である)は更に、他の多くの生のベクターと比較すると安全性が高く、なぜならL.モノサイトゲネス(L.monocytogenes)はペニシリンを初めとする大部分の抗生物質に対して非常に高い感受性を示すためである。更にL.モノサイトゲネス(L.monocytogenes)は、例えばサルモネラ エスピー(Salmonella sp.)のようなグラム陰性ベクターが示すエンドトキシンからの毒性に関連する問題は有さない。ワクチンとして既に広く用いられているベクター(例えば、ワクシニア(Vaccinia)もしくはBCG)による効果的な追加免疫を妨げることになる先取り免疫は、これまでにワクチン開発に用いられていないL.モノサイトゲネス(L.monocytogenes)については問題になりそうもない。無発病性であるが依然として防御的であるL.モノサイトゲネス(L.monocytogenes)の突然変異体株も潜在的ワクチン候補物として検査に利用可能である。
モデルマウス系を用いることで、L.モノサイトゲネス(L.monocytogenes)が腫瘍細胞により発現される蛋白質に対する免疫応答を誘導することができることが現在見いだされている。この免疫応答により、健常で免疫化されたマウスへの腫瘍細胞の転移の拒絶がもたらされ、かつ腫瘍成長が既に開始されてしまっているマウスにおける腫瘍成長に変化が与えられる。図3〜7を参照されたい。
組換えL.モノサイトゲネス(L.monocytogenes)を含むワクチンがCT26(マウス結腸直腸癌)およびRENCA(マウス腎臓癌)の成長に対する特異的防御免疫を誘導する能力を調査した。予備実験ではL.モノサイトゲネス(L.monocytogenes)を工学的に操作して、主要分泌性リステリア(Listeria)蛋白質であるリステリオリシンO(listeriolysin O)(LLO)(これはヘモリシン(hemolysin)遺伝子の産物である)との融合蛋白質として、A/PR/8/34から核蛋白質(NP)を分泌させるようにさせた。LLOは通常はL.モノサイトゲネス(L.monocytogenes)により宿主小胞内で発現および分泌され、かつその細菌が細胞質内へ逃げ込むのに必要とされる。NP分泌性L.モノサイトゲネス(L.monocytogenes)組換え体が、3つのマウス株からの大半のインフルエンザ特異的T細胞による認識のためには抗原プロセシングのクラスI回路を標的とする能力を調査した。LLO−NP融合蛋白質が3つのMHCクラスIハプロタイプ(このハプロタイプに対してA/PR/8/34の応答が限定される)(すなわち、Kd、Db、およびKk)による呈示のための適切なプロセシングを受けることが決定された。様々な用量のLM−NPでのBalb/cマウスの免疫化により強力な抗−NP CTC応答がもたらされることが示された。
更に進んだ実験ではマウスを3つの群に分割した。一つの群には野生型L.モノサイトゲネス(L.monocytogenes)のLD50の1/10での免疫化を施し、もう一つの群には滅菌食塩水での免疫化を施し、そして3番目の群にはインフルエンザ核蛋白質を分泌するように形質転換させた組換えL.モノサイトゲネス(L.monocytogenes)ワクチンベクター(LM−NP)での免疫化を施した。2週間後に各群に類似のブースター免疫を投与した。この免疫化スケジュールはインフルエンザ核蛋白質に対する強力なCTC応答を産生するように決定した。最終免疫後2週間目に各群の動物を、L.モノサイトゲネス(L.monocytogenes)ベクターを形質転換するのに用いたのと同一のインフルエンザ核蛋白質遺伝子(各々、CT26−NPもしくはRENCA−NP)でか、あるいは親株CT26もしくはRENCAのいずれかの殺腫瘍性用量で皮下注射による攻撃誘発を施した。腫瘍成長をモニターした。図3および4に示されるように、ワクチンとしてLM−NPを投与され、かつNPを発現する適切な腫瘍細胞での攻撃誘発を施された動物は更に進んだ腫瘍形成から保護された。CT26−NP群では25日後に、その動物の内の6匹が検出可能な腫瘍成長は全く示さず、3匹が5.0mmを下回る腫瘍を有し、そして1匹が9.0mmの腫瘍を有した(図4を参照されたい)。RENCA−NP群ではそれらの動物の内、腫瘍成長のいずれかの徴候を示したものは皆無であった(図3を参照されたい)。それとは対照的に他の群の全マウスは、1.5cmと3.0cmとの間の腫瘍を生じた(図1および2を参照されたい)。
