Description
本発明は、何らかの理由で個体内において存在が望ましくない自己タンパク質を除去するため、および/または低く調節するために免疫器官を利用するための新規の方法に関する。それらは、疾病および/またはその他の望ましくない症状もしくは疾病の徴候の原因となるタンパク質となり得る。その様なタンパク質は、ワクチン接種により特異的に誘発される、体内を循環する自己抗体により除去される。本発明は、その様な自己ワクチン(autovaccines)を開発するための方法を説明する。
〔発明の背景〕
生理学的には、脊椎動物の免疫系は、微生物のような感染物による身体への侵入に対する防御機構として働く。外来タンパク質は、体内を循環する特異性が極めて高い抗体により、細網内皮系を経由して効果的に除去され、ウイルスおよび細菌は、抗体、細胞傷害性Tリンパ球、ナチュラルキラー細胞、補体などを含む、複雑な一連の細胞性機構及び体液性機構の攻撃を受ける。この戦闘を主導するのがTヘルパー(TH)リンパ球であり、これは抗原提示細胞(APC)と協力してサイトカインの複雑なネットワークを通して免疫的防御を調節する。
THリンパ球は、APCの表面上に提示されるタンパク質抗原を認識する。しかしながら、それらは、それ自体では抗原そのものを認識できない。その代わりにそれらは、「処理された(processed)」(断片化された)タンパク質抗原(いわゆるT細胞エピトープ)と主要組織適合遺伝子複合体のクラスII分子との2つの構成成分からなる複雑なリガンドを認識するものと思われる(O.Werdelin et al.,Imm.Rev. 106,181(1988))。最終的にはこの認識により、Bリンパ球による元のタンパク質抗原に対して特異的な抗体の産生を、THリンパ球が特異的に補助することが可能となる(O.Werdelin et al.,supra)。特定のT細胞は、特定の抗原−MHC結合体のみを認識し、もう1つのMHC対立遺伝子の遺伝子産生物により提示される、同一もしくは別の抗原を認識することはない。この現象を、MHC拘束と称する。
通常、個体自身のタンパク質(いわゆる自己タンパク質)は、免疫器官により攻撃されることはない。したがって、本発明に関して説明される事象は、一般的には当該個体にとって有益であるのだが、しかしまれな事例においては過程がうまく進行せず、免疫系はその個体自身の構成要素に向かって働き、それにより自己免疫疾患を導くことがある。
幾つかの自己タンパク質の存在は、昂進して疾病の症状を誘発する状態においては不適当なものとなる。例えば、高水準のIgEクラスのイムノグロブリンは、タイプIアレルギーの誘発にとって重要であることが知られており、腫瘍壊死因子α(TNFα)は、癌患者及び他の慢性疾患を病んでいる患者における悪液質の原因となり得ることが知られている(H.N.Langstein et al.,Cancer Res. 51,2302-2306,1991)。TNFαは、炎症過程においてもまた重要な働きをする(W.P.Arend et al.,Arthritis Rheum.33,305-315,1990)。性ホルモン依存性の癌においてはホルモンが、特定の状態においては望ましくないタンパク質の、別の事例である。それ故、本発明は、その様な自己タンパク質に対する自己ワクチンを開発するための方法を提供する必要が存在する。
自己タンパク質の断片もまたAPCにより提示されるが、しかし、通常はその様な断片はTヘルパーリンパ球からは無視されるか、これによっては認識されない。これが、一般的に個体がその血清中の自己抗体を含まないことの主な理由である。
