免疫応答の刺激は、ホスト免疫系により異物であると認識された抗原の存在に依存する。サルモネラ・エンテリカ(Salmonella enterica)およびマイコバクテリウム・ブービス(Mycobacterium bovis)BCGなどの細菌抗原は、ファゴソーム中に残留し、CD4T細胞を、主要な組織適合性II群分子による抗原提示により刺激する。対照的に、リステリア・モノサイトジェネス(Listeria monocytogenes)などの細菌抗原は、ファゴソームから細胞質中に進出する。リステリア・モノサイトジェネスのファゴリソソーム逃散は、リステリア抗原の主要な組織適合性I群抗原提示を促進する独特な機構である。この逃散は、孔形成スルフヒドリル活性化細胞溶解素であるリステリオリシン(listeriolysin)O(LLO)に依存する。

本発明は、選択された抗原の、リステリオリシンOの非溶血性切断型への融合による、選択された抗原の免疫原性を増強する方法に関する。ここで、リステリオリシンOの非溶血性切断型への抗原の融合により、抗原単独と比較して、増強された免疫原性を有する抗原が得られることが見出された。抗原に融合されたリステリオリシンOのこの切断型は、抗原単独と比較して、細胞媒介免疫性および抗腫瘍免疫性を良好に可能にする。さらに、これらの融合タンパク質は、リステリア・モノサイトジェネスにより発現される必要はなく、むしろ、タンパク質発現および単離のためにルーチンに用いられる他のベクターおよび細胞系から発現され、単離されることができる。

対照的に、LLOのプロモーター、シグナル配列および最初の7個のアミノ酸残基に融合したE7抗原を含む短い融合タンパク質Lm−AZ/−E7は、無効な抗腫瘍免疫治療であった。この短い融合タンパク質は、PEST様配列の直前で終了し、これを含まない。実験の最初の組において、HPV−E7抗原を、リステリア・モノサイトジェネスにおいて発現させた。E7を発現したリステリア・モノサイトジェネス組み換え体を、hlyプロモーターの制御の下に、およびhlyシグナル配列の導入と共に、E7遺伝子の染色体統合により作成して、遺伝子生成物の分泌を確実にした。

相同性組み換えによる染色体中への統合の部位は、Lm病原性に必須ではない領域中であった。このための図式を、図1に示す。このタイプの形質転換を用いた利点は、プラスミド中に含まれて、エレクトロポレーションの後に好首尾な形質転換体について選択されるCAT遺伝子が、第2の組み換え事象の間に切り取られるため、得られる組み換え体が安定に形質転換され、薬剤選択マーカーを含まないことである。得られた組み換え体Lm−E7からの抗原の発現および分泌を、ウエスタンブロットにより実証した。さらに、Lm−E7の治療的効果を最適化した。例えば、ワクチンを経口的に送達することによって、非経口的に、および組み合わせた保護および腫瘍誘発の前にLm−E7での初回刺激および次にLm−E7を腫瘍誘発の後に投与することを必要とする退行モードと比較して、最良の結果が達成されることが見出された。

表1は、3種の異なる腫瘍細胞系、TC−1、C3およびEL−4/E7においてこのモデルにおいて観察された、最適化された抗腫瘍効果についてのさらなる詳細を提供する。細菌を、腫瘍誘発の14日および7日前並びに腫瘍誘発の7日および14日後に、経口的に送達した。同様のプロトコルでの腹腔内への106個の細菌の送達により、長期の防御は与えられなかった。しかし、Lm−E7を経口的に送達した際には、より良好な防御が観察された。さらに特に、この措置で、約50%の動物が、永久に腫瘍非保有のままであり、免疫化は、すべての動物において腫瘍成長を顕著に遅延させた。

腫瘍非保有であったTC−1またはEL−4/E7腫瘍細胞を投与された動物を、それぞれ60日においてTC−1で、または40日においてEL−4/E7で再誘発した。Lm−Gagで免疫化した、各々の群における2匹の動物は、腫瘍を成長させた一方、Lm−E7で免疫化した動物は、実験の終了(TC−1の場合において124日およびEL−4/E7について54日)まで腫瘍非保有のままであった。