LM−NPが既存の腫瘍の退縮および涸渇を生じる能力も証明した。腫瘍細胞(CT26もしくはRENCA細胞のいずれか)は皮下注射によりマウスに導入した。測定可能な腫瘍形成の後、それらのマウスを3つの個別の群に分配した。第一群のマウスにはLM−NPを投与し、第二群のマウスには野生型リステリア モノサイトゲネス(Listeria monocytogenes)を投与し、そして第三群のマウスには更なる処置は一切施さなかった。群1および群2のマウスには各々、LM−NPもしくは野生型リステリア モノサイトゲネス(Listeria monocytogenes)のいずれかの後続ブースターを投与した。図6および7に示されるように、LM−NPワクチンを投与されたマウスのみが、腫瘍が最早可視できなくなる地点までの腫瘍成長の退縮を示した。
本発明のワクチンは、腫瘍特異的抗原がその癌に関して知られているものであるか、あるいはその癌について同定されていることを必要とする。多数のこのような抗原が同定されている。これらの抗原には、白血病における抗原bcr/abl、子宮頸癌に関連する発癌ウイルスにおけるHPVE6およびE7、黒色腫におけるMAGE1およびMZ2−E、ならびに乳癌および膵臓癌におけるMUC−1が含まれる。しかしながら本開示の際には当業者には明白になるであろうように、本発明はいずれかの腫瘍抗原に適用可能である。
例えば、慢性骨髄性白血病(CML)抗原p210bcr-abl(これは、CML患者の内の90〜95%に発現される)はその独特なジャンクショナル配列のため腫瘍特異的抗原となっている。p210bcr-ablのJ領域(joining region)からのオリゴヌクレオチドを分泌するL.モノサイトゲネス(L.monocytogenes)組換え体を、細菌性染色体内への外来性遺伝子の直接挿入を可能にし、かつLLOシグナル配列を用いる遺伝子産物の分泌を可能にさせるいずれかの技術により構築する。その後にこの組換え体L.モノサイトゲネス(L.monocytogenes)を、単独もしくは薬剤学的に適切な担体の存在下のいずれかでワクチンとして投与してp210bcr-ablのレトロウイルス発現により誘導されるCMLを防ぐことができる。本開示の際には、当業者はこのアプローチを他の腫瘍抗原に日常的作業により拡張させることができるだろう。
多数の巨大ウイルス蛋白質を分泌する非常に安定な形質転換体が、当業者には決まりきった技術を用いて日常的作業により産生されている。組換え体L.モノサイトゲネス(L.monocytogenes)を産生するためには数々の技術が知られている。
例えば、DP−L967の構築の際のトランスポゾン挿入の結果としてのリステリア(Listeria)染色体内への組込みがSunら、Infection and Immunity 1990、58、3770−3778、により記載されている。トランスポゾン突然変異誘発は安定なゲノム挿入突然変異体を形成することができるという利点を有するが、外来性遺伝子が挿入されているゲノム内での位置が未知となるという欠点を有する。
prfA含有性ベクター内への遺伝子のクローニングおよびprfA(−)リステリア(Listeria)を補うためのこのプラスミドの使用がDP−L2028を構築するのに用いられている。DP−L2028は腫瘍保護実験に用いられるインフルエンザNP発現性株である。