とは言え、自己タンパク質に対する抗体を人工的に誘発することは可能である。上記のように、自己タンパク質を、例えば破傷風トキソイドまたはキーホール・リンペット・ヘモシアニン(KLH)のような適当な大きさの外来キャリヤータンパク質に共有結合させた複合体により、誘発が可能である。タルワーら(G.P.Talwar et al,Int.J.Immunopharmacol.14,511-514,1992)は、ヒト柔毛膜性生殖腺刺激ホルモンと破傷風トキソイドとの複合体からなるワクチンを用いることにより、女性の生殖を阻害することができた。そのような自己免疫性複合体を、ヒトおよび動物モデルにおいて治療に用いたことの別の事例もまた存在する(D.R.Stanworth et al.,Lancet 336,1279-1281(1990))。APCにおいてその様な複合体が加工処理(プロセシング)される間に、必要なTHリンパ球刺激性エピトープが外来タンパク質から提供され、最終的にキャリヤータンパク質に対してと同様に自己タンパク質に対しても抗体誘発が導かれる。しかしながら、この原理を用いる上での一つの欠点は、自己タンパク質に対する抗体応答が、共有結合しているキャリヤータンパク質によるエピトープの遮蔽により限定されうるということである。もう一つの欠点は、外来のキャリヤータンパク質に対して、望ましくない極めて強力な抗体応答を同時に誘発することにより、アレルギー的副作用を引き起こす危険性が大きくなることである。
他の研究者は、T細胞エピトープであると予測される一本鎖ペプチドを化学的に、キャリヤーとしてのハプテン作用を有するデカペプチドである自己ペプチド[D−Lys6]GnRHに複合化し、その特定のT細胞エピトープについての、MHC拘束を伴う自己抗体応答の誘発を達成した(S.Sad et al., Immunology 76,599-603,1992)。この方法は、巨大キャリヤータンパク質への複合化と比較して、より効果的であると思われる。しかしながら、この方法では、適切なMHC分子を発現する個体群においてしか抗体を誘発しないであろう。このことは、かなり多数の異なるT細胞エピトープを自己ペプチドに複合化しなければならず、そうすれば結局は、自己ペプチドの表面上のB細胞エピトープを混乱させるであろうことを意味する。さらに、タンパク質の徹底的な複合化は、免疫原性に関して反対の効果を有することがあり(国際特許出願WO87/00056号)、ペプチドのT細胞エピトープの露出された表面は、抗原を処理する間にタンパク質分解酵素により破壊されることがあり(S.Mouritsen,Scand.J.Immunol. 30,723,1989)、したがって、上記方法は不適当なものとなる。また、そのような多複合的(multi-conjugated)自己ペプチドの正確な構造は、化学的にも薬学的にも良好に定義できないであろう。
最近、分子量の大きいデキストラン分子にB細胞エピトープを(およびT細胞エピトープをも任意に)化学的に結合させた複合体による、B細胞の自己免疫反応を破壊するための改善された方法が提示された(WO93/23076、1993年11月25日公開)。しかしながら上記の欠点は、この方法についてもまた当てはまる。
組換えDNA技術を用いて(EP−A2−0343460)、広く認識される強力なT細胞エピトープを、B細胞エピトープを示す抗原構造を有する外来ペプチドに連結させることができる。例えばワクチンのような免疫原性組成物の調製において、ヘルパーT細胞(THリンパ球)反応性エピトープおよび好ましくはB細胞反応性エピトープを組み込んでいる免疫原性ペプチドまたは抗原性ペプチドを含むペプチジル樹脂複合体を使用することもまた示唆された。好ましくはポリアミド樹脂上で、固相合成により複合体を調製する。(WO90/15627)。意図するところは当該ペプチドに対する抗体応答を増幅することであるが、免疫系の自己寛容を破壊する目的でこれを行うことができることは提案されなかった。自己タンパク質に対する自己抗体を誘発するためにこれらの方法を用いるに当たって、応答のMHC拘束についての上記限定に関して、同一の規則(rules)が当てはまることが直観的に予想されるであろう。しかしながら驚くべきことに、免疫優性T細胞エピトープを含むことが知られている対応数のペプチドを用いての1以上のペプチド断片の置換により、自己タンパク質類似体を産生し、前記置換を行うことにより起源の自己タンパク質の3次構造全体を本質的に維持するものである本発明に従った方法を用いての自己タンパク質の調節により、用いられたT細胞エピトープ挿入物が免疫感作されたマウスのMHC分子に制限されないという事実にもかかわらず、TNFαに対して自己抗体応答を、等しく高速であり、且つ、より強力にさえ誘発することが可能となる(以下例3参照)。