いくつかの実験において、それぞれ、配列番号40および21に示すように、HISタグ化CR1aおよびnCR1aを用いた。
HEK 293T細胞(プレートあたり7x106細胞)を、15cm培養プレートにおいて、10%ウシ胎児血清(FBS、Sigma、カタログ番号F9665)を補充した、高グルコースを含む17mlのダルベッコの修飾イーグル培地(DMEM、Sigma、カタログ番号D469)中、5%CO2インキュベーター中、37℃で一晩増殖させた。細胞が60%集密に到達したら、これらを、86.4μlの7.5mMポリエチレンイミン(PEI、Polysciences、カタログ番号24765−2)、150mM NaCl(Fisher Scientific UK Ltd、カタログ番号1073592)を用い、シグナルペプチド(配列番号7)に連結されたCR1a(配列番号2)またはnCR1a(配列番号5)のいずれかを発現する14.4μgプラスミドで一過性にトランスフェクションした。陰性対照のため、14.4μlのTris−EDTA緩衝剤をプラスミドDNAの代わりに用いた。トランスフェクション5時間後、10%FBSを含有するトランスフェクション培地を、2%FBSを補充した高グルコースを含む17.5mlの新鮮なDMEM(本明細書において、以後、発現培地と称する)で置き換えた。発現培地をトランスフェクション24、48、72および140時間後に収集した。80μlの0.5Mフェニルメタンスルホニルフルオリド(PMSF、Sigma、カタログ番号P7626)を、収集した100mlごとの発現培地に添加し、そして4℃で保存した。24時間後に収集した発現培地を、以下に記載する拡散および機能研究に用いた。
ヒト細胞からのタンパク質発現を図1Aおよび1Bに示す。組換え発現され、そして精製されたCR1aタンパク質は、2つの糖型で細胞から分泌されることが見出された。グリコシル化を除去する酵素であるPNGアーゼFでの処理は、2つのバンドをより小さい見かけの分子量の1つのバンドに減少させた(1A)。タンパク質の非グリコシル化型(nCR1a)を発現させた。ウェスタンブロッティングによって、nCR1aが酵素的に脱グリコシル化されたタンパク質と同じ位置に移動する単一のバンドを生じることが立証された(1B)。
ヒトHEK293細胞から発現され、そして分泌されたCR1aおよびnCR1aを、C3bの因子I仲介性分解に関する補因子として作用する能力に関して試験した。
McHargら, J Vis Exp (2015) 1−7、上記に記載されるように、ドナーの眼から単離した濃縮ブルッフ膜の黄斑部を、ウッシングチャンバー(Harvard Apparatus、米国ハムデン)に載せた。載せたら、5mm直径の黄斑部は、2つの同一の区画間の唯一のバリアであり、すなわち、液体はブルッフ膜を通過しなければならない(図4)。ブルッフ膜の両側を2mlのPBSで、室温で5分間洗浄した。実験の前に、次のチャンバーに漏出することなく、1つのチャンバー中に2mlの液体を保持する能力によって、ブルッフ膜の構造的完全性を試験した。組換えタンパク質(CR1aおよび/またはnCR1a、上記を参照されたい)を含有する2mlの発現培地をチャンバー(以後、試料チャンバーと称する)に添加し、そして2mlの新鮮なPBSをもう一方のチャンバー(以後、拡散物/拡散チャンバーと称する)に添加した。拡散タンパク質の勾配が生じることを回避するため、磁気スターラーバーで各区画を穏やかに攪拌しながら、ウッシングチャンバーを室温で24時間放置した。
結果を図3Aに示す。CR1aは、24時間後、拡散チャンバー中に存在することが見出された。
本発明記載のポリペプチドの発現レベルを比較して、最適な配合物があるかどうか、すなわちグリコシル化ポリペプチドが非グリコシル化ポリペプチドよりも高いレベルで発現するかどうかを評価する。ポリペプチドのグリコシル化状態は、発現レベルに最小限の影響を有するであろうと予期される。