本開示に基づき当業者により理解されるように、リステリア エスピー(Listeria sp.)内で腫瘍抗原を発現させるためには数々のアプローチが採用されてよい。ある例は、例えばリステリオリシンO(Listeriolysin O)もしくはPI−PLCのような選択された腫瘍抗原とリステリア(Listeria)蛋白質との融合蛋白質を作製することである。他の方法は、リステリア(Listeria)プロモーターの下流を利用するリステリア(Listeria)の分泌蛋白質(例えば、ヘモリシンもしくはホスホリパーゼ)のために、あるシグナル配列の使用を介する。外来性抗原を発現するのには、様々なL.モノサイトゲネス(L.monocytogenes)遺伝子のプロモーターが用いられてよい。それに加え、これらの遺伝子を外来性抗原との融合蛋白質を作製するのに用いてよい。例えば、遺伝子 hly、actA、plcA、plcB、およびmpl(これらはリステリア(Listeria)蛋白質であるヘモリシンをコードする)、actA(宿主細胞のアクチンの組立てにとって必要であり、かつ細胞間の細菌撒種にとって必須である表面蛋白質)、ホスファチジルイノシトール特異的ホスホリパーゼ、ホスホリパーゼC、およびメタロプロテイナーゼの各々のためのプロモーターを使用することができる。
これらの組換え体を産生するための一層好ましい方法は、温度感受性プラスミドでの相同組換えによるリステリア(Listeria)染色体内への組込みである。この方法を用いて目的の蛋白質を分泌する安定な形質転換体を産生することができる。トランスポゾン突然変異誘発を用いる場合とは異なり、挿入部位は既知となる。この方法は、目的のいずれかの遺伝子のL.モノサイトゲネス(L.monocytogenes)の染色体内への日常的挿入を考慮しており、そしてこのL.モノサイトゲネス(L.monocytogenes)染色体はその後にL.モノサイトゲネス(L.monocytogenes)プロモーターの制御下で発現される。このようなプロモーターの一つであるヘモリシンプロモーターは、豊富に合成されかつ分泌される蛋白質であるLLOをコードするリステリア(Listeria)遺伝子hlyの発現を調節する。LLOシグナル配列の導入は細菌細胞壁の外側での発現蛋白質の分泌を考慮してのことであることが示されている。L.モノサイトゲネス(L.monocytogenes)のこれら安定な組換え体の構築は、その生物体の成長および撒種にとって必要な細菌遺伝子の破壊を伴うことのない挿入部位として作用することができる染色体領域を利用する(Camilli et al.、Mol.Microbiol. 1993、8、143−157)。この相同領域が、大腸菌(E.coli)およびL.モノサイトゲネス(L.monocytogenes)の両方で機能する温度感受性プラスミドであるシャトルベクターpKSV7内に組み込まれる。その後に、その中心近くのEcoRI部位を用いて、二分されたこの領域間に一連のDNA断片を挿入する。ポリリンカーの添加後に、このリステリア(Listeria)ヘモリシン遺伝子のプロモーター配列を挿入する。そのシグナル配列の正しいプロセシングを確認するためには、そのプロモーターと共に、LLO蛋白質の最初の26のアミノ酸(シグナル配列)および4つの追加的アミノ酸についての下流配列情報が含まれる。合成される全転写物の安定かつ調節されたを確認するために、ヘモリシン遺伝子の転写プロセシング配列も含まれる。これらのヘモリシン調節配列を用いていずれかの隣接下流遺伝子の豊富な合成および分泌を亢進させる。
本発明のワクチンを単独もしくは薬剤学的に許容される担体と組み合わせてのいずれかで、腫瘍特異的抗原に対する免疫応答を誘導するのに有効な量で、ある宿主に投与することができる。「宿主」によっては癌細胞を保持することが可能ないずれかの生物体、好ましくはヒトを含むことが意味される。「有効量」によっては、T細胞における免疫応答を誘導することが可能な腫瘍特異的抗原を発現することが可能な組換えL.モノサイトゲネス(L.monocytogenes)の濃度(この濃度により、この抗原を含む細胞が撲滅されるであろう)が意味される。このような量は本開示の際には当業者により日常的な方法により決定され得る。「薬剤学的に許容される担体」は、限定されるわけではないが、滅菌された蒸留水、食塩水、リン酸緩衝液、もしくは重炭酸緩衝液を含むことが意味される。選択される薬剤学的に許容される担体および用いられる担体の量は投与の様式に依存するであろう。投与は、経口投与、非経口投与、鼻内投与、筋肉内投与、静脈内投与、直腸内投与、腹膜内投与、もしくは様々なよく知られる投与経路の内のいずれか一つであってよい。投与経路は、様々な腫瘍に従って選択されてよい。例えば、消化管の癌の治療のためには経口投与が用いられてよい。結腸直腸癌の治療のためには直腸内免疫化が用いられてよい。卵巣もしくは膵臓の癌の治療のためには腹膜内投与が用いられてよい。本発明のワクチンは、経口投与のためにはエレキシル剤、カプセル剤、もしくは懸濁物、あるいは非経口投与もしくは血管内投与のためには滅菌液の形態で投与されてよい。本ワクチンは凍結してか、4℃下でか、室温でか、もしくは凍結乾燥させて保存してよい。
