本発明において細胞株抗原/免疫原の再現性のある産生
方法が与えられる。
腫瘍を初期に検出し診断できることに対する大きなニー
ズが存在する。さらに局所的治療または手術の前に腫瘍
を局所的に食い止めることができることは好ましい。
キシン(cytotoxin)または放射性アイソトープに結合
した抗体で腫瘍が死滅させられる。腫瘍を標的とするこ
れらの物質は腫瘍細胞を死滅させる力が強く、また腫瘍
でのサイトトキシン濃度が高く、従って通常の細胞増殖
抑制性の治療の副作用を軽減する。
腫瘍(すなわち上皮起源の腫瘍組織)は、異なる種類の
サイトケラチンの形で腫瘍マーカーを含有することが知
られている。19個の異なるサイトケラチンが性状解析さ
れており、すべて蛋白でできている。これらのサイトケ
ラチンは、細胞内でいわゆる中間体繊維を形成する。サ
イトケラチン対の8と18は単純な上皮細胞中にしばしば
見られる。

性サイトケラチン断片は、回りの体液(例えば血液、
尿、腹水および胸水)に漏れ出る。
地中に放出されるサイトケラチン断片が、モノクローナ
ル抗体を産生させるのに使用される。しかし実際の腫瘍
マーカーが正確にわかっていないため、この方法は再現
性がなく特異性が低い。また特定されていない細胞物質
が検査の標準物質として用いられているため、この試験
は定量性と特異性において信頼性が低い。
ケラチンがクロマトグラフィーで精製され、酵素で消化
されて「そのアルファらせん中心部分」が得られ、これ
が次にクロマトグラフィーで精製されて、免疫試験およ
びモノクローナル抗体産生の抗原として使用される。こ
こでは抗原/免疫原は再現性のある方法で得られる。こ
れらの公知の方法は体液中のサイトケラチン断片を検出
し、腫瘍が属する細胞カテゴリー/型を性状解析するの
に充分である。しかしヨーロッパ特許出願公開公報(A
l)337057で得られるモノクローナル抗体は、試験すべ
き試料をまず可溶化する必要がある。
ーナル抗体療法で腫瘍を治療することに対するニーズが
いまだに存在する。また腫瘍が進行性であるか否か、ま
たは治療を評価するためにモノクローナル抗体療法を続
けるか否かを決めるために、先行技術よりも簡便で高感
度な検査に対するニーズが存在する。本発明はこのよう
なニーズを満たすものである。

えば、DU145(ATCC、no.HTB81)、HeLa(ATCC、no.CCL
2)など)。
8、18および19の分子量は分子量標準物質に対してそれ
ぞれ53、45および41Kdであった。これらのサイトケラチ
ンについてこのパターンはすでに報告されている(モ
ル、フランケ、シラー(Moll R.,Franke WW.,Schiller
DL.)ら、Cell、31巻、11−24頁、1982年)。

化学的に行ってもよい(例えばキモトリプシン、V8プロ
テアーゼ、ペプシン、TPCKトリプシン、BrCN、部分的加
水分解、BNPSサクトール(Saktole)など)。本発明に
おいては、使用した精製法のため断片が凝集せずモノマ
ー型で維持される。凝集物はエピトープをマスクしてし
まうため、断片が抗体産生に使用される場合、これは重
要である。
ケラチン8、18および19はそれぞれ調節可能な方法で、
キモトリプシンとサイトケラチンの重量比約1:50−1:10
00でキモトリプシンにより断片化されるが、好適な重量
比は1:400(CYK8)、1:100(CYK18)および1:75(CYK1
9)である。酵素の活性は蛋白1mg当り40−60単位であ
る。サイトケラチンの濃度は0.2mg/mlである。サイトケ
ラチン溶液と酵素溶液はそれぞれ、別々に37℃の水浴中
で5分プレインキュベートされる。次に酵素溶液がサイ
トケラチン溶液に加えられ、混合物は37℃の水浴中でさ
らに5分インキュベートされる。20%のSDSを少量加
え、95℃の湯浴中で最後に5分インキュベートして消化
を停止させる。最終のSDS濃度は2%になるであろう。

ラチン断片は、PB中の0.1%SDSに対して透析され、次に
標準的方法(FCAのマウス1匹当り10μgの断片(s.
c.)、FIAのマウス1匹当り10μg、次にFIA中のマウス
1匹当り1μgで2回で、これらはすべて1週間の間隔
で行われる)に従いBalb/cマウス中で免疫される。融合
までの日にマウスに3回追加免疫をする。

