インターロイキン−2(IL−2)は、リンパ球の産生、生存およびホメオスタシスにおいて中心的な役割を果たしている。これは、133個のアミノ酸を有し、その機能に必要な四次構造を形成する4つの逆平行の両親媒性アルファヘリックスからなる(Smith, Science 240:1169-76 (1988); Bazan, Science 257:410-13 (1992))。

IL−2プロドラッグには、IL−2アゴニストポリペプチド(サイトカイン部分)、担体(担体部分)、およびIL−2アンタゴニスト(マスキング部分)が含まれうるものであって、前記IL−2アゴニストポリペプチドは、前記担体に、直接またはリンカー(例えば、切断可能なまたは非切断性ペプチドリンカー)を介して融合され、前記IL−2アンタゴニストは、前記IL−2アゴニストポリペプチドまたは前記担体に、切断可能なペプチドリンカーを介して結合されている。本発明のIL−2プロドラッグにおいて、前記IL−2アゴニストポリペプチドは、野生型IL−2ポリペプチド、例えば、野生型ヒトIL−2ポリペプチド(配列番号1)、またはIL−2変異タンパク質、例えば、ヒトIL−2に由来するIL−2変異タンパク質であってもよい。前記IL−2変異タンパク質は、野生型IL−2と比較して、CD25または三量体高親和性IL−2Rに対して顕著に減少した親和性を有しうる。ある実施態様において、IL−2変異タンパク質は、野生型IL−2と比較して100倍、300倍、500倍、1,000倍、または10,000倍低い、高親和性IL−2Rに対する結合親和性を有する。特に示されていない限り、本明細書に記載のIL−2およびIL−2変異タンパク質における全ての残基番号は、配列番号1のナンバリングに従うものである。
本開示のIL−15プロドラッグにおいて、前記IL−15アゴニストポリペプチドは、野生型IL−15ポリペプチド、例えば、野生型ヒトIL−15ポリペプチド(配列番号2)、あるいは野生型IL−15と比較してIL−15RαまたはIL−2Rβ(CD122)に対して減少した親和性を有するIL−15変異タンパク質、例えば、ヒト野生型IL−15に由来するIL−15変異タンパク質であってもよい。
前記サイトカインアンタゴニスト、すなわち、本発明のプロドラッグのマスキング部分は、前記プロドラッグにおけるサイトカイン部分に結合し、前記サイトカイン部分をマスクし、その生物学的機能を阻害するペプチドまたは抗体もしくは抗体断片を含みうる。
本発明のプロドラッグの担体部分は、抗原結合部分、または抗原結合部分ではない部分であってもよい。前記担体部分は、サイトカインアゴニストポリペプチドの血清半減期などのPKプロファイルを改善させ、体内の標的部位(例えば、腫瘍部位)に対してサイトカインアゴニストポリペプチドの標的としうる。
前記担体部分は、抗体またはその抗原結合断片、またはイムノアドヘシンであってもよい。ある実施態様において、前記抗原結合部位は、2つの重鎖および2つの軽鎖を有する全長抗体、Fab断片、Fab’断片、F(ab’)2断片、Fv断片、ジスルフィド結合Fv断片、単一ドメイン抗体、ナノボディ、または単鎖可変断片(scFv)である。ある実施態様において、前記抗原結合部分は、二特異性抗原結合部分であり、2つの異なる抗原または同一抗原上の2つの異なるエピトープに結合することができる。前記抗原結合部分は、さらに強力な相乗的な治療効果を前記サイトカインアゴニストポリペプチドに供しうる。