好ましい態様では、本発明のワクチンは宿主に対し、単独もしくは腫瘍の形成を阻害もしくは抑制するためのもう一つの癌療法と組み合わせてのいずれかで投与される。
従って、本発明のワクチンを用いて、家族性の遺伝的特質もしくは所定の種類の癌に罹りやすくする他の状況(例えば、夫がパピローマウイルスを有する女性の子宮頸癌)という理由で、癌について高い危険率にさらされている人を防御することができる。それに加え、外科手術、通常の化学療法、もしくは放射線療法による腫瘍成長の退縮後の癌の免疫療法として、本ワクチンを用いることができる。このような治療の後に、腫瘍抗原を発現する組換えL.モノサイトゲネス(L.monocytogenes)を投与することができる。このワクチンにより産生される腫瘍抗原に対するCTC応答は残存性転移癌を破壊し、かつ癌からの緩解を長期化させるであろう。本発明のワクチンを用いて既に定着している腫瘍の成長に変化を及ぼすことができるとも考えられる。
以下の実施例は本発明を説明することのみを目的とし、かつ本発明を制限することは意図されない。
実施例
実施例1
リステリオリシンO(Lysteriolysin O)(LLO)の最初の420アミノ酸、ならびに幾つかの上流調節配列を伴うそのプロモーターをコードする配列をL.モノサイトゲネス(L.monocytogenes)染色体DNA(野生型株10403s)からPCR増幅させ、そしてNPをコードするPCR増幅させたDNA(これはプラスミドpAPR502に由来する)にこれを連結させた(Young,J.F.、U.Desselberger、P.Graves、P.Palese and A.Shatzman、”Cloning and Expression of influenza virus genes”、The Origin of Pandemic Influenza Viruses、W.G.Laver、eds.、Elsevier、New York、1983、p.129)。この構築作業により、融合連結部位に2つの追加的アミノ酸が添加されているインフレーム融合物がもたらされた。この融合物をシャトルプラスミドpAM401内にクローン化させたが、このシャトルベクターは、グラム+クロラムフェニコール耐性遺伝子およびグラム−テトラサイクリン耐性遺伝子を含むグラム+およびグラム−の両方の細菌内で複製することが可能である(Wirth,R.、F.Y. An and D.B.Clewell、J.Bacteriol.1986、165、831)。得られるプラスミドpDP1659を野生型L.モノサイトゲネス(L.monocytogenes)(株10403s)内に電気穿孔により組込ませてL.モノサイトゲネス(L.monocytogenes)株DP−L1659を取得した。この組換え株は、抗−LLOポリクローナル抗血清および抗−NPモノクローナル抗体を用いる培養物上清内の分泌蛋白質のウエスタン(Western)ブロット分析により決定したところ、予想サイズ(105kD)の融合蛋白質を作製および分泌することが可能であることが明白となった。多重コピープラスミド内でLLOプロモーターの制御下に置かれる融合遺伝子の存在により、染色体によりコードされるLLOの分泌減少がもたらされたが、それは小胞からの細菌の移出もしくはその後の細胞質内成長を妨げる程度のものではなかった。しかしながらこの株はクロラムフェニコールの非存在下では安定ではなくなっていた。
