Abstract
Description
心臓トロポニンTは、心疾患患者において広く用いられるバイオマーカーである。心疾患患者におけるその有用性は、近年、Westermannら(Nature Reviews/Cardiology, vol 14(2017) 473−483)によって概説されてきている。cTnTの使用は、急性心筋梗塞(AMI)と推測される患者においてよく確立されているが、トロポニン測定はまた、他の急性および非急性セッティングでも用いられている。AMIと推測される患者において、迅速な治療およびさらなる診断評価を可能にするため、早期の意思決定が非常に重要である。
したがって、本発明は、一連の増幅反応を実行することによって、抗体の可変鎖をコードするポリヌクレオチドのライブラリーを生成する新規方法に関する。これらのポリヌクレオチドライブラリーを、ターゲット抗原に対する増加したアフィニティなどの改善された特性を持つ抗体を同定するための選択法において用いてもよい。

本発明にしたがって、本発明の方法によって選択される抗体は、親抗体のアフィニティより高い、ターゲット抗原に関するアフィニティを有する。この改善されたアフィニティは、親抗体に比較して、少なくとも20%のt/2の増加によって示されてもよい。t/2の測定を、例えば実施例6に記載するように行ってもよい。

一般的に、当該技術分野に周知であるように、より低いKD値は、より高いまたは改善されたアフィニティに対応する。1つの態様において、突然変異体抗cTnT抗体は、5.8E−10MのKDを有する親抗体のKDと等しいかまたはそれより低い結合アフィニティを有する。

親抗体可変重鎖は、ネズミ起源のものである(配列番号34)。それぞれ、HCCDR1、HCCDR2および/またはHCCDR3の単一アミノ酸ランダム化を目的として、突然変異HCCDRを含むライブラリーを構築した。第一の工程において、各々、4つの異なる親抗体フレームワーク領域の1つをコードする4つのDNA断片を生成した。社内で、ポリメラーゼ連鎖反応によって、フレームワーク領域1、3および4を得て、フレームワーク領域2に相当する短い断片2(42bp)を、Metabion international AGで注文した(図1Aを参照されたい)。断片をゲル精製し、そして定量化した。これらのDNA断片の1つの100ngを、4つのアドオンPCR反応混合物の各々において、ポリヌクレオチドテンプレートとして用いた。親CDRと同じ数のコドンを含むポリヌクレオチドライブラリーの使用によってCDR領域を付加し、ここで前記ライブラリーのメンバーは、それぞれのHCCDRのそれぞれのコドン位各々に1つのNNKコドンを含むライブラリーメンバーを含むよう設計された。さらに、CDRライブラリー中のポリヌクレオチドは、それぞれのCDRに隣接するフレームワーク領域にハイブリダイズ可能な配列を含んだ。入れ子(nested)PCR増幅のため、末端プライマーを用いた。それによって(図1Bを参照されたい)、部分的に重複する配列を含む4つのDNA断片を生成した。FW1配列の3’端に、そしてFW4配列の5’端にハイブリダイズする末端プライマーを用いて、重複PCRを行って、4つの断片を直鎖DNAライブラリー構築物に連結した(図1Cを参照されたい)。典型的なPCR反応を、PCR等級の水で100μl反応混合物に満たし、該混合物は、MgSO4を含む10μlの10xPCR緩衝剤、200μM dNTP混合物、0.5μM順方向プライマーおよび逆方向プライマー、250ng DNAテンプレート、5単位のPwo DNAポリメラーゼを含有した。典型的なPCRは、94℃5分間の初期テンプレート変性で開始し、30サイクル(94℃2分間、60℃45秒間、72℃1分間)を使用し、そして72℃5分間の最終伸長工程を含有した。プライマー、テンプレートおよび断片配列を表1に列挙する。ライブラリー断片は、細胞不含系における転写および翻訳の成功に必要なすべての制御配列を含有した。当業者は、当該技術分野の方法によって、こうしたライブラリーを生成可能である。