本発明は、養毛剤に関するものであり、更に詳細にはD(L)−パントラクトンを有効成分とする、特に育毛効果、脱毛予防効果及び頭皮の角化異常(ふけ)予防効果にすぐれた養毛剤に関するものである。

【従来の技術】
現在までに判明している脱毛の原因は、頭皮機能の老化及び代謝のアンバランス、皮脂腺、毛根部の成長の機能退化、すなわち、毛乳頭の新陳代謝の低下などがあげられている。よって、脱毛を防ぎ、発毛を促進するには、脱毛の原因になっていると思われる頭皮、皮脂腺、毛根、毛母細胞などに薬剤を効果的に働きかけ正常な形に戻すことであり、従来より、血行促進物質あるいは細胞賦括化物が用いられている。血行促進物質としては、ビタミンE誘導体、トウガラシチンキ、ニコチン酸誘導体、頭皮の皮膚細胞の賦括化物質としては、朝鮮ニンジンエキス、センブリエキス、甘草等が知られている。しかし、従来より使用されている血行促進物質は、皮膚刺激が強くその配合量に制限があったり、二次的な副作用発現により連続使用が困難であったり、血行促進の持続時間が短時間という欠点がある。また、細胞の賦括化物も効果が充分発揮されない。

一方、パントラクトンは、光学異性体を有し、D−体、L−体、DL−体のいずれもが知られており、既知の物質であるが、こ(れら)のD(L)−パントラクトンが育毛効果、脱毛予防効果、及びふけ予防効果を有することは全く知られていない。

【発明が解決しようとする課題】
本発明の目的は、長期にわたる使用に十分耐えうる安全性を備え、育毛効果、脱毛予防効果、及びふけ予防効果に優れた養毛剤を提供することにある。

【課題を解決するための手段】
上記目的を達成するために本発明者らは、鋭意研究を重ねた結果、D(L)−パントラクトンが、育毛効果、脱毛予防効果、及びふけ予防効果にすぐれ、しかも安全性にすぐれていて、長時間使用しても薬害等の副作用がないことを確認し、これらの新知見に基づき本発明を完成するに至った。以下、本発明を詳述する。

本発明において有効成分として使用するパントラクトンは、工業的に生産されており容易に入手可能である。この物質は2位に不斉炭素を有するため、光学異性体が存在する。本発明に係る育毛効果はD−体、L−体、DL−体に認められるが、天然物中に存在するのはD−体であり、その性質も良く知られており、安全性の面よりD−体を用いるのが好ましい。しかしながら、他の異性体も、いずれも育毛料としての特性を具備しているため、これらの異性体も本発明の有効成分として充分に使用することができ、本発明は特定の光学異性体のみに限定するものではない。

本発明に係る養毛剤を調製するには、化粧品や外用剤調製の常法にしたがい、D(L)−パントラクトンを有効分として、常用される化粧品基剤や外用剤基剤を用いて養毛剤の各種剤型に製剤すればよい。

本発明に係る養毛剤を調製するに当り、有効成分であるD(L)−パントラクトンの養毛剤中への配合量は、総量を基準として、0.001〜20.00wt%であれば良く、より好ましくは1.0〜10.0wt%である。即ち、この配合量の下限未満では本発明の目的とする効果が充分に得られず、また上限を越えても、その増加分に見合った効果の向上は望めないものである。

本発明の発毛・育毛料で使用される他の原料成分としては、通常、化粧料に適用される炭化水素類、ロウ類、油脂類、エステル類、高級脂肪酸類、高級アルコール類、界面活性剤、保湿剤、香料、色素、防腐剤、抗酸化剤、紫外線防御剤、pH調整剤および本発明の目的に合った薬効成分など適宜選択され、調整される。更に、公知公用の発毛・育毛成分と併用することもできる。また本発明の養毛剤は、種々の剤型、例えば、ヘアトニック、ヘアローション、ヘアクリーム、ポマード、ヘアコンディショナー、シャンプー、リンス、ヘアジェル、ヘアミスト、ヘアフォーム等があげられる。

次に、本発明に係る養毛剤の作用効果を、試験例により詳述する。

【試験例1】
下記表1に示す配合からなる評価物質(サンプル1、2)について、マウスを用いる動物実験により育毛効果試験を行った。なお、D−パントラクトンを配合しない対照物質をコントロールとして用いた。

実験に供したマウスはddY系雄性マウスで5週令のものを選んだ。マウスは第一期発毛が生後32〜36日頃より始まり、生後55〜58日頃から休止期に入るとされている。5週令のマウスがちょうど第二期の成長初期にあたる。マウスは各サンプル毎に一群9匹を用いた。

マウスの背部左右をそれぞれ直径1.5cmの円形にバリカンで刈毛した後、除毛クリームで完全に除去し、1日放置後表1に示す評価物質(サンプル1、2)を1日1回左側に塗布するとともに右側には対照物質(コントロール)を塗布する。塗布量はともに0.1mlとした。

2週間投与終了後各マウスの除毛部よりアトランダムに約50本を抜毛し、各マウス毎に長い順に10本の毛長を10倍の倍率下に検鏡してノギスを用いて測定した。得られた結果を下記表2に示した。

上記結果から明らかなように、本発明に使用されるD−パントラクトンは育毛促進効果を有することが判明した。

【試験例2】
下記表3に示す実施例1の配合からなるヘアローションを用いて、人による発毛・育毛効果試験を行った。なお、対照として、実施例1の配合においてDL−パントラクトンを処方しなかった比較例1の配合を用いた。

薄毛症、脱毛症を訴える20名のパネラーに実施例1に示すヘアローションを連続6ヵ月使用させることによる発毛・育毛効果試験を実施した。使用方法は通常のヘアローション使用方法と同様一日2回、頭部にまんべんなく塗布する方法を用いた。評価は使用開始後1ヵ月目と3ヵ月目に行い、うぶ毛、硬毛が増えてきたと感じた対象者がどの程度増えたかを計測した。また、テスト終了時に頭皮の状態の改善やふけの発生の減少についても申告してもらった。得られた結果を下記表4に示した。

上記結果から明らかなように、本発明に係るDL−パントラクトンを含有するヘアローションには、うぶ毛を生じた人、硬毛を生じた人が増加していることから発毛効果が認められた。また、頭皮の状態も改善され、ふけの発生の減少を申告したものが11名存在した。特にふけ予防効果については5〜10日で改善傾向効果がみられた。このことから、本発明のDL−パントラクトンを含有するヘアローションには育毛効果も認められた。なお、使用テスト期間中にいずれのパネラーにも頭皮の状態の悪化、炎症性の反応等の皮膚障害は全く観察されなかったことから、本発明は安全性が高いことが確かめられた。