追加の実験を実施して、C57Bl/6マウスからの確立された皮下HPV−16不死化腫瘍の退行を誘発する能力を、Lm−E7とLm−LLO−E7とで比較した。これらの実験において、マウスを、4種の構築物、Lm−E7、Lm−Gag(発現した抗原以外はLm−E7と同質遺伝子)、Lm−LLO−E7またはLm−LLO−NPの1種を有する0.1LD50で腹腔内で免疫化した。Lm−LLO−NPは、Lm−LLO−E7と同質遺伝子であるが、インフルエンザ抗原を発現する。第2の免疫化を、14日において実施した。Lm−LLO−E7で免疫化した8匹のマウスのうち6匹は、これらの腫瘍が治癒し、腫瘍非保有を維持した。他の動物のいずれも、確立された腫瘍の退行を示さなかった。種々の免疫化プロトコルの下で、Lm−LLO−E7について、同様の結果が達成された。さらに、1つのみの免疫化が、直径5mmの確立されたTC−1を有するマウスを治癒すると例証された。

LLOを抗原に融合することにより、増強された免疫性が与えられるという発見の一般性を確認するために、Lm−E7に類似するLm−NPの異形を構築した。この組み換え体を、インフルエンザ核タンパク質を抗原としてE7で置き換えた以外は、図1に示すように製造した。新たなLm−NPの能力を、Lm−LLO−NP(米国特許第5,830,702号に記載されており、図2に示すように製造した)と比較した。これらの実験において、32匹のBALB/cマウスに、5×105のRENCA−NP腫瘍細胞を接種した。RENCA−NPは、インフルエンザ核タンパク質NP(米国特許第5,830,702号に記載されている)をレトロウイルス的に形質導入した腎細胞癌である。明白な巨視的腫瘍が10日後に成長した後に、各群における8匹の動物を、3種の構築物、Lm−NP、Lm−Gag(発現した抗原以外はLm−NPと同質遺伝子)およびLm−LLO−NPの1種を有する0.1LD50で腹腔内で免疫化した。1週間後に、動物に、第2の免疫化を施した。8匹の動物を、未処理のままとした。40日における実験の終了時に、未処置の群のすべてのマウスは、大きい腫瘍を有していたか、または死亡した。Lm−Gagを施された群における1匹のマウスおよびLm−NPを施された群における2匹のマウスのみが、腫瘍非保有であった。この実験は、抗原のLLOへの融合は、E7に限定されないことを示し、抗原の形態が重要ではないことを示唆する。

追加の実験を実施して、E7抗原およびリステリオリシンOの切断型を含む融合タンパク質の増強された治療効果を確認した。これらの実験において、リステリオリシンOの非溶血性切断型との融合タンパク質としてE7を発現するワクシニアベクターを構築した。ワクシニアのWR株を、受容体として用い、融合遺伝子を、リステリアプラスミドから切り取り、p75プロモーターの制御の下でpSC11中に挿入した。このベクターが、ワクシニア構築物Vac−SigE7LampおよびVac−E7について用いられる移送ベクターであり、従ってVac−LLO−E7との直接の比較を可能にするため、このベクターを選択した。このようにして、3種のワクシニア組み換え体のすべては、同一の早期/末期化合物プロモーターp7.5の制御の下で発現した。さらに、SC11は、TK選択およびβ−ガラクトシダーゼスクリーニングに対する組み換えウイルス疫病の選択を可能にする。

腫瘍拒絶の研究を、前に記載したものと同一のプロトコルに従って、TC−1を用いて実施した。処理を開始する前の異なる遅延を用いて、2種の実験を実施した。1つの実験において、処理を、腫瘍が直径約3mmであった際に開始した。76日において、Vac−LLO−E7で処理したマウスの50%が、腫瘍非保有であり、Vac−SigE7Lampマウスの25%が、腫瘍非保有である。

好ましい態様において、本発明の融合タンパク質は、リステリア・モノサイトジェネスのPEST様アミノ酸配列を含むリステリオリシンOの切断型または、他の原核生物から由来するPEST様アミノ酸配列および抗原のいずれかをコードするプラスミドを介して、組み換え的に生成する。しかし、抗原はまた、リステリア・モノサイトジェネスのPEST様アミノ酸配列または他の原核生物から由来するPEST様アミノ酸配列を含むリステリオリシンOの切断型に、化学的に共役することができる。本発明の目的においては、「抗原」によって、生来の抗原遺伝子または遺伝子生成物あるいは、同定されたT細胞エピトープを含むこれらの切断異形を含むことを意味する。