この観察結果についての理由は明らかではないが、しかし、自己タンパク質における突然変異領域内の新規のMHC結合部分の出現によるものと考えられる。しかしながら、例5に示された実験からは、恐らく上記の理由ではないであろう事が例証される。というのは、ユビキチンに移植された卵アルブミンT細胞エピトープの重複領域に対応する合成ペプチドは、この組換え分子の免疫原性が高いものであるH−2kマウスの、MHCクラスII分子のいずれに対しても強力には結合しなかった。
タンパク質の潜在的なMHCクラスII結合部分のほとんどは、通常は露出しておらず、抗原提示細胞により宿主T細胞に提示されない(S.Mouritsen et al,Scand.J.Immunol. 34,421,1991)。挿入されたT細胞エピトープのMHC拘束と抗体応答の制限との間の一致の欠如が観察されることから、恐らく自己タンパク質の異なる部分による、MHC分子への結合についての分子内競合の一般的な外乱によるものであろう。本発明の方法に従って、T細胞に対して、隠れている非寛容化された(non-tolerized)自己タンパク質の一部を提示することができ、その結果、タンパク質に対するB細胞の自己免疫寛容と同様にT細胞の自己免疫寛容も破壊される。本発明に合致し、下記の実施例すべてにおいて例証されるように、自己タンパク質の断片は、外来のT細胞エピトープに置換された。この欠失に続くほかのタンパク質断片による置換は、自己タンパク質の3次構造の不明確化を最小にするが、MHCクラスII結合自己断片(binding self-segments)の前記分子内競合の外乱についてもまた強く関与するであろう。したがって、この概念は、上記の先行技術の機構および方法とは明確に異なる。本発明にしたがった方法による挙動の操作機構とは無関係に、B細胞の自己免疫寛容を破壊するための従来の方法と比較して、それはより技術的に優れたものである。というのは、最小数の異なる外来T細胞エピトープの挿入物により、広範なMHC分子において抗体を誘発することが可能だからである。
このように、本発明は、本質的に3次構造が維持された自己タンパク質類似体を産生するように、1以上のペプチド断片の欠失および同時になされる対応数の外来のT細胞エピトープを挿入することにより適切に調節された(modulated)組換え自己タンパク質を注射することにより、改変されていない自己タンパク質に対する意味深い(profound)自己抗体応答が誘発されるという驚くべき事実を基礎とするものである。驚くべきことに、誘発された自己抗体応答のMHC拘束は、必ずしも挿入されたT細胞エピトープのそれに制限されない。TNFαにおいてと同様、良好に保存される(highly conserved)自己タンパク質のユビキチンにおいても最小の3次構造変化を誘発することにより、遺伝子工学的方法を用いて、12−15アミノ酸長を有する外来のT細胞エピトープを挿入した。これらの組換え自己タンパク質類似体を精製し、アジュバントに乳化し、マウスに注射した。僅かに1週の間に、これらのマウスからの血清中において、ユビキチンに対する自己抗体応答を検出できた。同様に処理され、注射された、改変されていない組換えユビキチンは、応答を誘発できなかった。
望ましくないタンパク質に対するワクチンを開発するためにこの原理を用いることにより、アレルギー的副作用を誘発する危険性が少なくなり、タンパク質の生物学的な活性を有する部分を除去するかまたは突然変異させることにより、TNFαのような毒性自己タンパク質を同時に無毒化できる。上記のエピトープ遮蔽効果は問題にはならず、キャリヤーのタンパク質またはペプチドに自己タンパク質を複合化させる従来の技術と比較して、ユビキチンに対する自己抗体をより早く誘発した。重大なことは、この方法によりさらに、個体のB細胞の自己免疫寛容と同様にT細胞の自己免疫寛容を一時的に破壊することが可能であるように思われ、それによる組換えタンパク質は、多くの異なるMHCクラスII分子を発現する大きな群(a large population)において自己免疫的であろう。