例えば、Hanes, J. & Pluckthun, A. Fabディスプレイ用の緩衝液を調製し、そして回転ローテーションで、4℃で一晩インキュベーションした。洗浄緩衝液、WB(60mM Tris;AcOHでpH7.5に調整、180mM NaCl、60mM酢酸マグネシウム、5%ブロッカーBSA、33mM KCl、200μg t−RNA、0.05%Tween20);ビーズ洗浄緩衝液BWB(100mM PBS、0.1%Tween20);停止緩衝液SB(50mM Tris、AcOHでpH7.5に調整、150mM NaCl、50mM酢酸マグネシウム、5%ブロッカーBSA(Pierce)、33mM KCl、0.5%Tween20、8.2mM酸化グルタチオン);溶出緩衝液(55mM Tris AcOHでpH7.5に調整、165mM NaCl、22mM EDTA、1mg BSA、5000U rRNA(5000U)、50μg tRNA)。
さらに、4つの対照試料をセットアップした。最初の2つの試料は、試料TおよびBGのmRNA単離後のDNA消化から得られ、そして潜在的に残っているDNAを増幅するPCRによって該試料を検証した。第三および第四は、PCR等級の水を用いた、RTの陰性対照および「RTに対するPCR」に関する陰性対照であった。
濃縮されたFab結合剤を単離するため、FabのヒトCH1ドメイン、ネズミVLドメインおよびヒトCLドメインを含有するphoATIR3−9bi Fab TN−T M7chim発現ベクター(図2を参照されたい)内に、ネズミ可変HCをクローニングした。発現ベクター内へのクローニングを可能にするため、リーダー配列Tir9に位置するBsiWI制限部位を、各選択プールに提供した。
詳細なBioacore分析のために、最適な突然変異Fab結合剤を明らかにするため、先の酵素連結免疫吸着アッセイ(ELISA)を実施した。ELISAセットアップを図3に示す。軌道振盪装置上で振盪することによって、ビオチン化組換え心臓トロポニンT(100nM)をストレプトアビジン−MTP96ウェルプレートにRTで1時間捕捉した。抗原トロポニンTを100μL IP緩衝液(PBS pH7.3、1%BSA)中で希釈した。続いて、マイクロプレート洗浄装置BioTek ELx405 Selectを用いて、ウェルを300μL 1x洗浄緩衝液(150mM NaCl、0.05%Tween20、0.2%Bronidox)で3回洗浄した。洗浄後、突然変異抗cTnT Fab結合剤を含有する未精製細胞抽出物をIP緩衝液で1:2に希釈し、そしてトロポニンTが捕捉されたウェルに移した。再び、ウェルを300μL 1x洗浄緩衝液で3回洗浄した。ヤギで産生された抗ヒトIgG(Fab特異的)−ペルオキシダーゼ標識抗体(検出抗体)を1:40000希釈(IP緩衝液中)で用いて、トロポニンT結合突然変異Fab断片を検出した。ウェルを再び300μLの1x洗浄緩衝液で3回洗浄して、未結合検出抗体を除去した。マイクロプレートを、RTで30分間、ウェルあたり100μLのABTSでインキュベーションした。405nmに設定したマイクロプレート読み取り装置BioTek Power wave XSで、光学密度を測定した。親抗cTnT抗体の野生型Fabを陽性対照として用いた。最初のヒットを同定し、そしてその未精製細胞抽出物を動力学分析に供した。
GE Healthcare T200装置上、詳細な動力学研究を37℃で行った。Biacore CM−5シリーズSセンサーを装置に搭載し、そして製造者の指示にしたがってあらかじめ調整した。システム緩衝液は、HBS−ET(10mM HEPES(pH7.4)、150mM NaCl、1mM EDTA、0.05%(w/v)Tween(登録商標)20)であった。試料緩衝液は、1mg/ml CMD(カルボキシメチルデキストラン、Fluka)を補充したシステム緩衝液であった。1つの態様において、抗ヒト抗体捕捉系をCM5バイオセンサー上で確立した。NHS/EDC化学を用いて、製造者の指示にしたがって、GAHF(ab’)2、(ヤギ抗ヒトF(ab’)2)(コード番号:109−005−097、ロット番号13.12.2005、Jackson Immuno Research)を固定した。10mM酢酸ナトリウム緩衝液(pH5.0)中の30μg/mlのGAHF(ab’)2を、フローセル1、2、3および4にあらかじめ濃縮し、そして10.000 RU GAHF(ab’)2で固定した。続いて、センサーを1MエタノールアミンpH8.5で飽和させた。