L.モノサイトゲネス(L.monocytogenes)株DP−L2028(これはインビボでは安定であり、かつ実施例2〜6に用いられた)を構築するためには、プラスミドpDP−1659を10403sからのprfA遺伝子を挿入することにより改変させ、そしてその後にこの改変株を用いてprfA−L.モノサイトゲネス(L.monocytogenes)突然変異体DP−L1075を形質転換させた。このことによりインビボおよびインビトロで安定にLLP−NP融合蛋白質を分泌するL.モノサイトゲネス(L.monocytogenes)株DP−L2028がもたらされた。
実施例2:LM−NPでのマウスの治療
120匹のBalb/cマウスを40匹づつの3群に分配した。一つの群には野生型L.モノサイトゲネス(L.monocytogenes)のLD50の1/10での免疫化を施し、もう一つの群には滅菌食塩水での免疫化を施し、そして第三の群にはインフルエンザ核蛋白質を分泌するように形質転換させた組換えL.モノサイトゲネス(L.monocytogenes)ワクチンベクター(LM−NP)での免疫化を施した。2週間後には各群に類似のブースター免疫を施した。この免疫化スケジュールはインフルエンザ核蛋白質に対して強力なCTC応答を生じるように決定された。最終免疫後2週間目には各群の動物に、L.モノサイトゲネス(L.monocytogenes)ベクターを形質転換させるのに用いたのと同一のインフルエンザ核蛋白質で予めトランスフェクトさせてあるCT26もしくはRENCA(各々CT26−NPもしくはRENCA−NP)のいずれかでの皮下投与による攻撃誘発を施した。各マウスには、殺腫瘍性用量の50倍である5×105の腫瘍細胞の投与を施した。これら6群の動物における腫瘍成長を2日毎にモニターした。この研究からの結果が図1〜4に示される。殺腫瘍性用量からのいずれかの防御を示した唯一の群は、ワクチンとしてのLM−NPの投与が施され、かつNPを発現する適切な腫瘍細胞での攻撃誘発が施された動物であった。CT26−NP群では25日後には、その動物の内の6匹は検出可能な腫瘍増殖を全く示さず、3匹は5.0mmを下回る腫瘍を有し、そして一匹は9.0mmの腫瘍を有した。RENCA−NP群では、腫瘍成長のいずれかの徴候を示した動物は皆無であった。それとは対照的に、他の群のマウスは全て1.5cmと3.0cmとの間の腫瘍を有する。
外来性NP遺伝子を維持する目的では、CT26−NPを通常は抗生物質G418上に維持する。LM−NP免疫化マウス内で生育した少数のCT26−NP腫瘍細胞は、G418の非存在下でNP遺伝子を欠失していた細胞であると考えられる。
実施例3:インビトロでNPを発現する腫瘍細胞CT26およびRENCAを死滅化させることができるLM−NPを用いてBalb/cマウスを免疫化させることにより作製されるCTL
マウスに0.1 LD50のLM−NPで免疫化を施した。2週間後にそれらのマウスを屠殺し、そしてインフルエンザで感染させた(A/PR8/34)脾臓細胞(図5A)か、もしくはNP蛋白質の免疫優性エピトープを呈示することが知られている合成ペプチド147−158(図5B)のいずれかを用いて脾臓細胞の初期培養物を開始させた。培養4日後に両集団の細胞溶解性活性をCT26−NP、RENCA−NP、ならびに親細胞株CT26およびRENCAに対して測定した。陽性対照が含まれていた(P815、そのペプチドの存在下もしくはA/PR8/34による感染を受けた際にH−2dに制限されるCTLによる溶解を効果的に受けることが知られている肥満細胞腫の腫瘍細胞)。図5Aが示すように、RENCA−NPおよびCT26−NP(ただし親株ではない)を、LM−NPで免疫化させかつA/PR8/34で拡張させることにより誘導させたNP特異的エフェクターにより溶解させた。図5Bでは、エフェクターをペプチドで拡張させた類似実験が類似の結果を示している。
実施例4:RENCA腫瘍成長の除去をもたらすLM−NPによる免疫化