表面プラズモン共鳴(SPR)を用い、それによってC3bをSPRチップ上に固定し、そしてポリペプチドを液相で用いて、本発明記載の精製ポリペプチドのC3bに対する結合動力学を試験する。会合および解離定数を直接測定し、そして相互作用に関するkD値を推測する。
CR1aのC3bタンパク質に関するアフィニティを、OctetRed96系(ForteBio、Pall Corp、米国)を用いて測定した。ビオチン化C3bタンパク質を0.2%PBSTで最終濃度0.4μg/mLに希釈し、そしてあらかじめ同じ緩衝液で20分間水和させておいた高精度ストレプトアビジン(SAX)バイオセンサー(ForteBio、Pall Corp、米国)上に、600秒間装填した。次いで、C3b装填センサーを0.2%PBSTで150秒間洗浄し(ベースライン)、そして30.0μg/mL〜2.6μg/mLの範囲の異なる濃度のCR1aを含有するウェル内に600秒間ディップし(会合)、その後、0.2%PBSTで600秒間洗浄した(解離)。会合および解離プロファイルを記録し、そしてForteBioデータ分析v9(ForteBio、Pall Corp、米国)で分析した。陰性対照、すなわち0.2%PBST含有ウェルを平行して用いて、センサーとの非特異的相互作用から生じる結合を減じた。動力学モードを用い、25℃で実験を実行し、そして試料プレートをあらかじめ3分間攪拌した。結合プロファイルを全体的に1:1(表面上の1つの結合部位に対して溶液中の1つの分析物)に適合させた。定常状態分析によって、4つの最低の利用可能な分析物濃度から、会合(0秒〜600秒)および解離(0秒〜100秒)のデータを用いてKDを決定した。固定C3bに対する天然可溶性C3b結合補体制御因子、因子H(FH)およびFHL−1に関する結合アフィニティもまた決定した。
ウッシングチャンバー実験(上述)を用いて、本発明記載のポリペプチドの拡散速度を比較して、タンパク質の配合による何らかの相違が生じるかどうかを見た。
随意に、好ましいシグナルペプチド、配列番号4に少なくとも80%の配列同一性を有するポリペプチド、および終結コドンをコードする、本発明記載の核酸を、AAVベクター内に挿入する。生じた発現ベクターを用いて、培養RPE細胞をトランスフェクションする。コードされるポリペプチドの発現および分泌を評価する。配列番号4に少なくとも80%の配列同一性を有するポリペプチドは、RPE細胞によって分泌され、そしてシグナルペプチドは、分泌されたポリペプチドから切断されていると予期される。
結果を図7Aに示す。ヒトAPRE−19細胞から分泌される機能性CR1aポリペプチドは、因子Iに関する補因子として作用して、C3bのiC3b(産物e)およびC3dg(産物f)への分解を導くことが見出された。FHL−1+FI+C3bを含有する反応は、C3bおよび産物iC3bのMW対照を提供した。
本発明のポリペプチドは、補体関連障害の治療または防止の方法において、使用を見出す。補体関連障害の一例は、眼の黄斑変性症、例えばAMDである。

レーザー誘導性脈絡膜血管新生モデル
このモデルは、マウスまたはラットの網膜にレーザー熱傷を適用し、例えばSchnabolkら Mol Ther Methods Clin Dev. 2018; 9: 1−11に記載されるように、脈絡膜血管新生を引き起こす。脈絡膜血管新生複合体のサイズを、フルオレセイン血管造影によって、または組織学によって測定してもよい。
例えば、Katschkeら, Sci Rep. 2018; 8(1):7348に記載されるように、補体活性化に部分的に依存する網膜色素上皮の変性を誘導するヨウ化ナトリウムを、マウスに静脈内注射する。組換えタンパク質としての硝子体内注射によって、またはCR1a cDNAを含有するAAVベクター、例えばAAV2を用いた網膜下送達によって、CR1aを送達する。CR1a処置マウスは、CR1a処置を受けていない対照マウスよりも、より少ない網膜変性を有することが見出される。