本発明に従ったワクチンは、上記のように調節され、リン酸カルシウム、サポニン、キル(quil)Aもしくは生分解性ポリマー適切なアジュバントを用いて製剤化された1以上の自己タンパク質類似体とからなる。この調節された自己タンパク質類似体は、GM−CSFまたはインターロイキン2のような適切な免疫学的に活性なサイトカインを伴う融合タンパク質として調製される。
自己ワクチンは疾患がないとき(i.a.)でも、例えばガン患者のような悪液質の患者の治療のためのTNFαまたはγ−インターフェロンに対するワクチン、またはアレルギー患者の治療のためのIgEに対するワクチンとなり得る。更に、慢性炎症疾患の患者の治療のためのTNFα、TNFβまたはインターロイキン1に対するワクチンともなり得る。
本発明は、以下の実施例において詳細に示される。
例1: ユビキチンへの外来T細胞エピトープの置換
例としてT細胞エピトープMP7を用いて、図1に本発明の手順の外観を示す。適切な制限酵素クローン化部位を有するように設計された2つの相補性オリゴヌクレオチドとして、MP7(MP7.1−CおよびMP7.1−NC)で表される遺伝子配列を合成した。そのMP7のアミノ酸配列は、PELFEALQKLFKHAYである(Mouritsen et al,Scand.J.Immunol. 30,723-730,1989)。このオリゴヌクレオチドを、従来の自動固相オリゴヌクレオチド合成により合成し、アガロースゲル電気泳動および低融点(low melting)アガロースを用いて精製した。求められるバンドをゲルより切り取り、既知量のオリゴヌクレオチドを混合し、1−2時間ゲルの理論的な融点より5℃低い温度(通常は約65℃)まで加熱し、37℃まで穏やかに冷却する。この温度で、ハイブリッド化されたオリゴヌクレオチドを、ユビキチン遺伝子の一部を含むベクター断片に連結した。それに続く、制限断片分析およびDNA配列決定を用いた陽性クローンの分析を、従来の方法により行った("Molecular Cloning",Eds.:T.Maniatis et al.2 ed.CSH Laboratory Press,1989)。
例2: 改変されたユビキチン類似体でのワクチン接種によるユビキチンに対する自己抗体の誘発
熱感受性λリプレッサー(抑制因子)により調節されるプロモーターの制御下で、外来のT細胞エピトープの、卵アルブミン(OVA)由来のOVA(325−336)およびニワトリ卵白ライソザイム(HEL)由来のHEL(50−61)をコードする配列を含む遺伝子を、大腸菌AR58において発現させた。ポリクローナル抗ユビキチン抗体およびウエスタン・ブロッティングを利用して、組換えユビキチンタンパク質の発現を確認した("Antibodies",Eds.:D.Harlow et al.,CSH Laboratory Press, 1988)。組換えタンパク質は、従来の方法を用いて精製された(Maniatis et al.,上掲)。
フロイントの完全アジュバントに乳化されたリン酸緩衝生理的食塩水(PBS)中の100μgのユビキチンまたはその類似体をマウス胸部(i.p.)に注射した。フロイントの不完全アジュバントに1:1に乳化された、上記と同じ量の抗原のブースター(追加抗原)注射を、14日目と28日目に胸部に実施した。それぞれの群において5匹のBalb/Cマウスを試験し、従来のELISA法を用いて、7日目、14日目、21日目、28日目、35日目および42日目に、抗ユビキチン抗体の存在について血液試料を調べた。
T細胞エピトープOVA(325−336)またはHEL(50−61)を含む2つの異なるユビキチン類似体のそれぞれに対する抗体応答により得られた結果を図2に示す。挿入されたOVA(325−336)エピトープのアミノ酸配列は、QAVHAAHAEINEであり、HEL(50−61)エピトープのアミノ酸配列は、STDYGILQINSRである。