この実験では腫瘍成長が開始してしまった後のLM−NPでの免疫化により腫瘍の退縮および涸渇が生じた。30匹のマウスに皮下注射により腫瘍細胞(5×105)を導入させた。13日目に(これは測定可能な腫瘍(5mm)がマウス内で生育した後である)、それらのマウスを10匹ずつの3群に分配した。10匹のマウスにはLM−NPを投与し、10匹のマウスには野生型リステリア モノサイトゲネス(Listeria monocytogenes)を投与し、そして10匹のマウスにはそれ以上の処置は一切加えなかった。23日目には、これらのマウスに再度、LM−NPもしくは野生型リステリア モノサイトゲネス(Listeria monocytogenes)のいずれかでの免疫化を施した。図6に示されるように、LM−NPワクチンを投与されたマウスのみが、10匹のマウスの内の9匹で腫瘍は最早可視化されない程度までの腫瘍成長の退縮を示した。
実施例5:CT26−NP腫瘍成長の停止をもたらすLM−NPによる免疫化
実施例4に記載される実験も、結腸直腸のCT26−NP腫瘍細胞を用いて実施された。CT26−NPは成長がかなり迅速な腫瘍であり、かつNPの発現の際には一層不安定にもなる。それにもかかわらずこの実験では、腫瘍成長が開始してしまった後のLM−NPによる免疫化が腫瘍成長を停止させることも見いだした。腫瘍細胞(5×105)を30匹のマウスに皮下注射により組込ませた。10日目には(これは測定可能な腫瘍(5mm)がマウス内で生育した後である)、それらのマウスを10匹ずつの3群に分配した。10匹のマウスにはLM−NPを投与し、10匹のマウスには野生型リステリア モノサイトゲネス(Listeria monocytogenes)を投与し、そして10匹のマウスにはそれ以上の処置は一切加えなかった。17日目には、これらのマウスに再度、LM−NPもしくは野生型リステリア モノサイトゲネス(Listeria monocytogenes)のいずれかでの免疫化を施した。図7に示されるように、LM−NPワクチンを投与されたマウスのみが腫瘍成長における変化を示した。しかしながら、RENCAを用いる場合とは異なり、成長の退縮は多くのマウスで観察されたのではなかった。これは17日目までに、CT26−NP腫瘍細胞の不安定性によりNP抗原を喪失した多くの腫瘍細胞がもたらされるためであってよい。
実施例6:CD8+ T細胞によりもたらされる腫瘍成長の阻害
この実験では30匹のマウスに、実施例2に記載されるのと同一のプロトコールを用いてLM−NPでの免疫化を施した。最終免疫化後10日目には、10匹のマウスを抗体2.43(CD8分子に特異的である)での免疫化によりCD8+ 細胞の涸渇状態にさせ;10匹のマウスをGK1.5(CD4分子に特異的である)での免疫化によりCD4+ 細胞を涸渇状態にさせ;そして10匹のマウスは完全なT細胞レパートリーを付随させたままの状態にしておいた。(CD8+ もしくはCD4+ T細胞の涸渇化のためのプロトコールは、A.Kruisbeek、Current Protocols In Immunology、Coligan et al.、eds、John Wiley & Sons,Inc.、1994、V.1、4.1.1−4.1.2)により記載されるものであった)。T細胞涸渇の後、これらのマウスに、マウス当たり5×105のCT26−NP細胞での皮下注射による攻撃誘発をもたらした。対照として、10匹のナイーブマウスにも同一用量での攻撃誘発を施した。図8に示されるように、CD8+ T細胞サブセットが涸渇した状態のマウス群は対照群のマウス(ナイーブマウス)に類似の腫瘍成長を示した。CD4+ T細胞サブセットが涸渇した状態のマウスは腫瘍成長に対する防御が低下していることを示し、このことによりCD4+ 細胞が腫瘍成長の制御におけるアクセサリー応答を行っていることが示され;そして完全なT細胞レパートリーを保持するマウスはLM−NPワクチンにより誘導される腫瘍成長に対する防御を示す。
Claims (3)
- 宿主において腫瘍特異的抗原に対する免疫応答を誘導するためのワクチンの製造における、腫瘍特異的抗原もしくはその断片に対するリステリオリシンO(LLO)蛋白質の融合物を発現することが可能なリステリア モノサイトゲネスの組換え体の使用。
- 腫瘍特異的抗原に対する免疫応答を誘導するための、腫瘍特異的抗原もしくはその断片に対するリステリオリシンO(LLO)蛋白質の融合物を発現することが可能なリステリア モノサイトゲネスの組換え体を含むワクチン。
- 薬剤学的に許容される賦形剤をさらに含む、請求項2記載のワクチン。