TC−1は、HPV−16 E6よびE7により不死化させ、活性化ヒトc−HA−rasを発現するpVEJBにより形質転換したC57BL/6マウスからの肺上皮細胞である。C3は、マウス胚細胞frp、HPV16の完全なゲノムで不死化させ、pEJ−rasで形質転換したC57BL/6マウスである。EL−4/E7は、E7によりレトロウイルス的に形質導入された胸腺腫EL−4である。

例2:Lm−GG/E7、Lm−AZ/E7およびVac−SigE7Lampの効能の比較
TC−1(1×105)またはC−3(5×105)腫瘍細胞を、マウス中に皮下的に移植し、7〜9日間放置して成長させ、この時までに、これらは明瞭になった(大きさ〜5mm)。次に、マウスを、3種の構築物、Vac−SigE7Lamp(107PFU)、Lm−E7(106CFU)またはLm−LLO−E7(107CFU)の1種で、腹腔内に免疫化した。7日および14日において、動物に、Lm−LLO−E7およびLM−E7を施した。生存するマウスを、43日において、105TC−1で再誘発した。

8匹のマウスの4つの群に、TC−1の105個の細胞を移植した。7日後、腫瘍は、大きさが約4mmであった。1つの群のマウスは、未処理であった。他の群の各々に、107PFUのVac−E7、Vac−LLO−E7またはVac−Sig−E7−lamp7のいずれかを施した。ブースター用量を、14日において投与した。

TC−1腫瘍細胞(2×105)を、96匹のC57BL/6マウスの左側に皮下移植し、7日間放置して成長させた。マウスを、8匹のマウスの群に分割し、各々の群を、以下のワクチンの1種で処理した:未処置(ワクチンなし);Vac SigE7Lamp、107PFU、腹腔内;Vac−LLO−E7、107PFU、腹腔内;またはLm−LLO−E7、107PFU、腹腔内。1週間後、動物に、ブースター免疫化を施した。腫瘍成長を、2日おきに、ノギス測定により追跡し、最も狭いおよび最も長い表面長さの平均として記録した。また、免疫パラメーターを決定した。

LLO−PEST−E7フラグメントを、SOEing PCRにより構築することができる。

段階3においては、LLO−PEST−E7を、酵素NheIを有するプラスミドから2時間にわたり消化し、続いてエタノール沈殿および酵素XmaIを一夜施す。pGG−49からのprfAフラグメントを、酵素SalIで、2時間消化し、続いて、エタノール沈殿および酵素XmaIによる沈殿を一晩施す。pDP−2028を、SalIおよびXbaIで2時間消化し、続いて、エタノール沈殿およびTris:EDTA(TE)中での再懸濁を施す。フラグメントを、4℃で一夜貯蔵することができる。

Claims (12)
- 抗原の免疫原性を増強するための方法であって、抗原に、リステリオリシンOの非溶血性切断型を融合させることを含む、前記方法。
- リステリオリシンの非溶血性切断型が、PEST様アミノ酸配列を含む、請求項1に記載の方法。
- PEST様アミノ酸配列が、配列番号1を含む、請求項2に記載の方法。
- リステリオリシンOの非溶血性切断型を、抗原に、リステリオリシンOの切断型をコードするプラスミドおよび抗原の組み換え発現により融合する、請求項1に記載の方法。
- 増強された細胞媒介免疫性および抗腫瘍免疫性を有する組成物であって、抗原に融合されたかまたは抗原内に包埋された、原核生物から由来するPEST様アミノ酸配列を含む、前記組成物。
- PEST様アミノ酸配列が、リステリア種から由来する、請求項5に記載の組成物。
- PEST様アミノ酸配列が、リステリア・モノサイトジェネスから由来する、請求項6に記載の組成物。
- PEST様アミノ酸配列が、配列番号1を含む、請求項5に記載の組成物。
- PEST様配列が、配列番号2、3、4、5、6および7からなる群から選択されたアミノ酸配列を含む、請求項5に記載の組成物。
- 動物における抗原に対する増強された細胞媒介または抗腫瘍免疫原性応答を引き起こす方法であって、動物に、請求項5に記載の組成物を投与することを含む、前記方法。
- 抗原に対する増強された細胞媒介または抗腫瘍免疫原性応答を引き起こすためのワクチンであって、請求項5に記載の組成物を含む、前記ワクチン。
- 補助剤をさらに含む、請求項11に記載のワクチン。