最初の抗原の注射からわずか1週間以内に、天然のユビキチンに対する明らかな抗体応答を検出することが可能であり、2週間以内に最大に達した。ウシ・イムノグロブリンとの共有結合によるユビキチン複合体により、ウサギにおいて産生された抗ユビキチン抗体は更に長い免疫感作期間の後に最大値に達した(データなし)。
例3: 適切に改変された腫瘍壊死因子(TNF)類似体を用いたワクチン接種によるTNFに対する自己抗体の誘発
DNAのポリメラーゼ連鎖反応(PCR)でのクローン化により、天然のネズミTNFαタンパク質(MR101)の構造的部分をコードする遺伝子を得た。MR103 TNFα類似体において、両親媒性のα−ヘリックスの置換として、クローン化TNFα配列におけるアミノ酸26−35を、卵アルブミン(OVA)H−2d制限化T細胞エピトープ配列325−334(QAVHAAHAET)に置き換える。TNFαのこの領域における置換により組換えタンパク質を無毒化する(X.Van Ostade et al., Nature 361,266-269,1993)。MR105 TNFα類似体において、クローン化TNFα配列におけるアミノ酸5−20を、ニワトリ卵白ラインザイム(HEL;hen eggwhite lysozyme)からのH−2k制限化T細胞エピトープであるアミノ酸配列81−96(SALLSSDITASVNCAK)に置き換える。MR106 TNFα突然変異株において、クローン化TNFα配列におけるアミノ酸126−140を、上記と同じエピトープであるアミノ酸配列81−95(SALLSSDITASVNCA)に置き換える。遺伝子構築物を図3に示す。T細胞エピトープをコードするDNAを伴うTNFα遺伝子の一部を交換するために、例1に記載された技術とは異なる技術を用いた。T細胞エピトープを誘発するために用いられる部位に隣接するTNFα遺伝子の一部のPCRリクローニング(再クローン化)を用いて突然変異配列を誘発することにより、MR105および106構築物を作成した。突然変異オリゴヌクレオチドプライマーは、T細胞エピトープをコードするDNA配列と同様にTNFαのDNA配列に相同なDNA配列もともに含んでいた。TNFα遺伝子のPCRリクローン化部分を次いで、適切な制限酵素により切断し、MR101遺伝子にクローン化した。「スプライシング・バイ・オーバーラップ・エクステンション(重複延長によるスプライシング)」PCR技術(R.M.Horton et al., Gene 77,61,1989)の変法により、MR103構築物を作製した。ここでは、それぞれがTNFα遺伝子の一部をカバーするように2つのPCR産生物を産生させ、加えて、それぞれのPCR産生物は、T細胞エピトープ配列の半分を含む。次いで、第2のPCRにおいて2つのPCR産生物を結合することにより、完全な突然変異TNFαを作製した。最終的には、完全な遺伝子構築物をタンパク質発現ベクターに挿入した。続いて、制限断片分析と従来の方法を用いたDNA配列決定とにより、すべての遺伝子構築物を分析した("Molecular Cloning",Eds,:T.Maniatis et al. 2.ed. CSH Laboratory Press,1989)。組換えタンパク質は、大腸菌において発現され、従来のタンパク質精製方法により精製された。
フロイントの完全アジュバントにエマルジョン化された100μgの半精製のMR103およびMR106を用いて、BALB/C(MHCハプロタイプH−2d)およびC3H(MHCハプロタイプH−2k)のマウスの群をそれぞれ、皮下注射により免疫感作させた。フロイントの不完全アジュバントを用いて、2週ごとに免疫感作を反復した。生物学的に活性を有するMR101に対して、全てのマウスが速やかに強力な抗体応答を発現した。これは、受動的に吸着された100%精製MR101を用いての直接ELISA法により測定された(図4)。MR101とPBSとのそれぞれにより免疫感作された対照マウスは、MR101に対する抗体反応を示さなかった。