標準法にしたがってキメラヒト/マウス抗体を得た。対応するベクターおよびクローニング法が、Norderhaugら J Immunol Methods. 1997 May 12;204(1):77−87に記載される。
実施例7にしたがって産生した抗体をサンドイッチイムノアッセイ(図4を参照されたい)中で試験した。IgGルテニウムコンジュゲートを生成し、そして親抗cTnT抗体と突然変異抗cTnT抗体の成績を比較するため、真正Roche Elecsysアッセイ、カタログ番号05092744190(Roche Diagnostics GmbH、ドイツ・マンハイム)中に含まれる元来の標準ルテニル化コンジュゲートの代わりに用い、そして比較した。突然変異mAbを、異なる標識化学量論で、ルテニウムにコンジュゲート化した。1つの態様において、ルテニウム標識モル比は、1:10抗体IgG:標識であった。抗cTnT抗体変異体由来のルテニウムコンジュゲートをElecsys R2試薬中で希釈し、そしてトロポニンT hsアッセイ明細を用いて、ブランク対照(普遍希釈剤、Id. 11732277122、希釈剤マルチアッセイ、Id. 03609987170、Roche Diagnostics GmbH、ドイツ・マンハイム)、トロポニンT hsCalSet由来のCal1およびCal2(Id. 05092752190、Roche Diagnostics GmbH、ドイツ・マンハイム)で、トロポニンT hsアッセイプロトコルを用いて、Cobas E170モジュール上で測定を行った。結果を図5に示す。それぞれ組み合わせ番号11および12に存在する突然変異を含む抗体は、親(非突然変異)抗体に比較して、改善されたシグナル対ノイズ比を示す。
Claims (15)
- 各々、既知の親CDRを含む関心対象の抗体のフレームワーク領域および少なくとも1つの隣接相補性決定領域(CDR)をコードするポリヌクレオチドのライブラリーを生成する方法であって、該親CDRが既知の親CDRポリヌクレオチド配列にコードされ、該方法は、
i)抗体の第一のフレームワーク領域をコードするポリヌクレオチドを提供し、
ii)(i)のポリヌクレオチドに関する第一のPCRプライマーを提供し、
iii)各々、要素A−B−Cからなるポリヌクレオチドの混合物を提供し、
ここで
A)は第一のフレームワーク領域にハイブリダイズ可能なポリヌクレオチドであり、
各B)は、親CDRポリヌクレオチド配列と同じ数のコドンを含むポリヌクレオチドライブラリーのメンバーであり、ここで、該ライブラリーのメンバーは、少なくとも1つのランダム化コドンを含むよう設計されており、そして
C)は第二のフレームワーク領域にハイブリダイズ可能なポリヌクレオチドである、
iv)要素(C)に関する第二のPCRプライマーを提供し
v)ポリヌクレオチド(i)〜(iv)に基づくPCRを実行し、それによってポリヌクレオチドのライブラリーを得る、
そしてここでこうしたPCRは、親CDRポリヌクレオチド配列の非存在下で行われる
ことで特徴づけられる、前記方法。 - 前記の第一のフレームワーク領域は、FW1またはFW4のいずれかであり、前記の第二のフレームワーク領域は、該第一のものがFW1であればFW2であるか、または該第一のものがFW4であればFW3であり、そして前記CDRは、該第一のフレームワーク領域がFW1であればCDR1であるか、または該第一のフレームワーク領域がFW4であればCDR3である、請求項1の方法。
- 前記の第一のフレームワーク領域がFW1であり、前記の第二のフレームワーク領域がFW2であり、前記の第一のプライマーがFW1に関する順方向プライマーであり、そして前記の第二のプライマーがFW2に関する逆方向プライマーであり、そして前記CDRがCDR1である、請求項1または2の方法。