Claims (48)
- 補体関連疾患または状態を治療するかまたは防止する方法における使用のための、C3bに結合可能なポリペプチドであって、配列番号4に少なくとも85%の同一性を有するアミノ酸配列を含み、そして450アミノ酸またはそれ未満の全長を有する、前記ポリペプチド。
- 補体関連疾患または状態を治療するかまたは防止する方法における使用のための、C3bに結合可能なポリペプチドをコードする核酸であって、前記ポリペプチドが、配列番号4に少なくとも85%の同一性を有するアミノ酸配列を含み、そして450アミノ酸またはそれ未満の全長を有する、前記核酸。
- 補体関連疾患または状態を治療するかまたは防止するための薬剤の製造における、C3bに結合可能なポリペプチドの使用であって、配列番号4に少なくとも85%の同一性を有するアミノ酸配列を含み、そして450アミノ酸またはそれ未満の全長を有する、前記ポリペプチドの前記使用。
- 補体関連疾患または状態を治療するかまたは防止するための薬剤の製造における、C3bに結合可能なポリペプチドをコードする核酸の使用であって、配列番号4に少なくとも85%の同一性を有するアミノ酸配列を含み、そして450アミノ酸またはそれ未満の全長を有する、前記ポリペプチドをコードする前記核酸の前記使用。
- 補体関連疾患または状態が眼の疾患または状態である、請求項1または請求項2記載のポリペプチド、あるいは請求項3または請求項4記載の使用。
- 眼の疾患または状態の治療または防止が、被験体の少なくとも1つの眼の細胞が該ポリペプチドを発現するかまたは含むように修飾する工程を含む、請求項5記載のポリペプチドまたは使用。
- 眼の疾患または状態の治療または防止が、被験体の少なくとも1つの眼の細胞が該ポリペプチドをコードする核酸を発現するかまたは含むように修飾する工程を含む、請求項5または請求項6記載のポリペプチドまたは使用。
- 眼の疾患または状態の治療または防止が、被験体の少なくとも1つの眼の細胞に、該ポリペプチドをコードする核酸を含むベクターを投与する工程を含む、請求項5〜7のいずれか一項記載のポリペプチドまたは使用。
- 少なくとも1つの目の細胞が網膜色素上皮(RPE)細胞である、請求項6〜8のいずれか一項記載のポリペプチドまたは使用。
- 疾患または状態が、C3bまたはC3b含有複合体、C3bまたはC3b含有複合体に関連する活性/反応、あるいはC3bまたはC3b含有複合体に関連する活性/反応の産物が、病理的に関与している疾患または状態である、請求項1〜9のいずれか一項記載のポリペプチドまたは使用。
- 疾患または状態が黄斑変性症である、請求項1〜10のいずれか一項記載のポリペプチドまたは使用。
- 疾患または状態が:加齢性黄斑変性症(AMD)、初期AMD、中間期AMD、後期AMD、地図状萎縮(「乾性」AMD)、「湿性」(血管新生性)AMD、脈絡膜血管新生(CNV)、緑内障、自己免疫ブドウ膜炎、糖尿病性網膜症、および早発性黄斑変性症(EOMD)の1つまたはそれより多くより選択される、請求項1〜11のいずれか一項記載のポリペプチドまたは使用。
- ポリペプチドが配列番号4に少なくとも95%の同一性を有するアミノ酸配列を含む、請求項1〜12のいずれか一項記載のポリペプチドまたは使用。
- X1がAまたはTであり、X2がPまたはLであり、そして/またはX3がGまたはRである、請求項1〜13のいずれか一項記載のポリペプチドまたは使用。

- ポリペプチドが分泌経路配列を含む、請求項1〜21のいずれか一項記載のポリペプチドまたは使用。
- 分泌経路配列が配列番号7を含むかまたは該配列からなる、請求項22記載のポリペプチドまたは使用。