ここで際立っていることは、異なるように制限されたT細胞エピトープを含むMR103およびMR106に、MHCハプロタイプの異なる両株のマウスが良好に反応したことから、移植されたT細胞エピトープに対応するようにはMR101に対する抗体応答は、MHC拘束がなされなかったことである(図4)。それらを一緒に取り上げれば、上記の結果は、(a)本発明に従った方法により産生された自己タンパク質類似体の、分泌された自己タンパク質に対する自己抗体を誘発する能力および(b)挿入されたT細胞エピトープのMHC結合能力から予想された以上に広範なMHC群における応答の誘発について、ここで記載された方法において向上した作用能力を例証した。アルデヒド複合化MR101により誘発された免疫応答と比較して、組換え自己タンパク質類似体MR103およびMR106対する免疫応答は、より強力であり、より高力価であった(例4参照)。
例4: 大腸菌タンパク質に複合化された非改変自己タンパク質と比較しての本発明に従った方法により産生された自己タンパク質類似体によるTNFαに対する自己抗体の誘発
大きなキャリヤータンパク質とともに小さな一本鎖のT細胞エピトープ・ペプチドをも間違いなく含んでいる、TNFαと大腸菌タンパク質との複合体を用いた場合に誘導される自己抗体応答に対して、本発明の方法によるTNFαに対する自己抗体の誘発を直接比較した。
0.5%ホルムアルデヒドを用いて、pH7.4のPBS中の大腸菌タンパク質に、半精製の組換えネズミTNFα(MR101)を複合化させた。該タンパク質の複合化を、SDS−PAGEにより確認した。続いて、これらの複合体をC3Hマウスの免疫感作に用いた。別群のC3Hマウスを、半精製の複合化されていない自己タンパク質類似体MR105によりワクチン接種した。約100μgの組換えTNFα類似体および複合体をフロイントの完全アジュバント中に1:1にエマルジョン化し、それぞれの群のマウスに皮下注射した。TNFαについてL929バイオアッセイにより判定したところ、MR105は生物学的に不活性である。続く免疫感作において、2週ごとにフロイントの不完全アジュバントを用いた。結局、ELISAで測定したところ、両群は、高度に精製された生物学的活性を有するMR101に対する自己抗体を発現したが、しかし、本発明の方法により産生された複合化されていないMR105類似体に対する免疫応答を、より早く、且つより高い力価で誘発した(図5)。
例5: ユビキチンに挿入された卵アルブミンT細胞エピトープにおいて、および自己タンパク質の重複配列を表すペプチドにおいて可能なMHCクラスIIの結合
室温で3日間、125Iでラベル化されたペプチド(10−100nM)をアフィニティ精製されたMHCクラスII分子とともにインキュベートすることにより、ペプチド−MHC複合体を得た(S.Mouritsen,J.Immunol. 148,1438-1444,1992)。以下のペプチドを結合の放射性標識マーカーとして用いた:Ek分子に強力に結合するHb(64−76)YおよびAk分子に強力に結合するHEL(46−61)Y。これらの複合体を、完全にすべての、免疫学的に関連があるMHCクラスII結合を阻害するのに十分に多量の、コールドな(放射性ラベル化されていない)ペプチド(>550μm)と共にインキュベートした。同一のペプチドを用いたにしても、3つの異なる重複ペプチドを用いたにしても、これらのペプチドは、ユビキチンの中に置換されたOVA(325−336)T細胞エピトープの全体と同様に隣接領域も表す(例2参照)。3種類のペプチドは、TITLEVEPSQAVHAA(U(12−26))、PSQAVHAAHAEINEKE(U(19−34))およびHAEINEKEGIPPDQQ(U(27−41))であった。G25螺旋カラムを用いてゲル濾過により遊離のペプチドから、8mMのクエン酸塩,17mMのリン酸塩,および0.05%NP−40(pH5)を含む反応緩衝液ならびにペプチド−MHCクラスII複合体を(2重に(in duplicate))分離した。除外された「ボイド」量および包含された量の放射活性を共に、ガンマ分光法で測定した。コールドなペプチドの添加により、(パーセントで)最大結合値の競合阻害を計算した。結果を表Iに示す。