他の非抗原結合担体部分が本発明のプロドラッグのために用いられてもよい。例えば、抗体Fcドメイン(例えば、ヒトIgG1、IgG2、IgG3、またはIgG4 Fc)、ポリマー(例えば、PEG)、アルブミン(例えば、ヒトアルブミン)、またはこれれらの断片、あるいはナノ粒子が用いられうる。
前記サイトカイン(例えば、IL−2またはIL−15)アゴニストポリペプチドは、前記担体部分に、ペプチドリンカーとともに、またはペプチドリンカーなしで融合されていてもよい。前記ペプチドリンカーは、切断されなくてもよい。ある実施態様において、前記ペプチドリンカーは、配列番号47〜51から選択される。特定の実施態様において、前記ペプチドリンカーは、アミノ酸配列GGGGSGGGGSGGGGS(配列番号49)を含む。
本開示のプロドラッグおよび変異タンパク質(すなわち、有効医薬成分またはAPI)を含む医薬組成物は、望ましい精製度を有するAPIを、1つまたはそれ以上の任意の医薬的に許容される賦形剤(例えば、Remington's Pharmaceutical Sciences, 16th Edition., Osol, A. Ed. 前記サイトカイン(例えば、IL−2またはIL−15)プロドラッグは、前記抗原結合ドメインによって結合している抗原により、疾患を治療するために用いることができる。ある実施態様において、前記IL−2またはIL−15プロドラッグは、癌を治療するために用いられる。ある実施態様において、前記IL−2またはIL−15プロドラッグは、例えば、薬物分子が抗菌薬または抗ウイルス薬である場合、感染症を治療するために用いられる。