- 前記の第一のフレームワーク領域がFW4であり、前記の第二のフレームワーク領域がFW3であり、前記の第一のプライマーがFW4に関する逆方向プライマーであり、そして前記の第二のプライマーがFW3に関する順方向プライマーであり、そして前記親CDRがCDR3である、請求項1または2の方法。
- 各々、第一および第二の既知の親CDRを含む関心対象の抗体のフレームワーク領域および2つの隣接相補性決定領域(CDR)をコードするポリヌクレオチドのライブラリーを生成する方法であって、第一および第二の親CDRが、第一および第二の既知のCDRポリヌクレオチド配列によってコードされ、該方法は、
i)抗体の第一のフレームワーク領域をコードするポリヌクレオチドを提供し、
ii)各々、要素A−B−Cからなるポリヌクレオチドの第一の混合物を提供し、
ここで
A)は該第一のフレームワーク領域にハイブリダイズ可能なポリヌクレオチドであり、
各B)は、該第一の親CDRと同じ数のコドンを含む第一のポリヌクレオチドライブラリーのメンバーであり、ここで、該ライブラリーのメンバーは、少なくとも1つのランダム化コドンを含むよう設計されており、そして
C)は第二のフレームワーク領域にハイブリダイズ可能なポリヌクレオチドである、
iii)要素C)に関する第一のPCRプライマーを提供し、

- 前記の第一のフレームワーク領域がFW3であり、前記の第二のフレームワーク領域がFW2であり、前記の第三のフレームワーク領域がFW4であり、該第一の親CDRがCDR2であり、該第二の親CDRがCDR3であり、要素C)に関する前記の第一のプライマーがFW2に関する順方向プライマーであり、要素C’)に関する前記の第二のプライマーがFW4に関する逆方向プライマーである、請求項5の方法。
- 要素B)において、親CDRポリヌクレオチド配列の1つのコドンまたは2つのコドンがランダム化されている、請求項1〜7のいずれか一項の方法。
- 抗体の可変鎖の1つのランダム化CDRまたは2つの隣接ランダム化CDRをコードする請求項1〜8のいずれか一項にしたがって得られるポリヌクレオチドのライブラリーであって、得られるライブラリーにおいて、他の(ランダム化)ポリヌクレオチド配列に対する親ポリヌクレオチド配列の比が、1つのCDRがランダム化されている場合は1:106またはそれ未満であり、そして2つのCDRがランダム化されている場合には1:107またはそれ未満である、前記ライブラリー。
- 抗体の可変鎖をコードするポリヌクレオチドのライブラリーを生成するための請求項9記載のライブラリーの使用であって、可変鎖が可変H鎖または可変L鎖より選択される、前記使用。
- 請求項3、4、6および7にしたがって生成されるライブラリーに基づいて重複PCRを実行することによって、抗体の可変鎖をコードするポリヌクレオチドのライブラリーを生成するための方法。
- 請求項11にしたがって得られる抗体の可変鎖をコードするポリヌクレオチドのライブラリーであって、可変鎖がランダム化CDR1、ランダム化CDR2およびランダム化CDR3を含み、そして得られるライブラリーにおいて、ライブラリー中の他のポリヌクレオチド配列に対する親ポリヌクレオチド配列の比が1:5x107またはそれ未満である、前記ライブラリー。
- 抗体が第一の可変鎖および第二の可変鎖を含む抗体ライブラリーを生成するための方法であって、請求項12記載の抗体の第一の可変鎖をコードするポリヌクレオチドライブラリーが発現され、そして前記抗体の第二の可変鎖と組み合わせられる、前記方法。
- 請求項13にしたがって生成されたライブラリーから、第一の可変鎖および第二の可変鎖を含む抗体を選択する方法であって、選択された抗体が既知の親CDRを含む親抗体に比較して、改善された結合特性を有する、前記方法。
- 選択された抗体が、親抗体に比較して少なくとも20%の解離複合体半減期t/2の選択された増加を示す、請求項14の方法。