EkおよびAkそれぞれへの放射性ラベル化されたペプチドHb(64−76)YおよびHEL(46−61)Yの結合についての完全な阻害は、同じペプチドのコールド・バージョンを用いてのみ達成し得たように思われた。これら極端な量のコールドなペプチドを用いることで、U(12−26)およびU(19−34)により結合に幾らかの阻害がみられたが、H−2k MHCクラスII分子へのこれらペプチドの親和性は、極めて低い傾向がある。
例6: 適切に改変されたTNFα類似体でのワクチン接種による糖尿病または炎症疾患の治療
例えば、破傷風毒素またはインフルエンザ・ヘマグルチニン(血球凝集素)に由来するT細胞エピトープをコードする適切な遺伝子断片を挿入することにより、TNFαをコードする遺伝子を改変する。例えば、大腸菌または昆虫細胞内で、適切な発現ベクターにおいてそのような遺伝子を発現する。従来の方法を用いて組換えTNFαタンパク質を精製した("Molecular Cloning",Eds.:T.Maniatis et al.2.ed.CSH Laboratory Press,1989)。
GMCSFまたはインターロイキン2のような免疫学的に活性を有するサイトカインに、そのような組換えタンパク質を任意に連結し得る。
組換えタンパク質は、適切なアジュバントを用いて製剤化し、抗TNFαワクチンとして、TNFαが病原として重要であるような疾患を患っている患者に投与し得る。ここで誘発された抗TNFα抗体は疾病に影響を与えるだろう。
例えば、TNFαが重要な役割を演ずると信じられているリウマチ様関節炎のような慢性炎症疾患が、前記疾患の1例である(F.M.Brennan et al.,Br.J.Rheumatol. 31,293-298,1992:に概説)。TNFαは、癌においてみられる悪液質状態において、およびAIDSのような慢性の感染症においてもまた重要な役割を演ずると信じられている(M.Odeh.J.Intern.Med. 228,549-556,1990に概説)。TNFが敗血症性ショックに関与することもまた知られている(B.P.Giroir,Crit.Care.Med., 21,780-789,1993:に概説)。その上、TNFαがタイプIIの真性糖尿病の発現において病原として働き得ることが示されている(CH Lang et al.,Endocrinology 130,43-52,1992)。

図4: 半精製されたMR103およびMR106によるBalb/cまたはC3Hマウスのワクチン接種によるTNFα自己抗体の誘発。ELISAにより抗体力価を測定し、マウス1個体からの強力な標準抗血清を参照値とした任意の単位値(arbitrary units)(AU)で表現した。プロットされた値は、動物5個体の平均力価を表す。最初の免疫感作のためのアジュバントとして、フロイントの完全アジュバントを用いた。続く14日のインターバルをおいての免疫感作はすべて、フロイントの不完全アジュバントを用いて行われた。PBS中の天然MR101を用いて平行して免疫感作されたマウスは、検出可能なTNFα自己抗体を発現しなかった(データなし)。検出できなかった抗体力価を力価値1と定めた。

Claims (18)
- 修飾された自己タンパク質を調製する方法であって、前記自己タンパク質は脊椎動物由来であり、前記方法は、分子生物学的手段によって、前記自己タンパク質における一以上のペプチドを対応する数のペプチドで置換することを含み、
ここで、
1)前記置換ペプチドは、前記脊椎動物に対して免疫優性の外来T細胞エピトープを含み、
2)前記置換は、未修飾の前記自己タンパク質における全体の三次構造を本質的に維持し、