63.治療を必要とするヒト対象における乳癌、肺癌、膵癌、食道癌、甲状腺髄様癌、卵巣癌、子宮癌、前立腺癌、精巣癌、大腸癌、直腸癌または胃癌、あるいは感染症の治療方法であって、実施態様61または62に記載の医薬組成物を前記ヒト対象に投与することを特徴とする方法。
発現プラスミドを、PEI(ポリエチレンイミン)を用いて3x106細胞/mlのFreeStyle HEK293細胞に2.5〜3μg/mlでコトランスフェクトさせた。Fcを基にしたIL−2プロドラッグについて、Fc−IL−2変異タンパク質融合ポリペプチドおよびFc−マスキング部分融合ポリペプチドの比率は、1:2の比率であった。抗体を基にしたIL−2プロドラッグについて、ノブ重鎖(IL−2アゴニストポリペプチドを含む)、ホール重鎖(マスキング部分を含む)、および軽鎖DNAの比率は、2:1:2のモル比であった。該細胞培養物を、トランスフェクション6日後に、9,000rpmにおける45分間の遠心分離、続いて0.22μMの濾過により回収した。
抗体を基にしたIL−2プロドラッグのタンパク質の精製は、1)プロテインAアフィニティークロマトグラフィー;2)Q Sepharose Fast Flow、および3)Capto MMC ImpResを含む、クロマトグラフィーの3ステップを用いることにより行った。Q FFは、25mM Trisおよび100mM NaClを含む緩衝液(pH7.5)により平衡にした。Capto MMC ImpResは、緩衝液A(20MM リン酸,30mM NaCl,pH6.2)を用いて平衡にし、緩衝液B(20MM リン酸,0.5M アルギニン,pH6.2)による10CV直線グラジエントを用いて溶出した。
SEC−HPLCは、Tosoh Bioscienceから注文されたTSKgel G3000SWXLカラム(7.8mm IDX 30cm,5μmの粒子径)を備えたHPLCシステムのAgilent 1100シリーズを用いて行った。100μl以下の試料を載せた。前記カラムを緩衝液(200mM K3PO4,250mM KCl,pH6.5を含有する)で流動した。流速は0.5ml/分であった。該カラムは室温で流動した。タンパク質溶出物を220nmおよび280nmの両方でモニターした。
10μlの培養上澄み液または20μgの精製タンパク質試料を、還元剤とともに、または還元剤なしで、Bolt(登録商標)LDSサンプル緩衝液(Novex)と混合した。該試料を70℃で3分間加熱し、次いでNuPAGE(登録商標)4−12%のBisTrisゲル(Invitrogen)に載せた。該ゲルをNuPAGE(登録商標)MOPS SDS泳動用緩衝液(Invitrogen)中で200ボルトにて40分間泳動し、クーマシーで染色した。
前記プロテアーゼ、ヒトu−プラスミノーゲン活性化因子(uPA)/ウロキナーゼ (R&D systems)またはヒトマトリプターゼ/ST14(R&D systems)を、それぞれ、81nMおよび250nMで前記前駆体分子に加え、37℃で終夜インキュベートした。
CTLL2細胞は、L−グルタミン、10% ウシ胎児血清、10% 非必須アミノ酸、10% ピルビン酸ナトリウム、および55μM ベータメルカプトエタノールを補充したRPMI1640培地中で増殖させた。CTLL2細胞を、培地(100ng/mlのIL−2を含む)中で5x104〜1x106細胞/mlにて接着させず、維持した。一般に、細胞を1週間に2回分注した。バイオアッセイのために、継代後48時間以上で細胞を使用することが最良であった。
PBS中で10μg/mlのIL−2タンパク質を96ウェルプレートに100μl/ウェルで撒種し、4℃で終夜コーティングした。該ウェルを、PBSで3回洗浄し、100μlの2% ミルク/PBSで1時間ブロッキングした。次いで、該ウェルをPBSにより3回洗浄し、3倍連続希釈した100μlのタンパク質試料を、前記ウェルに加え、室温で1時間のインキュベーションを行った。PBSで3回洗浄し、100μlのHRP抱合抗IgG抗体を加え、室温で1時間インキュベートした。次いで、前記ウェルをPBSにより3回洗浄し、検出試薬を加え、OD450nMを測定した。
IL−2RαβγまたはIL−2Rβγを発現する安定なHEK293細胞株を培養した。該細胞を非酵素細胞解離溶液で剥離した。細胞をカウントし、細胞密度をFACS洗浄緩衝液(PBS中の3% FBSを含有)で約3,000,000細胞/mlに調整した。50μlの細胞(150,000細胞)を96ウェルプレートの各ウェルに加えた。一次抗体または目的の抗体を発現する上澄み液を、所定の濃度で前記細胞に加えた。該プレートを氷上で1時間インキュベートした。該プレートをFACS洗浄緩衝液で3回洗浄した。蛍光を抱合させた二次抗体を該細胞(製造元の説明書による濃度)に加えた。該プレートを氷上で1時間インキュベートした。該プレートを再度洗浄した。PI染色溶液を0.1μg/mlで加え、該プレートを氷上で10分間インキュベートした。該細胞の蛍光をフローサイトメトリー装置で測定した。
クローディン18.2抗体、ADCCが高いクローディン18.2抗体、およびクローディン18.2抗体−IL−2の試料を、ヒトCLD18.2またはヒトCLD18.1を安定して発現するHEK293細胞に対してADCCを誘導するそれらの能力について分析した。
6週齢のBalb/cマウス(Taconic Biosciences)に、1X106個のCT26/18.2細胞を皮下注射した。7日後、腫瘍を、デジタルキャリパーを用いて測定し、腫瘍体積を算出した(V=(ab2)p/6[式中、bは、測定した2つの長さのうちの短い方の値である])。次に、マウスを、全ての群がほぼ同一の平均腫瘍サイズ(127.6mm3)を示すような治療群に無作為化した。次いで、マウスを、腹腔内注射により100μl中の10mg/Kgでのプラセボまたは試験物質で処理した。投薬を7、9、11、13、15、および18日目に行った。腫瘍を2〜3日ごとに測定し、腫瘍が2000mm3に達したらマウスを屠殺した。
ヒトIL−2(配列番号1)は、133アミノ酸のポリペプチドである。多くの変異体ヒトIL−2アゴニストポリペプチドを融合分子の一部として表し、それらの生物学的活性について試験した(表2)。必要に応じて、対のポリペプチドも示す。

IL−2のプロドラッグ基盤を構築するために、本発明者らは、ヒトIL−2受容体ベータサブユニットおよびガンマサブユニットを用いて、様々なIL−2アンタゴニスト(マスク)を設計した。典型的なマスクの設計を表3に記載し、これらには:
1)Fc断片のC末端に、1つのプロテアーゼ基質ペプチドを含む切断可能なペプチドリンカー(GGGGSGGGGSGGGGSLSGRSDNHGGGGS;配列番号18)を介して融合させた、IL−2Rβサブユニット細胞外ドメインの単一コピー(配列番号195);
2)FcのC末端に、2つのプロテアーゼ切断部位を含む切断可能なペプチドリンカー(GGGGSGGGGSGGGGSISSGLLSSGGSGGSLSGRSDNHGGGGS;配列番号38)を介して融合させ、IL−2Rガンマサブユニット細胞外ドメインの1コピーと融合させたIL−2Rβサブユニット細胞外ドメインの1コピー(配列番号196);
3)FcのC末端に、切断可能なリンカー配列番号38を融合させた、IL−2Rβサブユニット細胞外ドメインの単一コピー(配列番号197);
4)FcのC末端に、切断可能なリンカー配列番号38を介して融合させた、相互に結合させたIL−2Rβサブユニット細胞外ドメインの2コピー(配列番号198)
が含まれ、下線はプロテアーゼ基質配列を示す。