細胞1つにした、ハイブリドーマ細胞を増殖させ、安定
化し確立した。以下の特異性と反応性を有するモノクロ
ーナル抗体を産生する15個のクローンのすべてを確立し
た。
ELISA試験 pH9.0で吸着によりマイクロタイタープレートに係合
した、精製したサイトケラチン8、18および19に対する
得られたモノクローナル抗体をELISAで試験する時、あ
るモノクローナル抗体は1つ、2つまたは3つのサイト
ケラチンに対して特異性を有するようであるが、反応性
は異なる。

反応性はY軸上で吸光度(490nm)で表してある。図1
はサイトケラチン8(左の棒)、および18(右の棒)に
対する、ELISAでの15個の異なるモノクローナル抗体の
特異性と反応性を示す棒グラフである。

た。モノクローナル抗体との最初のインキュベートは室
温でわずか1時間であった。従って反応性の最も高いモ
ノクローナル抗体のみが選択された。2次抗体はHRPに
結合した抗マウスIgG抗体(すなわちダコパッツコード
(Dakopatts Code)P260)(1:1000希釈)であり、室温
でさらに2時間インキュベートし、基質o−フェニレン
ジアミンを加えた後、通常のELISA法により490nmでの吸
光度を読んだ。図1から明らかなように、8個のクロー
ンはサイトケラチン8に対して最も強い反応性を有して
いたが、他の7つはサイトケラチン18に対して強い反応
性を有していた。

断片に、7クローンについてはサイトケラチン18からの
大多数の断片に、そして5クローンについてサイトケラ
チン19からの大多数の断片にい対して良好な反応性と特
異性を示す。これは全サイトケラチンに対する15個のク
ローンの特異性と一致する(図1−2を参照)。

臓に対応する。24時間後においても血液はいくらかの標
識抗体を有する。放射性J131は抗体から離れ、尿中に分
泌され、抗体は身体により時間とともにアミノ酸に分解
される。

Claims (11)
- 【請求項1】調製用SDS−PAGEにより上皮腫瘍細胞から
サイトケラチンを精製し、サイトケラチン8、18及び19
に対応するバンドをゲルから溶出することを含む、サイ
トケラチン抗原/免疫原の再現性のある産生方法であっ
て、サイトケラチンを別々に消化して10−50Kdのサイズ
範囲のサイトケラチン8、18及び19からの断片の混合物
を産生することを特徴とする、上記方法。 - 【請求項2】消化は酵素的に行われることを特徴とす
る、請求項1に記載の方法。 - 【請求項3】消化は、キモトリプシン、V8プロテアー
ゼ、ペプシンおよびTPCKトリプシンからなる群から選択
される酵素により行われることを特徴とする、請求項2
に記載の方法。 - 【請求項4】サイトケラチンはキモトリプシン(40−60
U/mg蛋白)で、酵素:サイトケラチンの重量比が1:50か
ら1:1000で酵素消化されることを特徴とする、請求項3
に記載の方法。 - 【請求項5】酵素消化の酵素:サイトケラチンの重量比
は、サイトケラチン8については1:400、サイトケラチ
ン18については1:1000、そしてサイトケラチン19につい
ては1:75であることを特徴とする、請求項4に記載の方
法。 - 【請求項6】消化は、BrCN、加水分解またはBNPS−スカ
トールで、化学的に行われることを特徴とする、請求項
1に記載の方法。 - 【請求項7】産生された断片は第2の調製用SDS−PAGE
で精製され、サイトケラチン8、18又は19からの特定の
断片はゲルから溶出されることを特徴とする、請求項1
−6のいずれか一項に記載の方法。 - 【請求項8】10−50Kdのサイズ範囲の、サイトケラチン
8、18及び19からの断片を含む、請求項1から7のいず
れか一項に記載の方法により得られうる、サイトケラチ
ン断片混合物。 - 【請求項9】動物を請求項8に記載のサイトケラチン
8、18及び19からの断片の混合物を含む溶液で免疫し、
該動物から該断片に対する抗体を回収し、該サイトケラ
チン断片の大多数を認識する抗体を選択することを特徴
とする、サイトケラチン断片に対する抗体を産生する方
法。 - 【請求項10】マウスを請求項8に記載のサイトケラチ
ン8、18及び19からの断片の混合物を含む溶液で免疫
し、該マウスの脾臓からリンパ球を回収し、該リンパ球
をミエローマ細胞と融合させ、ハイブリドーマ細胞をク
ローン化し増殖させ、単一クローンを安定化し確立し、
該クローンからモノクローナル抗体を回収し、該サイト
ケラチン断片の大多数を認識する抗体を選択することを
特徴とする、サイトケラチン断片に対するモノクローナ
ル抗体の産生方法。 - 【請求項11】癌患者からの体液をテストすることによ
り上皮起源の腫瘍を検出する免疫化学的試験キットであ
って、請求項8に記載のサイトケラチン断片の混合物
と、請求項10の方法により産生される断片に対するモノ
クローナル抗体とを含むことを特徴とする、上記キッ
ト。