これにより、前記修飾された自己タンパク質は、前記修飾された自己タンパク質を前記脊椎動物に投与した後に、前記脊椎動物において、前記未修飾の自己タンパク質に対する抗体応答を誘発することができることを特徴とする方法。 - 請求項1に記載の方法であって、前記免疫優性の外来T細胞エピトープは、該T細胞エピトープの両側にある未修飾の自己タンパク質由来の隣接領域を維持するように挿入されることを特徴とする方法。
- 請求項1または2に記載の方法であって、前記免疫優性T細胞エピトープは、破傷風トキソイドまたはジフテリアトキソイドを起源とする方法。
- ヒトまたは動物における望ましくないタンパク質に対する自己ワクチンであって、請求項1〜3の何れかの方法に従って修飾された一以上の自己タンパク質を含有し、燐酸カルシウム、サポニン、キル(quil)Aおよび生分解性ポリマーのような薬学的に許容可能なアジュバントと共に製剤化されたことを特徴とするワクチン。
- 請求項4に記載の自己ワクチンであって、前記修飾された自己タンパク質は、GM−CSFまたはインターロイキン2のような免疫学的に活性な適切なサイトカイン類との融合タンパク質の形態で存在することを特徴とするワクチン。
- 請求項4に記載の自己ワクチンであって、癌患者のような悪液質感受性の患者を治療するための、TNFαまたはγインターフェロンに対するワクチンであることを特徴とするワクチン。
- 請求項4に記載の自己ワクチンであって、アレルギーを伴う患者を治療するための、IgEに対するワクチンであることを特徴とするワクチン。
- 請求項4に記載の自己ワクチンであって、慢性炎症性疾患を伴う患者を治療するための、TNFα、TNFβまたはインターロイキン1に対するワクチンであることを特徴とするワクチン。
- 請求項8に記載の自己ワクチンであって、リウマチ様関節炎または炎症性腸疾患を伴う患者を治療するためのワクチンであることを特徴とするワクチン。
- 請求項4または5に記載の自己ワクチンであって、真性糖尿病を治療するための、TNFαに対するワクチンであることを特徴とするワクチン。
- 免疫優性の外来T細胞エピトープを含む一以上のペプチドフラグメントを含んだ修飾された自己タンパク質であって、
該修飾された自己タンパク質は、未修飾の自己タンパク質が天然に存在する脊椎動物に導入されたときに、前記自己タンパク質と交差反応する抗体の産生を誘導し、
前記外来T細胞エピトープは、該T細胞エピトープの両側にある未修飾の自己タンパク質由来の隣接領域を維持するように挿入され、また前記修飾された自己タンパク質の全体の三次構造は、前記未修飾の自己タンパク質と比較して本質的に維持される、
自己タンパク質。 - 燐酸カルシウム、サポニン、キル(quil)Aおよび生分解性ポリマーのような薬学的に許容可能なアジュバントと共に製剤化された、請求項11に記載の修飾された自己タンパク質。
- GM−CSFまたはインターロイキン2のような免疫学的に活性な適切なサイトカイン類との融合タンパク質の形態で存在することを特徴とする、請求項11に記載の修飾された自己タンパク質。
- 癌患者のような悪液質感受性の患者を治療するための薬学的組成物の調製における、請求項11に記載の、または請求項1〜3のいずれか1項に従って調製された修飾された自己タンパク質の使用。
- アレルギーを伴う患者の治療のための薬学的組成物の調製における、請求項11に記載の、または請求項1〜3のいずれか1項に従って調製された修飾された自己タンパク質の使用。
- 慢性炎症性疾患を伴う患者の治療のための薬学的組成物の調製における、請求項11に記載の、または請求項1〜3のいずれか1項に従って調製された修飾された自己タンパク質の使用。
- リウマチ様関節炎または炎症性腸疾患を伴う患者の治療のための薬学的組成物の調製における、請求項11に記載の、または請求項1〜3のいずれか1項に従って調製された修飾された自己タンパク質の使用。
- 真性糖尿病の治療のための薬学的組成物の調製における、請求項11に記載の、または請求項1〜3のいずれか1項に従って調製された修飾された自己タンパク質の使用。