プロテアーゼ切断による活性化が高倍率となるIL−2アンタゴニストまたはマスクの改良型を探索するために、IL−2Rβ細胞外ドメインにおける多くの変異を構築した。これらの構築物は、HEK293細胞においてホモダイマーとして発現され、それらのIL−2との結合親和性を、上記に記載のELISA法によって測定し、表5で示す。単一の変異R15Y(配列番号199)、V75Q(配列番号202)、またはV75F(配列番号203)を有するIL−2Rβ細胞外ドメインは、ELISAアッセイにおいてIL−2に対する結合親和性を完全に喪失した。単一の変異S69H(配列番号201)またはE136Q(配列番号204)を有するIL−2Rβ細胞外ドメインは、pH7.4においてIL−2に対する結合親和性を喪失したが、pH6.4ではIL−2に対して2倍高い結合親和性を示した(表5)。二重変異E136Q/H138Rを有するIL−2Rβ細胞外ドメイン(配列番号205)は、pH7.4においてIL−2に対して野生型のものと同様の結合親和性を示したが、pH6.4でのIL−2に対するその結合親和性は2倍まで高くなった(表5)。変異D68Eを有するIL−2Rβ細胞外ドメイン(配列番号200)は、pH7.4およびpH6.4の両方においてIL−2に対して2倍高い結合親和性を示した(表5)。

サイトカインポリペプチドを抗体に融合することにより、サイトカインの疾患部位への標的とした送達が可能となる。しかしながら、免疫器官に豊富に存在しうる免疫細胞上に高親和性サイトカイン受容体が存在する場合、サイトカイン受容体への結合について顕著に競合する。この実験において、IL−2Rαに対する結合親和性が顕著に減少したIL−2変異タンパク質を抗体担体に融合させた。この種類の抗体IL−2プロドラッグは、抗体の標的となる疾患部位で活性化でき、顕著に改善したPKプロファイルと極めて高い疾患部位特異性を示しうる。

抗クローディン18.2抗体589A、589Aのフコシル化されていない型(af−589A)、およびIL−2変異タンパク質とaf−589Aとの融合体を、上記に記載されるように、ADCCアッセイにおいてそれらのインビトロ活性を試験した。Af−589Aは、そのN−グリカンにおいてフコースを有しないか、またはほとんど有せず、高まったADCC機能を示した。前記IL−2変異タンパク質には、変異T3A/C125S/R38S/F42A/Y45A/E62Aが含まれていた(配列番号10)。前記データは、前記IL−2変異タンパク質を589A抗体にさらに付加することによりそのADCC活性をさらに高めたことを示す(図9)。
インビボ抗癌効果試験を、589A−IL−2Eを抗PD−L1抗体と組み合わせて行った。前記プロドラッグおよびPD−L1抗体の両方を、10mg/kgで隔日にて皮下投与した。ヒトクローディン18.2をトランスフェクトしたCT26マウス腫瘍細胞を、Balb/cマウスに移植した。腫瘍サイズが約100mm3に達したら、前記マウスをそれらの腫瘍サイズに基づいて3つの群に無作為化した。各マウスは、緩衝液のプラセボ(群1)(図10、左上パネル)、10mg/kgの抗PD−L1抗体(群2)(図10、右上パネル)、または10mg/kgの抗PD−L1抗体と10mg/kgの589A−IL−2Eプロドラッグ(群3)(図10、下パネル)皮下を受容した。7、9、11、13、15、および18日目に投薬した。腫瘍サイズおよび体重を、実験期間中モニターした。
